Abstract

본 발명은 단백질 공학에 의해 생산된 신규의 변형 세팔로스프린 C 아실라제(CC아실라제), 이것을 코딩하는 DNA, 이 DNA를 함유한 발현 벡터, 이것을 사용하여 형질전환시킨 미생물 및 이 형질전환제를 배양하는 CC아실라제의 생산 방법에 관한 것이다.

Claims (7)

  1. 숙주 세포중에서 α-서브유니트 및 β-서브유니트로 처리되기 전의 전구체로서 하기 식으로 나타낸 변형 세팔로스프린 C 아실라제.
    A1-268-X1-Tyr-X2-A272-304-X3-A306-773
    여기서, A1-265은 천연 CC 아실라제의 Thr1내지 Gly268에서와 동일한 아미노산 서열이고, A272-304는 천연 CC 아실라제의 Gln272내지 Tyr304에서와 동일한 아미노산 서열이고, A306-773은 천연 CC 아실라제의 Val306내지 Ala773에서와 동일한 아미노산 서열이고, X1은 Met 또는 다른 아미노산이고, X2는 Ala 또는 다른 아미노산이고, X3은 Cys 또는 다른 아미노산이고, 단 X1이 Met이고 X3은 Cys 이외의 아미노산이다.
  2. 제1항에 있어서, X1이 Tyr이고, X2가 Ala이고, X3이 Ser인 변형 세팔로스포린 C 아실라제.
  3. 제1항의 세팔로스포린 C 아실라제를 코딩하는 DNA.
  4. 제3항의 DNA로 이루어진 발현 벡터.
  5. 제4항의 발현 벡터에 의해 형질전환된 숙주 세포.
  6. 제5항의 숙주 세포를 수정 영양 배지 중에서 배양하고, 배양 브로스로부터 변형 세팔로스포린 C아실라제를 회수하는 것으로 이루어지는, 변형 세팔로스포린 C 아실라제의 제조 방법.
  7. 하기 일반식(Ⅱ)의 화합물 또는 그의 염을 제5항의 형질전환체의 배양 브로스 또는 그의 처리 물질과 접촉시키는 것으로 이루어지는, 하기 일반식(Ⅰ)의 화합물의 제조 방법.
    상기 식 중, R1은 아세톡시, 히드록시 또는 수소이고, R2는 카르복실 아실이다.
