Abstract
본 발명은 열충격단백질의 에피토프를 포함하는 백신 및 이의 병용 요법에 관한 것으로, 본 발명의 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프인 폴리펩타이드를 포함하는 백신 조성물을 이용하면, 대다수의 환자에서 범용으로 사용가능하고, 산업화가 용이하며, 항종양효과가 현저하게 우수하여, 경제적으로 암을 예방 및 치료하는 데 사용할 수 있다.
Description
본 발명은 열충격단백질의 에피토프를 포함하는 백신 및 이의 병용 요법에 관한 것으로, 더욱 자세하게는 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프인 폴리펩타이드, 이를 포함하는 백신 조성물 및 상기 조성물을 이용하여 암을 치료 또는 예방하는 방법에 관한 것이다.
현대에 들어서 많은 질병들이 치료가 쉽게 가능하게 되었고, 치료가 불가능한 병은 거의 없어졌지만, 암은 다른 질병치료와는 달리 매우 힘들고 복잡한 치료가 요구되고 있으며, 그 복잡한 치료들 또한 완벽하게 효과적이지 못하다. 최근 면역치료 방법이 대두되고 있다. 면역치료는 환자 체내의 면역반응을 이용하여 암을 치료하는 방법이다. 이 면역치료 방법을 통하여 암의 예방까지도 꾀할 수 있다. 암 면역치료는 백신의 원리와 같이 암의 원인이 되는 항원을 투여하여 암에 특이적인 면역세포들을 활성화 시킨 후, 활성화된 면역세포들이 체내에서 암을 특이적으로 공격하게 하여 치료하는 방법이다. 또한, 암에 걸리지 않은 상태에서 암에 특이적인 항원을 몸 속에 투여하면, 활성화 되지 않았던 면역세포들이 암 특이적 기억면역세포로 활성화 되면서, 암에 걸렸을 시 암 세포를 특이적으로 공격하게 된다.
열 충격 단백질 90(heat shock protein 90: Hsp90)은 많은 "클라이언트"(client) 단백질의 단백질 접힘, 성숙 및 입체 형태 안정화를 위해 요구되는 ATPase-의존형 분자 샤페론이다(Young et al., 2000; Kamal et al., 2003). Hsp90은 성장 인자 수용체, 세포 주기 조절제 및 Akt와 유사한 신호전달 키나제, 안드로겐 수용체(AR) 또는 Raf-1을 비롯하여, CRPC에 연루된 수개의 단백질과 상호작용한다(Whitesell et al., 2005; Takayama et al., 2003). 종양 세포는 양성 세포에 비해서 더 높은 Hsp90 수준을 발현하고(Kamal et al., 2003; Chiosis et al., 2003), Hsp90 저해는 CRPC 및 기타 암에서 여기 상태의 표적(exciting target)으로서 생긴다. 많은 Hsp90 저해제는 겔다나마이신 및 그의 유사체, 또는 합성 화합물 등과 같은 천연 화합물을 비롯하여, 그의 ATP-결합 포켓을 표적화하면서 발달하였다. 이들 제제는 Hsp90 기능을 저해하여, 결장, 유방, PCa 및 기타 암의 임상 전 연구에서 아폽토시스를 유발하는 것으로 밝혀졌다(Kamal et al., 2003; Solit et al., 2003; Solit et al., 2002).
최근, 자가종양세포 유래 gp96 HSP peptide complex를 포함하는 HSPPC-96 (Oncophage®, Antigenics, Inc., New York, NY, USA)이 4기 전이성 신장암 환자에서 IL-2와 병합치료 시 우수한 치료 효과를 나타내며, 4기 흑색종 환자에서 기존 치료법과 비교한 3상 임상시험에서 생존기간 연장효과는 없으나, 안전성, 효과적인 면역반응 유도 및 장기간 면역기억 현상 등이 보고되고 있다. 그러나, 자가 종양세포 유래이므로, 종양세포 획득이 어려운 환자에서 적용 불가능하며, 백신 제조 원가가 비싸고 제조공정 및 효과에 대한 표준화가 어렵기 때문에 상업화가 어렵고, 종양 특이 단백질의 epitope이 알려져 있지 않아 면역반응에 대한 모니터링이 불가능하다는 어려운 문제점이 있다.
이에, 본 발명자들은 대다수의 환자에서 범용으로 사용할 수 있고, 항종양 효과가 우수한 백신 조성물을 찾고자 예의 노력한 결과, 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프인 폴리펩타이드를 포함하는 백신 조성물을 이용하면, 대다수의 환자에서 범용으로 사용가능하고, 산업화가 용이하며, 항종양효과가 우수하다는 것을 확인하고, 본 발명을 완성하게 되었다.
본 발명의 목적은 열충격단백질 90의 에피토프인 폴리펩타이드를 제공하는데 있다.
본 발명의 다른 목적은 상기 폴리펩타이드를 유효성분으로 포함하는 암의 치료 또는 예방을 위한 백신 조성물을 제공하는데 있다.
본 발명의 또 다른 목적은 상기 폴리펩타이드를 유효성분으로 포함하는 항암 조성물을 제공하는데 있다.
본 발명의 또 다른 목적은 상기 백신 조성물을 암 환자에 투여하는 것을 포함하는 암을 치료 또는 예방하는 방법을 제공하는데 있다.
본 발명의 또 다른 목적은 상기 폴리펩타이드를 특이적으로 인식하는 항체를 제공하는데 있다.
상기 목적을 달성하기 위하여, 본 발명은 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프인 폴리펩타이드를 제공한다.
본 발명은 또한, 상기 폴리펩타이드를 코딩하는 유전자, 상기 유전자를 포함하는 재조합 벡터 및 상기 유전자 또는 상기 재조합 벡터가 도입된 재조합 미생물을 제공한다.
본 발명은 또한, (a) 상기 재조합 미생물을 배양하여 상기 폴리펩타이드를 생성시키는 단계; 및 (b) 상기 생성된 폴리펩타이드를 수득하는 단계를 포함하는 상기 폴리펩타이드의 제조방법을 제공한다.
본 발명은 또한, 상기 폴리펩타이드를 유효성분으로 포함하는 암의 치료 또는 예방을 위한 백신 조성물 및 상기 백신 조성물을 암 환자에 투여하는 것을 포함하는 암을 치료 또는 예방하는 방법을 제공한다.
본 발명은 또한, 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프를 특이적으로 인식하는 항체를 제공한다.
본 발명의 열충격단백질 90의 에피토프를 포함하는 백신 조성물은 대다수의 환자에서 범용으로 사용가능하고, 산업화가 용이하며, 항종양효과가 현저하게 우수하므로, 경제적으로 암을 예방 및 치료하는 데 사용할 수 있다.

본 발명에서는 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90(heat shock protein 90: Hsp90)의 에피토프 폴리펩타이드를 포함하는 백신 조성물이 Th1 면역반응을 유도하며, 항종양 효과가 우수하다는 것을 확인하였다.
따라서, 본 발명은 일 관점에서, 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프인 폴리펩타이드에 관한 것이다.
본 발명에서 용어, "에피토프"는 특정 항체에 의해 인식되는 항원결합부위의 아미노산 잔기 세트, 또는 T 세포에서는 T 세포 수용체 단백질 및/또는 주요 조직적합성 복합체(Major Histocompatibility Complex, MHC)수용체에 의해 인식되는 잔기이다. 에피토프는 항체, T 세포 수용체 또는 HLA 분자에 의해 인식되는 부위를 형성하는 분자로, 일차, 이차 및 삼차 펩티드 구조, 또는 전하를 의미한다.
발명의 일 실시예에서는, Hsp 90 풀 시퀀스를 5개의 알고리즘 프로그램을 이용하여 가장 흔한 MHC class II 대립유전자(allele)에 결합 친화도(binding affinity)가 좋을 것으로 예상되는 15-mer 펩타이드 시퀀스 12개를 선정한 후, MHC class II 대립형질에 결합친화성이 좋고, 항종양 효과가 우수한 2종류의 에피토프를 획득하였다.
본 발명은 다른 관점에서, 상기 폴리펩타이드를 코딩하는 유전자에 관한 것이다.
본 발명에 있어서, 상기 유전자는 서열번호 3 또는 서열번호 4의 염기서열로 표시되는 것을 특징으로 할 수 있다.
본 발명은 또 다른 관점에서, 상기 유전자를 포함하는 재조합 벡터 및 상기 유전자 또는 상기 재조합 벡터가 도입된 재조합 미생물에 관한 것이다.
본 발명은 또 다른 관점에서, (a) 상기 재조합 미생물을 배양하여 상기 폴리펩타이드를 생성시키는 단계; 및 (b) 상기 생성된 폴리펩타이드를 수득하는 단계를 포함하는 상기 폴리펩타이드의 제조방법에 관한 것이다.
본 발명에서, 벡터는 적당한 숙주세포에서 목적 단백질을 발현할 수 있도록 적합한 조절 서열에 작동 가능하게 연결된 상기 목적 단백질을 암호화하는 폴리뉴클레오타이드의 염기서열을 함유하는 DNA 제조물을 의미한다. 상기 조절 서열은 전사를 개시할 수 있는 프로모터, 그러한 전사를 조절하기 위한 임의의 오퍼레이터 서열, 적합한 mRNA 리보좀 결합 부위를 코딩하는 서열 및 전사와 해독의 종결을 조절하는 서열을 포함할 수 있으며 목적에 따라 다양하게 제조될 수 있다. 벡터의 프로모터는 구성적 또는 유도성 일 수 있다. 벡터는 적당한 숙주 내로 형질전환 된 후, 숙주 게놈과 무관하게 복제되거나 기능할 수 있으며, 게놈 그 자체에 통합될 수 있다.
본 발명에서 사용되는 벡터는 숙주세포 중에서 복제 가능한 것이면 특별히 한정되지 않으며 당 업계에 알려진 임의의 벡터를 이용할 수 있다. 통상 사용되는 벡터의 예로는 천연 상태이거나 재조합 된 상태의 플라스미드, 파지미드, 코스미드, 바이러스 및 박테리오파지를 들 수 있다. 예를 들어, 파지 벡터 또는 코스미드 벡터로서 pWE15, M13, λMBL3, λMBL4, λIXII, λASHII, λAPII, λt10, λt11, Charon4A, 및 Charon21A을 사용할 수 있으며, 플라스미드 벡터로서 pBR계, pUC계, pBluescriptII계, pGEM계, pTZ계, pCL계 및 pET계를 사용할 수 있다. 본 발명에서 사용 가능한 벡터는 특별히 제한되는 것이 아니며 공지된 발현 벡터를 사용할 수 있다.
“발현 조절 서열 (expression control sequence)”이라는 표현은 특정한 숙주 생물에서 작동가능하게 연결된 코딩 서열의 발현에 필수적인 DNA 서열을 의미한다. 그러한 조절 서열은 전사를 실시하기 위한 프로모터, 그러한 전사를 조절하기 위한 임의의 오퍼레이터 서열, 적합한 mRNA 리보좀 결합 부위를 코딩하는 서열 및 전사 및 해독의 종결을 조절하는 서열을 포함한다. 예를 들면, 원핵생물에 적합한 조절 서열은 프로모터, 임의로 오퍼레이터 서열 및 리보좀 결합 부위를 포함한다. 진핵세포는 프로모터, 폴리아데닐화 시그날 및 인핸서가 이에 포함된다. 플라스미드에서 유전자의 발현 양에 가장 영향을 미치는 인자는 프로모터이다. 고 발현용의 프로모터로서 SRα 프로모터와 사이토메가로바이러스(cytomegalovirus) 유래 프로모터 등이 바람직하게 사용된다.
본 발명의 DNA 서열을 발현시키기 위하여, 매우 다양한 발현 조절 서열중 어느 것이라도 벡터에 사용될 수 있다. 유용한 발현 조절서열의 예에는, 예를 들어, SV40 또는 아데노바이러스의 초기 및 후기 프로모터들, lac 시스템, trp 시스템, TAC 또는 TRC 시스템, T3 및 T7 프로모터들, 파지 람다의 주요 오퍼레이터 및 프로모터 영역, fd 코드 단백질의 조절 영역, 3-포스포글리세레이트 키나제 또는 다른 글리콜분해 효소에 대한 프로모터, 상기 포스파타제의 프로모터들, 예를 들어 Pho5, 효모 알파-교배 시스템의 프로모터 및 원핵세포 또는 진핵 세포 또는 이들의 바이러스의 유전자의 발현을 조절하는 것으로 알려진 구성과 유도의 기타 다른 서열 및 이들의 여러 조합이 포함된다. T7 RNA 폴리메라아제 프로모터 Φ10은 대장균에서 단백질 NSP를 발현시키는데 유용하게 사용될 수 있다.

본원 명세서에 사용된 용어 “발현 벡터”는 통상 이종의 DNA의 단편이 삽입된 재조합 캐리어(recombinant carrier)로서 일반적으로 이중 가닥의 DNA의 단편을 의미한다. 여기서, 이종 DNA는 숙주 세포에서 천연적으로 발견되지 않는 DNA인 이형 DNA를 의미한다. 발현 벡터는 일단 숙주 세포내에 있으면 숙주 염색체 DNA와 무관하게 복제할 수 있으며 벡터의 수 개의 카피 및 그의 삽입된 (이종) DNA가 생성될 수 있다.
당업계에 주지된 바와 같이, 숙주세포에서 형질감염 유전자의 발현 수준을 높이기 위해서는, 해당 유전자가, 선택된 발현 숙주 내에서 기능을 발휘하는 전사 및 해독 발현 조절 서열에 작동가능하도록 연결되어야만 한다. 바람직하게는 발현 조절서열 및 해당 유전자는 세균 선택 마커 및 복제 개시점(replication origin)을 같이 포함하고 있는 하나의 발현 벡터 내에 포함되게 된다. 발현 숙주가 진핵세포인 경우에는, 발현 벡터는 진핵 발현 숙주 내에서 유용한 발현 마커를 더 포함하여야만 한다.
본 발명의 폴리펩타이드를 코딩하는 유전자를 발현시키기 위해 매우 다양한 발현 숙주/벡터 조합이 이용될 수 있다. 진핵 숙주에 적합한 발현 벡터에는, 예를 들어 SV40, 소 유두종바이러스, 아네노바이러스, 아데노-연관 바이러스(adeno-associated virus), 시토메갈로바이러스 및 레트로바이러스로부터 유래된 발현 조절 서열을 포함한다. 세균 숙주에 사용할 수 있는 발현 벡터에는 pBluescript, pGEX2T, pUC벡터, colE1, pCR1, pBR322, pMB9 및 이들의 유도체와 같이 E. coli에서 얻는 것을 예시할 수 있는 세균성 플라스미드, RP4와 같이 보다 넓은 숙주 범위를 갖는 플라스미드, λgt10과 λgt11, NM989와 같은 매우 다양한 파지 람다(phage lambda) 유도체로 예시될 수 있는 파지 DNA, 및 M13과 필라멘트성 단일가닥의 DNA 파지와 같은 기타 다른 DNA 파지가 포함된다. 효모 세포에 유용한 발현 벡터는 2μ 플라스미드 및 그의 유도체이다. 곤충 세포에 유용한 벡터는 pVL 941이다.
상술한 발현 벡터에 의해 형질전환 또는 형질감염된 숙주 세포는 본 발명의 또 다른 측면을 구성한다. 본원 명세서에 사용된 용어 “형질전환”은 DNA를 숙주로 도입하여 DNA가 염색체외 인자로서 또는 염색체 통합완성에 의해 복제가능하게 되는 것을 의미한다. 본원 명세서에 사용된 용어 “형질감염”은 임의의 코딩 서열이 실제로 발현되든 아니든 발현 벡터가 숙주 세포에 의해 수용되는 것을 의미한다.
본 발명의 숙주 세포는 하나 이상의 목적 단백질을 암호화하는 폴리뉴클레오타이드를 갖는 벡터가 도입된 재조합 미생물이거나, 하나 이상의 목적 단백질을 암호화하는 폴리뉴클레오타이드가 미생물에 도입되어 폴리뉴클레오타이드가 염색체에 통합되어 목적 단백질을 발현시키도록 형질이 감염된 재조합 미생물을 의미한다. 원핵 또는 진핵생물 세포일 수 있다. 또한, DNA의 도입효율이 높고, 도입된 DNA의 발현효율이 높은 숙주가 통상 사용된다. 대장균, 슈도모나스, 바실러스, 스트렙토마이세스, 진균, 효모와 같은 주지의 진핵 및 원핵 숙주들, 스포도프테라 프루기페르다(SF9)와 같은 곤충 세포, CHO 및 생쥐 세포같은 동물 세포, COS 1, COS 7, BSC 1, BSC 40 및 BMT 10과 같은 아프리카 그린 원숭이 세포, 및 조직배양된 인간 세포는 사용될 수 있는 숙주 세포의 예이다. COS 세포를 이용하는 경우에는 COS 세포에서 SV40 라지 T안티겐(large T antigen)이 발현하고 있으므로 SV40의 복제개시점을 갖는 플라스미드는 세포중에서 다수 카피(copy)의 에피솜(episome)으로 존재하도록 되고 통상보다고 발현이 기대될 수 있다. 도입된 DNA 서열은 숙주 세포와 동일한 종으로부터 얻을 수 있거나, 숙주 세포와 다른 종의 것일 수 있거나, 또는 그것은 어떠한 이종 또는 상동성 DNA를 포함하는 하이브리드 DNA 서열일 수 있다.
물론 모든 벡터와 발현 조절 서열이 본 발명의 DNA 서열을 발현하는데 모두 동등하게 기능을 발휘하지는 않는다는 것을 이해하여야만 한다. 마찬가지로 모든 숙주가 동일한 발현 시스템에 대해 동일하게 기능을 발휘하지는 않는다. 그러나, 당업자라면 과도한 실험적 부담없이 본 발명의 범위를 벗어나지 않는 채로 여러 벡터, 발현 조절 서열 및 숙주 중에서 적절한 선택을 할 수 있다. 예를 들어, 벡터를 선택함에 있어서는 숙주를 고려하여야 하는데, 이는 벡터가 그 안에서 복제되어야만 하기 때문이다. 벡터의 복제 수, 복제 수를 조절할 수 있는 능력 및 당해 벡터에 의해 코딩되는 다른 단백질, 예를 들어 항생제 마커의 발현도 또한 고려되어야만 한다. 발현 조절 서열을 선정함에 있어서도, 여러 가지 인자들을 고려하여야만 한다. 예를 들어, 서열의 상대적 강도, 조절가능성 및 본 발명의 DNA 서열과의 상용성 등, 특히 가능성있는 이차 구조와 관련하여 고려하여야 한다. 단세포 숙주는 선정된 벡터, 본 발명의 DNA 서열에 의해 코딩되는 산물의 독성, 분비 특성, 단백질을 정확하게 폴딩시킬 수 있는 능력, 배양 및 발효 요건들, 본 발명 DNA 서열에 의해 코딩되는 산물을 숙주로부터 정제하는 것의 용이성 등의 인자를 고려하여 선정되어야만 한다. 이들 변수의 범위내에서, 당업자는 본 발명의 DNA 서열을 발효 또는 대규모 동물 배양에서 발현시킬 수 있는 각종 벡터/발현 조절 서열/숙주 조합을 선정할 수 있다. 발현 클로닝에 의해 NSP 단백질의 cDNA를 클로닝 하려고 할 때의 스크리닝법으로서 바인딩법(binding법), 페닝법(panning법), 필름에멀션법(film emulsion 법)등이 적용될 수 있다.

레트로바이러스의 친화성은 외부 외피 단백질과 일체화함으로써 변할 수 있어 표적 세포의 종류를 확대시킬 수 있다. 렌티바이러스 벡터는 비분열 세포를 형질도입 또는 감염시켜서 고바이러스 역가를 생성하는 레트로바이러스 벡터이다. 표적 조직에 따라 레트로바이러스 유전자 잔달 시스템이 결정된다. 레트로바이러스 벡터는 6-10kb 외부 서열을 포장할 수 있는 시스 액팅 긴 말단 반복을 포함한다. 벡터의 복제와 포장을 위해 충분한 최소의 시스 액팅 LTR은 영구적인 트랜스젠 발현을 위해 치료 유전자가 표적 세포로 통합되도록 사용할 수 있다. 널리 사용되는 레트로바이러스 벡터는 쥐 백혈병 바이러스(MuLV), 긴팔원숭이 백혈병 바이러스(GaLV), 원숭이 면역결핍 바이러스(SIV), 인간 면역결핍 바이러스(HIV), 및 그들의 조합 바이러스를 포함한다(Buchscher et al., J. 수크로오스 포스포릴라아제 단백질을 일시적으로 발현하는 경우에는 아데노바이러스 기반 시스템을 더욱 많이 이용하며, 아데노바이러스계 벡터는 많은 세포들에서 고효율로 형질도입을 일으키지만 세포 분열을 필요로 하지 않는다. 상기 벡터를 이용하면 높은 역가와 높은 수준의 발현을 얻을 수 있고, 간단한 시스템에서 대량으로 생산될 수 있다. 또한 아데노 부속 바이러스(AAV) 벡터는 표적 핵산을 가진 세포로 형질도입하는데 이용되는데, 예를 들면 생체 외 상에서 핵산과 펩티드의 생산 및 생체 내 및 생체 외 상에서 유전자 치료에 이용되며(West et al., Virology., 160:38-47, 1987; 미국특허 제4,797,368호; WO93/24641; Kotin, HumanGene Therapy., 5:793-801, 1994; Muzyczka,J. Med., 1:1017-102, 1995; Malech et al., PNAS., 94:(22)12133-12138, 1997), PA317/pLASN는 유전자 치료에 사용된 최초의 치료 벡터로(Blaese et al., Science., 270:475-480, 1995) MFG-S 포장 벡터의 형질도입 효율은 50% 또는 그 이상을 보였다(Ellem et al., Immunol Immunother., 44(1):10-20, 1997; Dranoff et al., Hum.Gene Ther., 1:111-2, 1997).
재조합 아데노 연관 바이러스 벡터(rAAV)는 결함 있고 비병원성인 제2형 파르보바이러스 아데노 연관 바이러스를 기반으로 하는 유망한 대체 유전자 전달 시스템이다. 모든 벡터는 트랜스젠 발현 카세트 측면에 위치하는 AAV 145 bp 역위 말단 반복을 보유한 플라스미드로부터 유래한다. 형질도입된 세포의 게놈으로 통합에 기인하는 효율적인 유전자 전달 및 안정한 트랜스젠 전달은 상기 벡터 시스템의 큰 장점이다(Wagner et al., Lancet., 351:9117-17023, 1998; Kearns et al., Gene Ther., 9:748-55, 1996).
본 발명은 또 다른 관점에서, 상기 폴리펩타이드를 유효성분으로 포함하는 암의 치료 또는 예방을 위한 백신 조성물에 관한 것이다.
본 발명에서, 상기 백신 조성물은 Th1 면역반응을 유도한다.
본 발명에서, 용어 “Th1 세포”는 유전자 발현, 단백질 분비 및 기능적 활성 측면에서 특정되는 헬퍼 T 세포 림포사이트의 서브세트를 의미한다. 예를 들어, Th1 세포는 IL-2 및 IFN-γ를 합성하지만 IL-4, IL-5, IL-10 및 IL-13는 합성하지 않는 사이토카인 발현 패턴을 나타낸다. Th1 세포는 다양한 세포내 병원균에 대한 세포-매개 면역반응, 기관-특이적 자가면역 질환 및 지연성 과민반응에 관여한다.
본 발명에서 “CTLA-4(cytotoxic T-lymphocyte-associated protein 4)”는 면역조절 억제제로서 CD152(Cluster of Differentiation 152)로도 알려져 있고, 면역 관문으로 작용하는 단백질 수용체로서 면역 반응을 감소시킨다.
본 발명에서 “STING(STimulator of InterferoN Gene, 인터페론 유전자의 촉진제)”은 STING에 관련된 신호전달 경로를 활성화하여 인터페론을 포함하는 염증성 사이토카인의 생산을 유도하여 항암 효과를 나타내는 물질을 의미한다.

본 발명의 백신 조성물은 초기 암에 적용가능하다.
본 발명에서 용어, “항암 보조제”는 항암제의 항암효과를 증대시키고, 항암제의 부작용을 억제하거나 개선시키기 위하여 사용될 수 있으며, 항암제와 병용하여 환자에게 투여될 수 있다.
본 발명에서 용어, “예방”은 본 발명의 Hsp 90 에피토프 단백질을 발현하는 형질전환체 또는 상기 형질전환체를 유효성분으로 하는 조성물의 투여로 상기 암을 억제 또는 지연시키는 모든 행위를 말한다.
본 발명에서 용어, “치료”는 본 발명의 Hsp 90 에피토프 단백질을 발현하는 형질전환체 또는 상기 형질전환체를 유효성분으로 하는 조성물의 투여로 상기 암 또는 종양이 불사화되지 않고, 분열을 멈추거나 이롭게 되는 모든 행위를 말한다.
본 발명에 있어서, 상기 조성물은 서열번호 1 및/또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프를 모두 포함하는 것을 특징으로 할 수 있다.
본 발명에서, 용어 “다중 펩타이드 백신”은 상술한 폴리펩타이드 에피토프가 2개 이상 포함되어 있는 백신을 나타내기 위하여 정의된 용어이다.
본 발명에 있어서, 상기 조성물은 면역 항암제를 추가로 포함하는 것을 특징으로 할 수 있다.
본 발명에 있어서, 상기 면역 항암제는 CTLA-4(cytotoxic T-lymphocyte-associated protein 4), PD-1(Programmed cell death protein 1), LAG-3(Lymphocyte Activation Gene-3), TIM-3(T-cell Immunoglobulin and Mucin-domain containing-3), TIGIT(T-cell Immunoreptor with IG and ITIM domain) 및 VISTA(V-domain Ig Suppressor of T cell Activation)로 구성된 군으로부터 선택된 어느 하나에 대한 면역관문 억제제이고, 보다 바람직하게는 CTLA-4 억제제이지만 이에 제한되지는 않는다.

본 발명에서, 용어 “면역 항암제”는 암 자체를 공격하는 기존 항암제와는 달리 인공면역 단백질을 체내에 주입하여 면역체계를 자극함으로써 면역세포가 선택적으로 암세포만을 공격하도록 유도하는 치료약제로서, 암세포가 획득한 면역억제 또는 면역회피 기전을 극복하기 위하여 면역체계의 종양 인지능력 또는 파괴능력을 회복 또는 강화시키는 기전의 항암제이다. 상기 면역항암제는 면역관문 억제제, 면역세포 치료제 및 면역바이러스 치료제를 포함하지만 이에 제한되지는 않는다.
본 발명에서, 용어 “면역관문 억제제”는 일부 암세포가 체내 면역세포인 T 세포의 면역체크포인트를 활용하면서 면역을 회피하는 경우에, T 세포 억제에 관여하는 면역체크포인트 단백질(Immune Checkpoint Protein)의 활성화를 차단하여 T 세포를 활성화시켜 암세포를 공격하는 작용을 하는 면역항암제의 일종으로서, CTLA-4 억제제, PD-1 억제제, PD-1 억제제, LAG-3 억제제, TIM-3 억제제, TIGIT 억제제 및 VISTA 억제제를 포함하지만 이에 제한되지는 않는다.
에피토프 펩타이드를 백신 조성물로 이용하는 경우, 상기 유효성분은 단독으로 이용되기 보다는 약학으로 허용된 담체에 혼입시킨 형태로 이용하는 것이 적합하다. 여기서, 약학으로 허용된 담체는 제약 분야에서 통상 사용되는 담체, 부형제 및 희석제를 포함한다.
본 발명의 백신 조성물에 이용할 수 있는 약제학적으로 허용된 담체는, 이에 제한되는 것은 아니지만, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로스, 메틸 셀룰로스, 폴리비닐피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다.
본 발명의 백신 조성물은 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀전, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 또는 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 제제화할 경우에는 통상 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 그러한 고형 제제는 유효성분에 적어도 하나 이상의 부형제, 예를 들면 전분, 칼슘 카르보네이트, 수크로스, 락토오스, 젤라틴 등을 섞어 조제될 수 있다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용될 수 있다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데, 일반적으로 사용되는 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수용성용제, 현탁제, 유제, 동결건조 제제 및 좌제가 포함된다. 비수용성용제, 현탁제로는 프로필렌 글리콜, 폴리에틸렌 글리콜, 올리브유와 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 트윈(tween) 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다. 특히, 액상 제제의 경우에는 박테리아 포획 필터 등을 통한 여과에 의해 살균제 등을 혼입시켜 살균하는 것이 바람직하다. 이렇게 살균된 조성물은 예를 들면 동결건조에 의해 고형화시킬 수 있으며, 사용시에 이를 무균수 또는 무균 희석액에 용해시킨다.
본 발명에 따른 백신 조성물은 소, 쥐, 마우스, 가축, 개, 인간 등의 포유동물에 다양한 경로로 투여될 수 있다. 투여의 모든 방식이 예상될 수 있는데, 예를 들면 경구, 정맥, 근육, 피하, 복강내 주사에 의해 투여될 수 있다.
본 발명에 따른 백신 조성물의 투여량은 동물의 연령, 체중, 성별, 신체 상태 등을 고려하여 선택된다. 별다른 부작용 없이 동물에 면역보호반응을 유도하기에 필요한 양은 면역원으로서 사용된 에피토프 및 부형제의 임의 존재에 따라 다양할 수 있다. 일반적으로 각각의 용량은 본 발명의 폴리페타이드의 면역원량의 멸균 용액 ㎖ 당 0.1 내지 1000㎍의 단백질, 바람직하게는 0.1 내지 100㎍을 함유한다. 백신 조성물의 경우에는 필요에 따라 초기 용량에 이어 임의로 반복된 항원 자극을 수행할 수 있다.
본 발명은 또 다른 관점에서, 상기 백신 조성물을 암 환자에 투여하는 것을 포함하는 암을 치료 또는 예방하는 방법에 관한 것이다.
본 발명에 있어서, 상기 백신 조성물은 면역항암제와 병용하여 투여하는 것을 특징으로 할 수 있으며, 항체치료제와 병용하여 투여하는 것을 특징으로 할 수 있다.
발명의 백신 조성물과 병용하여 사용되는 면역항암제 또는 항체치료제는 상호 동시에 또는 시간 차를 두고 순차적으로 투여될 수 있으며, 적절한 시간 및 주기에 따라 선택될 수 있다.
본 발명에 있어서, 상기 백신 조성물은 면역항암제와 병용하여 투여할 수 있다. 상기 면역 항암제는 CTLA-4(cytotoxic T-lymphocyte-associated protein 4), PD-1(Programmed cell death protein 1), LAG-3(Lymphocyte Activation Gene-3), TIM-3(T-cell Immunoglobulin and Mucin-domain containing-3), TIGIT(T-cell Immunoreptor with IG and ITIM domain) 및 VISTA(V-domain Ig Suppressor of T cell Activation)로 구성된 군으로부터 선택된 어느 하나에 대한 면역관문 억제제이고, 보다 바람직하게는 CTLA-4 억제제이지만 이에 제한되지는 않는다.
본 발명에 있어서, 상기 백신 조성물은 항암 보조제를 추가로 병용 투여할 수 있고, 상기 항암 보조제는 STING(STimulator of InterferoN Gene) 아고니스트일 수 있으나 이에 제한되지는 않는다.
본 발명에 있어서, 상기 암은 예컨대 편평세포 암(squamous cell cancer, 예를 들어, 상피의 편평세포 암), 소형 세포 폐암, 비-소형 세포 폐암, 폐암, 복막암, 결장암, 담도 종양, 비인두암, 후두암, 기관지암, 구강암, 골육종, 담낭암, 신장암, 백혈병, 방광암, 흑색종, 뇌암, 신경 교종, 뇌종양, 피부암, 췌장암, 유방암, 간암, 골수암, 식도암, 대장암, 위암, 자궁경부암, 전립선암, 난소암, 두경부암 및 직장암으로 이루어진 군으로부터 선택된 1종 이상인 것을 특징으로 할 수 있으나, 이에 한정되는 것은 아니다.
또한 본 발명은 서열번호 1 및/또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프를 포함하는 항암 조성물을 제공한다.
상기 조성물은 상기 에피토프 이외에 유효성분의 안정화를 위한 다른 성분이 더 포함될 수 있다.
본 발명에서 용어, "주사" 또는 "투여"는 투여대상의 나이, 성별, 체중 등에 따라 투여량이 달라질 수 있고, 투여경로, 질병의 정도, 성별, 체중, 나이 등에 따라서도 백신의 투여량이 달라질 수 있다.
본 발명에서, 콘카나발린 A(concanavalin A; 양성대조군, Positive control)는 T세포를 활성화시키지만 B세포는 활성화시키지 않는다(불용화하면 B세포도 활성화된다). 또 여러 암세포에서 정상세포에 비하여 Con A에 대한 높은 응집성을 나타내기 때문에 암세포막 구조의 특이성을 연구하는 수단으로 이용되고 있다.
본 발명은 또 다른 관점에서, 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프를 특이적으로 인식하는 항체에 관한 것이다.
본 발명에 있어서, 상기 항체는 단클론 또는 다클론 항체인 것을 특징으로 할 수 있다.
본 발명에서, 용어 “항체”는 면역계 내에서 항원의 자극에 의하여 만들어지는 물질로서, 면역글로불린이라고도 하며, 특정한 항원과 특이적으로 결합하여 림프와 혈액을 떠돌며 항원-항체반응을 일으킨다. 항체가 특이적 항원에 대한 특이성을 나타내는 반면에, 면역글로불린은 항체, 및 항원 특이성이 결여된 항체 유사 물질 모두를 포함한다. 후자의 폴리펩티드는 예컨대 림프계에서는 낮은 수준으로 생산되며 골수종에 의해서 증가된 수준으로 생산된다. 본 발명에서는 Hsp 90 에피토프 단백질의 일부를 포함하는 유전자 서열의 항원에 대한 항체일 수 있으며, 바람직하게는 서열번호 1의 아미노산 서열, 서열번호 2의 아미노산 서열 또는 서열번호 1 및 서열번호 2의 아미노산 서열이 융합된 형태의 항원에 대한 항체일 수 있다.
본 발명에서 유효성분과 결합하여 사용된 "치료학적으로 유효한 양"이란 용어는 대상 질환을 예방 또는 치료하는데 유효한 양을 의미하며, 본 발명의 조성물의 치료적으로 유효한 양은 여러 요소, 예를 들면 투여방법, 목적부위, 환자의 상태 등에 따라 달라질 수 있다. 따라서, 인체에 사용 시 투여량은 안전성 및 효율성을 함께 고려하여 적정량으로 결정되어야 한다. 동물실험을 통해 결정한 유효량으로부터 인간에 사용되는 양을 추정하는 것도 가능하다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예 1: in silico 알고리즘을 이용한 Hsp 90 MHC class II 항원결정기 발굴
열충격단백질 90(Heat shock protein 90; Hsp 90)에서 사람에서 가장 흔한 MHC class II 대립형질(allele)에 결합친화성이 좋을 것으로 예상되는 15-mer 펩타이드 시퀀스를 선정하기 위하여, SYFPEITHI(Institute for Cell Biology, Heidelberg, Germany), Propred(Institute of Microbial Technology, Chandigarh, India), MHC-Thread(University of Aberdeen, AFPberdeen, United Kingdom), Average Binding matrix method, Rankpep(Harvard, Boston, MA, USA)의 컴퓨터 알고리즘을 이용하였다(도 1).
사람에서 가장 흔한 MHC class II 대립형질은 DRB1*0101, DRB1*1501, DRB1*0301, DRB1*0401, DRB1*0404, DRB1*0405, DRB1*0701, DRB1*0802, DRB1*0901, DRB1*1101, DRB1*1201, DRB1*1302, DRB3*0101, DRB4*0101, DRB5*0101이다.
그 결과, Hsp 90 15-mer 펩타이드 시퀀스를 12개를 획득하였다(표 1). 상기 획득한 15개의 펩타이드 시퀀스는 펩타이드 합성회사에 의뢰하여 펩타이드로 합성하였다.
이 후, 서열번호 1 및 2의 아미노산 서열로 표시되는 2가지(p485, p527)의 다중 펩타이드 시퀀스를 선정하였다.

실시예 2: 마우스 모델에서 Hsp 90 다중 펩타이드 백신의 면역원성 확인
마우스모델에서 Hsp 90 다중 펩타이드 백신의 면역원성을 확인하기 위하여, FVB 마우스 (6~8주령)를 각 군당 5마리씩으로 생리식염수(PBS)와 면역 보조제(immune adjuvant, CFA/IFA) 군 또는 Hsp 90 펩타이드와 면역보조제 군으로 나누고, 10일 마다 3회씩 마우스 등에 피하주사로 투여하고 마지막 투여로부터 10일 후 마우스의 비장을 적출하여 비장세포를 분리하고 인터페론감마(IFNγ)-고착화효소항체법(EezymeLinked Immuno-SPOT assay, ELISPOT)을 이용하여 Hsp 90 다중 펩타이드 특이 T 세포반응을 관찰하였다.
마우스 비장 세포 준비 및 동결 시, 기계장치 및 물질은 표 2에 나타난 바와 같다.

마우스 T 세포 배지(Mouse T Cell Media)는 500mL RPMI-1640(L-Glut + Hepes) [Mediatech Cellgro], 5mL 페니실린 - 스트렙토 마이신 용액(penicillin-streptomycin solution)[Mediatech Cellgro, #30-002-CI], 50mL 열에 의해 불활성화된 태아 소 혈청(heat-inactivated fetal bovine serum)[SAFC Biosciences, 확인된 경우, 다른 것도 사용가능], 0.5mL 1000X 2-메르캅토에탄올(2-mercaptoethanol)[GIBCO, #21985]를 이용하였고, 플라스틱웨어(plasticware)로 필터링(Filtering)하였다. 동결 배지(Freezing Media)는 45ml heat-inactivated FBS 및 5ml DMSO를 포함하며, 필터링 후 4℃조건에서 보관하였다. ACK 용해 버퍼(Lysis Buffer)(RBC solution)는 1L ddH2O, 8.29g NH4Cl (final 150mM) 및 1g KHCO3 (최종 농도 10mM)를 포함하고, pH 7.2-7.4이며, 필터링하였다.
멸균 기술(sterile technique)은 어세이(assay)의 각 단계(step)마다 사용하였다. 모든 실험자들은 조직 배양 후드(tissue culturehood)에서 시행하였으며, 20개가 넘는 비장세포(splenocyte)를 준비해야 한다면 9-12단계는 건너 뛰었다.
적출한 비장에서 비장세포를 분리하는 방법은, Mouse T cell 배지를 50ml 튜브에 적당량 덜어서 37℃ 항온수조(water bath)에서 가열한 후, p100 dish에 3ml 배지와 신선한 비장(fresh spleen)만 남기고 버렸다. 이후, 비장을 칼날을 이용하여 잘게 자른다. 50ml tube에 셀 스트레이너(cell strainer)를 끼고 잘게 잘린 비장과 배지를 넣어주었다. 1.5ml tube끝으로 비장을 셀 스트레이너 위에서 갈아준 후 10ml warm T cell media로 셀 스트레이너에 분주하여 세포들을 걸러내었다. 또한, p100 dish의 세포도 수득하였다. 이후, 원심분리기를 이용하여 1,200rpm조건에서 8분 동안 원심분리하였다. 상층액은 조심스럽게 제거한 후, 10ml warm Mouse T cell media 첨가하여 펠렛(pellet)을 리서스펜딩(resuspend)한다. 이후, 원심분리기를 이용하여 1,200rpm조건에서 8분 동안 원심분리한 뒤, 상층액을 조심스럽게 제거하였다. ACK Lysis Buffer5ml로 서스펜딩(suspend)한 후에 5분 동안 배양(incubation)하였다. 이후, Mouse T cell media 10ml을 첨가하고, 원심분리기를 이용하여 1,200rpm조건에서 8분 동안 원심분리하고, 상층액을 조심스럽게 최대한 완전히 제거하였다. 그 후, warm mouse matis media 10ml을 첨가하고, 15ul를 새로운 튜브로 옮겨 세포 카운팅(cell counting)하였다. 원심분리기를 이용하여 1,200rpm조건에서 8분 동안 원심분리하고, 상층액을 조심스럽게 제거하였다.
같은 날에 비장세포(splenocyte)를 사용할 경우, 세포에 원하는 농도를 맞춰서 배지를 넣어준 후 suspension하였다. 비장세포를 동결할 경우, 2ml의 freeze media를 넣고 resuspend한 후 2개의 tube에 1ml씩 할당(aliquot)하였다. 그 후에 -80도 냉동고(freezer)에 overnight으로 보관한 뒤 그 다음날 LN2로 옮겨주었다.
인터페론감마(IFNγ)-고착화효소항체법(EezymeLinked Immuno-SPOT assay, ELISPOT)을 수행 시, 기계장치 및 물질은 표 3에 나타난 바와 같다.

Mouse T Cell Media는 상기와 동일한 것을 사용하였다.
(Day 0)에 Prewet MAIPS 96-웰 플레이트(well plate)에 35% 에탄올 30ul 1분 배양(incubation) 후, 1xPBS 200ul 씩 분주하여 3회 세척하였다(Ultra purewater과 에탄올은 Merck를 사용하여 35% 에탄올을 제조하였고, PBS는 1xPBS를 구입하여 사용하였다). 항 마우스 IFNγ 항체(anti-mouse IFNγ antibody)(1mg/ml, stored at 4℃, green cap)를 1xPBS를 이용하여 10ug/ml로 희석하였다(10ul/ml). 제조한 solution을 각각의 well에 50ul씩 분주하였다. 96-웰 플레이트 당 5ml의 양이 필요하다. 플레이트 뚜껑을 닫고 랩으로 감싼 후 4℃조건에서 16~24시간 동안 배양하였다(이 때, 반드시 랩을 씌워야한다).
(Day 1)에 플레이트를 꺼내온 후 멀티채널 피펫(multichannel pipette)을 이용하여 항체(antibody)를 제거하였다. 1xPBS 200ul 씩 분주하여 3회 세척하였다. 마지막 세척에서 plate에 막(membrane) 손상 없이 PBS를 완전히 제거하였다. Mouse T cell media 200ul씩 각각의 well에 분주하여 blocking 한다. 37℃ 인큐베이터(incubator)에서 2시간 동안 배양하였다. plate 뚜껑과 안쪽에 라벨링하였다.

펩타이드의 농도를 mouse T cell media에 2배로 만든 후, 웰 당 100ul씩 분주 할 수 있게 만들고 +1개로 만들었다(표 4).

이후, 블랏킹 배지(blocking media)를 제거하고, 상기 제조한 항원 샘플(antigen sample)을 서로 적절한 웰에 100ul씩 분주하였다. 동결 또는 신선한 비장세포를 어세이(assay)에 사용하였다. 비장세포의 분리방법은 상기 SOP IV-101(Mouse Splenocyte Preparation and Freezing protocol)를 참고하여 실험을 진행하였다. Mouse T cell media에 ml당 3.0x106~3.5x106의 최종농도로 resuspend 하였다. 멀티채널 피펫을 이용하여 카운팅한 세포를 웰 당 100ul (3.0x105~3.5x105)씩 분주하였다. 단, no antigen 웰은 mouse T-cell media 100ul씩 분주하였다. 37℃ 인큐베이터에 넣고 70~72 시간 동안 배양하였다. 세포가 반응하면 배지 색이 진한 노란색으로 바뀌는 것을 확인하였다(Day 1 end).
(Day 4)에는 70-72 시간 동안 배양한 후 37℃ 인큐베이터에서 꺼내어 플레이트의 배지를 멀티채널 피펫으로 조심스럽게 제거하였다. 1x PBS 200ul씩 2회 세척하고, 1x PBS+0.05% tween-20 200ul씩 3회 세척하였다. 단, 마지막 세척은 조심스럽게 1x PBS+0.05% tween-20을 완전히 제거하였다.
바이오티닐화된 항-마우스 IFNγ 항체(biotinylated anti-mouse IFNγ antibody)(1mg/ml, stored at 4℃, yellow cap)를 1x PBS+0.05% tween-20을 이용하여 5ug/ml로 희석하였다(5ul/ml). 제조한 용액을 각 웰에 50ul씩 분주하였다. 96-웰 플레이트 당 5ml의 양이 필요하다. 플레이트 뚜껑을 닫고 랩으로 감싼 후 4℃조건에서 16~24시간 동안 배양하였다(반드시, 랩을 씌어야 함) (Day 4 end).
(Day 5)에는 배양 후 플레이트의 배지를 멀티채널 피펫으로 조심스럽게 제거하였다. 1x PBS 200ul 씩 분주하여 4회 세척하였다. 마지막 세척 시, 플레이트에 막(membrane) 손상없이 PBS를 완전히 제거하였다. 1x PBS에 스트렙타비딘-HRP(streptavidin-HRP)을 1:250으로 희석(4ul/ml)하였다. 제조한 용액을 각각의 웰에 50ul씩 분주하였다. 96-웰 플레이트 당 5ml의 양이 필요하다. 플레이트 뚜껑을 닫고 랩으로 감싼 후 상온(room temperature)에서 60분 동안 배양하였다(HRP 반응시키는 동안에 ABC kit를 상온에 꺼내 놓는다).
배양 후 플레이트의 배지를 멀티채널 피펫으로 조심스럽게 제거하였다. 1x PBS 200ul 씩 분주하여 4회 세척하였다. 마지막 세척 시, 플레이트에 막(membrane) 손상없이 PBS를 완전히 제거하였다. 플레이트의 밑부분을 분리하여 PBS의 물기를 휴지로 제거하였다.
ABC kit development solution(큰 bottle 1ml에 작은 것 한방울 mix) 50ul 씩 각 웰에 분주하였다. 96-웰 플레이트 당 5ml의 양이 필요하다. positive control wells의 color를 확인하였다. Development 시간을 길게는 45분을 넘기지 않는다. 보통은 5~10분 사이에 spot이 보이나, stop이 보이지 않으면 20~30분까지 늘린다. stop reaction은 흐르는 차가운 수돗물 조심스럽게 세척하였다. 어두운 곳 상온에 자연 건조시켰다(이때, Stop이 light-sensitive 하여서 빛에 노출되지 않도록 한다).
그 결과, FVB 마우스 모델의 HSP90 펩타이드 반응에서 Hsp 90 다중 펩타이드 백신군이 대조군에 비해 인터페론감마(IFN-γ) 분비에 의한 특이(specific) 반응성이 높게 나타나는 것을 확인하였다(도 2).
실시예 3: 마우스 모델에서 HSP90 다중 펩타이드 백신의 항종양 효과 확인
항종양 효과를 확인하기 위하여, 마우스 유방암 세포주(mouse mamary cancer cell-FVB/N-Tg(MMTVneu)-202Mul 마우스 유래 HER-2 과발현 유방암 세포주)를 이용하였다. MMTVneu 형질전환 마우스 암컷 6주령을 각 군당 10마리씩으로 생리식염수와 면역 보조제 군과 HSP90 펩타이드와 면역 보조제 군으로 나누고, 10일 마다 3회씩 피하주사를 투여하고 마지막 투여로부터 10일 후 마우스 유방암 세포주를 마우스 옆구리에 피하 주사하여 접송하고 종양의 성장을 3~4일 간격으로 관찰하며 종양크기를 측정하여 HSP90 다중 펩타이드 백신의 항종양 효과 확인하였다. 실험방법은 실시예 2와 동일하다.
실험 결과, 마우스 모델에서 대조군에 비해 Hsp90 다중 펩타이드 백신군의 종양크기가 현저하게 작아지는 것을 확인하였다(도 3a). 또한, HSP90 펩타이드 반응에서 HSP90 다중 펩타이드 백신군이 대조군에 비해 인터페론감마(IFN-γ) 분비에 의한 특이(specific) 반응성이 높게 나타나는 것을 확인하였다(도 3b).
따라서, 본 발명의 Hsp90 다중 펩타이드 백신 조성물은 면역원성을 나타내며, 항종양 효과가 우수하다는 것을 알 수 있다.
실시예 4: 마우스 모델에서 HSP90 다중 펩타이드 백신 / STING agonist / 면역조절 억제제(CTLA-4) 병용 투여의 항종양 효과 확인
4.1 종양 형성 및 항종양 측정
6주령의 암컷 MMTVneu 형질전환 마우스 옆구리에 마우스 유래 유방암 세포 5 X 105개를 피하 주사하였다. 10일 후에 1개 대조군(PBS+면역보조제; immune adjuvant; Complete Freund’s Adjuvant, Incomplete Freund’s Adjuvant), 4개의 실험군으로서 1군 HSP90 다중펩타이드 백신군(HSP90 백신+immune adjuvant), 2군(HSP90 백신+CTLA-4 억제제; HSP90백신+CTLA-4 억제제+immune adjuvant), 3군(STING Agonist+CTLA-4 억제제; STING agonist+CTLA-4 억제제+immune adjuvant), 4군(HSP90 백신+STING agonist+CTLA-4 억제제)을 10일 간격으로 주사하였다. HSP90 백신 및 STING agonist (DMXAA, 5,6-dimethylxanthenone-4-acetic acid, Invivogen제품)는 3회 피하 주사 및 복강 주사하였고, anti-mouse CTLA-4 (CD152, Bioxcell 제품)는 주 2회씩 실험이 끝날 때까지 복강주사를 실시하였다. 실험 기간 동안, 종양의 성장을 3~4일 간격으로 측정하였다. 마지막 투여 10일 후 마우스의 비장을 적출하여 비장세포를 분리하고 인터페론감마(IFNγ)-고착화효소항체법(EezymeLinked Immuno-SPOT assay, ELISPOT)을 이용하여 HSP90 다중 펩타이드 특이적인 T 세포 반응을 확인하였다. 마우스 비장 세포 준비 및 동결, ELISPOT 실험방법은 실시예 2와 동일하게 실시하였다.
4.2 HSP90 다중 펩타이드 백신 효과의 생물학적 표지자 측정
HSP90 다중 펩타이드 백신 효과의 생물학적 표지자로서 혈중 HER2 ECD(Human Epidermal growth factor Receptor type2 ExtraCellular Domain) 변화를 효소면역측정법 (EezymeLinked Immunosorbent assay, ELISA)에 의해 분석하였다. 구체적인 분석 방법은 다음과 같다.
가. 실험 재료
a. 탄산염 완충액 (Carbonate buffer) : 1 리터 비커에 0.8g Na2CO3과 1.47g NAHCO3를 넣고 1차 증류수 400ml을 넣고 용해시킨 후, pH 9.6을 맞추고 1차 증류수를 500ml까지 채운다. 필터링 후 4℃에서 보관하였다.
b. 희석 완충액 (Diluent buffer; 1X PBS / 1% BSA) : 10g BSA를 10x PBS에 100ml에 넣고 녹으면 1리터의 볼륨까지 1차 증류수를 추가하였다. filtering 후에 4℃에서 보관하였다.
c. 블랏킹 완충액 (Blocking Buffer ; 1X PBS / 5% BSA): 50g BSA를 10xPBS에 100ml에 넣고 녹으면 1리터의 볼륨까지 1차 증류수를 추가하였다. filtering 후에 4℃에서 보관하였다.
d. 세척 완충액 (Wash Buffer; 1xPBS / 0.1% Tween-20): 100ml 10xPBS, 1ml Tween-20 및 1차 증류수 899ml를 혼합하여 제조하였다.
e. 2차 항체 : Goat anti-mouse IgG (H+L) HRP 2차 항체는 ThermoFisher scientific사(cat# 62-6520, 용량 1ml)로부터 구입하였다. 4℃에서 보관하였고, 사용 시에 세척 완충액을 사용하여 1:10,000으로 희석하였다.
f. 마우스 IgG 표준 항체: 마우스 IgG 표준 항체는 Sigma 사(cat# I-4506)로부터 구입하였다. 구입 후에, 항체는 150 mM NaCl를 희석 완충액으로 사용하여 최종 농도 2 mg/ml로 만들어서 -20℃에 보관하였다.
g. TMB 용액: TMB 용액 Sigma사(cat# T0440)에서 구입한 후, 4℃에서 보관하였다.
h. 1N HCl 반응정지액(stop solution) : 1차 증류수 481.8 ml에 38% HCl 18.2 ml를 조심스럽게 천천히 조금씩 첨가하여 스톡 용액(stock solution)을 제조하여 상온에 보관하였다.
나. 실험 방법 및 결과
하기의 단계를 거쳐 ELISA 실험을 진행하였다. 단계 1 및 단계 2는 얼음 조건에서 실시하였다.
탄산염 완충액을 A 튜브에는 2.5ml, 나머지 튜브에는 1ml씩 분주한다. 그리고 A 튜브에 마우스 IgG 스톡 용액 (2mg/ml) 10ul를 첨가한다. 보텍싱을 실시하고 A 튜브에서 1ml을 B 튜브로 옮겨서(1:1 dilution) B->C, C->D 순서로 L까지 1:1로 희석을 실시하였다(단계 1) .
신선한 탄산염 완충액을 이용하여 1ug/ml 농도의 HSP90 재조합 단백질을 제조하였다. 상기 HSP90 재조합 단백질은 Coat column #1 ~ #10에 well 당 50ul씩 분주하였다. column #11에는 carbonate buffer만을 분주하였고, Microseal로 plates를 덮어준 후 4℃에서 하룻밤 동안 항원-항체 반응을 실시하였다(단계 2). 200ul 세척 버퍼를 사용하여 3회 세척하였고, 3회 세척시에는플레이트를 페이퍼 타올에 조심히 털어서 세척 버퍼를 완전히 제거하였다(단계 3). 그 다음 블랏킹 용액을 웰당 100ul씩 분주하고 상온에서 1~2 시간 동안 반응시켰다(단계 4). 상기 단계 3과 같은 세척을 실시하였다(단계 5). 새로운 1.5ml 튜브에 희석 버퍼: 세럼을 1:1 비율로 혼합하여 보텍싱 후 각각의 웰에 50ul씩 분주하였다(단계 6). 플레이트 컬럼 #11 및 #12에 희석 버퍼 50ul를 분주하였다(단계 7). 마이크로실( microseal)을 사용하여 플레이트를 덮어준 후, 4℃에서 하룻밤 동안 반응시켰다(단계 8). 상기 단계 3과 같은 세척을 실시하였다(단계 9). HRP 컨쥬게이트 항체를 희석 버퍼를 사용하여 1:10,000로 희석하였고, 웰당 50ul씩 분주한 다음 상온에서 45 분 동안 교반하면서 반응시켰다(단계 10). 상기 단계 3과 같은 세척을 실시하였다(단계 11). TMB 용액을 상온으로 꺼내어 준비해 둔 다음, TMB 용액을 각각의 웰당 100ul씩 분주하였고, 빛이 없는 상온에서 20분 동안 반응시켰다.(단계 12). 반응정지액을 각각 웰당 50ul씩 분주하였다(단계 13). 마이크로플레이트 리더 기기를 사용하여 450nm에서 형광세기를 측정하였다(단계 14).
HSP90 다중 펩타이드 백신 투여 후 HER2 ECD 혈중 농도 변화를 효소면역측정법(EezymeLinked Immunosorbent assay, ELISA)으로 측정한 결과, 본 발명에 따른 HSP90 백신을 사용한 결과 HER2 ECD가 낮게 측정되었다.
4.3 HSP90 다중 펩타이드 백신의 병용 투여에 의한 효과
실험 결과, 마우스 모델에서 대조군, 1군(HSP90 다중 펩타이드 백신군), 2군(HPS90 다중 펩타이드 백신+CTLA-4 억제제) 및 3군(STING agonist+CTLA-4 억제제)에 비해 4군(HSP90 다중 펩타이드 백신+STING agonist+CTLA-4 억제제)을 병합 치료하는 경우 종양크기가 현저히 감소하는 것을 확인하였다(도 4a). 또한, 대조군에 비해 모든 실험군에서 HER2-ECD 반응성이 감소하는 것을 확인하였다(도 4b). HSP90 펩타이드 반응은 4군(HSP90 다중 펩타이드 백신+STING agonist+CTLA-4 억제제)에서 인터페론감마(IFN-γ) 분비에 의해 더 많은 특이(specific) 반응성을 나타내었다(도 4c). 이를 통해, 본 발명의 HSP90 펩타이드 백신을 STING agonist / CTLA-4 엑제제를 병합 치료함으로써 HSP90 펩타이드 백신의 효과가 증감하므로 HSP90 펩타이드를 백신으로 사용할 수 있음을 알 수 있다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.

- 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프인 폴리펩타이드.
- 제1항의 폴리펩타이드를 코딩하는 유전자.
- 제2항에 있어서, 서열번호 3 또는 4의 염기서열로 표시되는 것을 특징으로 하는 유전자.
- 제2항의 유전자를 포함하는 재조합 벡터.
- 제2항의 유전자 또는 제4항의 재조합 벡터가 도입된 재조합 미생물.
- 다음 단계를 포함하는 제1항의 폴리펩타이드의 제조방법:
(a) 제5항의 재조합 미생물을 배양하여 제1항의 폴리펩타이드를 생성시키는 단계; 및
(b) 상기 생성된 폴리펩타이드를 수득하는 단계.
- 제1항의 폴리펩타이드를 유효성분으로 포함하는 암의 치료 또는 예방을 위한 백신 조성물.
- 제7항에 있어서, 상기 조성물은 서열번호 1 및 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프를 모두 포함하는 것을 특징으로 하는 백신 조성물.
- 제7항에 있어서, 상기 조성물은 면역보조제를 추가로 포함하는 것을 특징으로 하는 백신 조성물.
- 제 9항에 있어서, 상기 면역항암제는 CTLA-4(cytotoxic T-lymphocyte-associated protein 4), PD-1(Programmed cell death protein 1), LAG-3(Lymphocyte Activation Gene-3), TIM-3(T-cell Immunoglobulin and Mucin-domain containing-3), TIGIT(T-cell Immunoreptor with IG and ITIM domain) 및 VISTA(V-domain Ig Suppressor of T cell Activation)로 구성된 군으로부터 선택된 어느 하나에 대한 면역관문 억제제인 것을 특징으로 하는 백신 조성물.
- 제 9항에 있어서, 상기 조성물은 항암 보조제를 추가로 포함하는 것을 특징으로 하는 백신 조성물.
- 상기 항암 보조제는 STING(STimulator of InterferoN Gene) 아고니스트인 것을 특징으로 하는 백신 조성물.
- 제 1항의 폴리펩타이드를 유효성분으로 포함하는 항암용 조성물.
- 서열번호 1 또는 2의 아미노산 서열로 표시되는 열충격단백질 90의 에피토프를 특이적으로 인식하는 항체.
