Abstract
La presente divulgación proporciona métodos y composiciones relacionados con la modificación de células efectoras inmunes para aumentar la eficacia terapéutica; en algunas modalidades, se proporcionan células efectoras inmunes m odificadas para reducir la expresión de uno o más genes diana endógenos, o para reducir una o más funciones de una proteína endógena para potenciar las funciones efectoras de las células inmunes; en algunas modalidades, las células efectoras inmunes se modifican adicionalmente mediante la introducción de transgenes que confieren especificidad de antígeno, tales como los receptores de células T (TCR) exógenos o receptores de antígenos quiméricos (CAR); también se proporcionan métodos para tratar un trastorno proliferativo celular, como un cáncer, utilizando las células efectoras inmunes modificadas descritas en el presente documento.
