트롬빈 억제 활성을 갖는 폴리펩티드 및 그의 제조방법

제2도는 HV1형 히루딘 분비플라스마, pHph208를 제조하는 방법을 나타낸다.
제3도는 히루딘을 코딩하는 DNA서열을 함유하는 하이브리드 플라스미드 p3009를 제조하는 방법을 나타낸다.
제4도는 히루딘을 분비하는 플라스미드, pNPH141를 제조하는 방법을 나타낸다.

Claims (14)

  1. 하기 아미노산 서열을 갖는, 트롬빈 억제 활성을 갖는 폴리펩티드의 전구체,
  2. 하기 아미노산 서열을 갖는, 트롬빈 억제 활성을 갖는 폴리펩티드,
  3. 제1항에 기재된 트롬빈 억제 활성을 갖는 폴리펩티드의 전구체를 코딩하는 하기의 DNA 서열.
  4. 제2항에 기재된 트롬빈 억제 활성을 갖는 폴리펩티드의 전구체를 코딩하는 하기의 DNA 서열.
  5. (a)프로모터 영역 및 리보소옴 결합 부위를 코팅하는 영역을 함유하는 DNA서열, (b) DNA서열(a)의 3'말단에 결합 가능한, 제1항에 기재된 전구체를 코딩하는 DNA서열, 및 (c)숙주에서 복제가능한 벡터DNA를 함유하는 분비 플라스미드.
  6. 제5항에 있어서, DNA서열(a)내의 여역이 바실루스 아밀로리퀴파시엔스(Bacillus amyloliquifaciens)의 중성 프로테아제 유전자의 것이 분비 플라스미드.
  7. 제6항에 있어서, (a)와 (b)의 결합 DNA 서열이 하기와 같은 분비 플라스미드.
  8. 제5항 내지 제7항중 어느 한 항에 있어서, 벡터 DNA가 바실루스속의 세균에서 복제 가능한 플라스미드인 분비 플라스미드.
  9. 제8항에 있어서, 프라스미드가 pUB110인 분비 플라스미드.
  10. 제9항에 있어서, 분비 플라스미드가 pNPH141인 분비 플라스미드.
  11. 제5항 내지 제10항에 기재된 분비 플라스미드 중의 하나를 사용하여 형질 전환된 형질전환체.
  12. 제11항에 있어서, 형질전환된 미생물이 바실루스 서브틸리스 (Bacillus subtilis)인 형질전환체.
  13. 제12항에 있어서, 바실루스 서브틸리스가 바실루스 서브틸리스 FERM BP-2403인 형질전환체.
  14. 제12항 또는 제13항의 형질 전환체를 배양하고 배양 상충액으로부터 트롬빈 억제 활성을 갖는 폴리펩티드를 회수하는 단계를 특징으로 하는, 트롬빈 억제 활성을 갖는 폴리펩티드의 제조방법.
