Werkwijze voor de bereiding van een in water oplosbaar complex van a-6-deoxy-5-hydroxytetracycline en natriumtetrametafosfaat.
De onderhavige uitvinding heeft betrekking op de bereiding van een nieuw in water oplosbaar complex van a-6-deoxy-5-hydroxytetracycline en natriumtetrametafosfaat, alsmede op de solvaten ervan met een lagere alcohol en water.
5 Dit nieuwe derivaat, d.w.z. doxycyclinetetrametafosfaatna-triumcomplex, biedt aanmerkelijke voordelen ten opzichte van a-6-deoxy-5-hydroxytetracycline, gewoonlijk met doxycycline aangeduid, alsmede van de reeds bekende zouten en complexen ervan, in het bijzonder met betrekking tot zijn niet-interfe-10 rentie met de natuurlijke bactericidewerking van het menselijke serum, welke interferentie een van de belangrijkste nadelen van de al sinds langere tijd als anti-bacteriemiddelen bekende tetracyclineverbindingen, is.
Enkele andere voordelen van het doxycycline-15 tetrametafosfaatnatriumcomplex zijn zijn uitstekende biologische beschikbaarheid, zeer breed anti-bacteriëel spectrum, lage dosering, een enkele dagelijkse toediening, handhaving van bloed- en weefselniveaus gedurende 24 h of langer, verminderde nevenwerkingen op de maag, uitstekende stabiliteit in de 20 maagsappen, alsmede zeer langdurige stabiliteit in (¾ kristal-lijne vorm.
Allerlei tetracyclineadditiezouten en -fos-faatcomplexen zijn reeds in de literatuur beschreven.
In het Amerikaanse octrooischrift 2.791.609 25 wordt het tetracyclinehexametafosfaat beschreven, dat een complex is met een geringe oplosbaarheid en dat hoge bloedniveaus veroorzaakt. Zijn analoog echter, d.w.z. doxycyclinehexameta-fosfaat, dat een zeer geringe oplosbaarheid in water heeft, leidt tot dermate lage bloedniveaus, dat het geen enkele klini-30 sche betekenis heeft.
In het Amerikaanse octrooischrift 3.053.892 wordt de bereiding van complexen van metafosforzuur en tetracycline beschreven, in een moleculaire verhouding van 1-4 HPO^ per mol tetracycline.
35 In het Amerikaanse octrooischrift 3.068.264 -----^ 8105309 , - 2 - 5' ^ worden complexen beschreven van fosforzuur, aluminium en tetracycline. Deze complexen zijn oplosbaar bij een pH tussen 6,5 en 8,0, maar zijn vrijwel onoplosbaar bij een pH tussen 3 en 5. De moleculaire verhouding tussen tetracycline-aluminium-meta-5 fosfaat is 1 : 4 : 12 of 1:4:8. Deze complexen kennen geen klinische toepassing vanwege het feit, dat het aluminium "in vivo" interfereert met de antibacteriële werking van het tetracycline.
In het Nederlandse octrooischrift 152.248 en 10 de Nederlandse octrooiaanvrage 74.08875 wordt o.a. de bereiding onder watervrije omstandigheden beschreven van polymeta-fosfaatcomplexen van a-6-deoxy-5-hydroxytetracycline en de bereiding van de nieuwe hexametafosfaatcomplexen toegelicht.
Terwijl bij de werkwijze volgens het Ameri-15 kaanse octrooischrift 2.791.609 de reactie in waterig medium wordt uitgevoerd, worden volgens alle andere octrooischriften de boven genoemde complexen in watervrij medium bereid.
De onderhavige uitvinding heeft betrekking op de bereiding van polymetafosfaatcomplexen van alkalimetalen en 20 van a-6-deoxy-5-hydroxytetracycline, waarin de polymerisatie-graad van het metafosforzuur (HP03)n in het complex gelijkmatig is en wel een tetrameer vormt, d.w.z. n = 4.
Overeenkomstig de werkwijze volgens het Nederlandse octrooischrift 152.248 en de Nederlandse octrooiaanvra-25 ge 74.08875 worden de polymetafosfaatcomplexen van alkalimetalen en van doxycycline bereid in een strikt watervrij medium.
De werkwijze volgens de onderhavige aanvrage wijkt in zoverre af van de werkwijze volgens het Nederlandse octrooischrift 152.248 dat de reactie wordt uitgevoerd in aan-30 wezigheid van een nauwkeurig gecontroleerde hoeveelheid water, in plaats van in een watervrij medium, onder oplevering van tetrametafosfaatcomplexen in plaats van hexametafosfaten. Wanneer de isolering wordt bewerkstelligd door toevoeging van isopropanol, slaat het nieuwe complex neer in de vorm van een 35 solvaat met de formule (<^2H24N2°8) 3' (HP<V 3 *NaP03 *^31^011,6H2°
Door uitvoering van de boven aangeduide pre-cipitatie met absolute alcohol in plaats van isopropanol, kristalliseert het nieuwe complex in de vorm van het ethano- 8105309 ...................- . - - -- -· - - ..... - - - — -..... .............

De hoeveelheid water, aanwezig in het reac-tiemengsel bedraagt tenminste 2 mol per mol te reageren doxycycline, maar mag niet hoger zijn dan 4 mol.
5 Het metafosforzuur wordt volgens één der be kende werkwijzen bereid, bij voorkeur direct vóór de bereiding van het complex. De bereiding uit fosforpentoxide en een stoechiometrische hoeveelheid water in een inert oplosmiddel, zoals chloroform, heeft de voorkeur. Zodra het metafosforzuur 10 is gevormd, wordt het reactiemengsel verdund met methanol, teneinde het reactiemedium te homogeniseren, waarna water wordt toegevoegd en daarop het doxycycline in de vorm van het monohydraat. Men voegt 3 mol doxycyclinemonohydraat toe aan telkens 4 molequivalenten metafosforzuur. De snelheid van het 15 toevoegen van het doxycyline aan de metafosforzuuroplossing moet op een zodanige manier worden geregeld, dat het reactiemengsel in oplossing wordt gehouden. De eventuele toevoeging van methanol kan het volledig oplossen van het doxycyline, dat in achtereenvolgende porties wordt toegevoegd, bevorderen.
20 Daarop wordt een methanolische oplossing van natriumhydroxide toegevoegd in de verhouding van 1 mol per 4 molequivalenten van het aanwezige fosforzuur. De toevoeging van isopropanol aan het reactiemengsel leidt tot kristallisatie van het nieuwe tetrametafosfaatnatriumcomplex in de vorm van een solvaat met 25 isopropanol en water.
Het tetrametafosfaatnatriumcomplex wordt bij orale toediening zeer snel geabsorbeerd en zijn concentratie in het serum is zelfs na 24 h duidelijk boven de minimale concentratie met genezende werking.
30 In de onderstaande tabel zijn de serumniveaus verkregen na orale toediening aan twee groepen van 12 gezonde vrijwilligers van een enkele orale dosis van 100, resp. 200 mg weergegeven. De toediening vond plaats in de vorm van 1 kapsu-le voor de eerste groep en 2 kapsules voor de tweede, welke 35 kapsules het doxycyclinetetrametafosfaatnatriumcomplex en lactose als excipiens bevatten. Bloedmonsters werden getrokken naO, 0,5, 1, 2, 3, 4, 5, 6, 12 en 24 h na toediening. De serumconcentratie werd microbiologisch bepaald.

Deze waarden zijn aanzienlijk hoger dan de in de wetenschappelijke literatuur gepubliceerde waarden, zowel ten opzichte van het doxycycline als ten opzichte van de reeds bekende complexen en toont een uitstekende biologische be-15 schikbaarheid aan.
Het belangrijkste voordeel echter;"van het doxycyclinetetrametafosfaatnatriumcomplex vergeleken met doxycycline en de bekende derivaten ervan ligt in het feit dat het nieuwe complex volgens de onderhavige uitvinding een sterk 20 verminderde interferentie vertoont met de natuurlijke bacteri-cidewerking van het menselijke serum.
Het is algemeen bekend dat tetracyclinen, in het bijzonder doxycycline, zowel "in vitro" als "in vivo" interfereren met de antibacteriele werking van het serum 25 (Forsgren, A. en Gnarpe, H.: Antimicrobial Agents and Chemotherapy, 711-715 (1973)^; Forsgren, A. en Gnarpe, H.: Nature New Biology,' '244, 82-83 (1973) .
Het effect van het nieuwe complex dat het onderwerp vormt van de onderhavige uitvinding, op de bactericide-30 werking van het menselijke serum, werd vergeleken met die van doxycyclinehydrochloride.
Het aantal in het menselijk serum overlevende bacteriën werd bepaald in verloop van de tijd, parallel met identieke preparaten welke het complex en andere preparaten, hoeveelheden 35 die doxycyclinehydrochloride in gelijke - bevatten. Het aantal levende bacteriën (E. coli) werd in het menselijk serum 8105309 - 5 - i?*-__ bepaald op de tijdstippen 0 en 2 h na incubatie, zonder toevoeging van het antibioticum, alsmede in aanwezigheid van doxycyclinehydrochloride (10 ^ig/ml) en van het doxycycline-tetrametafosfaatnatriumcomplex (10 ^ig/ml). Zonder toevoeging 5 van het antibioticum nam het aantal overlevende bacteriën af 6 3 van 2 x 10 tot 5,5 x 10 , terwijl in het experiment met doxy- 5 cycline het aantal levende bacteriën 5 x 10 was en in het experiment met het doxycyclinetetrametafosfaatnatriumcomplex 3 dit na 2 uur incubatie 2,5 x 10 bedroeg.
10 Het feit dat de interferentie van het doxy- cyclinemolecuul met de bactericidewerking van het serum zeer sterk is, terwijl het nieuwe doxycyclinetetrametafosfaat-natriumcomplex niet ermee interfereert na 2 h, heeft een fundamenteel belang; de genezingsduur wordt verkort vanwege het 15 feit, dat de bactericidewerking van het serum volledig kan werken, met als resultaat dat de mogelijkheid van vorming "in vivo" van resistente stammen ook tot een minimum wordt teruggebracht.
De solvaten met isopropanol/water en met 20 ethanol/water hebben een identieke microbiologische en farmacologische activiteit en bezitten een uitstekende stabiliteit in massa bij kamertemperatuur.
VOORBEELD I
Onder goed roeren werd 100 ml methanol toege-25 voegd aan 4,33 g metafosforzuur, dat vers was bereid uit fos-förpentoxide in 100 ml chloroform. Na roeren gedurende 15 min werden 1 ml water en 20 g a-6-deoxy-5-hydroxytetracycline-monohydraat (961^ig/mg) op zodanige wijze in porties toegevoegd, dat de oplossing helder bleef. De oplossing werd daarop 30 gefiltreerd, gevolgd door de toevoeging van 0,574 g natriumhydroxide, opgelost in 17,5 ml methanol. Na roeren gedurende 15 min werd daarop 430 ml isopropanol toegevoegd. Het zo gevormde kristallijne produkt werd afgefiltreerd, gewassen met isopropanol en gedroogd bij 35-40°C. Het verkregen produkt met 35 de formule 3. (HPO^) ^.NaPO^ . ^C^HyOH. 6^0 had een gewicht van 21 g en vertoonde de volgende eigenschappen: UV-absorptie: E^cm 238 bij 349 nm en 307 bij 266 nm, in methanol bevattende 1 % HCl. Specifieke rotatie: 8105309 " 6 " ii /α/ = -86° (c = 0,5 in methanol bevattende 1 % HC1) . pH van een 1 % waterige oplossing: 2,8. Doxycycline-analyse: 70,21 %. Oplosbaarheid in water: 335 mg/ml.

5 Voorbeeld I werd herhaald, waarbij echter het isopropanol werd vervangen door absolute ethanol, waarbij het nieuwe complex neersloeg in de vorm van een solvaat met ethanol en water volgens de formule (C22H24N2°8^ 3* ^HP03^ 3’Na]?03* 1%C2H50H.6H20.

Claims (6)
1. Werkwijze voor de bereiding van een in water oplosbaar complex van a-6-deoxy-5-hydroxytetracycline en van natriumtetrametafosfaat met een sterk antibacteriële wer-5 king, in de vorm van een solvaat met de formule (C22&24®2®%* 3' (HPO^Jg.NaPO^.^C^H^OH. eK^O, waarbij de polyme-risatiegraad van het metafosforzuur in het complex gelijkmatig is(n=4), met het kenmerk, dat vers bereid metafosforzuur tot reactie wordt gebracht met a-6-deoxy-5- 10 hydroxytetracyclinemonohydraat in de verhouding van 4 HPO^ tot 3 mol “-6-deoxy-5-hydroxytetracycline, bij kamertemperatuur in een medium, dat bestaat uit chloroform, methanol en een gecontroleerde hoeveelheid water, waarna een kwart van de zuur-functies van het zo verkregen tetrametafosfaatcomplex wordt 15 geneutraliseerd met een methanolische oplossing van natriumhydroxide en het complex wordt gekristalliseerd door toevoeging van isopropanol.
2. Werkwijze volgens conclusie 1, met het kenmerk, dat het polymetafosforzuur wordt be- 20 reid uit fosforpentoxide en water in een stoechiometrische / verhouding in chloroform.
3. Werkwijze volgens conclusie 1, met het kenmerk, dat het polymetafosforzuur, bereid volgens conclusie 2, wordt verdund in methanol dat 2-4 mol 25 water per mol te reageren a-6-deoxy-5-hydroxytetracyclinemono-hydraat bevat.
4. Werkwijze voor de bereiding van een in water oplosbaar complex van a-6-deoxy-5-hydroxytetracycline en van natriumtetrametafosfaat met een sterk antibactericide- 30 werking in de vorm van een solvaat met ethanol en water, met het kenmerk, dat het complex wordt gekristalliseerd door de toevoeging van watervrije ethanol.
5. Polymetafosfaatcomplex van alkalimetaal en van doxycycline, met het kenmerk, dat het de 35 formule 3· (HPC>3) 3 . (NaP03) . l^C^OH. 6H20 heeft.
