Abstract

Se describe en la presente un método para producir un animal quimérico no humano que expresa un gen humano de NKX2- 5, HANDII, TBX5 o una combinación de los mismos que comprende: a) generar un NKX2-5, HANDII, TBX5 o una combinación de los mismos de una célula animal no humana, en la que ambas copias del gen NKX2-5, HANDII, TBX5 no humano o una combinación de las mismas llevan una mutación que impide la producción de proteína funcional NKX2-5, HANDII, TBX5 o una combinación de las mismas en el animal no humano; b) crear un NKX2-5, HANDII, TBX5 o una combinación de los mismos blastocito no humano mediante transferencia nuclear de células somáticas que comprende fusionar un núcleo del NKX2- 5, HANDII, TBX5 o una combinación de las mismas células animales no humanas de a) en un no nucleado enucleado ovocito humano y activación del ovocito para dividirse para formar un NKX2-5, HANDII, TBX5 o una combinación de los mismos blastocito no humano no humano; c) introducir células madre humanas en el NKX2-5, HANDII, TBX5 o combinación de blastocito nulo no humano de b); y d) implantar el blastocito de c) en un animal no humano sustituto pseudopreñado para generar un animal no humano quimérico que expresa NKX2-5, HANDII, TBX5 humano o una combinación de los mismos.
