Abstract
본 발명은, 약물의 송달 기술에 이용 가능한 신규 코폴리머를 제공하는 것. 보다 상세하게는 종양을 표적으로 한 약물 송달 캐리어용의 신규 코폴리머를 제공한다. 본 발명은, 다음 식 (A), (B) 및 (C)로 표시되는 구조 단위로 이루어지는 코폴리머에 관한 것이다. Description
본 발명은, 약물 송달 기술에 이용 가능한 신규 코폴리머에 관한 것이다. 보다 상세하게는 종양을 표적으로 한 약제 송달 캐리어용의 코폴리머, 당해 코폴리머에 항암제 등의 생리 활성 물질을 담지시킨 의약용 조성물, 당해 조성물을 함유하는 의약품에 관한 것이다.
최근 들어, 약물을 질환 부위에 효율적이며 동시에 안전하게 송달하는 기술로서, 드럭 딜리버리 시스템(Drug delivery system, DDS)에 관한 연구가 활발히 이루어지고 있다. 그 중에서도, 질환 부위의 구조적인 특성을 이용하여 약물 집적의 선택성을 높이는 기술로서, 나노 입자를 약물 송달 캐리어로 한 DDS의 수요가 높아지고 있다.
예를 들어 고형암 조직에 있어서는, 신생 혈관(종양 혈관)의 구조가 정상 혈관과 비교하여 미성숙이기 때문에, 혈관 내피에 수백 nm 정도의 세포 간극이 생겨 물질의 투과성이 높다. 이 구조적 특징에 의해 나노 입자를 포함하는 고분자량체는, 종양 혈관을 선택적으로 투과하여 고형암 조직에 집적된다는 것이 알려져 있다. 또한, 고형암 조직에서는 고분자의 배출에 관여하는 림프계가 기능 부전을 일으키고 있기 때문에, 침투한 나노 입자는 조직 내에서 지속적으로 체류된다 (Enhanced permeability and retention effect, EPR 효과). 일반적인 저분자 약제는 혈관 세포의 막투과에 의해 혈관 밖으로 누출되기 때문에, 비선택적으로 조직에 분포되어 고형암 조직에 집적되지 않는다. EPR 효과의 방법론에 의하면, 나노 입자를 이용한 약물 송달은, 조직으로의 분포가 혈관 내피 세포 간극의 투과성에 의해 지배되기 때문에, 약물의 분포에 고형암에 대한 조직 선택성의 향상을 가져온다. 그 때문에 EPR 효과는, 고형암을 표적으로 한 나노테크놀로지 응용 의약품(나노 약물)의 개발에서의 유력한 학술적 근거가 되고 있다.
EPR 효과에 있어서의 약물의 송달 과정은 혈류를 통하고 있고, 또한 나노 입자의 혈관 외 누출 프로세스는 수동적이라고 생각되고 있다. 따라서, 고형암에 대한 나노 입자의 집적을 최대화하기 위하여는, 약물 송달 캐리어가 되는 나노 입자의 구성 성분에 대하여 장기간의 혈중 체류에 견딜 수 있는 분자 디자인을 부여하는 것이 중요하다. 그 때문에 약물 송달 캐리어에는, 혈액 구성 성분과의 비특이적 상호 작용, 간, 비장, 및 폐에 있어서의 세망내피계(reticuloendothelial system, RES)에 의한 이물 인식, 신장에 있어서의 사구체 여과와 같은 장벽을 회피하는 능력이 요구된다. 또한, 이러한 장벽은, 입자 직경이나 생체 적합성 고분자에 의한 표면 수식과 같은 입자 특성의 최적화에 의해 극복할 수 있다는 것이 알려져 있다. 예를 들어 약물 송달 캐리어의 입자 직경은, 신장 클리어런스의 역치인 약 6nm보다도 크고, RES에 의한 인식을 모면할 수 있는 200nm보다도 작은 것이 바람직하다.
또한, 약물 송달 캐리어의 입자 직경은, 질환 부위에 있어서의 조직 침투성에도 영향을 끼친다는 것이 알려져 있다. 예를 들어 동등한 혈중 체류성을 나타내는 입자 직경 30nm, 50nm, 70nm 및 100nm의 약물 내포 나노 입자의 제암 활성이 비교 검토되어 있고, 입자 직경 30nm의 약물 내포 나노 입자는 질환 부위 심부까지 도달한다는 점에서 가장 높은 치료 효과를 나타낸다는 것이 밝혀져 있다(비특허문헌 1). 따라서, 고형암을 표적으로 한 약물 송달 캐리어용의 나노 입자의 입자 직경은, 신장 클리어런스를 회피할 수 있는 범위에서 가능한 한 작은 직경인 것이 바람직하다고 생각된다.
약물 송달 캐리어용의 나노 입자로서는 리포솜, 에멀션, 또는 나노파티클 등의 콜로이드 분산체를 사용하는 방법, 알부민 등의 생물 유래 원료를 사용하는 방법, 천연 다당류 등의 천연 고분자를 사용하는 방법, 혹은 합성 고분자를 사용하는 방법이 개발되어 있다. 그 중에서도 합성 고분자는, 구성 성분이 되는 모노머와 합성 방법을 적절하게 선택함으로써, 입자 직경이 정밀하게 제어된 나노 입자를 조제하는 것이 가능하기 때문에, 약물 송달 캐리어의 구성 성분으로서 범용되고 있다.
예를 들어 친수성 세그먼트와 소수성 세그먼트로 이루어지는 양친매성 블록 공중합체의 약물 송달 캐리어로서의 이용 방법에 대하여 개시되어 있다. 해당 블록 공중합체는 분자간의 소수성 상호 작용 등을 구동력으로 하여 수성 매체 중에서 자발적으로 회합하고, 코어-쉘형 나노 입자(고분자 미셸)를 형성한다. 해당 고분자 미셸의 소수성 세그먼트에는 저분자 약제를 내포 또는 결합하는 것이 가능하고, 얻어진 약물 내포 고분자 미셸은 높은 혈중 안정성을 나타냄과 함께, EPR 효과를 통한 고형암으로의 선택적인 집적에 의해 저분자 약제의 용액 투여와 비교하여 높은 제암 활성을 가져온다는 것이 알려져 있다(특허문헌 1). 그러나 고분자 미셸은, 복수 분자의 회합체이므로, 입자 직경 약 30nm 정도가 조제 가능한 하한값이며, 신장 클리어런스의 영향을 회피할 수 있는 입자 직경 10nm 부근에서의 미세한 사이즈 제어는 곤란하다.
한편, 합성 고분자에 의해 형성되는 나노 입자 중, 단일 가닥 내의 화학 가교, 소수성 상호 작용, 이온 결합 등을 구동력으로 하여 입자를 형성하는 것(이하, 싱글 체인 나노파티클(single chain nanoparticle; SCNP)이라고 약기한다)은 입자 직경 20nm 이하의 작은 직경인 나노 입자를 형성한다고 알려져 있다(비특허문헌 2). 따라서, SCNP는 약물 송달 캐리어로서의 유용성이 기대되지만, 그 입자 직경을 정밀하게 제어하는 기술은 지금까지 발견되지 않았다.

본 발명은, 종양을 표적으로 한 약제 송달 캐리어용의 코폴리머를 제공하는 것을 과제로 한다. 보다 상세하게는 약물의 혈중 체류성 및/또는 종양 집적성을 향상시키기 위하여 이용 가능한 약물 송달 캐리어용의 코폴리머를 제공하는 것을 과제로 한다.
본 발명자들은, 상술한 과제를 해결하기 위하여 예의 검토하던 중, 아크릴산 유도체의 3원 공중합체가 수중에서 SCNP를 형성하는 특성을 발견하였다. 또한, SCNP를 20nm 이하의 10nm 정도의 미소한 스케일에 있어서의 정밀한 입자 직경 제어가 가능한 것 이외에, 종양 집적성이 높은 약물 송달 캐리어용의 신규 폴리머를 창제하는 것에 성공하였다. 또한, 해당 폴리머에 항암제를 담지시켜, 대장암 피하 이식 모델 마우스에 투여했더니, 우수한 항종양 효과를 확인하였다.

후술하는 실시예로부터 명확한 바와 같이, 본 발명 코폴리머의 자기 회합에 의해 얻어진 SCNP에 항암제를 담지시킨 것은, 마우스 담암 모델에 있어서 종양 증대 억제 효과를 나타냈으므로, 악성 종양의 치료제로서 적용이 가능하다. 본 발명의 코폴리머 자기 회합에 의해 얻어진 SCNP에 항암제를 담지시킨 것은, 저용량으로 높은 종양 증대 억제 효과를 갖는다는 점에서, 약리 작용의 증강과 부작용의 억제의 양립이 가능한 악성 종양의 치료제를 제공할 수 있다.

본 명세서에 있어서 "의약 조성물"이란, 질환의 진단, 예방 또는 치료에 사용할 수 있는 유효 성분(약물, 생리 활성 물질)이 본 발명의 코폴리머에 정전적 상호 작용, 수소 결합, 소수적 상호 작용 또는 공유 결합과 같은 작용 등에 의해 담지되어 있는 것을 의미한다. 담지의 형태로서는, 코폴리머가 나노 입자를 형성하고 있을 경우, 약물이 입자 표면에 존재하고 있는 형태나 약물이 나노 입자 내에 내포되어 있는 형태, 또는 이러한 조합 형태를 들 수 있다.
본 발명의 코폴리머는 여러 가지 공지된 방법에 의해 제조할 수 있다. 제조 방법은 특별히 제한되는 것이 아니지만, 예를 들어 이하에 기재하는 기본적인 고분자의 합성 방법에 따라 제조할 수 있다.

본 반응은, 모노머 (I)에 연쇄 이동제 (II)와 개시제를 반응시켜, 폴리머 (III)을 제조하는 공정을 나타낸다. 본 반응은 무용매로 행하거나, 또는 메탄올, 에탄올, 1-프로판올, 2-프로판올 등의 알코올류, 디에틸에테르, 테트라히드로푸란, 1,4-디옥산 등의 에테르류, 벤젠, 톨루엔, 크실렌 등의 방향족 탄화 수소류, 디클로로메탄, 클로로포름, 1,2-디클로로에탄 등의 할로겐화 탄화 수소류, N,N-디메틸포름아미드, N,N-디메틸아세트아미드, N-메틸피롤리돈, 아세토니트릴, 아세트산 에틸 등의 용매 중에서 행할 수 있고, 톨루엔, 크실렌 등의 방향족 탄화 수소류를 용매로서 사용하는 것이 바람직하다. 연쇄 이동제로서는, 2-(Dodecylthiocarbonothioylthio)-2-methylpropionic acid(DDMAT), Cyanomethyl dodecyltrithiocarbonate(CDTTC), 2-Cyano-2-propyldodecyl trithiocarbonate(CPDTTC), 4-Cyano-4-[(dodecylsulfanyl-thiocarbonyl)sulfanyl]pentanoic acid(CDSPA) 등을 사용할 수 있고, DDMAT를 사용하는 것이 바람직하다. 연쇄 이동제를 사용하여 중합했을 경우, 본 발명의 코폴리머는, 연쇄 이동제의 구조의 일부 또는 전부가 부분적으로 결합된 구조를 취한다. 코폴리머가 연쇄 이동제의 구조를 포함하는 경우, 적당한 방법에 의해, 그 구조를 제거해도 된다. 개시제로서는, 2,2'-Azobis-isobutyronitrile(AIBN), 1,1'-Azobis(cyclohexanecarbonitrile) (ACHN), 2,2'-Azobis-2-methy1b?utyronitrile(AMBN), 2,2'-Azobis-2, 4-dimethylvaleronitrile (ADVN) 등의 아조계 중합 개시제를 사용할 수 있고, AIBN을 사용하는 것이 바람직하다. 반응 온도는 0 내지 300℃, 바람직하게는 0 내지 150℃, 보다 바람직하게는 실온 내지 100℃이며, 반응 시간은 1분 내지 48시간, 바람직하게는 5분 내지 24시간이다. 본 반응에 있어서, 구조가 다른 모노머 (I)의 공존 하에 반응을 행함으로써 랜덤 공중합한 코폴리머를 제조할 수 있다.
제조한 본 발명의 코폴리머는, 고분자 화학의 분야에 있어서 일반적으로 알려져 있는 폴리머의 단리, 정제 방법에 의해 정제할 수 있다. 구체적으로는, 예를 들어 추출, 재결정, 황산 암모늄이나 황산 나트륨 등에 의한 염석, 원심 분리, 투석, 한외 여과법, 흡착 크로마토그래피, 이온 교환 크로마토그래피, 소수성 크로마토그래피, 순상 크로마토그래피, 역상 크로마토그래피, 겔 여과법, 겔 침투 크로마토그래피, 어피니티 크로마토그래피, 전기 영동법, 향류 분배 등이나, 이러한 조합 등의 처리 조작을 들 수 있다.
본 발명의 코폴리머는, 여러 가지 생리 활성 물질(약물)을 수송하기 위한 담체로서 이용할 수 있다. 예를 들어 본 발명의 코폴리머에 종양 치료약을 담지(내포)한 의약 조성물은, 후술하는 시험예에서 확인된 바와 같이, 종양의 증식을 억제 하기 때문에, 예를 들어 대장암, 십이지장암, 위암, 췌장암, 간암, 폐암, 자궁암, 난소암 등 여러 가지 암질환에 대한 예방 및/또는 치료제로서 사용할 수 있다. 또한, 종양 집적능이 높기 때문에, 종양의 진단약, 조영제로서 사용할 수 있다.
본 발명의 코폴리머를 약물 수송 담체로서 사용할 때, 그 투여량 및 투여 회수는, 투여 형태, 환자의 연령, 체중, 치료해야 할 증상의 성질 혹은 중증도 등을 감안하여 적절히 선택하면 되고, 투여량이나 투여 회수는 한정되어야 하는 것은 아니지만, 약물을 내포하는 폴리머를 주사제에 의해 정맥 내 주사할 경우, 성인 1인(60kg)당에 대하여 예를 들어 1회의 투여에 있어서 0.12mg 내지 12000000mg의 양을 투여하는 것이 바람직하고, 1.2mg 내지 1200000mg의 양을 투여하는 것이 보다 바람직하고, 12 내지 120000mg의 양을 투여하는 것이 특히 바람직하다.
본 발명의 의약 조성물은, 본 발명의 코폴리머와 약물을 혼합함으로써 제조할 수 있다. 바람직하게는, 본 발명의 코폴리머와 약물을 혼합하여, single chain nanoparticle를 제조하거나, 본 발명의 코폴리머 single chain nanoparticle를 제조하고 나서 약물을 혼합하면 된다. single chain nanoparticle는, 공지된 방법에 의해 제조할 수 있다.

약물로서는, 여러 가지 암 질환에 사용하는 것으로서는, 페메트렉세드나트륨, 마이토마이신 C, 블레오마이신 염산염, 아클라루비신 염산염, 암루비신 염산염, 에피루비신 염산염, 독소루비신 염산염, 피라루비신 염산염, 이리노테칸 염산염, 에토포사이드, 도세탁셀, 노기테칸 염산염, 파클리탁셀, 비노렐빈 타르타르산염, 빈블라스틴 황산염, 옥살리플라틴, 카필조밉, 카보플라틴, 시스플라틴, 템시로리무스, 트라벡테딘, 풀베스트란트, 보르테조밉, 미토크산트론 염산염, 미리플라틴, 엠탄신, SN-38, 모노메틸 오리스타틴 E, 모노메틸 오리스타틴 F, 스타우로스포린 등을 들 수 있다. 이러한 약물을 본 발명의 의약 조성물로 제조할 경우, 약물은 프리체로서 코폴리머에 담지되어 있어도 된다.
본 발명의 의약 조성물 투여 경로는, 치료 시에 가장 효과적인 것을 사용하는 것이 바람직하고, 경구 투여 제제, 주사제 또는 경피 투여 제제 등의 비경구 투여 제제로 투여 가능한데, 예를 들어 동맥 내 주사, 정맥 내 주사, 피하 주사, 근육 내 주사, 복강 내 주사 등의 비경구 투여가 바람직하고, 동맥 내 주사 및 정맥 내 주사가 보다 바람직하다. 투여 회수는 한정되어야 하는 것은 아니지만, 예를 들어 1주일 평균당, 1회 내지 수회 투여하는 것을 들 수 있다.
투여 경로에 적합한 각종 제제는, 제제상 통상 사용할 수 있는 부형제, 증량제, 결합제, 습윤제, 붕괴제, 윤활제, 계면 활성제, 분산제, 완충제, 보존제, 용해 보조제, 방부제, 교미 교취제, 무통화제, 안정화제, 등장화제 등의 제제 첨가물 등을 적절히 선택하여, 통상법에 의해 제조할 수 있다.
상기 각종 제제에 포함될 수 있는 제제 첨가물은, 의약적으로 허용되는 것이라면 특별히 한정되는 것은 아니다. 이러한 제제 첨가물의 예로서, 정제수, 주사용수, 주사용 증류수, 의약적으로 허용되는 유기 용제, 콜라겐, 폴리비닐알코올, 폴리비닐피롤리돈, 카르복시비닐폴리머, 알긴산 나트륨, 수용성 덱스트란, 카르복시메틸스타치나트륨, 펙틴, 크산탄검, 아라비아 고무, 카제인, 젤라틴, 한천, 글리세린, 프로필렌글리콜, 폴리에틸렌글리콜, 바셀린, 파라핀, 스테아릴알코올, 스테아르산, 인간 혈청 알부민, 만니톨, 소르비톨, 락토오스 등을 들 수 있다. 사용되는 첨가물은, 각종 제제에 따라 적절히 선택하고, 단독 또는 조합하여 사용할 수 있다.
또한, 주사제는, 비수성의 희석제(예를 들어 폴리에틸렌글리콜, 올리브유 등의 식물유, 에탄올 등의 알코올류 등), 현탁제 또는 유탁제로서 조제할 수도 있다. 주사제의 무균화는, 필터에 의한 여과 멸균, 살균제 등의 배합에 의해 행할 수 있다. 또한, 주사제는, 용시 조제의 형태로서 제조할 수 있다. 즉, 동결 건조법 등에 의해, 무균의 고체 조성물로 만들고, 사용 전에 주사용수, 주사용 증류수 또는 다른 용매에 용해하여 사용할 수 있다.
실시예
이하, 본 발명을 실시예에 의해 더욱 구체적으로 설명한다. 이 실시예는 예시를 위하여 제공된 것이며, 본 발명의 실시 형태를 한정하는 것은 아니다.
[실시예 1] poly[(benzyl acrylate)-co-(poly(ethylene glycol)methyl ether acrylate)-co-(1-ethoxyethyl acrylate)]의 제조
(1) 1-Ethoxyethyl acrylate(EEA)의 합성
아르곤 분위기 하에서, Ethyl vinyl ether(28.725mL)을 칭량하여 덜고, 빙냉 하에서 Phosphoric acid(50mg)를 첨가하였다. 그 후, Acrylic acid(17.15mL)를 첨가하고, 실온에서 48시간 교반하였다. Hydrotalcite(3g)를 첨가하여, 다시 2시간 교반하고, 반응을 정지하였다. 셀라이트 여과 후, 이배퍼레이션에 의해, 미반응된 Ethyl vinyl ether를 제거하였다. 중합 금지제로서 Phenothiazine를 500ppm이 되게 첨가하고, 수소화 칼슘과 함께 감압 증류함으로써 정제하였다(증류 온도 28-32℃). 얻어진 1-Ethoxyethyl acrylate는 유리 바이알에 분취하고, -30℃에서 보관하였다.
13C NMR (400MHz, CDCl3), δ, ppm:15.29(-OCH2CH3), 21.16(-COOCH(CH3)), 64.98(-OCH2-), 96.73(-COOCH(CH3)), 128.84(CH2CH-), 131.43(CH2CH-), 166.00(-COO).
(2) poly[(benzyl acrylate)-co-(poly(ethylene glycol)methyl ether acrylate)-co-(1-ethoxyethyl acrylate)]의 합성
100mg의 2-(Dodecylthiocarbonothioylthio)-2-methylpropionic acid(DDMAT)을 칭량하여 덜고, Toluene 17.3mL에 녹여 DDMAT/Toluene stock solution(DDMAT 농도로서 5.78mg/mL)으로 하였다. 마찬가지로, 22mg의 2,2'-Azobis(2-methylpropionitrile)(AIBN)를 칭량하여 덜고, Toluene 17.3mL에 녹여 AIBN/Toluene stock solution(AIBN 농도로서 1.27mg/mL)으로 만들었다. 별도로, poly(ethylene glycol)methyl ether acrylate(mPEGA, 에틸렌글리콜의 반복수의 평균값(n)은 9이다.) 1.296g, Benzyl acrylate(BnA) 0.394g, 1-Ethoxyethyl acrylate 0.039g, DDMAT/Toluene stock solution 1.73mL 및 AIBN/Toluene stock solution 1.73mL를 첨가하고, 70℃의 유욕에서 중합을 행하였다. 90분 경과 후, 중합을 정지한 후, 반응 용액을 재침전법 또는 메탄올에 대하여 투석함으로써 코폴리머를 회수하였다. 얻어진 코폴리머는 기본적으로 점조체이기 때문에, 재침전법에 대해서는, 빈용매(헥산/아세트산 에틸=7/3[v/v])를 첨가한 원심관에 반응 용액을 적하하고, 원심 분리(2,000×g, 5min)에 의해 회수하는 조작을 3회 반복하고, 최종적으로 진공 건조를 하여, poly[(benzyl acrylate)-co-(poly(ethylene glycol)methyl ether acrylate)-co-(1-ethoxyethyl acrylate)]를 1.223g 얻었다.
얻어진 코폴리머에 대하여, NMR을 사용하여 측정한 1H-NMR 스펙트럼으로부터 각 모노머의 중합도, 및 수평균 분자량(MN,NMR)을 해석한 결과, mPEGA(n=9)의 중합도는 102, BnA의 중합도는 94, EEA의 중합도는 9이며, MN,NMR은 65,900이었다. 또한, 얻어진 코폴리머에 대하여, GPC를 사용하여 분자량 분산도(Mw/Mn)를 측정한 결과 1.53이었다.

[실시예 2 내지 68]
실시예 1에서 사용한 모노머(mPEGA, BnA, EEA)의 종류, 투입량, 반응 온도, 중합 시간을 적절히 변경하고, 실시예 1과 동일한 방법을 사용함으로써, 하가 표에 나타내는, 조성 비율이나 평균 분자량이 다른 폴리머를 제조하였다.
[실시예 69]
poly[(benzyl acrylate)-co-(poly(ethylene glycol) methyl ether acrylate)-co-(acrylic acid)]의 제조
실시예 1에서 얻은 poly[(benzyl acrylate)-co-(poly(ethylene glycol) methyl ether acrylate)-co-(1-ethoxyethyl acrylate)]를 실온에서 0.5N HCl로 처리함으로써, ethoxyethyl기를 탈리하여 카르복실기를 갖는 3원 공중합체 1.176g를 얻었다. 얻어진 3원 공중합체의 수 중에 있어서의 Z평균 입자 직경, 및 다분산 지수를 동적 광산란법(Dynamic light scattering, DLS)에 의해 측정한 결과, 8.5nm(다분산 지수 0.14)이었다.

[실시예 70]
(1,2-diaminocyclohexane)platinum(II) 내포 SCNP의 제조 방법
(1,2-diaminocyclohexane)platinum(II)(이하, DACHPt라고 약기한다)의 Cl(H2O)체(DACHPt·Cl·H2O) 65.28mg을 정제수 20mL에 용해하고, 70℃에서 2시간 교반하였다. 이 용액 5mL에 대하여 실시예 69에서 얻은 3원 공중합체 287.4mg를 첨가하고, 50℃에서 밤새 교반하였다. 교반 종료 후, 반응 용액을, 정제수를 외액으로 하여 투석 정제하여, DACHPt 내포 SCNP을 5mL 얻었다. 정제 후에 얻어진 DACHPt 내포 SCNP의 Pt 함량은 유도 결합 플라즈마 발광 분석(Inductively coupled plasma-atomic emission spectrometry, ICP-AES)에 의해 측정하고, 720μg/mL(DACHPt로서 1.14mg/mL)였다. 별도로, DACHPt 내포 SCNP 200μL을 동결 건조하고, 고형분 농도를 산출한 후, Pt함량과의 비를 취해 폴리머당의 Pt 결합량을 산출한 결과, 3.4mol/mol이었다. 또한, 얻어진 DACHPt 내포 SCNP의 Z평균 입자 직경, 및 다분산 지수를 동적 광산란법(Dynamic light scattering, DLS)에 의해 측정한 결과, 8.7nm(다분산 지수 0.14)이었다. DACHPt 내포 전후의 SCNP의 입자 직경을 도 3에 나타낸다. SCNP의 입자 직경은, DACHPt 내포 전후에서 거의 변동되지 않았다. 결과를 아래 표에 정리하였다.

[비교예 1]
옥살리플라틴 용액의 조제
엘플래트 점적 정주액 50mg ((주) 야쿠루트 본사) 1mL을 5.9(w/v)% 글루코오스 용액 5.58mL에 첨가하고, 옥살리플라틴으로서 760μg 함유 5(w/v)% 글루코오스 용액을 조제하였다.
[시험예] 약효 시험
자성 누드 마우스(BALB/c-nu nu/nu, 7주령; 일본 찰스·리버(주))에 마우스 대장암 세포주 C26(American Type Culture Collection)을 피하 이식한 담암 모델을 약효 시험에 사용하였다.
CO2 인큐베이터 내에서 계대 배양한 마우스 대장암 세포주 C26을 액체 배지(Dulbecco's Modified Eagle's Medium-high glucose, Sigma- Aldrich)에 현탁하고, 한마리당 세포수로서 1×106/100μL이 되도록 누드 마우스의 등부 피하에 주사하였다. 그 후 약 1주일 누드 마우스를 사육한 후, 종양 체적의 평균값이 약 30mm3으로 생육되었을 때 약제의 투여를 시작하였다. DACHPt 내포 SCNP(실시예 70의 코폴리머를 사용하여 조제한 DACHPt 내포 SCNP)를 미정맥 내 투여하고(격일 3회), 종양 체적으로부터 항종양 효과를 평가하였다(1군 4 내지 5마리). 비교로서 옥살리플라틴 용액(비교예 1)을 사용하여 마찬가지로 투여하였다. 각 제제의 투여량은, 옥살리플라틴 용액에 대해서는 투여 가능한 최고 용량으로서 8mg/kg(Pt 환산으로 3.9mg/kg), DACHPt 내포 SCNP에 대하여는, Pt 환산으로 3mg/kg으로 하였다.
종양 체적의 경시 변화를 도 4에 나타냈다. DACHPt 내포 SCNP의 경우, 투여 14일 후에 T/C=0.4였다[T/C:약물 투여군 (T)와 Control군 (C)의 종양 체적의 비교]. 옥살리플라틴 용액(비교예 1)의 경우, 투여 14일 후에 T/C=1.1이었다. 또한, 투여 14일 후에 있어서 DACHPt 내포 SCNP은 Control과 비교하여 유의미하게 종양의 증대를 억제하는 것을 확인하였다(student's t-test). 이상의 결과는, DACHPt 내포 SCNP은 옥살리플라틴 용액과 비교하여 우수한 항종양 효과를 갖는다는 것을 나타내고 있다.

- 다음 식 (A), (B) 및 (C)로 표시되는 구조 단위를 갖는 코폴리머:
[식 중, R1, R2 및 R3은 동일하거나 상이하며 수소 원자 또는 C1-3 알킬기를 나타내고, R4는 C1-3 알킬기를 나타내고, R5는 수소 원자, C1-18 알킬기, 치환기를 가질 수 있는 3 내지 8원 시클로알킬기, 아다만틸기, 치환기를 가질 수 있는 C6-18 아릴기 또는 치환기를 가질 수 있는 5 내지 10원 헤테로아릴기를 나타내고, X1, X2 및 X3은 동일하거나 상이하며 산소 원자, 황 원자 또는 N-R7을 나타내고, R6은 수소 원자, 탈리기 또는 링커를 나타내고, R7은 수소 원자 또는 C1-3알킬기를 나타내고, m은 1 내지 100의 정수를 나타내고, n은 0 내지 3의 정수를 나타낸다]. - 다음 일반식 (1) 내지 (3):
