갈대 추출물의 갈락토미세스 발효물의 피부 진피층의 콜라겐 형성세포인 인간 섬유아세포(human fibroblast)에 대한 세포독성효과을 조사하였다. 구체적으로 DMEM 배지에서 배양한 인간 섬유아세포를 96웰-플레이트에 각 웰당 10,000개 세포씩 씨딩(seeding)하였다. 여기에 각각 상기 실험예 1의 갈대 추출물과 갈대 추출물의 갈락토미세스 발효물을 각각 0.125%부터 8%까지 첨가하고 72시간동안 배양하였다. 배양 후 살아있는 세포를 염색시키는 Easytox kit 시약을 처리하였고, 1시간 후 ELISA reader를 이용하여 450nm에서 흡광도를 측정하였다(도 3). 갈대 추출물의 갈락토미세스 발효물에 의한 피부 진피층의 콜라겐 형성세포인 인간 섬유아세포(human fibroblast)의 증식능을 측정함으로써 이의 주름개선효과를 시험하였다. 구체적으로 DMEM 배지에서 배양한 인간 섬유아세포를 96웰-플레이트에 각 웰당 10,000개 세포씩 씨딩(seeding)하였다. 여기에 양성대조군으로 피부 주름개선효과가 있는 것으로 알려진 아스코르브산(ascorbic acid)을 최적농도인 0.005%로 희석하여 첨가하고, 실험군으로 상기 실험예 1의 갈대추출물 및 갈대추출물의 갈락토미세스 발효물을 각각 3%로 희석하여 첨가하고 상기 실시예 3의 방법으로 섬유아세포의 증식능을 조사하였다(도 4).
