Description
본 발명은 유두종(乳頭腫) 비루스 게놈(genome)으로부터 유도되어 결정된 DNA 서열에 관한 것으로서, 보다 상세히 설명하자면 DNA 재조합체를 복제시킬 수 있는 적당한 숙주 세포에 DNA 재조합체를 도입시킬 경우, 상기 DNA 서열이 대응하는 단백질 형태로 발현될 수 있는 방식으로 상기 DNA 서열로 변형시킨 벡터를 포함한 상기 DNA 재조합체에 관한 것이다. 또한 본 발명은 정제시켜서 면역 조성물의 제조에 사용할 수 있는 단백질에 곤한 것이다.
더욱 구체적으로 본 발명은 1985년 11월 29일자에 제85.402362.9호로 출원된 유럽 출원에서 IP-2(현재 HPV-33으로 재명명됨)로 명명된 유두종 비루스의 DNA 생성물에 관한 것이다. 상기 비루스의 DNA를 함유하는 플라스미드는 I-450으로 CNCM(파리에 소재하는 빠스뙤르 연구소의 "Collection nationale de Culture de Micro-Organismes")에 기탁되었다(위 HPV-33 비루스는 1987년 12월 29일자로 한국 종균 협회에 기탁 의뢰하였으나, 인체에 유해한 균주라는 이유로 인해 수탁이 거부되었음).
유두종 비루스는 파포바(papova) 비루스과의 성원으로서, 공유 결합으로 폐쇄된 환상 DNA 분자로 구성된 약 7,900 염기쌍(bp)의 게놈을 갖는다. 인체 유두종 비루스(HPV)는 그들의 DNA 서열 상동성(6)에 기초하여 분류되며, 현재 약 40종류가 기재되어 있다. HPV 생물학과 인체 질병에 있어서 이들의 관련에 대한 상당한 통찰은 분자 생물학의 기술을 적용함으로써 달성되었다. 인체 암에서 HPV의 가능한 역할은 생식기 우췌(33)의 악성 전환으로 인한 종양에서 HPV DNA의 탐지 후 밝혀지게 되었다. 경부암종으로부터 2가지 HPV 게놈, HPV-16과 HPV-18(3,11)의 클로닝(cloning)은 사회 경제적으로 매우 중요한 이 분야의 연구를 자극하였다. 이 비루스드른 시험된 악성 생식기 종양의 70% 이상에서 발견되었으며, 다수의 나머지에서 HPV-16과 관련된 서열이 탐지되었다(3,16,33). 이중에는 최근에 감소된 긴축(stringeney)(1)조건 하에서 프로브(probe)로서 HPV-16을 사용하여 침습성 경부암으로부터 클로닝시킨 HPV-33이 있다. 본 연구소에서 본 발명자드른 HPV-33의 DNA 서열을 결정하였으며 이것과 HPV-16과의 관계를 기재하였다. 유두종 비루스 중에서 HPV-33은 SV40(4,14)의 인핸서와 크게 닮은 78bp의 종렬 반복을 가지므로 독특하다.
본 발명은 특정 DNA 서열의 동정을 가능하게 하는 하기 HPV-33 게놈의 클로닝법에서 나온 것으로서, 더욱 구체적으로 HPV-33의 게놈은 혼성화 프로브를 제공하여 특히 인체 조직에서 HPV-33과 관련된 유두종 비루스의 DNA 탐지에 유용하고, 이로써 양성 반응은 침습성 경부암의 숙주에서의 가능한 발육에 관련될 수 있다.
이후로 관계 도면을 참고하여 기재한다.
제1a도 및 제1b도는 HPV-33의 뉴클레오티드 서열임. 환상 게놈상의 위치 1은 HPV-6b와 정열시킴으로써 발견된 "Hpa-유사" 서열에 해당한다. 제2도는 HPV-33 게놈에 있어서 주 해독 프레임(frame)의 배치임. 주 해독 프레임은 다른 HPV 서열과 비교함으로써 동정되었으며, 정지 코돈은 수직 막대로서 표시된다. 또한 특정 제한 부위(S, Sma I ; E, EcoRv ; B2, Bgl II ; B1, Bgl I)의 위치 및 초기와 후기 전사에 대한 가능한 폴리아데닐화 신호(PA)가 나타나 있다. 이들 외에, 6개의 다른 잠재적 PA 부위(AATAAA)가 위치 862, 1215, 1221, 2666, 5837 및 6239에서 탐지되었다.
제3도는 비암호화 영역의 주요 특징임. 위치 7500에서 114까지 비암호화 영역의 부분을 나타냄. 78bp의 종렬 반복은 굵은 윗줄로 그었으며 SV40 인핸서의 Z-DNA 생성 요소를 닮은 영역들을 나타낸다. 잠재적 프로모터(promoter) 요소는 별표로 표시되었고, 3부의 12bp의 팔린드롬은 2줄의 점선 사이에 넣었다.
적합한 서열은 전체 단백질을 암호화하는 서열, 더욱 구체적으로, 이후 표 1에 관련된 개방 해독 프레임을 갖는 각각의 뉴클레오티드 서열이다.
DNA 서열 분석을 행하는 조건은 이후에 표제 "재료 및 방법"에 기재한다. 이 서열 분석으로부터 이끌어 낸 결론은 표제 "검토"에 나타내었다.
재료 및 방법
DNA 서열 분석
본 연구에서 서열화시킨 HPV-33의 근원은 pBR322 유도체 중에 클로닝시킨 Bgl II 선형화 BPV-33 게놈으로 되는 플라스미드 p15-5(1)이었다. 임의의 DNA 단편(400-800bp)의 라이브러리(library)를 본질적으로 상기한 바와 같은 p15-5의 초음파 거리 및 말단 수복(28) 후, M 13mp8 (17)중에서 제조하였다. DNA 서열화는 비긴(Biggin) 등(2)의 변형 방법을 사용하여 디데옥시 사슬 종결법(19, 20)에 의해 행하였다. 대부분의 서열은 이 방법으로 유도되었으나 비암호화 영역 부분은 M13 라이브러리(>300클론) 중에 없거나 또는 소량인 것으로 밝혀졌다. 이 영역의 서열은 스미쓰(Smith)의 방법(24)을 사용하여 P15-5로부터 직접 얻었다. 간단하게, 2개의 "상보적" M13 클론으로부터 단리시킨 제한 단편은, 제한 효소를 사용하여 선형화시키고, 이어서 엑소뉴클레아제 III으로 처리(22℃에서 1시간 동안 200 단위/pmol DNA)한 p15-5로 부터 제조한 주형 상에서 DNA 합성을 개시하는데 사용하였다.
컴퓨터 분석
DNA 서열을 컴퓨터 언어로 번역해서, 비. 커드론(B.Caudron)에 의해 변형된 쉬타덴(Staden)(26, 27)의 프로그램으로 분석하였다. DNA 또는 단백질 서열의 최적 정렬은 월버(Wilbur)와 립맨(Lipman)(31)에 의해 개발된 연산을 사용하여 얻었다.
결과 및 검토
HPV-33의 게놈 배열
M13 숏건(shortgun) 클로닝/디데옥시 서열화법에 의해 결정된 HPV-33의 완전한 7909 뉴클레오티드 서열은 제1도에 나타나 있다. 각 위치는 평균 6.5회 서열화하였다. 다른 유두종 비루스 서열에 대한 규약에 따라 번호는 비암호화 영역에서 Hpa I의 인식 서열을 닮은 부위에서 시작한다.
넌센스 코돈(제2도 참조)의 배치에 대한 분석은 다른 모든 서열화된 유두종 비루스에서와 같이 8개의 주 개방 해독 프레임이 동일한 가닥 위에 위치함을 나타낸다. 코튼테일(cottontail)종 토끼의 유두종 비루스 및 원형 소의 유두종 비루스인, BPV-1과 함께 HPV-33과 HPV형 1a, 6b 16(5, 7, 8, 3, 21, 22)의 공통적인 몇몇 특징은 초기 영역, 즉 E1 및 E2의 가장 큰 개방 해독 프레임 사이의 오버랩 및 E2를 암호화한 부분 안에서 E4 함유부를 포함한다. 흥미롭게도 BPV-33의 분자 클로닝에서 사용되는 Bgl II 부위는 E1/E2 오버랩 안에 위치한다. BPV-1을 제외한 모든 유두종 비루스의 공통적인 다른 특성은 L1과 L2 해독 프레임 사이의 오버랩이다. L1 다음은 BPV1(15, 29)에서 유추하여 확실하게 복제 기점 및 각종 전사 조절 요소를 함유하는 892bp의 비암호화 영역이 존재한다. HPV-33 게놈의 주요 특성을 표 1에 요약하였다.
HPV-16과 뉴클레오티드 서열 비교
HPV-16은 완전히 서열화된 항문-생식기부의 종양으로부터 단리시킨 유일한 기타 종양원성(腫瘍原性) 유두종 비루스이다(22). HPV-33의 전체적인 특징은 HPV-16의 E1 해독 프레임이 차단되었다는 것을 제외하고는 HPV-16의 특징과 유사하다. E5의 암호화 서열을 제외한 HPV-33의 암호화 서열 모두는 HPV-16에 있어서 이들의 대응부보다 약간 짧다. 이것은 L1과 E6 사이의 비암호화 영역(제2도 참조)이 76염기쌍이 더 길고 그에 의하여 게놈을 거의 일정한 크기로 유지한다는 사실에 기여할 수 있다. 개방 해독 프레임을 한 쌍씩 비교할 경우(표 2), E1, E2, E6, E7, L1 및 L2는 65-75% 사이의 상동성을 나타낸 반명 E4 및 E5에 있어서는 보다 벗어남(약 50% 상동성)을 알게 되었다. 이러한 발견은 이전에 행한 헤테로듀플렉스 분석을 확증한다(1). 유두종 비루스 E1 유전자 생성물의 비교 실험(8)은 폴리펩티드가 그 서열이 매우 다양한 NH2-말단 소편 및 잘 보존된 1차 구조의 COOH- 말단부로 구성됨을 나타내었다. 완전한 서열 상동성을 갖는 가장 긴 신장부, 33개의 뉴클레오티드(위치 1275-1307,제1도 참조)는 폴리펩티드의 가변성 영역을 암호화하는 부위의 E1 해독 프레임의 5'-말단 근처에서 발견된다. 여러가지 다른 완전한 동질성을 갖는 부위(19-28 뉴클레오티드)를 E1 이외에서, 또한 E2, L2 및 L1에서 탐지하였다. 이 서열들 중 다수는 HPV-1a 및 HPV-6b와 같은 다른 HPV의 게놈에서 발견되지 않기 때문에, 이것은 대응하는 올리고뉴클레오티드를 제조해서, 잠재적으로 종양 발생 병변으로부터 생검(生檢) 물질을 선별하기 위한 진단용 혼성화 프로브로서 사용할 수 있는 가능성을 높인다.
잠재성 유전자 생성물
유두종 비루스 유전자 생성물은 순수하게 구조적인 역할을 하는 것으로 여겨지는 생성물 L1과 L2 비루스의 증식과 잔존에 필요한 생성물로 나누어질 수 있다. HPV-33, 16 및 6b로부터의 주 해독 프레임의 가능한 생성물의 비교 결과를 표 2에 요약하였다. 예측한 바와 같이 종양원성 HPV-33 및 16, 특히 제안된 E1, E6, E7, L2 및 L1 단백질 사이에 강한 동일성이 있다. 보존적 치환이 2종류의 L1 폴리펩티드 사이의 상동을 포함할 경우, 90%까지 증가는 대응하는 캡시드(capsid)가 반드시 항원적으로 관련되어야 함을 암시한다. 이와 대조적으로, 상당히 약한 상동은 분석을 양성(良性) 생식기 우췌 형성 HPV-6b를 포함하기 위해 확대시킬 경우에 탐지된다(표 2). HPV-16 단백질과 HPV-6b 단백질과의 비교는 HPV-33 단백질과의 비교에서 보다 약간 더 상동임이 나타났으며, 더 밀접한 진화 관계를 나타낸다.
비암호화 영역
HPV-33의 비암호화 영역은 여러 독특한 특성을 나타내며, HPV-16에서 그의 상동성과 약간 닮았을뿐이다. L1 정지 코돈 사이에 위치하며, 후기 전사의 추정되는 폴리아데닐화 신호를 포함하는 부분은 T+G가 대단히 풍부한(79%) 223bp(위치 7097-7320, 제1도 참조)의 신장 부분이다. 이 소편에는 2복제수의 19dp 직접 반복(1개의 착오 정합을 가짐)과 7복제수의 모티프 TTGTRTR(R은 A 또는 G임)이 함유된다. 후자는 또한 이것이 HPV-16의 대응하는 영역에서 7배가 발견되었으며 이것은 복제에 관련된 단백질의 인식 부위를 나타냄을 의미한다. 초기 복제 포크는 BPV-1 게놈(29)의 이 영역에 위치하며 엡스타인-바아(Epstein-Barr) 비루스의 복제 기점은 반복 서열의 일군으로 구성됨(32)을 유의해야 한다.
조사된 모든 유두종 비루스 게놈의 비암호화 영역에서 독점적으로 생기는 12bp의 팔린드롬(ACCG…CGGT)은 최근에 다트만(Dartmann) 등(9)에 의해 보고되었다. 3복제 수가 HPV-33 게놈에서 발견되었으며(제3도 참조), 이들은 HPV-16의 비암호화 영역에서 동일한 위치를 점유한다. 초기 프러모터에 대한 예상 조절 부위로서 팔란드롬의 역할이 제안되었으며(4, 9, 15), HPV-33 과 같은 HPV의 비암호화 영역이 프로모터 특이성 활성화 인자 Sp1(12)에 대한 인식 부위의 군집 배치를 나타내지 않는다는 본 발명자들의 발견에 의해 간접적으로 지지되었다. 이것은 다른 파포바 비루스, SV40(12, 14)의 환경과는 직접 대조된다.
HPV-33의 가장 현저한 특징은 추정되는 복제 기점의 200bp 이후에 위치된 완전한 78bp의 종렬 반복서열이다(제3도 참조). 이 크기 또는 서열의 다른 반복 서열은 다른 유두종 비루스의 게놈에서는 기재되지 않았다. HPV-33의 추정되는 초기 프로모터는 종렬 반복 서열로부터 약 300bp 아래로 위치하며, 특징적인 프로모터 요소(4)를 동정할 수 있다(제3도 참조). HPV-33 게놈에서 78bp의 반복 서열의 크기, 위치 및 배치는 이들이 비루스 전사의 인핸서로서 작용할 수 있는 것을 암시한다. 72, 73 및 68bp의 종렬 반복은 SV40(4, 14)의 초기 프로머터 근처, 몰로니(moloney)종 쥐의 육종 비루스(10)의 LTR 중에 및 BK 비루스 게놈(23)에서 위치하며, 시스-활성 방식으로 Pol II 의존 프로모터로부터 전사를 증강시킴을 나타낸다. SV40 인핸서(14, 30)의 돌연변이유발 및 특징적인 전사 활성화제의 서열 비교로부터 교감 인핸서 서열을 유도하였다. 이 구조는 78bp의 반복 서열에서는 탐지할 수 없었으나 잠재성 Z-DNA 형성 영역은 노출되었다. Z-DNA는 진핵성 프로모터에 대한 유력한 조절 분자로 여겨지며, Z-DNA 항체 결합 부위는 SV40 인핸서(18) 내에서 나타났다. 이 항체가 결합하는 서열은 또한 비록 단일의 착오정합을 가진 것이나, 추정되는 HPV-33 인핸서에서 발견된다(위치 7520-7527, 7599-7606, 제1, 3도 참조).
제안된 HPV-33 인핸서는 잘 특성화된 인핸서 또는 기타 유두종 비루스 조절 영역에 대해 신장되지 않은 서열 상동성을 나타낸다. 그러나, 최근에 인핸서와 유사한 요소가 BPV-1의 비암호화 영역에 위치한다는 것과, 이것이 활성화를 위해 E2 생성물을 필요로 한다는 것이 증명되었다.이 발견은 78bp의 종렬 반복부가 인핸서 작용을 가질 수 있고 HPV-33과 16의 E2 생성물 사이의 비교적 낮은 상동성(표 2)이 대응하는 인핸서/조절 영역에 대한 특이성을 반영한다는 본 발명자들의 제안을 뒷받침한다.

본 발명은 더욱 구체적으로 E6, E7, E1, E2, E4, E5, L2, L1의 개방 해독 프레임에 대응하는 서열에 관한 것이다.
본 발명은 또한 다른 유두종 비루스의 탐지에도 유용하며, 예를들면 L1에 대응하는 개방 해독 프레임의 일부 또는 전부를 함유하는 프로브 또는 보다 비루스 특이적인, 즉 이에 대응하는 개방 해독 프레임의 일부 또는 전부를 함유하는 것인 혼성화 프로브로서 이 서열들의 용도에 관한 것이다.
본 발명은 또한 유두종 비루스의 서브 그룹(sub-group)을 탐지하는 기타 프로브, 특히 중요한 부류의 질병과 관련될 수 있는 비루스, 즉 종양과 관련된 비루스 탐지용 프로브에 관한 것이다. 상기 프로브 중 하나의 예를 통해서, 이것은 제1a도, 1b도에서 뉴클레오티드의 번호 매김에 의하여 뉴클레오티드 1275와 1307 사이에 위치하는 서열을 함유해야 한다.
본 발명은 또한 적합한 표지, 즉 방사성 효소 또는 면역 형광 표지로 표지된 상기 DNA 서열 모두에 관한 것임을 말할 필요도 없다.
비루스 게놈으로부터 유도되고, 항체에 의해 인식될 수 있는 화학적 기에 의해 변형된 뉴클레오티드를 운반하는 DNA들도 본 발명의 일부를 형성한다. 이와 같은 DNA들은 적절히 변형된 뉴클레오티드 존재하에서 닉 트랜스레이션에 의해 제조될 수 있음은 공지되어 있다. 이 DNA들은 특히 구하고자 하는 상보적 사닥을 함유하는 DNA 제제에 혼성화될 경우, 상기한 항체에 의해 탐지될 수 있는 유용한 혼성화 프로브를 형성한다.
본 발명은 또한 그 자체로 진단 방법에 관한 것이다. 적합한 방법을 이후에 예시한다.
여러가지 혼성화 방법을 사용할 수 있다. 예를들면 스포트 혼성화 방법은 DNA의 변성 후, DNA의 부분표본을 필름 지지체(니트로셀룰로오스 또는 진스크린플러스)에 침착, 프로브를 지닌 통상의 조건 하에서 각 필름의 혼성화 및 혼성화 필름을 방사선 사진 필름에 접촉 노출함으로써 방사성 혼성체의 탐지를 포함한다. 또 다른 가능성은 제한 효소로 DNA를 처리하여 생긴 DNA 단편의 아가로스겔 전기 영동 분리, 알칼리 변성 후 단편의 필름(니트로 셀룰로오스 또는 진스크린플러스)으로의 전이 및 프로브의 상이한 혼합물을 사용하는 통상의 조건 하에서 이들의 잡종 형성을 포함하는 복제된 배양체 혼성화법이다. 방사성 혼성체의 형성은 홍성화 지지체 필름을 방사선 사진 필름에 접촉 노출시킴으로써 다시 탐지된다.
예를들면, 본 발명에 의한 프로브는 병소를 찰과시켜 얻은 세포의 생검 또는 카르노이(Carnoy) 혼합물(에탄올, 클로로포름, 아세트산 6 : 3 : 1)로 고정시키고, 파라핀 중에 포함시킨 생검 부분으로 되는 제제에 있어서 관련된 비루스(또는 그의 DNA)의 탐지에 사용할 수 있다.
상기 뉴클레오티드 서열은 벡터에 삽입시켜서, 적합한 세포 숙주에 도입될 경우, 전사를 제공할 수 있으며, 필요에 따라서 숙주의 세포 추출물로부터 단리시될 수 있는 대응하는 단백질을 생성하는 상기 DNA 서열의 번역을 제공할 수 있는 변형된 벡터를 제공한다. 당업계의 숙련된 지식 안에서 특히, 이로써 형질 전환시키고자 하는 숙주와 관련하여 적합한 벡터를 선택하는 것은 명백하다. 벡터는 예를들면 대응하는 원핵세포(또는 이스트 세포)에서 벡터를 복제하고 벡터가 운반하는 DNA 서열의 발현을 가능케하는 인정된 능력에 따라서 선택되는 플라스미드 또는 파지(phage)로 된다.
본 발명은 또한 상기 정의한 개방 해독 프레임 중의 어느 하나 또는 그 일부에 대응하며 진핵 세포, 특히 온혈 동물의 세포 중에서 삽입체의 발현을 허용하도록 적절히 DNA 서열로 되는 삽입체를 함유하는 DNA 재조합체에 관한 것이다. 적합한 DNA 재조합체는 상기 삽입체가 선택된 세포의 폴리머라제에 의해 인식되는 비루스 또는 진핵 세포의 프로모터의 조절 하에 놓여 있으며, 또한 상기 삽입체의 하류단의 적합한 폴리아데닐화 부위로 되는 유전자 구조물이다.
예로서, 본 발명은 SV40 비루스의 게놈으로부터 유도한 프로모터의 조절 하에 놓인 상기 개방 해독 삽입체 중의 어느 하나를 함유하는 DNA 재조합체에 관한 것이다. 이와 같은 DNA 재조합체-또는 벡터-는 고등 진핵 생물 세포, 특히 포유 동물 세포(예를들면 베로(Vero) 세포)의 형질전환에 사용할 수 있다. 본 발명은 또한 유사한 벡터에 삽입될 경우, 상기한 전체의 뉴클레오티드 서열에 의해 암호화된 단백질과 유사한 면역학적 특성을 갖는 대응하는 단백질의 일부에 대해 암호화할 수 있는 상기 동정된 DNA 서열 일부에 관한 것이다. 면역학적 특성의 유사성은, 관련 숙주에 의해 생성된 대응 폴리펩티드의 능력에 의해 확인될 수 있으며 이 폴리펩티드는 상기한 전체 DNA 서열을 함유하는 벡터로 미리 형질전환시킨 세포에 의해 생성된 단백질에 대해서 미리 형성시킨 항체에 의해 인식된다.
본 발명은 또한 적어도 부분 합성적으로 얻을 수 있는 상기한 것과 관련된 임의의 뉴클레오티드 서열에 관한 것으로서, 그 중에서 뉴클레오티드는 유전 코드의 구조 안에서 변할 수 있으며, 본 발명은 이 변화가 이와 같이 변형된 뉴클레오티드 서열에 의해 암호화된 폴리팹티드 서열의 실질적인 변형을 일으키지 않는 정도에 관한 것임은 물론이다.
본 발명은 또한 상기한 방법으로 얻을 수 있는 정제된 단백질 또는 폴리펩티드에 관한 것임은 이미 전술한 검토에서 나온다. 적합한 숙주에서 생성될 경우, 이 폴리펩티드는 예를들면 숙주의 세포벽을 파열시킨 후 세포로부터 얻거나 또는 상기 세포 배지에서 분비될 경우 사용되는 세포의 DNA 제조합 체계에 의존하여, 상기 세포의 배지로부터 얻을 수 있다. 이어서, 얻은 폴리펩티드를 통상적이 정제 방법에 의해 정제시킬 수 있다. 본문에서 "정제된"이란 SDS-PAGE에서 전기영동시킬 경우 정제시킨 단백질이 예를들면 웨스턴 블럿(Western blot)에 의해 단일 탐지 밴드를 얻는 순도의 수준을 의미한다는 것을 이해해야 한다.
예를들면 대장균(E.coli)에서 상기 DNA 서열의 발현의 결과로서 얻은 비루스 단백질, 더욱 구체적으로 구조 단백질은 유두종 비루스에 감염된 환자의 조직 시료 중에서 탐지되는 유두종 비루스에 대한 항체의 시험관 내 탐지에 사용할 수 있다.
L1 및 L2 개방 해독 프레임으로부터 추정되는 펩티드 서열을 갖는 유전적으로 처리된 단백질이 특히 적당하다. 중요한 다른 펩티드는 E6*단백질(E6 별표)로서, 그의 합성은 접합(splicing)에 의해 일어날 수 있으며, HPV 33 서열(더욱 구체적으로 1a도 참조)의 뉴클레오티드 229(공여체 부위)와 404(수용체 부위)사이에 위치한 뉴클레오티드 서열에 의해 암호화되며, 또한 그 부위는 HPV 33의 E6*개방 해독 프레임에서 추정되는 접합 부위를 나타낸다. 이러한 단백질의 생성 조건에 관해서는 슈나이더-가르디케(Schneider-Gardicke) 및 슈바르츠(Schwartz)의 출판물, Embo, J., 5, 2285-2292를 참조할 수 있다.
이 정제시킨 폴리펩티들은 또한 생물학적 유체, 특히 환자의 혈청 또는 조직 배양체 중에서 비루스 폴리펩티드의 존재를 시험관 내에서 진단하는데 사용할 수 있는 대응하는 항체의 생성에 사용할 수 있다. 상기 예에서와 같이, 본 발명은 상기 정의한 폴리펩티드, 특히 동일한 항체에 의해 인식되거나 또는 반대로 완전한 단백질을 인식하는 항체의 생성을 생체 내에서 유발할 수 있는 폴리펩티드의 일부에 관한 것이다.
본 발명은 또한 구체적으로 상기 명세서에서 언급된 DNA 영역에 의해 암호화되었으며, 특별한 중요성이 발견된 특별한 펩티드에 관한 것이다.
본 발명은 추가로 상기한 상이한 펩티드의 발현을 유도하는 뉴클레오티드 서열을 함유하는 DNA 재조합체로 형질전환되고, 적합한 배양 배지 중에서 배양될 경우, 상기 펩티드를 유효하게 생성할 수 있는 숙주 세포에 관한 것이다.
최종적으로 본 발명은 더욱 구체적으로는 상기 정제시킨 폴리펩티드를 사용하여 표준 방법으로 면역시킨 토끼와 같은 동물 및(또는) 또한 임의의 공지된 방법으로 미리 제조한 하이브리도마(hybridoma)로부터 얻을 수 있는 항체에 관한 것이다. 구조 단백질에 대해 유도된 항체(폴리클로날 항체 및 모노클로날 항체)들이 특히 중요하다. 이 항체들은 비루스 감염의 탐지에 유용하다. HPV-33의 L1, L2 및 E6* 단백질을 인식하는 항체들이 특히 중요하다. L2에 대해 특이성인 항체들은 유사한 L2 단백질을 암호화하는 HPV 33 서열을 갖는 특정 비루스의 시험관 내 탐지용 진단도구를 제공한다. L1에 대해 특이성인 항체들은 HPV-33이 속하는 비루스 군의 탐지에 유용하다. E6* 단백질에 대해 특이성인 항체들은 비루스가 유발하는 상기 비루스 감염의 종양원성 특성을 탐지하는데 유용하다.
본 발명은 또한 특히 중요한 유전자간 서열, 특별한 78bp의 서열에 관한 것이다. 이 서열은 진핵성 벡터중에, 특히 프로모터의 상부 위치와 유전자의 전사 또는 구하고자 하는 뉴클레오티드 서열 전사가 관련 숙주 내에서 개시되는 부위의 하부에서 가능한 삽입체로서 특히 중요하다.

- DNA 단편이 HPV-33의 게놈 DNA로부터 유도되고 제1a도 및 제1b도에서의 뉴클레오티드의 번호 매김과 관련하여 아래에 정의된 뉴클레오티드 말단 사이에서 확장되는 단편들로 이루어진 군 중에서 선택된 개방 해독 프레임으로 구성되는 DNA 삽입체와 함께 벡터를 숙주내에서 복제시키는 것을 특징으로 하는 DNA 삽입체를 함유하는 재조합 DNA의 제조 방법.76-556543-864867-28112728-38083326-35753842-40794198-56115516-8091
