Abstract

본 발명은 체성 배아 현탁 배양물과 체성 배아를 포함하는 전-배발생물 현탁 배양물(PEMs)에 관한 것으로 PEMs는 거의 유사한 배수도를 갖는 것이며, 이들로부터 식물이 유도된다. 또한 본 발명은 상술한 체성 배아를 수득하는 방법에 관한 것이다.

제 1 도는 임펠러 또는 진동 혼합기의 사용과 PEMs(수)/ℓ의 관계를 도시한 것이다.

Claims (20)

  1. 전-배발생물(pro-embryogenic mass;PEM) 형성을 유발하는데 충분한 효과적인 양의 오옥신 또는 오옥신 혼합물을 포함하는 액체 식물조직 배양배지와 비-켈러스 이식(explant) 물질을 접촉시키는 것을 특징으로 하는 상기 이식 물질로부터 생존성을 지닌 체성 배아(somatic embryos)를 산출할 수 있는 PEMs 형성을 촉진시키는 방법.
  2. 하기 단계 ⅰ) 내지 ⅳ)를 포함하는 현탁 배양물에서 거의 같은 배수도의 체성 배아를 수득하는 방법; ⅰ)전-배발생물(PEM) 형성의 유발을 촉진하는데 충분한 효과적인 양의 오옥신 또는 오옥신 혼합물을 포함하는 액체 배지와 접촉된 상태로 이식물질을 배양하는 단계 ; ⅱ)액체 배지를 포함하는 적당한 오옥신과 접촉하여 상기 단계 ⅰ)에서 수득된 PEMs의 수를 중식시키는 단계 ; ⅲ)상기 단계 ⅱ)에서 수득된 PEMs를 수집하고 및 거의 오옥신 부재의 액체 배지에 PEMs를 접촉시키는 단계 ; 및 ⅳ)상기 단계 ⅲ)에서 발생된 PEMs에서 유도된 체성 배아를 수집하는 단계.
  3. 제 2 항에 있어서, 생존성 있는 PEMs가 풍부한 선택된 크기의 PEMs 분획을 상기 단계 ⅲ)에 사용하는 것을 포함하는 방법.
  4. 제 1 항 내지 제 3 항중 어느 한 항에 있어서, 상기 액체 배지가 약 5g/1 내지 90g/1 농도의 탄수화물 에너지 제공원을 추가로 포함하는 것을 특징으로 하는 방법.
  5. 제 4 항에 있어서, 상기 탄수화물 에너지 제공원 농도가 약 20g/1 내지 60g/1 범위에 있는 것을 특징으로 하는 방법.
  6. 제 4 항 또는 제 5 항에 있어서, 상기 탄수화물 에너지 제공원이 수크로오스, 글루코오스 및 라피노스군으로부터 선택되는 것을 특징으로 하는 방법.
  7. 제 1 항 내지 제 6 항중 어느 한 항에 있어서, 상기 오옥신 함유 PEM 현탁물을 진동 혼합기내에서 진동시키는 것을 특징으로 하는 방법.
  8. 제 7 항에 있어서, 상기 진동 혼합기가 거의 수직 평면에서 진동되는 것을 특징으로 하는 방법.
  9. 제 1 항 내지 제 8 항중 어느 한 항에 있어서, 상기 이식물질이 원형질체, 줄기, 잎, 꽃잎, 하배축 단면, 정단 분열 조직, 접합성 배아 자체, 괴경, 유관속, 내초, 자방 또는 꽃밥 필라멘트에서 유도되는 것을 특징으로 하는 방법.
  10. 거의 모두 같은 배수도를 갖는 체성 배아를 포함하는 비-다우커스 식물물질에서 유래한 체성 배아 현탁 배양물.
  11. 제10항에 있어서, 주로 이배체 체성 배아를 포함하는 것을 특징으로 하는 체성 배아 현탁 배양물.
  12. 제10항에 있어서, 주로 사배체 체성 배아를 포함하는 것을 특징으로 하는 체성 배아 현탁 배양물.
  13. 제10항 내지 제12항중 어느 한 항에 있어서, 상기 체성 배아가 쌍자엽 또는 단자엽 이식 물질에서 발생된 PEMs로부터 유도되는 것을 특징으로 하는 현탁 배양물.
  14. 제10항 내지 제13항중 어느 한 항에 있어서, 상기 체성 배아가 시클라멘, 호리병박, 라이코페르시콘, 알리엄, 베고니아, 베타스, 앵초속 식물, 브라시카스, 고추, 시코륨, 게베라, 임페이션, 락투카스, 오리자스, 펠라고늄, 페투니아, 비올라, 및 제아스로 이루어진 군으로부터 선택되는 것을 특징으로 하는 현탁 배양물.
  15. 제14항에 있어서, 쌍자엽 이식물질인 것을 특징으로 하는 현탁 배양물.
  16. 제15항에 있어서, 시클라멘, 호리병박, 베타스, 브라시카스, 비올라, 펠라고늄, 및 고추로 이루어진 군으로부터 선택된 것을 특징으로 하는 현탁 배양물.
  17. 거의 이배체인 세포로 이루어진 PEMs를 포함하는 현탁 배양물.
  18. 거의 모두 같은 배수도를 갖는 비-다우커스 체성 배아.
  19. 제18항에 있어서, 거의 모두 이배체 체성 배아 또는 거의 모두 사배체 체성 배아를 포함하는 것을 특징으로 하는 체성 배아.
  20. 제 2 항 내지 제19항에 의한 체성 배아에서 유래한 식물.
