Abstract
La presente invención se refiere a un método para determinar el porcentaje de glactosa recubierta en una glicoproteína, que comprende: (a) preparar una glicoproteína en un medio para análisis, con lo cual se obtiene una preparación proteica; (b) preparar muestras de proteína enzimáticamente tratada, y un control de la proteína preparada que comprende: dividir la preparación de proteína en una pluralidad de muestras de proteína; (i) no agregar enzima a por lo menos una muestra de proteína como un control antecedente de muestra de medio, con lo cual se obtiene la muestra A; (ii) agregar al menos ß-galactosidadas a por lo menos una muestra de proteína, con lo cual se obtiene la muestra B; (iii) agregar al menos ß-galactosidada y asialidasa a por lo menos otra muestra de proteína, con lo cual se obtiene la muestra C; e incubar la pluralidad de muestras de proteína que comprenden las muestras A, B y C; (c) cuantificar la cantidad de galactosa en cada una de las muestras A, B y C usando cromatografía HPAEC-PAD, en donde la cantidad de galactosa en la muestra A refleja la cantidad de galactosa en el medio, en donde la cantidad de galactosa en la muestra B refleja la cantidad de galactosa no recubierta con un residuo de ácido siálico en la preparación de proteína además de la cantidad de galactosa del medio, y en donde la cantidad de galactosa en la muestra C refleja la cantidad de galactosa recubierta con un ácido siálico además de la cantidad de galactosa del medio y no recubierta por ácido siálico; y (d) determinar el porcentaje de galactosa recubierta en la glicoproteína con base en las cantidades de galactosa en la muestra A, muestra B y muestra C, en donde el método tiene la linealidad de R2= 0.99.
