Abstract
본 발명은 ROS 생성을 억제함으로써 SHC-1/p66 관련 질환의 하나 이상의 증상을 치료하는 방법 또는 상기 치료를 위한 약제의 제조 방법에 관한 것이다.
Description
본 발명은 노화 관련 질환에 대처하기 위한 SHC-1/p66의 억제 방법에 관한 것이다.
20세기 하반기 동안 불임이 줄고 평균 수명이 20년 연장됨에 따라 세계 인구의 평균 연령이 증가 추세에 있다(참조. 예, Centers for Disease Control and Prevention, Morbidity and Mortality Weekly Report, February 13, 2003, 52(06): 101-106). 2차 세계대전 이후 20년 동안 많은 국가에서의 임신 증가(즉, 베이비 붐(baby boom))와 더불어, 이들 요소들은 2010 - 2030의 기간 동안 65세 이상의 인구 수를 증가하는 결과를 낳게 할 것이다. 전 세계적으로, 평균 수명은 2050년까지 10년이 더 연장될 것으로 예상된다. 노년층 인구의 증가로 인해 공공 건강 시스템과 의료 및 사회적 서비스에 대한 요구가 증가되게 된다. 노년층에 불균형적으로 영향을 미치는 만성 질병은 신체적 장애, 삶의 질 저하 그리고 건강 및 장기 요양 비용의 증가를 초래한다.
노화의 증상과 이들 증상의 생화학적 및 신경학적 근거 사이의 연관성을 밝히기 위한 연구가 지속되어 왔으며, 특히, 수명의 연장 및 다양한 노화 관련 질환으로부터의 보호를 위한 여러 생물학적 경로가 밝혀졌다(Trinei et al., "P66Shc Signals to Age" Aging (2009) v. 1(6) pages 503-510).
본 발명에 기재된 방법들은 노화의 증상에 대한 생화학적 및 신경학적 근거에 관련된 것이다. 특히, 본 발명에 기재된 방법들은 SHC-1/p66 경로, 활성산소종(reactive oxygen species; ROS) 및 이로 인한 산화적 스트레스, 간 기능 및 병리학, 및 운동 조절(motor control)에 대한 긍정적 효과를 보여주고 있다.
SHC-1/p66과 노화
분자량 66kD의 Src 상동성 콜라겐(Src Homology and Collagen; SHC) 아종(isoform) 단백질 패밀리가 결핍된 생쥐는 p66-풍부 생쥐(p66 proficient littermates)에 비해 수명이 30% 더 길다는 것이 보고된 바 있다. p66KO 생쥐는 수명이 길고, 표현형질상 정상이고, 가임(fertile)이며, 건강한 것으로 보여진다 (Migliaccio et al., "The p66shc adaptor protein controls oxidative stress response and the life span in mammals. Nature. 1999; 402:309-313; see also Alam et al., Endocr. Relat Cancer. 2009 5 March; 16(1): 1.) SHC 단백질들은 어댑터 분자(adapter molecules), 즉, 활성화된 성장 인자 수용체(RTKs)의 하부(doswnstream)에 위치한 거대-연구 관점 분자 복합체(macro-Research Perspective molecular complexes)의 조립(assembly)과 관련된 신호발송(signaling) 성분으로 알려져 있다. 인슐린 신호 전달(signaling)에 있어서 SHCs의 역할도 보고된 바 있다 (Giorgetti et al."Involvement of Src homology/collagen (SHC) proteins in signaling through the insulin receptor and the insulin-like-growth-factor-I-receptor." Eur. J. Biochem. 1994; 223:195-202). 따라서, 상기 p66KO 생쥐는 인슐린/IGF 신호발송의 유전적 감쇠(genetic attenuation)에 의한 수명 연장을 갖는 최초의 포유동물의 한 예(example)로 볼 수 있다.
오히려, p66와 장수(longevity)의 관계는 예기치 못한 방향으로 진전되어 하만(Harman)의 "노화에 대한 자유 급진 이론(free radical theory of aging)" (Harman, D.,"A biologic clock: the mitochondria?" Journal of the American Geriatrics Society 1972; 20:145-147.)을 지지하는 가장 강력한 논쟁 중 하나가 되었으며, 실제로, p66-결핍 생쥐 및 세포는 현저히 감소된 수준의 활성산소종(ROS) 및 산화적 스트레스에 대한 저항력 증가를 나타낸다.
p66shc는 다른 SHC 단백질과는 전혀 다른 기능을 갖고 있다: 산화촉진적(prooxidant) 및 세포자살적(apoptogenic) 자극에 대한 수에 따라, p66shc는 미토콘드리아로 전위(translocate)되어 직접 활성산소종(ROS)을 생산하는 것으로 밝혀졌다.

Oncogene. 1996; 13:633-41)으로도 명명된 글리신 및 프롤린 잔기가 매우 풍부한 영역을 포함하는 동일한 아미노산 서열을 공유한다. p66Shc의 특징은 N-말단(N-terminus)에 있는 추가의 CH영역(CH2)이다(Pelicci, G. et al., "A novel transforming protein (SHC) with an SH2 domain is implicated in mitogenic signal transduction." Cell. 1992; 70: 93-104. Migliaccio, E. et al., "Opposite effects on the p52shc/p46shc and p66shc splicing isoforms on the EGF receptor-MAP kinase-fos signaling pathway. EMBO J. 1997; 16:706-716).
활성산소종(Reactive Oxygen Species), 세포적 스트레스(Cellular Stress) 및 노화(Aging)
활성산소종(ROS) 슈퍼옥사이드(superoxide) 및 과산화 수소는 많은 생리학적 및 병리생리학적(pathophysiological) 과정에서 중요한 시그널링 기능을 수행한다. 세포 노화(cell senescence) 및 유기체의 노화(organismal age)도 예외가 아니다 (Afanas'ev, Igor, Oxidative Medicine and Cellular Longevity, V.3 (2010), Issue 2, pages 77-85). 활성산소종(ROS)은 세포자살(apoptosis)의 잠재적 유도체로서 세포자살 프로그램 자체를 수행한다 (Giorgio et al. "Electron Transfer between Cytochrome c and p66Shc Generates Reactive Oxygen Species that Trigger Mitochondrial Apoptosis," Cell Vol. 122, 221-233, July 29, 2005).
특히, SHC-1/p66는 활성산소종(ROS) 시그널링에 의해 자극되며, 반대로 활성산소종(ROS)은 SHC-1/p66에 의해 분비된다.
그 결과, SHC-1/p66은 당뇨-유도 산화적 스트레스 및 산화제-의존적 신장 상해(renal injury)의 기저를 이루는 분자 단계의 메커니즘으로 이해되어 왔다(Menini et al., Diabetes 55:1642-1657 (2006)). 더욱이, SHC-1/p66 는 고혈당 관련 혈관 내피세포 기능 장애(high glucose associated endothelial dysfunction), 죽종형성(atherogenesis), 당뇨적 신증(diabetic nephropathy) 및 심근병증(cardiomyopathy)과 같은 당뇨 관련 증상에 연루되어 왔다(참조: Francia et al., J. Mol. Med. (2009) 87:885-891 and Berniakovich et al. JBC Vol. 283 No. 49, pp. 34283-34293, December 5, 2008).
노화와 간(Aging and the Liver)
노화된 간은 어린 간에 비해서 세포의 퇴화가 증가된다. 특히, 노화된 간은 세포 메커니즘의 손상 및 미토콘드리아의 기능 및 형태(morphology)에 있어 특이적 변화를 야기한다(Sastre et al. "Aging of the Liver: Age-Associated Mitrochondrial Damage in Intact Hepatocytes," Hepatology November. (1996) pp. 1199-1205 and Koch et al., "Role of the life span determinant P66shcA in ethanol-induced liver damage," Laboratory investigation (2008) 88, 750-760)). 예를 들면, 미토콘드리아의 크기가 커지는 반면, 포도당 생성(glucogenesis) 및 케톤 형성(ketogenesis)은 감소된다. 간세포의 퇴화는 풍선확장(ballooning) 또는 "거품형(foamy)"세포 또는 지방증(steatosis)(지방 변화, 지방 퇴화 또는 지방질 퇴화로도 불림)이라는 증거를 통해 병리학적으로 탐지될 수 있다.
풍선확장된(ballooned) 세포들은 인근 간세포들에 비해 통상적으로 2 내지 3 배로 크며, H&E 염색 단면에서 성긴 투명 세포질을 갖는 특징이 있다. 이들은 이들의 세포질 및 핵에 의해 지방세포-유사(adipocyte-like) 세포로부터 분화될 수 있다: (주변에 핵을 갖는 지방세포-유사들과는 달리) 풍선확장된 세포들은 중앙에 핵을 갖고 있다. 또한, 풍선확장된 세포들은 (작은) 소견(pyknotic nuclei) 또는 핵붕괴(karyorrhexis) 가 진행 중인(즉, 붕괴 과정에 있는) 세포를 갖는다. 지방세포-유사 세포들은 투명한 세포질이나 공포(vacuolated)를 갖는 것인 반면에, 풍선확장 퇴화가 진행 중인 세포질은 성긴(wispy)/거미줄 같다.
지방증(steatosis)은 세포 내에서 지방질의 비정상적 보유(retention)를 나타내는 과정이다. 지방증은 중성지방(triglyceride fat)의 정상적 합성 및 제거 과정에 있어서의 손상을 나타내는 것으로서, 지방증은 산소 유리기(oxyradicals)에 의해 야기되는 간세포의 팽윤과 연관되어 왔다 (Del Monte, "Swelling of hepatocytes injured by oxidative stress suggests pathological changes related to macromolecular crowding" Medical Hypotheses (2005) 64, 818-825).
노화가 운동기능(Motor Skills) 및 운동력저하(Motor Deficits)에 미치는 영향
노년에 들어서면서 감각 운동 조절(sensory motor control) 및 기능의 저하가 나타난다. 미세 운동 조절, 보행 및 균형의 저하는 노년층 성인의 일상적이고 독립적인 활동의 수행에 영향을 미친다(Yankner BA, Lu T, Loerch P. The aging brain. Annu Rev Pathol 2008;3:41-66.; Twohing, J. P. et al., "Age-dependent maintenance of motor control and corticostriatal innervation by death receptor 3." J. Neurosci. 2010. 30:3782-3792.). 이들 운동력 저하의 원인은 중추신경계의 퇴화 및 감각 수용체, 근육, 및 말초신경의 기능의 변화와 같이 다인성인(multi-factorial) 것이다.
노년층 성인의 운동 수행력 결핍(motor performance deficits)은 신경근육계 시스템뿐만 아니라 중추신경계 및 말초신경계의 기능 장애에 기인하는 것으로 생각된다(Seidler, R. et al., "Motor control and aging: Links to age-related brain structural, functional, and biochemical effects."Neuroscience and Biobehavioral Reviews 30 (2010) 721-733). 젊은층 성인에 비하여, 운동 수행력 결핍은 협조 장애(coordination difficulty) (Seidler, RD et al.,. "Changes in multi-joint performance with age." Motor Control. 2002;6(1):19-31.), 동작 가변성(variability of movement)의 증가(Contreras-Vidal et al., "Elderly subjects are impaired in spatial coordination in fine motor control," Acta Psychol (Amst). 1998 Nov;100(1-2):25-35; Darling et al., "Control of simple arm movements in elderly humans." Neurobiol Aging, 1989 Mar-Apr;10(2):149-57), 움직임의 둔화(slowing of movement)(Diggles-Buckles V. "Age-related slowing. In: Stelmach GE, Homberg V, editors." Sensorimotor impairment in the elderly. Norwell, MA: Kluwer Academic; 1993.), 및 균형 및 보행상 장애(Tang PF, Woollacott MH. "Balance control in the elderly." In: Bronstein AM, Brandt T, Woollacott MH, editors. Clinical disorders of balance, posture and gait. London: Arnold; 1996.)를 포함한다.
이들 결핍은 노년층 성인이 일상적 삶의 기능적 활동을 수행함에 있어 부정적 영향을 초래한다. 노년층 성인에 있어 낙상(falls)이 부상 및 발병(morbidity)의 주요 원인이 되므로, 보행 및 균형상 문제는 특히 중요한 문제점으로 대두된다: 낙상을 경험한 노년층 성인의 20-30%는 보통 또는 심각한 부상으로 이해 이동이 제한되고 삶의 질이 저하된다. 연령의 증가에 따라 운동 지속(movement duration)에서 뚜렷한 증가가 다양한 업무에서 나타난다. 연령이 증가함에 따라 움직임은 최대 15-30%까지 느려진다 (cf. Diggles-Buckles, 1993). 노년층 성인은 움직임의 속도 보다는 움직임의 정확성에 치중하는 편이므로 이러한 둔화된 움직임은 일편 전략적인 것으로 볼 수도 있다. 정보 처리 속도의 둔화는 또한 신경 소음(neural noise) 및 기타 시냅스 변화(synaptic changes)의 증가에 따라 운동성 수행에 비특이적 글로벌 형태의 영향을 미칠 수도 있다.
더욱이, 보행 패턴의 변화와 노화 간에 연관성이 있음이 밝혀졌다. 특히, 노령 인구에 있어서, 보행은 짧아지고 느려진다(Wolfson, L., et al., (1990) "Gait assessment in the elderly: A gait abnormality rating scale and its relation to falls." J. Gerontol. 45: M12-19). 인간의 보행에 있어서 이러한 인식적 변화는 설치류에서 연령-의존형 보행과 연관 지어져 (See Klapdor, K. et al., (1997) "A low-cost method to analyze footprint patterns." J. of Neuroscience Methods. 75: 49-54 and Hilber et al., (2001) "Motor skills and motor learning in lurcher mutant mice during aging." Neuroscience. 102:615-623).
본 발명의 일 목적은 활성산소종(ROS)을 저해함으로써 SHC-1/p66 관련 질환의 하나 이상의 증상을 치료하는 방법 또는 상기 치료를 위한 약제를 제조하는 방법을 제공하는 것이다.
일 실시예에서 본 발명은 화학식 I 의 단량체 단위 및/또는 이의 약학적으로 허용 가능한 염, 용매화물 또는 프로드러그(prodrug)를 함유하는 고분자 조성물을 필요로 하는 포유동물에 투여함으로써 SHC-1/p66-의존형 질환의 하나 이상의 증상을 치료하는 방법에 대한 것으로서,
화학식 I
상기 화학식 I 에서 R1 및 R2는 각각 독립적으로 수소, 알킬, 또는 아실이고; R3, R4, R5, R6 및 R7은 각각 독립적으로 수소, 히드록시, 알콕실, 또는 아실이며; R8는 수소 또는 당 성분(saccharide moiety)이고; 상기에서 상기 단량체의 중합 수가 2 내지 30이며, 상기 고분자의 분자량은 600 내지 10,000이다.
일 실시예에서 본 발명은 노화, 당뇨 및 허혈 후 재관류 손상(reperfusion injuries after ischemia)으로 이루어진 그룹에서 선택된 SHC-1/p66 관련 질환의 하나 이상의 증상 치료용 약제의 제조를 위한 고분자 조성물의 용도에 관한 것으로서, 상기 고분자 조성물은 화학식 I 의 단량체 단위 및/또는 이의 약학적으로 허용 가능한 염, 용매화물 또는 프로드러그(prodrug)를 함유한다.
일 실시예에서 본 발명은 SHC-1/p66의 발현 또는 활성에 의해 초래되는 질환의 하나 이상의 증상을 치료하는 방법에 대한 것이다. 일 구체적 실시예에서, "SHC-1/p66 관련 질환"은 SHC-1/p66의 발현 또는 활성을 감소시킴으로써 개선된다. 상기 질환은 노화, 당뇨 및 허혈 후 재관류 손상을 포함한다. 본 발명의 방법은 세포변성(cellular degeneration), 간세포 팽윤(hepatocyte swelling), 미토콘드리아 기능 이상(mitochondrial dysfunction), 노화 관련 운동 결핍(age-related motor deficits), 및 보폭 감소(reduced stride length)와 같은 노화 관련 증상들; 고혈당 관련 혈관 내피세포 기능 장애(high glucose associated endothelial dysfunction), 죽종형성(atherogenesis), 신증(nephropathy) 및/또는 심근병증(cardiomyopathy)과 같은 당뇨의 하나 이상의 증상; 및 활성 산소종(ROS) 및 세포사(cell-death)의 증가가 존재하는 것을 포함하는 허혈 후 재관류 손상의 하나 이상의 증상을 치료하는 실시예에 관한 것이다.
일 실시예에서 본 발명의 방법은 BEL-X 및/또는 이의 약학적으로 허용 가능한 염, 용매화물 또는 프로드러그(prodrug)를 함유하는 조성물을 투여하는 것에 대한 것이다. 일 실시예에서 상기 조성물은 50 내지 1,500 mg/kg/일의 용량으로 투여된다.
이하 구체적 실시예를 통하여 첨부된 도면과 더불어 본 발명을 더욱 상세히 설명하고자 한다.
도 1: 본 발명의 일 실시예에 따른 한약재(Herbal drug) BEL-X가 노령의 생쥐의 간에 미치는 치료 효과.
도 1A 내지 도 1C는 본 발명의 일 실시예에 따라 어린 생쥐(2개월, 도 1A)로부터 얻은 포르말린에 고정되고 파라핀에 봉합된(embedded) 간 샘플을 헤마톡실린 및 에오신으로 염색한 슬라이드; 노령의 무처리 생쥐 슬라이드(20개월, 도 1B); 및 p.o. 1000 mg/kg/일 농도의 BEL-X로 처리한 노령의 생쥐의 슬라이드(20개월, 도 1C)이다.

인간 간암세포(hepatoma cells) Huh7를 과산화수소(H2O2)로 처리하여 ROS 생성을 유도하였다 (도 4A 및 4C). 세포를 과산화수소로 유도 후 약물 BEL-X로 처리하였다. 도 4A 및 4C는 현미경으로 관찰한 결과 각 슬라이드에 존재하는 세포 수가 비슷하였다. 상기 세포 중, 도 4B는 면역형광법(immunofluorescences)으로 관찰한 결과 과산화수소 유도 후 다량의 ROS 생성을 나타낸 반면, 약물 BEL-X로 처리한 세포는 ROS 생성이 아주 적어서 BEL-X가 ROS 생성을 감소시켜 세포를 보호할 수 있다는 것을 나타내었다(도 4D).
도 5: 본 발명의 일 실시예에 따른 세포 내 ROS의 감소 탐지. 인간 간암세포 Huh7, 인간 직장암세포 HRT 및 정상 인간 표피 세포 WS1을 포함한 다양한 세포를 테스트하였다. 괄목할만한 것은, 모든 BEL-X 처리한 테스트 세포들이 과산화수소로 유도한 경우 ROS 생성량이 유의성 있게 감소됨을 보여주었다.
도 6a 내지 6c는 본 발명의 일 실시예에 따라 모시플(Boehmeria nivea(L.) Gaud)의 프로안토시아니딘(proanthocyanidins)의 정제(purification)를 13C 자기공명분광(magnetic resonance spectroscopy)을 통한 결과를 보여준다.
도 7a 내지 7b는 본 발명의 일 실시예에 따라 프로안토시아니딘의 두 단량체 간의 결합을 나타낸다.
도 1A 내지 도 1C는 본 발명의 일 실시예에 따라 어린 생쥐(2개월, 도 1A)로부터 얻은 포르말린에 고정되고 파라핀에 봉합된(embedded) 간 샘플을 헤마톡실린 및 에오신으로 염색한 슬라이드; 노령의 무처리 생쥐 슬라이드(20개월, 도 1B); 및 p.o. 1000 mg/kg/일 농도의 BEL-X로 처리한 노령의 생쥐의 슬라이드(20개월, 도 1C)이다.

인간 간암세포(hepatoma cells) Huh7를 과산화수소(H2O2)로 처리하여 ROS 생성을 유도하였다 (도 4A 및 4C). 세포를 과산화수소로 유도 후 약물 BEL-X로 처리하였다. 도 4A 및 4C는 현미경으로 관찰한 결과 각 슬라이드에 존재하는 세포 수가 비슷하였다. 상기 세포 중, 도 4B는 면역형광법(immunofluorescences)으로 관찰한 결과 과산화수소 유도 후 다량의 ROS 생성을 나타낸 반면, 약물 BEL-X로 처리한 세포는 ROS 생성이 아주 적어서 BEL-X가 ROS 생성을 감소시켜 세포를 보호할 수 있다는 것을 나타내었다(도 4D).

상기 화학식 I 에서 R1 및 R2는 각각 독립적으로 수소, 알킬, 또는 아실이고; R3, R4, R5, R6 및 R7은 각각 독립적으로 수소, 히드록시, 알콕실, 또는 아실이며; R8는 수소 또는 당 성분(saccharide moiety)이다.
일 실시예에서 본 발명은 영양소(nutrient), 의약식품(nutriceutical), 건강식품(health food) 또는 보충제(supplement)의 형태인 화학식 I의 단량체 단위를 포함하는 고분자 조성물을 포함하는 조성물을 포함한다.
상기 화학식 I에 있어서, 고분자 화합물의 단량체 단위는 하기 형태(features)를 하나 또는 그 이상 가질 수 있다: R1 및 R2는 독립적으로 수소이고, R3 및 R7은 각각 수소이고, R4, R5 및 R6 는 히드록시 또는 알콕실이고, R8는 수소일 수 있다.

상기 단량체 단위의 중합 수(polymerized number)는 2 내지 30이 될 수 있고, 더욱 바람직하기로는 3 내지 20이다. 평균분자량은 바람직하게는 600 내지 10,000이다.
일 실시예에서 본 발명의 (BEL-X를 포함하는)조성물은 화학식 I의 단량체 단위의 하나 이상의 혼합물을 함유할 수 있다. 일 실시예에서 상기 고분자는 동종 중합체(homopolymers)일 수 있다. 일 실시예에서 상기 조성물은 서로 다른 동종 중합체의 혼합물을 포함할 수 있다. 일 실시예에서 상기 고분자는 화학식 I의 범위에 속하는 다중 이종 단량체(multiple different monomer) 화합물을 포함하는 이종 중합체(heteropolymers)일 수 있다.
"분리 제조물(isolated preparation)"이란 천연 원료 또는 합성 혼합물로부터 분획된(partitioned) 상기 고분자 화합물을 하나 또는 그 이상 함유하는 조성물을 의미한다.
"알킬(alkyl)"이라는 용어는 1 내지 10개의 탄소 원자를 포함하는 직쇄 또는 분지형 탄화수소를 의미한다. 알킬기의 예로는 메틸, 에틸, n-프로필, i- 프로필, n-부틸, i- 부틸, 및 t- 부틸을 들 수 있으나 이에 한정되지는 않는다. 상기 고분자 화합물의 단량체는 비대칭 중심을 포함한다. 따라서, 이들 단량체는 라세미체(racemates), 라세미 혼합물(racemic mixtures), 단일 광학이성질체(single enantiomers), 개별 부분입체 이성질체(individual diastereomers) 및 부분입체 이성질체 혼합물(diastereomeric mixtures) 로 나타날 수 있다. 이들 이성체 형태가 고려된다. 일 실시예에서 상기 단량체는 C4 위치에 (R) 또는 (S) 광학 이성체(optical isomers)를 포함할 수 있다.
본 발명의 또 다른 면으로서, 본 발명은 약학적으로 허용 가능한 담체 및 상기 분리된 제제를 혼합하여 수득한 약학적 조성물을 필요로 하는 환자(subject)에게 투여함으로써 SHC-1 발현, 간세포 팽윤 억제, 활성산소종(ROS)발생 억제 방법에 관한 것 또는 노화관련 운동기능 장애의 발생 또는 심화를 감소시키는 방법에 관한 것이다.
일 실시예에 있어서 본 발명은 SHC-1 발현의 억제, 간세포 팽윤, 활성산소종(ROS)생성 억제 또는 노화관련 운동기능 장애의 발생 또는 심화를 감소시키는 약제의 제조를 위하여 고분자 조성물을 사용하는 것을 포함하며, 상기 고분자 조성물은 화학식 I 의 단량체 단위 및 또는 약학적으로 허용 가능한 염 또는 프로드러그를 함유한다.
또한, 본 발명의 범위에는 SHC-1/p66 관련 질환의 치료 방법 또는 상기 개선/치료/촉진을 위한 약제의 제조에 있어서 상기 분리 제조물 사용하는 것을 포함한다.
이와 다르게, 본 발명의 조성물은 영양소(nutrient), 의약식품(nutriceutical), 건강식품(health food) 또는 보충제(supplement)의 형태의 식품으로 투여할 수 있다.
본 발명의 많은 실시예를 이하 기재하였다. 기타 본 발명의 특징, 목적 및 장점은 이들 기재 및 청구항들로부터 명확해 질 수 있을 것이다.
상기 화학식 I의 고분자 화합물은 SHC-1/p66 관련 질환의 환자를 치료 및 활성산소종(ROS)의 생성을 억제하기 위해 투여할 수 있다.
본 발명에 기재된 조성물은 식물에서 추출되거나 또는 인공적 수단을 사용하여 개질 또는 합성할 수 있다. 일 실시예에서, 상기 식물은 콩과(Leguminosae), 돌나물과(Crassulaceae), 콤브레타과(Combretaceae), 박주가리과(Asclepiadaceae), 장미과(Rosaceae), 꿀풀과(Lamiaceae), 마디풀과(Polygonaceae), 진달래과(Ericaceae), 침엽수(Pinaceae), 포도과(Vitaceae) 또는 쐐기풀과(Urticaceae)에 속하는 식물일 수 있으며, 바람직하기로는 쐐기풀과에 속하는 모시풀(Boehmeria nivea (L.) Gaud)이다. 상기 식물의 추출 부분은 뿌리, 줄기, 잎 및/또는 열매 부분일 수 있다.
추출 방법들은 미국 특허출원 공개번호 제2010/0168221호 및 제2011/0158933호에 기재되어 있으며, 이들 내용 전체는 이하 참조문헌으로 명확히 반영되어 있다. 상기 추출액은 선택적으로 추가 부분 정제 또는 완전 정제될 수 있다.
"감소(reduce)" 또는 "억제(inhibit)"라는 용어는 증상의 심화 또는 발생을 낮추는 것을 포함한다. 일 실시예에서, 감소 또는 억제는 연령과 일치된(age-matched) 비처리 대조군 및/또는 비처리 젊은 대조군과 비교하여 백분율로 측정한다. 일 실시예에서, 증상의 발생은 연령과 일치된 대조군과 비교하여 최소한 25%, 50%, 또는 75%가 감소할 수 있다. 일 실시예에서, 특정 증상의 심각성은 최소한 25%, 50%, 또는 75% 내지 99%를 포함하여 그 이상이 될 수 있다. 증상의 심각서의 100% 감소는 상기 증상이 더 이상 존재하지 않거나 탐지할 수 없음을 의미한다. 일 실시예에서, 감소 또는 억제는 어린 생쥐와 비교하여 상기 증상의 발생 또는 심각성의 "배수(fold)"증가로 측정된다. 바람직하기로는, 상기 배수 증가는 어린 생쥐와 비교하여 상기 증상의 발생 또는 심각성이 최소한 25% 이상 증가하는 것을 의미한다.

유의성 이라는 용어는 처리된 및 미처리된 그룹 또는 기타 연령 그룹을 비교하는 통계적 방법을 사용함을 의미한다.
"SHC-1/p66 관련 질환의 치료"란 SHC-1/p66의 발현 또는 활성에 영향 받는 질병의 하나 이상의 증상을 치료하는 것을 의미할 수 있다. 특히, "SHC-1/p66 관련 질환"은 SHC-1/p66의 발현 또는 활성을 감소시킴으로써 그 증상을 개선시킬 수 있는 질환이다. 상기 질환은 노화, 당뇨, 및 허혈 후 재관류 손상을 포함한다.
특히, 본 발명의 방법들은 세포변성(cellular degeneration), 간세포 팽윤(hepatocyte swelling), 미토콘드리아 기능 이상(mitochondrial dysfunction), 노화 관련 운동기능 장애(age-related motor deficits), 및/또는 보폭 감소(reduced stride length)와 같은 하나 또는 그 이상의 노화 관련 증상; 고혈당 관련 혈관 내피세포 기능 장애(high glucose associated endothelial dysfunction), 죽종형성(atherogenesis), 신증(nephropathy), 또는 심근병증(cardiomyopathy)과 같은 당뇨의 하나 또는 그 이상의 증상; 및 활성산소종(ROS) 및 세포사(cell-death)의 증가의 존재를 포함하는 허혈 후 재관류 손상의 하나 이상의 증상의 치료에 관한 것이다.
"연령관련 운동기능 장애(Age-Related Motor Deficits)"는 운동적 활동 속도의 감소, 보폭의 감소, 또는 운동적 활동 범위의 감소를 포함한다. 연령관련 운동기능 장애는 연령관련 운동 기능 장애 시발점(onset)의 지연 또는 연령관련 운동기능 장애의 심각성의 감소일 수 있다. 일 실시예에서, 연령관련 운동 기능 장애 시발점은 인간에 있어서 최소한 5년, 10년, 20년, 30년, 40년, 또는 50년이 지연된다. 일 실시예에서, 연령관련 운동 기능 장애의 심각성의 감소는, 연령과 일치된 비처리 대조군의 운동기능 장애와 비교하여, 운동적 활동 및/또는 운동적 활동의 범위를 최소한 50%-100% (각 범위 포함) 향상시킬 수 있다.

제형(Dosage Form)
본 발명에 따른 방법을 실시하기 위하여, 상기 기재된 고분자 화합물 또는 이들의 구성 단량체를 하나 또는 그 이상 함유하는 조성물은 비경구, 경구(예, p.o.), 비강, 직장, 국소, 구강(buccally)을 통해 투여할 수 있다. 이하 사용된 “비경구(parenteral)”라는 용어는 피하지방 (subcutaneous), 피내 주사(intracutaneous), 정맥(intravenous), 근육내(intramuscular), 관절내(intraarticular), 동맥내(intraarterial), 활액내(intrasynovial), 흉골내(intrasternal), 척수관(intrathecal), 병변내(intralesional) 또는 두 개내(intracranial) 주사 뿐 아니라 기타 여하한 적절한 인퓨전(infusion) 기술을 이용한 주사를 의미한다.
멸균 주사용 조성물은 무독성의 비경구로 허용 가능한 1,3-부탄디올(1,3-butanediol)과 같은 희석제나 용매에 있는 용액 또는 현탁액(suspension)일 수 있다. 사용할 수 있는 허용 가능한 비히클 및 용제는 만니톨과 물이다. 또한, 고정유(fixed oils)는 통상적으로 용매 또는 현탁용 매질(예: 합성 모노- 또는 디글리세리드(diglycerides))로 사용된다. 올리브유 또는 캐스터 오일, 특히 이들의 폴리옥시에틸레이트화된 버전(polyoxyethylated versions)과 같은 천연 약학적으로 허용 가능한 오일과 같이, 올레익산 및 이의 글리세리드 유도체와 같은 지방산은 주사제로서 유용하다. 이들 오일 용액이나 현탁액은 장쇄 알코올 희석제나 분산제, 카르복시메틸 셀룰로오스(carboxymethyl cellulose), 또는 유사한 분산제를 포함할 수 있다. 기타 자주 사용되는 트윈(Tweens)이나 스판(Spans)과 같은 계면활성제 또는 기타 유사 유화제나 또는 약학적으로 허용 가능한 고체, 액체, 또는 기타 제제(dosage forms)의 제조에 일반적으로 사용되는 생체이용률 증가제(bioavailability enhancers)도 포뮬레이션(formulation) 목적으로 사용할 수 있다.
경구 투여용 조성물은 캡슐제, 정제, 에멀전, 및 수용성 현탁액, 분산액, 및 용액을 포함하여 여하한 경구 허용 가능한 제제를 사용할 수 있다. 정제의 경우, 흔히 사용되는 담체로는 락토스 및 옥수수 전분을 들 수 있다. 마그네슘 스테아레이트와 같은 윤활제도 또한 통상적으로 첨가된다. 캡슐제로 경구 투여의 경우, 유용한 희석제로는 락토스 및 건조된 옥수수 전분을 들 수 있다. 수용성 현탁액이나 에멀전을 경구로 투여할 경우에는, 주요 성분은 오일상(oily phase)을 유화제 또는 현탁제와 혼합하여 현탁 또는 용해할 수 있다. 필요시, 특정 감미제, 풍미제, 또는 색소제(coloring agents)를 첨가할 수 있다.
비강 에어로솔이나 흡입용 조성물은 약학적 포뮬레이션 업계에 공지된 기술을 사용하여 제조할 수 있다. 예를 들면, 상기 조성물은 벤질 알코올이나 기타 적절한 보존료, 생체이용률을 증대하기 위한 흡수 촉진제, 플루오로카본, 및/또는 기타 당업계에 공지된 용해제 또는 분산제를 사용하여, 식염수 용액으로 제조할 수 있다. 하나 또는 그 이상의 주요 성분을 함유하는 조성물은 직장 투여(rectal administration)용 좌제(suppositories)로 투여할 수 있다.
본 발명은 50 내지 1,500 mg/kg/일 의 용량으로 투여할 수 있다. 일 실시예에서, 본 발명의 조성물은 250 내지 1,000 mg/kg/일의 용량으로 투여할 수 있다.
보폭 증가를 위하여 바람직한 투여 용량은 50 내지 1000 mg/kg/일이다.
간염세포 팽윤의 감소를 위한 바람직한 투여 용량은 100 내지 1000 mg/kg/일이다.
상기 약학적 조성물에 포함되는 담체는 상기 조성물의 주요 성분과 양립할 수 있는(compatible)(바람직하기로는, 상기 주요 성분을 안정화시킬 수 있는) “허용 가능한(acceptable)”것이어야 하며, 치료 대상에 해롭지 않아야 한다. 하나 또는 그 이상의 용해제를 주요 화합물의 전달을 위한 약학적 부형제로 사용할 수 있다. 기타 담체로는 콜로이드 실리콘 산화물(colloidal silicon oxide), 마그네슘 스테아레이드(magnesium stearate), 셀룰로오스(cellulose), 소디움 라우릴 설페이트(sodium lauryl sulfate) 및 D&C Yellow # 10 을 들 수 있다.
이와 다르게, 본 발명의 조성물은 영양소, 의약 식품, 건강 식품 또는 보조제로서 음식을 통해 투여할 수 있다.
화합물은 생체 내 또는 실험실 내에서 시험할 수 있다. 예를 들면, 본 발명의 화합물은 예비적으로 실험실 내 검사를 통하여 산화적 스트레스와 관련된 생물학적 활성을 테스트함으로써 스크린 할 수 있다. 상기 예비적 스크린에서 높은 약효를 보이는 화합물은 당업계에 널리 알려진 생체 내 방법을 통해 추가 평가하여, 이들 화합물이 노화와 관련된 유전자인 SHC-1/p66의 발현이나 전사(transcription)에 대한 감소 또는 억제 활성을 평가할 수 있다.

실시예들
실시예 1: Production and Characterization of BEL-X
프로안토시아니딘 함유 모시풀(Boehmeria nivea (L.) Gaud) 추출액의 제조
제법 1
모시풀의 뿌리 및 줄기를 세척 후 자연환경에서 건조하였다. 건조된 모시풀을 5 mm 두께의 절편으로 자른 후 4 °C에 저장하였다. 상기 저장된 모시풀을 분쇄기(grinder)로 분쇄 후 20메쉬로 스크린하였다. 스크린된 파우더에 95% 에탄올(1:10, w/v)을 첨가한 후, 가열 후 2시간 환류하고(2회 반복), 실온으로 냉각시켰다. 상기 가열 후 실온으로 냉각시킨 추출액을 원심분리 백(centrifuge bag)에 넣고 원심분리에 의해 여과하였다. 여과된 용액을 40 °C 미만에서 진공 건조기(vacuum evaporator)를 사용하여 농축한 후 동결건조제를 사용하여 동결건조하였다. 동결건조된 추출액은 프로안토시아니딘 성분을 함유한 약학적 조성물임이 밝혀졌다.

제법 1에서 4°C에 저장한 모시풀을 분쇄기로 분쇄 후 20 메쉬로 스크린하였다. 상기 스크린된 파우더(20메쉬 미만)를 취하여 알오 물(reverse osmosis wate; RO water, 1:10, w/v)에 첨가한 후, 가열 후 2시간 환류하고(2회 반복), 실온으로 냉각시켰다. 상기 가열 후 실온으로 냉각시킨 추출액을 에탄올 수용액(95-50%)에 넣은 후 혼합하였다. 상기 추출액을 냉각 및 침전시킨 후, 상층액을 원심분리 백에 넣고 원심분리에 의해 여과하였다. 여과된 용액을 40°C 미만에서 진공 건조기(vacuum evaporator)를 사용하여 농축한 후 동결건조제를 사용하여 동결건조하였다. 동결건조된 추출액은 프로안토시아니딘 성분을 함유한 약학적 조성물임이 밝혀졌다.
모시풀 추출액의 정제
제법 1
용매추출-1
프로안토시아니딘을 함유한 모시풀 추출액을 헥산(1:10 w/v)에 넣고 가열 후 6시간 환류하여 추출액 내에 있는 지질을 제거하였다. 상기 고체 추출액을 70% 메탄올 수용액 및/또는 0.3% 비타민 C 용액에 용해시킨 후 40°C 미만에서 진공 건조기(vacuum evaporator)를 사용하여 농축하여 용매를 제거하였다. 그 후, 상기 추출액을 클로로포름(추출액: 클로로포름=1:1, v/v)에 넣고 30분간 볼텍스(vortex) 하였다(복수 추출(multiple extractions)함). 상기로부터 수득한 수층(water layer)을 에틸 아세테이트(추출액: 에틸 아세테이트 =1:1, v/v)에 넣고 30분간 볼텍스(vortex) 하였다(복수 추출(multiple extractions)함). 상기로부터 수득한 수층을 40°C 미만에서 진공 건조기(vacuum evaporator)를 사용하여 농축 후 동결건조제를 사용하여 동결건조하였다.
제법 2
용매추출-2
프로안토시아니딘을 함유한 모시풀 추출액을 물/에탄올 용액에서 용해시킨 후 40°C 미만에서 진공 건조기(vacuum evaporator)를 사용하여 에탄올을 제거하고, 헥산(1:10 w/v)에 넣고 30분간 볼텍스(vortex)하여(복수 추출(multiple extractions)함) 추출액 내에 있는 지질을 제거하였다. 상기로부터 수득한 수층(water layer)을 에틸 아세테이트(수층: 에틸 아세테이트 = 1:1, v/v)에 넣고 30분간 볼텍스(vortex) 하였다(복수 추출(multiple extractions)함). 상기로부터 수득한 수층(water layer)을 1-부탄올(1-butanol)(1:1, v/v)에 넣고 30분간 볼텍스(vortex) (복수 추출(multiple extractions)함)하였다. 상기로부터 수득한 수층(water layer)을 진공 건조기(vacuum evaporator)를 사용하여 40°C 미만에서 농축 후 동결건조제를 사용하여 동결건조하였다.
제법 3
겔 침투 크로마토그라피(Gel Permeation Chromatography)
부분 정제된 프로안토시아니딘을 함유한 제법 1의 모시풀을 용출액(elute)에 대한 상이한 극성을 갖는 용액들을 이용하여 겔 침투 크로마토그라피(gel permeation chromatography) (4 cm 직경 x 45 cm 길이 Sephadex LH-20)를 통하여 분리하고 그 안에 있는 불순물을 제거하였다. 상기 부분 정제된 물질 2.5 g을 0.5 ml의 95% ethanol에 용해한 후 겔 침투 크로마토그라피 칼럼에 넣고 여러 일련의 용매를 사용하여 지속적으로 용출하였다. 상기 서로 다른 용매에 의해 용출된 용액들을 회수하였다. 상기 용매들은 각각 300 ml의 95% 에탄올, 300 ml의 95% 에탄올 /methanol (1/1. v/v), 300 ml의 메탄올, 300 ml의 50% 메탄올 수용액 및 300 ml의 50% 아세톤 수용액, 300 ml의 아세톤이었다. 300 ml의 95% 에탄올로 용출한 용출액을 제외하고, 기타 다른 모든 용출액은 진공 건조기(vacuum evaporator)를 사용하여 40°C 미만에서 농축 후 동결건조제를 사용하여 동결건조하였다. 동결건조된 물질은 즉시 사용할 수 있도록 -20 °C에 저장하였다. 상기 부분 정제 및/또는 정제된 프로안토시아니딘을 함유한 동결건조된 모시풀 추출액의 물리적 및 화학적 특성을 분석하였다. 상기 동결건조된 용출된 물질은 부분 정제된 및/또는 정제된 프로안토시아니딘 성분을 함유하였다.
13C 및 1H 핵자기 공명 분광법(Nuclear Magnetic Resonance Spectroscopy)
정제된 프로안토시아니딘 샘플을 13C 핵자기 공명 분광법 및 1H 핵자기 공명 분광법을 통하여 탐지하였다. 13C 핵자기 공명 분광법 결과는 도 6a 내지-6c에 나타내었고, 145.2-145.7 ppm에서 오직 단일 더블렛-더블렛(doublet-doublet)피크를 보였고 다른 피크는 없었다. 따라서, 상기 단량체는 시아니딘(cyanidin)을 갖고 있지만 델피니딘(delphinidin)은 갖고 있지 않음을 보였다, 즉, B 고리는 3개의 OH기를 가졌고. 이는 EGA/MS 분석결과와 동일하였다. 도 6에서, R1=H 또는 OH 이고 R2=H, OH 또는 OCH 이다.
13C 핵자기 공명 분광법 및 1H 핵자기 공명 분광법에 의하면, 도 7a 및 7b에 나타난 바와 같이, 프로안토시아니딘의 인접 단량체 사이의 결합은 주로 C4, C8 탄소-탄소 결합, C4, C6 탄소-탄소 결합, 및 C2, C7 탄소-탄소 결합 단위에서 발생하였다.
실시예 2: BEL-X의 효과
동물 군집(Animal population): 수컷 및 암컷 C57BL/6 계통의 생쥐를 사용하였으며, 총 4개 그룹으로 분류하였다: (1) 수컷 비처리군 (대조군), (2) 9-20 개월령 수컷 생쥐, BEL-X 처리군, (3) 암컷 비처리군 (대조군), 및 (4) 9-20 개월령 암컷 생쥐, BEL-X 처리군.
조성물의 제조 및 투여용량(Preparation and Dosage of Composition): 제법 3의 BEL-X을 증류수에 용해 후 매일 투여용 바늘(feeding needle)을 사용하여 1000 mg/kg/일의 양을 매일 p.o.로 생쥐에 투여하였다.
20개월령에 생쥐를 희생시킨 후 간조직 및 혈청을 회수한 후 병리학적 및 생물학적 분석을 하였다.
A. 노화된(aged) 간에 대한 Bel-X의 효과
생쥐를 희생시키는 시점에서 생쥐의 체중 및 간의 무게를 측정하였다. 간을 회수 후 헤마톡실린 및 에오신 염색(H & E staining)을 하였다. 간세포 팽윤은 고정 및 염색된 간 단편(sections)에 대하여 현미경 평가 및 병리조직학적 평가를 하였다.
결과는 도 1A, 1B 및 1C에 나타내었다. 어린 생쥐(2개월령)의 간과 노령(20개월령)의 간을 비교한 결과, 노령 간은 간세포 팽윤을 나타내었다(도 1A 내지 1B 비교 요망). 어린 생쥐의 간의 간세포 팽윤을 Bel-X로 처리한 노령 간과 비교한 결과, 처리된 군은 최소한의 간세포 팽윤 또는 간세포 팽윤이 전혀 없었고, 이는 어린 생쥐의 간과 유사하였다 (도 1C). 이러한 결과는 표1을 통하여 더욱 자세히 나타내었다.

구체적으로, 수컷 대조군(비처리) 생쥐는, 12마리를 평가하였는데, 그 중 9마리는 간세포 팽윤 및 퇴화(degeneration)를 보였다. 따라서, "간 비정상 비율(liver abnormal ratio)"은 "평가된 총 생쥐 수/간세포 팽윤을 나타내는 생쥐 수"를 백분율로 나타낸 것이다. 비정상 비율이 높을수록 간세포 팽윤율이 더 높았다.
표1은 BEL-X 처리가 수컷 및 암컷 생쥐 군집에서 모두 간세포 팽윤의 발생을 50 내지 75 %나 유의성 있게 감소됨을 보여준다.
B. RT qPCR에 의한 SHC-1/p66의 탐지
실시간 RT- PCR : 토탈 RNA 추출은 Trizol RNA 분리 프로토콜에 기재된 바와 같이 냉동 생쥐 간을 이용하여 수행한다. cDNA 합성에는 랜덤 프라이머들과 SuperScript II 키트를 사용한다. 목적 유전자 SHC1 아형(isoform) p66 염분제거(salt-free) 프라이머 정방향 프라이머, 5'-CGGAATGAGTCTCTGTCATCGCTGGA (서열 번호: 1); 역방향 프라이머 5'-CGCCGCCTCCACTCAGCTTGTT (서열 번호: 2) 및 내부 조절 보조관리 유전자(internal control house-keeping gene) GAPDH 정방향 프라이머, 5'--GAAGGTGAAGGTCGGAGT (서열 번호: 3), 역방향 프라이머 5'- GAAGATGGTGATGGGATTTC (서열 번호: 4)를 제조하였다. 다양한 간조직에서 목적 유전자의 발현을 탐지하기 위해, 1μL의 토탈 cDNA를 유리 모세관(glass capillaries)에 있는 9 ml 의 실시간 PCR 마스터믹스(mastermix) (Roche Molecular Biochemicals)에 첨가하였다. 4단계 실험 프로토콜을 사용하였다: (i) 변성 프로그램(95°C, 20초); (ii) SHC-1/p66에 대한 증폭 및 정량 프로그램을 60회 반복 수행하거나 GAPDH는 40회 반복 수행(SHC-1/p66는 95°C, 20초; 62°C, 20초 또는 GAPDH 는 60°C, 20초; SHC-1/p66 및 GAPDH는 72°C, 20초; 82°C, 20초로 단일 형광 측정); (iii) 용융 곡선 프로그램(melting curve program) (60-95°C 에서는 초당 0.1°C 의 가열률(heating rate) 및 지속적 형광 측정); (iv) 40°C 까지의 냉각 프로그램.
RT-PCR 생성물은 LightCycler Software 3.5 (Roche Molecular Biochemicals)을 사용하여 CT 값으로 계산한다. GADPH 유전자는 참고용으로 사용한다. SHC-1/p66의 상대적 발현 수준은 비교 CT법(Schmittgen, T. D. & K.J. Livak. (2008) "Analyzing real-time PCR data by the comparative CT method." Nature Protocol. 3: 1101-1108.)을 사용하여 분석한다.
결과: 도 2에서 보는 것과 같이, BEL-X 처리하지 않은 생쥐의 SHC-1/p66 발현과 비교할 때, BEL-X는 SHC-1/p66의 발현을 유의성 있게 감소시킨다. p66-결핍 생쥐 및 세포는 활성산소종(ROS)의 수준을 감소시켰고 산화적 스트레스에 대한 저항력을 높였으므로, SHC-1/p66 발현의 감소는 또한 활성산소종(ROS)을 저하시키고 산화적 스트레스도 감소시킨다.
C. 노화 BEL-X가 SHC-1/p66 단백질에 미치는 영향
웨스턴 블로팅 (Western blotting): 간조직에 10x 부피의 RIPA를 첨가한 후 균질화하고 조직 찌꺼기를 원심분리하여 제거한 후, 얻어진 용액은 SDS-PAGE로 단백질을 분리하기 위하여 사용하였다. SDS-PAGE 상의 단백질을 PVDF 멤브레인(Millipore)으로 옮긴 후 특정 항-SHC-1/p66 및 GAPDH 항체와 더불어 각각 배양하였다. 특정 단백질 발현은 UVP Biospectrum을 사용하여 탐지 및 분석하였다.
결과: 도 3은 노령의 생쥐의 SHC-1/p66가 어린 생쥐의 것에 비해 단백질 농도가 거의 2배나 더 높은 것을 보여준다. 그러나, BEL-X로 처리한 노령의 생쥐는 간에서 SHC-1/p66 단백질이 유의성 있게 낮았다(단백질 농도가 단지 약 64% 증가됨).
실시예 3: 운동 기능 평가(Motor Skills Evaluation)
동물: 수컷 및 암컷 C57BL/6 계통 생쥐를 사용하였으며, 총 4개 그룹으로 분류하였다: (1) 수컷 비처리군 (대조군), (2) 12-20 개월령 수컷 생쥐로 BEL-X 처리군, (3) 암컷 비처리군 (대조군), 및 (4) 12-20 개월령 암컷 생쥐로 BEL-X 처리군. BEL-X을 증류수에 용해 후 매일 투여용 바늘(feeding needle)을 사용하여 250 mg/kg/일의 양을 매일 p.o.로 생쥐에 투여하였다.
운동 기능 시험: 보폭 패턴에 있어서 연령-의존성 변화를 노화 중의 인간의 걸음걸이 패턴과 비교되어 왔다(Wolfson L, Whipple R, Amerman P, Tobin JN. (1990) Gait assessment in the elderly: A gait abnormality rating scale and its relation to falls. J Gerontol. 45:M12-19.). 따라서, 보정된 보폭 시험 방법을 사용하여(Klapdor, K. et al., (1997) A low-cost method to analyze footprint patterns. Journal of Neuroscience methods. 75:49-54; Hilber, P. and J. Caston. (2001) Motor skills and motor learning in lurcher mutant mice during aging. Neuroscience. 102:615-623.) 다양한 연령의 생쥐의 보폭을 측정한다. 생쥐의 발은 음식 색깔로 염색되고, 발자국은 40x10 cm 종이로 평가한다. 보폭길이는 각 생쥐에 대하여 최소한 3번 분석한다.
결과: 도 2에 보이는 바와 같이, 노령의 생쥐는 수컷과 암컷 모두에서 어린 생쥐보다 보폭이 유의성 있게 짧았다. 그러나, Bel-X로 처리한 노령의 생쥐는 어린 생쥐와 보폭이 거의 동일하였다. 따라서, 250 mg/kg/일의 Bel-X 처리는 노령의 대상(subject)이 보폭 길이를 유지하는데 도움이 되었다.

생쥐의 수명을 사람의 수명과 비교해 보면, 생쥐가 인간보다 훨씬 빨리 노화된다는 것은 명백하다. 잭슨 실험실(The Jackson Laboratory)의 보고에 따르면, 3 내지 6 개월령 생쥐는 20 내지 30세의 사람과 거의 동등하며, 16 내지 24 개월령 생쥐는 56내지 69세의 사람과 거의 비슷하다. 따라서, Bel-X 처리를 하면 56내지 69세의 사람에게 20 내지 30세의 사람의 보폭을 제공하는 것으로 볼 수 있다.
실시예
4:
활성산소종
(
ROS
)
생성에 대한 치료의 효과
세포 배양: 인간 간암세포 (Huh7)는 MEM에서 유지하고, 인간 직장 선암종 세포(adenocarcinoma cells)(HRT-18) 및 인간 피부 섬유아세포(fibroblastoid cells)(WSI)는 DMEM에서 유지한다. 배양 배지(medium)는 1% 페니실린/스트렙토마이신 혼합물과 1% 비필수 아미노산, 1% GlutaMAX-I, 1 mM 피루빈산 나트륨(sodium pyruvate), 및 10% 소태아혈청(fetal bovine serum)을 첨가한다. 상기 세포는 37 -C, 5% CO2 인큐베이터에서 배양한다.
활성산소종 ( ROS ) 유도: 세포는 24-웰 플레이트에 분주하고 24시간 배양한 후 800 mM H2O2를 배양 배지에 1시간에 걸쳐 첨가하여 상기 세포에서 활성산소종(ROS)을 유도한다.
BEL -X 처리: 약물 BEL-X의 효과를 시험하기 위하여 상기 세포를 과산화수소(H2O2)로 유도 후 BEL-X로 24시간 동안 처리하였다.
활성산소종 ( ROS ) 탐지: 10 mM CM-H2DCFDA를 상기 세포에 첨가한 후 상기 세포를 37 -C에서 45분간 배양하였다. 형광 프로브(fluorescent probe) 인큐베이션 후, 상기 세포를 PBS로 두 번 세척하고, 활성산소종(ROS) 생성을 형광현미경으로 직접 관찰하거나 또는 용해된 세포 및 분광광도계(spectrophotometer)를 사용하여 형광 강도로써 활성산소종(ROS) 생성을 측정한다.
결과: 도 4B 내지 4D에서 보는 바와 같이, 활성산소종(ROS) 생성을 유도하기 위하여 인간 간암세포 Huh-7 세포를 과산화수소로 유도한 경우 다량의 활성산소종(ROS) 생성을 보여준다(도 4B의 녹색 형광). 그러나, 활성산소종(ROS) 생성을 유도된 세포 중 Bel-X로 처리된 세포는 활성산소종(ROS) 생성이 아주 적음을 보여준다(도 4D의 녹색 형광).
더욱이, 도 5에서 보는 바와 같이, Bel-X는 과산화수소에 의한 활성산소종(ROS) 생성을 세 가지 세포 유형(인간 간암세포 Huh7, 인간 직장암 세포 HRT-18 및 인간 정상 피부 세포 WS1)에서 모두 유의성 있게 감소시켰다. 따라서, 이들 실험결과를 이전의 평가결과와 비교해 볼 때, Bel-X는 SHC-1에 효과가 있고 이는 다시 산화적 스트레스 경로에 영향을 미치게 된다. 따라서, Bel-X는 간, 직장 및 피부 세포 등 다양한 세포에서 산화적 스트레스를 감소시킨다.
