Claims (14)

  1. 글리코실화되지 않고, 최소한 약간의 분있간 디술파이드 결합이 있고, 약 50,000달톤의 분자량의 갖는 폴리펩티드로서 비-환원 변성 폴리아크릴아미드 겔 전기 영동 시스템에서 이동함을 특징으로 하는 입자크기가 약 16 내지 약 25mm인 HBsAg의 면역원형.
  2. 제1항에 있어서, HBsAg의 실제적인 정제형.
  3. 황원자의 90% 이상이 디술파이드 결합을 형성하며 그중 적어도 일부가 분자간 디술파이드 결합이며 100,000달톤 이상의 분자량의 갖는 폴리펩티드로서 비-환원 변성 폴리아크릴아미드 겔 전기 영동 시스템에서 이동하지 않으며, pH2에서 불안정하고 펩신에 대하여 불안정하며 포스파티딜이노시톨을 함유하나 콜레스테롤은 함유하지 않는 HBsAg 입자의 글리코실화 되지 않은 면역원형.
  4. 제3항에 있어서, HBsAg의 실제적인 정제형.
  5. 제1항에 있어서, 시험관 내에서, 형질전환된 진핵성 숙주에 의해 발현된 HBsAg를 pH8.5 또는 그 이상에서 처리하여 생성시킨 HBsAg의 면역원형.
  6. 제3항에 있어서, 시험관내에서, 형질전환된 진핵성 숙주에 의해 발현된 HBsAg를 분자간 디술파이드 결합을 형성하는데 적용하는 화학적 수단으로 처리하여 생성시킨 HBsAg의 면역원형.
  7. 제6항에 있어서, 화학적 수단이 티오시아네이트, 요소 또는 약 11 내지 12.5의 pH로 처리하는 방법인 HBsAg의 면역원형.
  8. 시험관내에서, 형질전환된 진핵성 숙주에 의해 발현된 HBsAg를 pH8.5 또는 그 이상의 pH에서 처리함을 특징으로 하여, 글리코실화 되지 않고, 최소한 약간의 분자간 디술파이드 결합이 있고, 약 50,000달톤의 분자량을 갖는 폴리펩티드로서 비-환원 변성 폴리아크릴아미드 겔 전기 영동 시스템에서 이동함을 특징으로 하는 입자크기가 약 16 내지 약 25mm인 HBsAg의 면역원형을 제조하는 방법.
  9. 시험관내에서, 형질전환된 진핵성 숙주에 의해 발현된 HBsAg를 분자간 디술파이드 결합을 형성시키기에 적합한 화학적 수단으로 처리함을 특징으로 하여, 황원자의 90% 이상이 디술파이드 결합을 형성하며 그중 적어도 일부가 분자가 디술파이드 결합이며, 100,000달톤 이상의 분자량을 갖는 폴리펩티드로서 비-환원 변성 폴리아크릴아미드 겔 전기 영동 시스템에서 이동하지 않으며 pH2에서 불안정하고 펩신에 대하여 불안정하며 포스파티딜이노시톨을 함유하나 콜레스테롤은 함유하지 않는 HBsAg 입자의 글리코실화 되지 않는 면역원형을 제조하는 방법.
  10. 제9항에 있어서, 화학적 수단이 티오시아네이트, 요소 또는 약 11 내지 약 12.5의 PH로 처리하는 방법인, HBsAg의 면역원형을 제조하는 방법.
  11. 비전하 공유 결합의 우세로 소수성 기질에 페닐 또는 탄화수소 치환체가 결합되어 형성된 기질에 HBsAg를 통과시킴을 특징으로 하여 효모 세포에서 생성된 HBsAg로부터 발열성 물질 및 효모 단백질을 제거하는 방법.
  12. 제13항에 있어서, 치환체가 탄소수 4 내지 10의 알킬아민인 방법.
  13. 제13항에 있어서, 알킬아민이 부틸아민, 헥실아민, 옥틸아민 또는 데실아민인 방법.
  14. 제13항에 있어서, 알킬아민이 n-부틸아민인 방법.
