Abstract
본 발명은 크실로글루칸-특이 엔도-(1,4)-β-D-글루카나제 활성을 갖는 효소를 암호화하는 서열 및 그의 기능적 동등 서열에 관한 것이다. 또한, 상기 서열로 이루어지는 벡터, 상기 서열이 도입된 변종 식물, 식물 특성의 변경방법, 및 재조합 DNA 기법에 의해 상기 정의한 활성을 갖는 효소의 제조방법에 관한 것이다.

제1 및 2도는 조(組)(제1A도) 또는 친화 정제된(제1B 또는 2도)항 크실로글루카나제 항체로 탐침화한 면역 블롯의 사진이고, 제 3 도는 항 크실로글루카나제 항체(레인 1 내지 3)를 사용하여 면역 침전시킨 한련종자 RNA의 시험관내 번역 산물(레인 5 및 6)의 SDS-PAGE 분석 사진이고,제 4 도는 한편 종자에서 크실로글루카나제 발현 수준 대 시간의 관계를 나타내고, 제 5 도는 한편 cDNA클론의 제한 효소 분석 결과를 나타낸다.
Claims (13)
- 제 9 도(서열 번호1)에 나타낸 서열의 뉴클레오티드 35 내지 919로 이루어지는, 크실로글루칸- 특이 엔도-(1,4)-β-D-글루카나제 활성을 갖는 효소를 암호화 하는 뉴클레오티드 서열 또는 그의 기능적 동등 서열.
- 제 1 항에 있어서, 5′-비번역 영역을 더 포함하는 서열.
- 제 1 항 또는 제 2 항에 있어서, 3′-비번역 영역을 더 포함하는 서열.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 한련(Topaeolum majus L.)으로부터 얻는 서열을 포함하는 서열.
- 제 1 항 내지 제 4 항 중 어느 한항에 서열로 이루어지는 벡터.
- 제 1 항 내지 제 4 항 중 어느 한항의 서열리 도입된 식물 전체 또는 그의 일부.
- 제 1 항 내지 제 4 항 중 어느 한항의 서열을 도입하는 것으로 이루어지는 식물 전체 또는 그의 일부의 특성의 변경방법.
- 제 7 항에 있어서, 상기 변경되는 특성에 크기, 조직 또는 성숙 속도가 포함되는 방법.
- 제 7 항 또는 제 8 항에 있어서, 과실 또는 채소의 특성이 변경되는 방법.
- 제 7 항 내지 제 9 항 중 어느 한 항에 있어서, 토마토 식물 전체 또는 그의 일부의 특성이 변경되는 방법.
- 제 1 항 내지 제 4 항 중 어느 한 항의 서열을 적합한 벡터에 삽입하는 단계; 상기 벡터로 적당한 숙주세포를 형질 전환시키는 단계; 효소가 발현되는 적합한 배양 조건에서 상기 형질 전환 숙주 세포를 성장시키는 단계; 배양 배지 및(또는) 숙주 세포로부터 효소를 얻는 단계로 이루어지는 크시로글루칸-특이 엔도 -(1,4)-β-D-글루카나제 활성을 갖는 효소의 제조방법.
- 제11항에 있어서, 숙주 세포가 미생물인 방법.
