대부분의 조직에서, 세포는 콜라겐, 라미닌, 및 피브로넥틴과 같은 단백질을 포함하는 세포외 기질(ECM)에 둘러싸여 있다. 포유류가 노화할 때 및 일부 질병 상태에서, 신체의 특정 부위 내의(예를 들어, 윤활 관절, 유리체액, 척추 디스크, 피부 등 내의) 세포외 기질은 변성되어, 다양한 형태의 관절염, 실명 등과 같은 바람직하지 않은 증상을 일으킬 수 있다.

연골 변성

1: 펩티도글리칸의
합성
시약
펩티드 GAHWQFNALTVRGGGC(GAH) 및 WYRGRLGC(WYRGRL)를 진스크립트(Genscript, 뉴저지주 피스카터웨이)로부터 구입했다. N-[β-말레이미도프로피온산] 히드라지드, 트리플루오로아세트산 염(BMPH)을 피어스(Pierce, 일리노이주 록퍼드)로부터 구입했다.
방법
제I형 콜라겐 및 히알루론산 결합성 펩티도글리칸의 합성은 앞에서 기술되었다(예를 들어, 문헌 [Bernhard J.C., Panitch A. Acta Biomater 2012; 8: 1543-1550], 및 문헌 [Paderi J.E., Panitch A. Biomacromolecules 2008; 9: 2562-2566] 참조). 본원의 합성 펩티도글리칸을 이에 기술된 방법에 따라 제조하고 다음과 같이 변성했다.
황산 콘드로이틴(10 mg/mL)을 0.1 M 아세트산 나트륨 완충액 pH 5.5 내의 메타-과아이오딘산 나트륨으로 산화했다. 산화도는 메타-과아이오딘산 나트륨의 농도에 의해 제어했다. 헤테로이관능성 크로스링커 BMPH를 약 40-배 몰 과량의 BMPH를 황산 콘드로이틴에 1xPBS pH 7.2 완충액에서 실온에서 2 시간 동안 반응시킴으로써 관능화된 황산 콘드로이틴에 컨쥬게이션했다. 관능화도는 과량의 BMPH으로부터 CS-BMPH의 정제 도중 BMPH의 소비를 측정함으로써 판단했다. 컨쥬게이션의 최종 단계에서, 펩티드 WYRGRLGC 및 GAHWQFNALTVRGGGC를 물에 용해시키고 각각을 콘드로이틴 위의 BMPH 반응기의 수의 절반 몰 당량으로 첨가했다. 예를 들어, 평균 약 22 BMPH 관능기를 갖는 황산 콘드로이틴은 약 11 WYRGRLGC 펩티드 및 약 11 GAHWQFNALTVRGGGC 펩티드로 컨쥬게이션했다. 두 펩티드를 모두 함유하는 혼합물을 2 시간 동안 실온에서 반응시키고 이어서 초순수에서 정화 및 동결건조했다.
모든 중간체를 AKTA 퓨리파이어(Purifier) FPLC(GE 헬스케어) 및 폴리아크릴아미드 비즈(Bio-Rad Labs)로 패킹된 컬럼을 사용하여 크기 배제 크로마토그래피에 의해 정제했다. 최종 생성물을 세파덱스(sephadex) G-25 비즈로 패킹된 컬럼을 사용하여 유사하게 정제했다. 합성 후, 합성 펩티도글리칸은 동결건조되고 -80 ℃에서 연장된 기간 동안 보관될 수 있다.

2: 루브리신
모방
루브리신 모방 펩티도글리칸의 연골 표면 사이의 마찰력을 감소시키는 능력을 다음과 같이 시험했다. 연골 마개를 레오미터의 판에 부착시키고, 제2 연골 마개를 레오미터의 회전 기구에 부착시켰다. 두 연골 표면을 5 N 힘으로 맞대고, 이어서 전단력을 가했다. 토크를 측정하고 표면 사이의 마찰력을 계산하기 위해 사용했다.

작은 연골 마개를 테이프 스트립에 부착하고 구조 헤드(20 mm)에 둠.

10 ㎕의 히알루론산을 큰 마개에 첨가하고 모든 마개가 닿을 때까지 구조 헤드를 낮춤. 수화를 유지하도록 20 mL의 HBSS를 페트리 접시에 첨가.

단계를 실행하고 프롬프터가 뜨면 파일명 설정.

HBSS가 증발되었는지 종종 확인. 증발되었으면, 더 첨가.

단계가 끝나면, 데이터 분석을 열고 파일을 염.
10. 파일명을 우클릭하고 표를 선택. 샘플 포인트가 있는 표가 나타날 것임. 각 샘플에 대한 다른 단계를 위해서는, 끌어내릴 수 있는 탭이 나타나고 한 번에 한 단계를 선택할 수 있고 값이 화면에 나타날 것임.

도 1은 손상되지 않은 연골(아그레칸 손실없음)의 경우의 마찰력을 나타내고 본원에 따른 루브리신 모방 펩티도글리칸을 사용하면 연골 표면 사이의 마찰이 증가함을 나타낸다. 도 2는 손상된 연골(골관절염을 모의하는 아그레칸 손실 있음)의 경우의 마찰력을 나타내고 본원에 따른 루브리신 모방 펩티도글리칸이 연골에 첨가되면 연골 표면 사이의 마찰이 감소됨을 나타낸다. 따라서, 본원에 따른 루브리칸 모방 합성 펩티도글리칸이 두 연골 표면 사이에 적용되면, 마찰력이 더 높음 진동 전단에서 감소하여, 빠른 움직임 도중 연골을 보호할 수 있음을 시사한다.

방법

Claims (29)
- a) 글리칸;
b) 약 1 내지 약 80 콜라겐 결합성 펩티드(들); 및
c) 약 1 내지 약 80 히알루론산 결합성 펩티드(들)를 포함하고,
b) 및 c)의 펩티드가 글리칸에 공유 결합된, 세포외 기질 결합성 합성 펩티도글리칸. - 제1항에 있어서, 글리칸이 덱스트란, 콘드로이틴, 황산 콘드로이틴, 데르마탄, 황산 데르마탄, 헤파란, 헤파린, 케라틴, 황산 케라탄, 또는 히알루론산인, 합성 펩티도글리칸.
- 제1항에 있어서, 글리칸이 황산 데르마탄인, 합성 펩티도글리칸.

i) WYRGRLGC, RRANAALKAGELYKSILYGC, RLDGNEIKRGC, AHEEISTTNEGVMGC , GCGGELYKSILY, NGVFKYRPRYFLYKHAYFYPPLKRFPVQGC, CQDSETRTFY, TKKTLRTGC, GLRSKSKKFRRPDIQYPDATDEDITSHMGC, SQNPVQPGC, SYIRIADTNITGC, SYIRIADTNIT, KELNLVYT, KELNLVYTGC, GSITTIDVPWNV, GELYKSILYGC 및 GSITTIDVPWNVGC로 이루어지는 군에서 선택되는 아미노산 서열, 또는
