Description
본 발명은 신규의 유기화합물과, 보다 자세하게는 중추신경계에 영향을 주는 약품으로서 관심을 모으는 신규의 4-치환 피페라진-2-카르복실산, 그들의 제조방법, 그들을 함유하는 제약조성물 및 인간이나 동물신체에 가해지는 치료법에 사용되는 화합물에 관한 것이다.
각종 아미노산은, 그들이 중추 신경계의 특정 수용체 부위의 작용에 영향을 미칠 수 있다는 발견에 따라 최근에 관심을 모으게 되었으며, 중추 신경계 기능 부전으로부터 초래되는 각종 무의식 근육 작용 및/또는 정신 및/또는 감정분열의 조절에 사용될 수 있는 화합물을 확인할 목적으로, 이들 수용체 부위와 관련된 특이 작용을 갖는 물질의 확인에 그 주목을 끌고 있다.
이제 우리는, 그러한 작용과 연관하여 전에는 연구된 바 없는 유형의 구조를 갖는 4-치환 피페라진-2-카르복실산이, 중추 신경계의 특정 수용체 부위에서 전기 작용의 억제제로 작용한다는 것을 발견하였다.
따라서 본 발명은 하기 일반 구조식(1)의 화합물을 제공한다:

본 발명 화합물에서 X가 C3쇄를 나타냄이 바람직한데, 그러한 화합물은 보다 활성인 화합물이기 때문이다. 그러나 X가 C1, C2, C4, C5또는 C6탄소의 직쇄인 화합물도 관심의 대상이다.
관심을 모으는 그외의 화합물은 X가 불포화 잔기인 화합물인데, 여기서는 탄소-탄소 불포화 부위가 한 개 있는 C3쇄를 함유하는 화합물에 특히 관심이 집중된다. 우리가 채택한 합성법은 올레핀형 이중결합 한 개를 함유하는 화합물의 생산에 가장 적합한데, 여기서 이중결합은 피페라진 고리와 가장 가까운 2개의 탄소원자 사이에 또는 피페라진 고리에서 가장 먼 2개의 탄소원자사이에 위치할 수 있다.
X잔기내의 수소원자중 적어도 한 개가 수소의 중 동위원소인 화합물은 추적자 실험에서 특히 중요하다. 그러한 방사성 화합물은 기 X내에 올레핀형 이중결합을 함유하는 본 발명 화합물을 삼중수소로 수소화시킴으로써 가장 쉽게 제조되기 때문에, 보통 2개의 중수소 원자를 함유한다.

R6및/또는 R7가 아릴기인 경우, 어떤 때는 화합물의 친수성/지방친화성 평형에 영향을 미치는 특성 때문에 나프틸 잔기와 같은 다핵 아릴 잔기가 중요하긴해도, 바람직한 것은 페닐기이다. 화합물의 친수성/지방 친화성 평형은 존재하는 할로겐 치환체의 존재 또는 부재에 의해 영향을 받을 수 있는데, 여기서 중요한 것은 플루오로, 클로로 또는 브로모 치환제이다.
기 Y에 대한 포스폰산 잔기 이외에 본 발명의 화합물은 설폰산, 설핀산, 카르복실산 또는 붕소산 잔기를 함유할 수 있으며, 전술한 바와 같이 이들 산의 잔기는 유리산 또는 에스테르 형태로 존재할 수 있고, 화합물이 유리산 또는 에스테르 형태로 표현되는데 있어서는 유사한 고려방법이 적용된다(후자의 경우 에스테르화 잔기의 선택에 적용됨).
Y에 대한 그외의 다른 기로서 페놀 또는 우라실, 또는 히단로인, 또는 바르비투르산 또는 이속사졸 또는 옥사디아졸리딘디온 잔기 또는 유사한 전기음성 부위를 제공하는 그외의 기가 있다.
본 발명 화합물내의 R4잔기는 화합물의 친수성/지방 친화성 평형에 영향을 주기 위해서도 도입된다. 본 발명의 가장 간단한 화합물의 경우, R4는 수소이지만, 그 외의 다른 구조로서 R4가 탄소 6개까지, 바람직하게는 1-4개를 함유하는, 예컨대 메틸, 에틸, n-프로필 또는 n-부틸과 같은 알킬 또는 알콕시기인 화합물이 있다. 본 발명의 화합물이 염기성 및 산성 작용기 모두를 함유하는 경우, 염기성 및 산성 작용기 모두 혹은 둘중의 하나는 염의 형태인 본 발명 화합물로 제조될 수 있다. 따라서 배합상의 이유로, 생리적으로 허용가능한 수용성 염(예:나트륨 염)형태의 2-카르복실산 잔기 및/또는 산 잔기 Y를 제조하는 것이 때로 바람직하다. COOR과 Y가 유리산 혹은 그의 에스테르나 그의 염 형태인 본 발명 화합물은 염기성 아미노기의 염 형태로 제조될 수 있으며, 여기서 중요한 염은 염산, 초산, 숙신산, 주석산 또는 구연산과의 염과 같이 생리적 허용가능한 산부가염이다.
본 발명의 화합물은, C2및 아마도 다른 곳에서 비대칭 중심을 항상 갖는다. 본 발명의 화합물은 라세미 혼합물 및, C2가 실질적으로 완전 R 배열이거나 실질적으로 완전히 S 배열인 화합물을 포함한다.
본 발명의 화합물은 하기 구조식(Ⅲ)의 피페라진-2-카르복실산 또는 에스테르를 하기 구조식(Ⅳ)의 화합물과 반응시킴으로써 제조할 수 있다 :
상기 식에서, R,R1,R2,R4,R5,X 및 Y는 앞서 정의한 바와 같으며, Z은 할로겐기 또는 P-토실기와 같은 반응성기를 나타낸다. 이러한 축합반응은 염기성 조건하에 실시되는 것이 보통이다. R4-X-Y가 함께 아실기를 나타내거나 함유하는 경우, 이 화합물은 염기성 조건하에 피페라진을 상응되는 할로겐화아실 또는 그외의 활성화 아실 전구체와 반응시킴으로써 제조될 수 있다.

Z가 활성 할로겐기, 바람직하게는 염소 또는 브롬인 중간체(Ⅳ)를 사용하는 것이 편리하지만, 전술한바와 같이 염기성 질소원자 상의 반응을 위해 그외의 합성법을 채택함으로써 피페라진 고리상에 XR4Y를 도입시킬 수 있다.
중간체(Ⅲ)은, 상응되는 피라진 카르복실산을 수소화시킴으로써 얻을 수 있다. 이러한 환원은 C2라세미 화합물을 생성시킬 수 있다. 그러나 개개의 이성체가 필요할 때는, 중간체(Ⅳ)의 반응에 앞서 수소화 생성물을 분해시키거나, 혹은 최종 축합생성물을 분해시킬 수 있다.
염 형태의 본 화합물을 생산해야할 경우, 염의 생성은 (Ⅲ)과 (Ⅳ)와의 반응 이후에 일어나는 것이 보통이지만, 몇몇 축합 반응은 염 생성물을 직접 생성시킬 수 있다.
본 발명의 다른 특색에 따라, 구조식(Ⅰ)의 화합물 및 제약상 허용가능한 희석제 또는 담체로 구성되는 제약 조성물이 제공된다. 본 발명의 화합물은 중추 신경계에 작용하며, 비경구적 또는 경구적으로 투여될 수 있다(예 : 급성 치료시에 정맥내 투여, 또는 만성 치료시에 피하 혹은 경구투여), 생리학상 허용가능한 유화제와 결합된 낮은 물-용해도의 조제물이 축적 투여에 사용될 수는 있으나, 본 발명의 화합물은 수용성염 조제물의 형태로서 적합한 부형제 내에 임상적 이용을 위해 배합된다.
본 발명의 화합물이 혈 뇌벽을 통과해야 한다고 생각되어지므로, 본 발명의 화합물은, 소요의 치료 효과를 위해 뇌 안에 이루어져야 할 양보다 상당 과량으로 투여될 필요가 있으며, 이것은 본 발명 조성물내 활성 화합물의 농도에 영향을 줄 수 있다. 이같은 생각은, 통상적인 투약 용량이 환자에게 체중 1kg당 약 200mg은 제공하도록 하는 농도로 본 조성물내에 활성 화합물이 함유되어야 한다는 것을 시사하고 있다. 그러나 본 화합물을 정맥내 또는 피하로 투여해야할 경우에는, 보다 활성인 화합물 및/또는 친화성/지방친화성의 평형이 잘된 물질을 위해 약 1-20mg/체중 범위의 용량이 기대된다.
본 발명의 다른 특색에 따라, 인간이나 동물 신체에 가해지는 치료법에 사용되는 구조식(Ⅰ)의 화합물이 제공된다. 보다 구체적으로, 본 발명의 화합물은, 흥분성 아미노산에 대한 포유동물 중추 뉴런(neurone)의 반응에 반작용하며, 또한 중추 신경계내에 자발적으로 일어난 시냅스 활동을 약화시킨다는 것이 발견되었다. 아미노산 수용체는, 뇌속의 많은 흥분성 시냅스의 흥분을 중개하거나 조절한다. 본 발명의 화합물은 아미노산 소용체를 포함한 비정상 중추신경계 작용을 수정할 수 있고, 그 결과 본 화합물은, 그러한 비정상이 발생하는 경우에 유익한 조정을 할 수 있다는 중요성을 갖는다.

[실시예 1]
3-((±)-2-카르복시피페라진-4-일)프로필-1-포스폰산(CCP)의 합성
(a) 에틸 3-((±)-2-카르복시피페라진-4-일)-프로프-2-엔일-1-포스포네이트
수산화나트륨(7.2g,0.18몰)을 함유하는 물(50ml)과 피페라진-2-카르복실산(8g,0.06몰)의 용액에다 디에틸 3-브로모-프로프-2-엔일-1-포스포네이트(15.75g,0.06몰)를 첨가한다. 산출된 용액을 교반시킨후 12시간 동안 65-70°하에 유지시킨다. 이 용액을 냉각시킨 후 묽은 염산을 사용하여 pH 7로 중성화시키고, Dowex AG-50H+수지를 적용시킨다. 닌히드린-양성의 수성 용출액을 결합하고 감압하에 증발시키면 에틸 3-((±)-2-카르복시피페라진-4-일)프로프-2-엔일-1-포스포네이트 9g이 백색 고체 형태로 얻어지는데, 이것은 더 정제하지 않은 채로 다음 단계에 이용된다.
(b) 에틸 3-((±)-2-카르복시피페라진-4-일)프로필-1-포스포네이트
에틸 3-((±)-2-카르복시피페라진-4-일)프로프-2-엔일-1-포스포네이트(5.0g)와 7M 수성 암모니아 용액(50ml)과의 용액을, 실온 및 대기압에서, 수소가 더 이상 취하여 지지않을 때까지 목탄(1g)상의 5% 팔라듐으로 수소화시킨다. 산출된 용액을 감압하에 증발시켜 에틸 3-((±)-2-카르복시피페라진-4-일)프로필-1-포스포네이트(4.8g)를 얻는다. 에탄올로부터 재결정화시키면 융점 140-4°인 백색 고체가 얻어진다.

(c) 3-((±)-2-카르복시피페라진-4-일)-프로필-1-포스폰산
에틸 3-((±)-2-카르복시피페라진-4-일)-프로필-1-포스포네이트(4.7g)의 용액을 6M 염산(100ml)에 체워넣고, 산출된 용액을 환류하여 2시간동안 끓인다. 용액을 증발건조시킨 후 Dowex AG-1-초산 염 수지 컬럼에 적용시키기 위해, 최소 용량의 물속에 잔사를 집어넣는다. 생성물인 3-((±)-2-카르복시피페라진-4-일)-프로필-1-포스폰산을 0.1M 초산을 사용하여 수지로부터 용출시킨다. 용출액을 증발시킨 후 수성 에탄올로부터 잔사를 재결정화시키면 융점 174-5°인 백색 고체 3.9g이 얻어진다.

[실시예 2]
3-((±)-2-카르복시피페라진-4-일)-프로판-1-설폰산(CPS)의 합성
나트륨 3-브로모-프로판-1-설포네이트(2.25g,0.01몰)을 피페라진-2-카르복실산(1.3g,0.01몰)과 수성 수산화나트륨(0.9g NaOH/30ml H2O)과의 용액을 첨가한다. 산출된 용액을 12시간동안 60-65°에 유지시킨다. 반응혼합물을 Dowex AG-1-초산염 수지에 통과시킴으로써 생성물인 3-((±)-2-카르복시피페라진-4-일)-프로판-1-설폰산을 분리해 낸다. 컬럼 여액과 수성세척액을 증발시키고, 50% 수성 에탄올로부터 조생성물을 재결정화시키면, 융점 190-191°인 백색결정성 고체 형태의 3-((±)-2-카르복시피페라진-4-일)-프로판-1-설폰산(500mg)이 얻어진다.

[실시예 3]
2-((±)-2-카르복시피페라진-4-일)-프로피온산의 합성
피페라진-2-카르복실산(1.3g, 0.01몰)과 2-클로로프로피온산(1.1g,0.01몰)을 수성 수산화나트륨 용액(0.9g NaOH/20ml H2O)에 용해시킨후, 산출된 용액을 12시간동안 60-65°로 유지시킨다. 0.3M 초산으로 용출시키면서 Dowex AG-1-초산염 수지상에서 이온 교환 크로마토그라피시킴으로써 생성물을 분리해낸다. 50% 수성 에탄올로부터 재결정화시키면 융점 173-4°인 백색 고체 형태의 2-((±)-2-카르복시피페라진-4-일)-프로피온산(520mg)이 얻어진다.

[실시예 4]
4-(4-브로모벤조일)-피페라진-2,3-디카르복실산(BBP)의 합성
피페라진-2,3-디카르복실산(1.74g/0.01몰)과 수성 수산화나트륨(1.2g/20ml)과의 빙냉 용액에 30분에 결쳐 4-브로모벤조일 클로라이드(2.2g/0.01몰)을 첨가한다. 산출된 용액을 0°에서 2시간동안 교반시킨후 실온으로 하룻밤 동안 가온시킨다. 이 용액을 Dowex AG-1-초산염 수지의 컬럼에 적용시키고, 3M 초산 용출액의 닌히드린 양성 분액을 결합한 후 증발건조 시킨다. 50% 수성 에탄올 용액으로부터 잔사를 재결정화시키면, 융점 218-20°인 백색 고체 형태의 4-(4-브로모벤조일)-피페라진-2,3-디카르복실산(800mg)이 얻어진다. 본 발명의 화합물은 중추 신경계내 흥분성 아미노산 수용체에서 길항작용을 갖는다. 이들 수용체에는 몇가지 다른 유형이 있는데 그들 모두 또는 그들중 몇몇은 중추신경기능과 밀접하게 연관되어 있다. 신경 과학문헌에 기술된 3가지 유형의 수용체는, N-메틸-D-아스파르테이트(NMDA), 카이네이트(K) 및 키스쿠알레이트(Q) 수용체로 알려져 있다. 본 발명의 화합물은 이들 수용체에서 다른 작용을 하는데, 어떤 것은 NMDA 수용체에서 보다 유효하고 다른 것들은 K 또는 Q 수용체에서 더 유효하다. 흥분성 아미노산 수용체에서 작용하는 화합물은 중성 아미노산 전달자 물질의 작용에 영향을 줌으로써 중추신경계의 전기작용에 영향을 미칠 수 있다.
아미노산 수용체에서 물질이 신경세포(뉴런)에 작용하는 것을 평가하기 위하여, 물질을 척수대 뉴런(뇌의 신경세포와 유사한 특성을 가짐)상에서 시험할 수 있다. 전형적으로, 개구리 또는 4-8일된 쥐에서 분리한 척수대가 사용되며, 화합물이, 흥분성 아미노산 또는 구심성 섬유의 전기 자극에 의해 유도된 척수 뉴런의 작용에 영향을 미치는 능력을 시험한다. 표2는 전에 알려진 흥분성 아미노산 길항물질에 관한 본 화합물의 상대적 능력을 제시해준다.
전에 알려진 가장 유력한 NMDA 길항물질은 2-아미노-5-포스포노펜탄산(AP5)과 2-아미노-7-포스포노헵탄산(AP7)이며, γ-D-글루타밀글리신(γDGG)는 전에 알려진 카이네이트 및 키스쿠알레이트의 길항물질중 가장 유력한 것의 하나이다. 본 화합물 3-((±)-2-카르복시피페라진-4-일)-프로필-1-포스포네이트(CPP)은 NMDA 및 폴리시냅스 척수 흥분의 길항물질로서 AP5 또는 AP7에 비하여 보다 더 유력하다는 것을 표 2로부터 알 수 있다. 본 화합물 4-(4-브로모벤조일)-피페라진-2,3-디카르복실산(BBP)는 카이네이트 및 키스쿠알레이트-유도 분극소실의 길항물질로서 γDGG에 비해 보다 더 유력하며, 모노시냅스 흥분의 저하제로서도 보다 더 유력하다.
NMDA 수용체 길항물질은 항경련 작용을 갖는다고 알려졌다. 공지의 흥분성 아미노산 길항물질중에서 이전에 가장 유력한 것을 AP7이었다. 쥐에서의 3-메르캅토 프로피오네이트-유도 경련에 대한 시험을 할 때 CPP는 AP7에 비하여 상당히 높은 항경련 활성을 갖는다는 것이 표 3에 나와 있다.

1 : 4-8일된 쥐에서 꺼낸 척수대내의 운동성 뉴런의 모노시냅스(MS) 및 폴리시냅스(PS) 흥분을 배근의 전기적 자극에 의해 유도해내고, 상응되는 복근으로부터 기록한다. 도표는 두 성분의 저해율에 관한 것이다(오실로프래프 추적 및 도표 기록 추적으로부터 각기 측정).
2 : 개구리(a) 또는 4-8일된 쥐(b)의 척수대한에서의 아미노산-유도 운동성 뉴런 분극소실을, 과융 매체내에 연속 존재하는 NMDA(5μM), 카이네이트(2.5μM) 또는 키스쿠알레이트(2.5μM)에 의해 유지시키고, 본 발명 및 표준 물질이 이들 분극소실을 역전시키는 상대적 능력을, 0.1μM-1mM의 농도로 과융 매체내에 2분간 첨가할 때 그의 효과로부터 평가한다. 각 물질을, 3가지의 모든 동근에 대해 효과적이라고 알려진 표준 길항물질로서의 γDGG(10μM-1mM)와 비교한다. 도표는, 동일한 역전도를 이루기 위한 동근의 상대적 농도를 제시해준다. 매체는 테트로도톡신(0.1μM)을 함유한다.

Claims (10)
- 하기 구조식(Ⅲ)의 피페라진-2-카르복실산 또는 에스테르를 하기 구조식(Ⅳ)의 화합물과 반응시키는 것으로 구성되는, 하기 구조식(I)의 화합물 또는 생리학적으로 허용가능한 그의 염의 제조방법.상기식에서, X는 C1-C6직쇄의 포화 또는 불포화된 히드로카르빌기인데, 기 R4와 Y는 이 사슬내의 임의 위치에 위치하며, X내의 수소원자중 한 개 또는 그 이상은 수소의 중 동위원소일수 있으며 ; R4는 수소이거나, 또는, 알킬, 알콕시, 히드록시, 아릴, 아릴옥시, 아르알킬, 아르알콕시, 아르알킬아미노 또는 모르폴리노기인데 여기서 상기 기들중의 알킬 또는 아릴부위는 한 개 또는 그 이상의 할로겐기로 치환될 수 있고 ; 또는 R4는 X의 탄소원자 1개 이상와 함께 고리원자가 5-6개인 카르보시클릭 또는 헤테로시클릭 고리를 형성하며 ; Y는 산성기 또는 이것의 에스테르 또는 염, 혹은 페놀, 우라실, 히단로인, 바르비투르산, 이속사졸 또는 옥사디아졸리딘디온 잔기인데, 기 안에 한개 또는 그 이상의 전기음성 부위를 생기게 하며 ; 또는 R4-X-Y가 카르복실아실기를 나타내며 ; R은 수소 또는 알킬, 할로알킬, 아릴, 할로아릴, 아르알킬 또는 할로아르알킬기이며 ; R1은 수소 또는 알킬, 할로알킬, 아릴, 할로아릴, 아르알킬 또는 할로아르알킬기이며 ; 같거나 다른 R2, R3및 R5는 각각 수소 또는 알킬, 히드록시, 알콕시, 카르복시, 알킬옥시카르보닐, 할로, 아릴, 할로아릴 또는 아릴옥시카르보닐기이고 ; 혹은 R2및 R3는 그들의 결합되어 있는 탄소원자가 함께 고리 시스템을 형성거나 또는 R3화 R4가 함께 및/또는 R3와 X가 함께 한 개 또는 그 이상의 고리시스템을 형성하며, Z은 반응성기를 나타낸다.
