Abstract
본 발명은 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 유효성분으로 함유하는 전립선암 예방 또는 치료용 조성물에 관한 것이다.
본 발명에 따른 조성물은 두 유효성분의 병용에 따라 전립선암 세포 사멸에 현저한 시너지 효과를 가져 전립선암의 예방 또는 치료에 효과적이다.

Description
본 발명은 전립선 암의 예방 또는 치료용 조성물에 관한 것이다.
전립선암은 남성의 질병율 및 사망율의 주된 요인이다. 전립선암은 폐암 다음으로 미국 남성의 암 사망 요인 2위를 차지하고 있으며, 우리나라에서도 최근 발생이 급격히 증가하여 현재 성인 암 중 발생 증가율 1위이다. 전립선암 발병율은 1980년대 후반에 급격하게 증가하였는데, 평균 수명의 증가 및 식생활 양식의 서구화 등으로 발병률이 증가하고 있는 추세이며, 상기 암으로 인한 사망률 역시 빠르게 증가하고 있다. 또한, 전립선 암은 수술 후 재발율이 매우 높은 암으로, 암의 완치에 큰 어려움을 겪고 있다.
현재 활발히 수행되고 있는 전립선 암의 치료 방법으로는 외부 방사선 치료와 안드로젠 차단술 등이 있다. 그러나, 두 치료법 모두 심혈관 질환의 위험성 증가, 골다공증, 체중 증가, 뇌인식 저하, 요도협착발병, 성욕 상실 및 불임, 골다공증과 관련한 골격 칼슘염의 감소 위험성 및 병적 골절의 위험성의 현저한 증가 등의 부작용이 발생하는 문제점이 인지되고 있다. 이러한 위험들을 고려할 때 안드로젠 차단 요법과 외부 방사선 요법의 치료적 가치가 의문시되고 있으며, 새로운 치료 요법에 관한 연구 개발이 요구되고 있다.
또한, 전립선암의 주어진 병태를 치료하기 위하여 두 약물 이상의 투여가 고려될 수 있으나, 다수의 잠재적인 문제를 야기할 수 있다. 두 약물 간의 생체 내 상호작용은 복잡하며, 두 약물이 신체 내로 도입되었을 때 각 약물은 다른 쪽 약물의 흡수, 분포 및 제거에 영향을 미칠 수 있고, 이에 따라 다른 쪽 약물의 효과를 변화시킬 가능성이 있다. 예컨대, 하나의 약물은 다른 쪽 약물 제거의 대사 경로에 관여된 효소의 생산을 저해, 활성화 등을 유도할 가능성이 있다. 따라서 두 약물이 동일 병태를 치료하기 위하여 투여될 경우 각각이 인간 대상체에서 다른 쪽 약물의 치료 활성을 보완할 수 있는 약물을 선택할 필요성이 있다. 그러나, 이러한 약물의 병용 투여를 위한 조합, 특히 특정 약물에 대한 내성을 가지는 암종의 내성을 제어하여 약물의 효과를 달성하기 위한 병용 투여 약물에 대한 연구개발이 매우 부족한 실정이다.
이러한 배경 하에 전립선암의 치료를 위한 새로운 치료 방법들에 대한 연구개발이 활발히 진행되고 있으며, 특히 약물의 병용 치료 요법과 관련된 연구 개발의 필요성이 절실히 대두되고 있다.
본 발명은 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 유효성분으로 포함하는 전립선암 예방 또는 치료용 약제학적 조성물을 제공하고자 한다.
또한, 본 발명은 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 유효성분으로 포함하는 전립선암 예방 또는 개선용 건강기능식품을 제공하고자 한다.
본 발명자들은 전립선 암 치료에 적합한 병용 치료제를 개발하기 위하여 연구를 진행하던 중 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)의 병용 투여가 전립선암의 예방 및 치료에 현저한 효과가 있음을 확인하고 본 발명을 완성하였다.
상기 목적을 달성하기 위하여, 본원 발명은 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 유효성분으로 포함하는 전립선암 예방 또는 치료용 약제학적 조성물을 제공한다. 본원 발명의 조성물은 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)의 병용에 의한 시너지 효과로 비티신 A (Vitisin A) 또는 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand) 각각의 처리보다 전립선암의 예방 또는 치료에 현저한 효과를 보이며, 특히 전립선 암세포 사멸에 우수한 효과를 보인다.
본원 발명에 있어서, 비티신 A (Vitisin A)란 하기 화학식 1의 구조를 가지는 화합물 또는 이의 이성질체이며, 보다 바람직하게 하기 화학식 2의 화합물이다.

비티신 A (Vitisin A)는 포도속 식물에서 분리 및 추출 가능한 스틸베노이드 화합물로 두 개의 레스베라트롤 dimer가 결합된 레스베라트롤 사합체(resveratrol tetramer)이다. 비티신 A (Vitisin A)는 당업계에 알려진 방법에 따라 포도속 식물에서 분리 및 추출하여 사용하거나, 합성하여 사용할 수 있다.
TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)은 Apoptosis를 유도하는 리간드의 기능을 하는 단백질이다. 종양 세포에서 Death receptor에 결합하여 Apoptisis를 유발하고 항암제로써 기능을 수행할 수 있다.
본 발명에 있어서, 전립선암이란 전립선에 발생한 암을 의미한다. 예컨대, 전립선의 샘 세포에서 발생하는 암인 전립선 샘 암종을 포함하며, 다른 종류의 암으로 전립선 육종, 전립선 소세포 암종 및 전립선 이행세포 암종을 포함한다.
특히, 본원 발명은 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)에 대해 내성을 가진 전립선 암에 대해 현저한 효과를 보인다. TRAIL의 반복 투여 등을 통해 전립선 암에 내성이 생기는 경우, TRAIL 단독 처리에 의하면 항암 치료의 효과를 가질 수 없는 반면, 본원 발명의 비티신 A (Vitisin A) 및 TRAIL 병용 처리는 비티신과 TRAIL의 시너지 효과에 의해 TRAIL에 대해 내성을 가진 전립선 암에 대해서 현저한 예방 및 치료 효과를 보인다.
본원 발명의 약제학적 조성물은 pro-apoptotic 단백질을 활성화한다. 예컨대, PARP, pro caspase 7 및 pro caspase 8 등의 pro-apoptotic 단백질들은 비티신 A 및 TRAIL 병용 처리에 의해 현저히 발현이 감소하며, Cleaved PARP, Cleaved caspase 3, 및 Cleaved caspase 7 등의 발현을 현저히 증가시킨다.
본원 발명의 약제학적 조성물은 Survival 단백질의 발현을 억제한다. 예컨대 bcl-2 및 bcl-xl의 발현은 비티신 A 및 TRAIL 병용 처리에 따라 크게 감소한다.
본원 발명의 약제학적 조성물은 Cell death 단백질들의 발현을 증가시킨다. 예컨대, 비티신 A 및 TRAIL 병용 처리에 따라 DR (Death Receptor 5)5의 발현량이 크게 증가하며, DcR1는 발현량이 감소한다.
본원 발명의 약제학적 조성물은 전립선 암세포에서 활성 산소종의 생성을 촉진하여 세포 사멸을 유도하며, 또한, Death Receptor 5(DR5)의 발현을 촉진하여 세포 사멸을 빠르게 유도한다.
본 발명의 조성물은 상기 유효성분을 조성물 총 중량에 대하여 비티신 A를 0.01 ~ 50 중량 % 및 TRAIL을 0.01 ~ 50 중량 % 함유할 수 있다. 또한, 비티신A 및 TRAIL 개별 투여량의 1, 2, 3 또는 4배 이상을 함유하거나 1/2, 1/3, 1/4 배를 함유할 수 있다.
본 발명의 약제학적 조성물은 약제학적으로 허용 가능한 첨가제를 추가적으로 포함할 수 있으며, 이때 약제학적으로 허용 가능한 첨가제로는 전분, 젤라틴화 전분, 미결정셀룰로오스, 유당, 포비돈, 콜로이달실리콘디옥사이드, 인산수소칼슘, 락토스, 만니톨, 엿, 아라비아고무, 전호화전분, 옥수수전분, 분말셀룰로오스, 히드록시프로필셀룰로오스, 오파드라이, 전분글리콜산나트륨, 카르나우바 납, 합성규산알루미늄, 스테아린산, 스테아린산마그네슘, 스테아린산알루미늄, 스테아린산칼슘, 백당, 덱스트로스, 소르비톨 및 탈크 등이 사용될 수 있다.
본 발명에 따른 약제학적으로 허용 가능한 첨가제는 약제학적 조성물 100중량부에 대해 0.1 ~ 90중량부 포함되는 것이 바람직하나 이에 한정되는 것은 아니다.
즉, 본 발명의 약제학적 조성물은 실제 임상 투여 시에 경구 및 비경구의 여러 가지 제형으로 투여될 수 있는데, 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 조성물에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(Calciumcarbonate), 수크로스(Sucrose), 락토오스(Lactose) 또는 젤라틴 등을 섞어 조제될 수 있다. 또한 단순한 부형제 이외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용될 수 있다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제 및 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함될 수 있다. 비수성용제, 현탁용제로는 프로필렌글리콜(Propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 약제학적 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여할 수 있으며, 비경구 투여시 피부 외용 또는 복강내주사, 직장내주사, 피하주사, 정맥주사, 근육 내 주사 또는 흉부 내 주사 주입방식을 선택하는 것이 바람직하다. 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설율 및 질환의 중증도 등에 따라 그 범위가 다양하다.
본 발명의 조성물의 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설율 및 질환의 중증도에 따라 그 범위가 다양하며, 일일 투여량은 비티신 A는 1~ 10000 ㎍/kg(체중) 이고, 바람직하게는 10~1000 ㎍/kg이며, TRAIL은 1~ 10000 ㎍/kg(체중) 이고, 바람직하게는 10~1000 ㎍/kg이다. 투여의 경우 하루 1 ~ 6 회 투여될 수 있다.
아울러, 본 발명은 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 유효성분으로 포함하는 전립선암 예방 또는 개선용 건강기능식품을 제공한다.
본 발명의 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다.
상기 식품의 종류에는 특별한 제한은 없다. 상기 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 첨가할 수 있는 식품의 예로는 각종 스프, 음료수, 차, 드링크제, 알코올음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강식품을 모두 포함한다.
상기 외에 본 발명의 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 건강기능식품은 천연 과일주스, 과일주스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 혼합하여 사용할 수 있다.
본 발명에 따른 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 유효성분으로 포함하는 조성물은 두 유효성분의 병용에 따라 전립선암 세포 사멸에 현저한 시너지 효과를 가져 전립선암의 예방 또는 치료에 효과적이다.
도 1은 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand) 병용 처리에 따른 전립선 암세포 사멸 효과를 확인한 결과이다.
도 2는 PC-3 세포에서 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand) 병용 투여에 따른 Sub-G1기 population 및 apopotosis 변화를 확인한 결과이다.
도 3은 DU 145 세포에서 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand) 병용 투여에 따른 Sub-G1기 population 변화를 확인한 결과이다.
도 4는 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand) 병용 투여에 따른 pro-apoptotic 단백질 및 survival 단백질의 발현 변화를 확인한 결과이다.

본 발명의 이점 및 특징, 그리고 그것들을 달성하는 방법은 상세하게 후술되어있는 실시예들을 참조하면 명확해질 것이다. 그러나 본 발명은 이하에서 개시되는 실시예들에 한정되는 것이 아니라 서로 다른 다양한 형태로 구현될 것이며, 단지 본 실시예들은 본 발명의 개시가 완전하도록 하고, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자에게 발명의 범주를 완전하게 알려주기 위해 제공되는 것이며, 본 발명은 청구항의 범주에 의해 정의될 뿐이다.
<실시예 1> 전립선암 세포의 배양 및 세포 생존율 확인
전립선암 세포인 PC3과 DU145 세포를 ATCC (American type culture collection:rockville, MD)에서 구입하여 열-비활성화륜, 2mM 글루타민, 100U/mL 페니실린 및 100μg/mL 스트렙토마이신이 첨가된 RPMI 1640배지에서 5% CO2에서 배양하였다.
세포 생존율 확인을 위하여, DU145 및 PC3 세포를 70 %로 배양하고, 비티신 A (Vitisin A) (출처 : 한국 화학연구원)와 TRAIL (출처 : Sigma)의 단독 또는 결합으로 처리하여 24 시간동안 추가 배양을 수행하고, MTT 분석법을 수행하여 세포 생존율을 측정하였다. 그 결과를 도 1에 나타내었다.
도 1에 나타낸 바와 같이, 비티신 A(Vitisin A) 또는 TRAIL 단독 처리시 PC3 세포 및 DU145 세포에서 세포 생존율에 큰 영향을 미치지 못하였으나, 비티신 A(Vitisin A) 및 TRAIL 병용 처리시에 세포 생존율을 크게 감소시켰다. 이는 두 약물의 병용 투여에 의한 전립선 암 예방 또는 치료에 대한 시너지 효과를 보여주는 것이다.
<실시예 2> 유세포 분석을 통한 Sub-G1기 세포 확인
Apoptosis가 일어남을 확인하기 위하여 유세포 분석을 통해 Sub-G1기의 세포를 확인하였다.
DU145 및 PC3세포를 배양 후, PBS로 한 번 세척한 다음 차가운 70% 에탄올로 고정한 후 4℃로 보관하였다. 100 μg/mL RNase A, 50 μg/mL PI, 0.1% (w/v) 시트르산 소듐 및 0.1% (v/v) NP-40를 함유한 차가운 PI 용액에 현탁시킨 후 30분 동안 추가로 배양하였다. Flow cytometer: FACS를 이용하여 세포의 sub-G1를 분석하였고, 셀퀘스트 프로그램 (CellQuest software)를 이용하여 상대 DNA 함량을 관찰하였다. 그 결과를 도 2 및 도 3에 나타내었다.
도 2는 PC3 세포의 유세포 분석 결과를 나타낸다. 도 2의 (A)는 Histogram을 나타내며, 도 2의 (B)는 세포 주기 분석에 의한 PC-3 전립선 암세포에서 sub-G1기 population에 대한 비티신 A 및 TRAIL의 효과를 나타내는 그래프이며, 도 2의 (C)는 Apoptosis가 일어난 세포 확인을 위한 유세포 분석 결과를 나타내며, 도 2의 (D)는 Apoptosis 유발에 대한 비티신 A 및 TRAIL의 시너지 효과를 보여주는 그래프이다. Untreated는 약물 처리가 없는 것을 의미하며, V4 또는 V8은 각각 Vitisin A 4 또는 8 uM 처리, T20 또는 T40은 각각 TRAIL 20 ng/ml 또는 40 ng/ml 처리를 말하며, V4T20, V8T20, V4T40은 각각 해당 농도의 병용 처리를 의미한다.
Histogram을 정리한 도 2의 (B)에서 확인되는 바와 같이, Vitisin A 및 TRAIL의 병용 처리는 이들 각각을 처리하였을 때와 대비하여 Sub-G1 population을 현저하게 증가시킴을 확인하였다. 또한, 도 2의 (D)에서 확인되는 바와 같이, Vitisin A 및 TRAIL의 병용 처리시에 Apoptosis가 현저히 증가함을 확인하였다.
도 3은 DU146 세포의 유세포 분석 결과를 나타낸다. 도 3의 (A)는 Histogram을 나타내며, 도 3의 (B)는 세포 주기 분석에 의한 DU145 전립선 암세포에서 sub-G1기 population에 대한 비티신 A 및 TRAIL의 효과를 나타내는 그래프이다. Histogram을 정리한 도 3의 (B)에서 확인되는 바와 같이, Vitisin A 및 TRAIL의 병용 처리는 이들 각각을 처리하였을 때와 대비하여 Sub-G1 population을 현저하게 증가시킴을 확인하였다.
상기 결과로부터 Vitisin A 및 TRAIL의 병용 처리에 의한 암세포 사멸의 시너지 효과를 확인하였다.
<실시예 3> 웨스턴 블랏을 통한 pro-apoptotic 및 survival 단백질의 발현 변화 확인
웨스턴 블랏을 통하여 pro-apoptotic 및 survival 단백질의 발현 변화를 확인하였다. PC3 세포에 Vitisin A 및/또는 TRAIL를 첨가하고 24 시간 배양 후 세포를 PBS로 세척하였다. 세척된 세포는 lysis 용액 (20mM 자당, 1mM EDTA, 20 μM Tris-CL, pH7.2, 1mM DTT, 10mM KCL, 1.5mM MgCL2, 5μg/mL 펩스타틴 A, 10 μg/ml 류펩틴 및 2 μg/mL 아프로티닌)에 첨가하여 용리하였다. 원심 분리 후에 상등액을 수집하여 단백질 농도를 측정한 후 30 μg의 분취액을 bio-rad 단백질 분석 키트을 이용하여 SDS-폴리아크릴아마이드 겔 상에서 분리하고 지시된 각각의 1차 항체 및 2차 항체를 처리한 후, ECL 검출 시스템으로 분석하였다. 검출 대상의 단백질은 PARP, Cleaved PARP, pro caspase 7, Cleaved caspase 7, pro caspase 8, pro caspase 3, Cleaved caspase 3, bcl-2, bcl-xl, DR5 및 DcR1이었으며, 분석결과는 도 4 및 도 5에 나타내었다.
도 4의 (A)는 pro-apoptotic 단백질들의 발현 변화를 웨스턴 블랏으로 확인한 결과이다. 도 4의 (A)에 나타낸 바와 같이, PARP, pro caspase 7 및 pro caspase 8 모두 비티신 A 및 TRAIL 병용 처리에 따라 크게 감소되는 반면, Cleaved PARP, Cleaved caspase 3, 및 Cleaved caspase 7 모두 증가되는 것이 확인되었다.
도 4의 (B)는 Survival 단백질인 bcl-2 및 bcl-xl의 발현 변화를 웨스턴 블랏으로 확인한 결과이다. 도 4의 (B)에 나타낸 바와 같이, Survival 단백질의 경우 비티신 A 및 TRAIL 병용 처리에 따라 크게 감소되는 것이 확인되었다.
도 5의 (A)는 Cell death 단백질들의 발현 변화를 웨스턴 블랏으로 확인한 결과이다. 도 5의 (A)에서 확인되는 바와 같이, 비티신 A 및 TRAIL 병용 처리에 따라 DR 5(Death Receptor 5)의 발현량이 증가하는 것이 확인되었으며, DcR1의 경우 발현량이 감소되는 것이 확인되었다.
도 5의 (B)는 비티신 A 및 TRAIL의 병용 처리에 따른 DR5 프로모터의 활성 변화를 보여주며, 병용처리에 따라 DR5 프로모터 활성이 각각을 처리한 경우와 대비하여 현저히 증가하는 것이 확인되었다.
상기 결과들을 종합하여 볼 때, 비티신 A 및 TRAIL의 병용 처리는 비티신 A 또는 TRAIL의 각각 처리와 대비하여도 현저히 Apoptosis를 활성화시키고, Survival 단백질의 발현을 감소시키며, Cell Death protein들의 발현을 크게 증가시키는 것을 보여주며, 이는 비티신 A 및 TRAIL의 병용 처리에 따른 시너지 효과를 확인시켜준다.
<실시예 4> 활성산소종 (ROS) 변화 확인
Vitisin A 및 TRAIL의 병용 처리에 따른 ROS 생성 변화 및 Cell death 단백질의 변화를 ROS 활성 측정 및 웨스턴 블랏을 통해 확인하였다.
PC3 세포에 Vitisin A 와 TRAIL를 단독 또는 공동 투여하여 24 시간 배양 후 20 μM 2070-diacetyl dichlorofluorescein(DCFDA)이 포함된 배지에서 37 ℃에서 20분간 반응시킨 후 Flow cytometer: FACS를 이용하여 활성산소종(ROS)을 측정하였다.
그 결과를 도 6에 나타내었다. 도 6의 (A)는 N-Acetyl-L-cystein(NAC)의 존재 또는 부재하에 ROS에 대한 비티신 A 및 TRAIL의 병용 처리 효과를 확인한 결과이다.
도 6의 (A)에서 확인되는 바와 같이, 비티신 A 및 TRAIL의 병용 처리는 활성산소의 발생을 각각의 개별 약물과 대비하여 크게 증가시킨 것을 확인하였다.
또한, 상기 PC3 세포는 상기 실시예 3의 방법과 동일하게 웨스턴 블랏을 통해 비티신 A 및 TRAIL 처리에 따른 Pro PARP 및 Cleaved PARP의 발현 변화를 확인되었으며, 그 결과를 도 6의 (B) 및 (C)에 나타내었다.
도 6에 나타낸 바와 같이, 비티신 A 및 TRAIL 병용 처리에 따라 Pro PARP의 발현이 현저히 감소하고 Cleaved PARP의 발현이 현저히 증가하는 것이 확인되었으나, N-Acetyl-L-cystein(NAC) 처리에 따라 활성산소 생성이 억제되어 Cleaved PARP의 발현이 억제되는 것이 확인되었다.
상기 결과로부터 본원 발명의 비티신 A 및 TRAIL 병용 처리가 전립선 암세포주에서 활성 산소 생성에 시너지 효과를 가져 전립선 암세포 사멸에 현저한 효과를 보이는 것을 확인하였다.
Claims (6)
- 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 유효성분으로 포함하는 전립선암 예방 또는 치료용 약제학적 조성물.
- 제1항에 있어서, 상기 전립선암은 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand) 내성을 가진 전립선암인 전립선암 예방 또는 치료용 약제학적 조성물.
- 제1항에 있어서, 상기 전립선 암은 전립선 샘암종, 전립선 육종, 전립선 소세포 암종 및 전립선 이행세포 암종으로부터 선택된 어느 하나 이상인 전립선암 예방 또는 치료용 약제학적 조성물.
- 제1항에 있어서, 상기 전립선암 예방 또는 치료는 전립선 암세포 사멸 촉진에 의한 것인 전립선암 예방 또는 치료용 약제학적 조성물.
- 제1항에 있어서, 상기 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)은 전립선 암세포에서 활성 산소종을 촉진하는 것인 전립선암 예방 또는 치료용 약제학적 조성물.
- 비티신 A (Vitisin A) 및 TRAIL (tumor necrosis factor-related apoptosis-inducing ligand)을 유효성분으로 포함하는 전립선암 예방 또는 개선용 건강기능식품.
