Claims (10)

  1. 유전 공학적으로 생성된 폴리펩파드를 함유하는 세균 세포를 pH 내지 2.0의 수성산에 현탁시키고 기계적으로 파괴시킨 다음에, 생성된 현탁액중에 존재하는 폴리펩티드를 분리 및 정제함을 특징으로 하여, 재조합 방법에 의해 생성된 폴리펩티드를 세균 세포로 부터 분리하는 방법.
  2. 제1항에 있어서, 세균 세포로 재조합 방법에 의해 생성된 산-안정성 인터페론을 함유하는 세균 세포를 사용하는 방법.
  3. 제1항에 있어서, 세균 세포로 재조합 방법에 의해 생성된 알파-인터페론을 함유하는 세균 세포를 사용하는 방법.
  4. 제1항에 있어서, 세균 세포로 재조합방법에 의해 생성된 IFN-α2(arg)를 함유하는 세균세포를 사용하는 방법.
  5. 제2항 내지 제4항중 어느 하나에 있어서, pH 2.5 내지 3.5에서 수행하는 방법.
  6. 제1항 내지 제5항중 어느 하나에 있어서, 생성된 전단력에 의해 세균 세포를 파괴시키는 방법.
  7. 제1항 내지 제6항중 어느 하나에 있어서, 세균 세포를 균질화기를 사용하여 파괴시키는 방법.
  8. 제7항에 있어서, 사용되는 균질화기가 울트라-투랙스형(ultra-Turrax type), 바람직하게는 디스팩스-리액터 3-6/6(Dispax-Reactor 3-6/6)의 균질화기인 방법.
  9. 제1항 내지 제8항중 어느 하나에 있어서, 사용되는 산이무기산, 또는 아세트산이나 프로피온산과 같은 알카노산인 방법.
  10. 제1항 내지 제9항중 어느 하나에 있어서, 사용되는 수성산이 pH 2.5의 묽은 염산, 또는 1% 아세트산인 방법.
