Abstract
본 발명은 방사성요오드화된 아미노폴리카르복실레이트-첨부 펩타이드 및 표적 M 제제의 접합체를 제조하고 정제하는 방법에 관한 것이다. 상기 방법은 (A) (i) 결합하지 않은 방사성요오드, (ii) 표적 제제에 접합되지 않은 방사성요오드화된 아미노폴리카르복실레이트-첨부 펩타이드, 및 (iii) 표적 제제에 접합된 방사성요오드화된 아미노폴리카르복실레이트-첨부 펩타이드를 포함하는 용액을 제공하는 단계; (B) 상기 용액을 음이온-교환 수지와 접촉시키는 단계; 및 (C) 상기 음이온-교환 수지와 용액을 함께 음이온-교화 수지 입자들을 트랩핑(trapping)할 수 있는 필터를 통해 통과시키는 단계를 포함한다.
Description
본 발명은 방사성면역검출(radioimmunodetection) 및 방사성면역치료(radioimmunotherapy)에서 사용되는 반응물의 정제에 관한 것으로, 보다 상세하게는 생체 내에서 증강된 안정성 및 종양 부위들에서 증강된 보존성을 가지는 방사성요오드 라벨된 접합체들의 정제에 관한 것이다.
방사성요오드화된 단클론 항체들은 Amer. J. Med. 1993; 94: 297-312에 게재되고 Goldenberg에 의해 요약된 암의 진단 및 치료에 중요하다. 이들을 사용하기 위하여 방사성요오드를 단클론 및 다클론 항체들로 화학적으로 도입하는 많은 방법들이 지난 삼십년간 개발되어 왔다. 요오드는 단백질의 방사성요오드화를 위하여 사용되는 응용화학이 비교적 쉽고, 방사성요오드가 유용한 물질 붕괴 특성들을 가지며 요오드의 동위원소들이 통상적으로 이용되기 때문에 이들 적용에서 방사성라벨로서 바람직하다. 여러 가지 응용화학들은 암 세포들을 표적하는 항체들에 요오드를 결합하도록 개발되어 왔다. 이들 응용화학들은 Wilbur, Bioconjugate Chemistry 1992; 3: 433-70에서 재검토되었다. 대부분 통상적인 결합 과정은 항체에 있는 관능기와 반응하는 친천자성(electrophilic) 방사성요오드 종 위치에서 준비된다. 클로라민-T(chloramin-T) 및 아이오도젠(iodogen)과 같은 반응물들은 친전자성 요오드를 생성하는데 사용된다. 단백질의 타이로신기는 일반적으로 요오드화 부위이다.
MAbs의 통상적인 방사성요오드화는 사용된 산화물질뿐만 아니라 일부 정제 방법을 이용하여 결합되지 않은 방사성요오드를 제거하는데 필요하다. 클로라민-T와 같은 완충액-가용성 산화물질을 사용하는 경우, 산화물질과 방사성요오드는 통상적으로 사용가능한 PD10® 컬럼과 같은 크기-배제 컬럼 상에서 크기-배제 크로마토그래피에 의해 통례적으로 제거된다.
직접적인 방사성요오드화 도표를 사용하는 주요 결점은 생체내 탈요오드화(deiodination) 현상이다. 생체 내에서 항체 내재화 및 리소좀 제조의 결과로서, 라벨된 단백질은 작은 펩타이드로 손상되고, 그 방사성요오드는 요오드타이로신(iodotyrosine)을 생성하거나 또는 저분자량 펩타이드 단편에 부착되는 요오드 형태로 세포로부터 방출된다. 이러한 발견들은 Geissler et al., Cancer Research 1992; 52: 2907-2915 및 Shih et al., J. Nucl. Med. 1994; 35: 899-908에 의해 보고되었다. 생체 내에서 표적 세포들에서 방사성요오드의 제거는 방사선진단에서 중요한 종양 대 비종양 식별을 감소시키고, 또한 방사선치료 효과에 상당한 영향을 주는 표적 세포들에서 방사성요오드의 잔류 시간을 감소시킨다.
세포내 보유되는 요오드 방사성라벨들의 설계를 통하여, 생체 내에서 탈요오드화의 현상을 극복하기 위한 여러 가지 접근들이 연구되었다. 이러한 라벨들은 "잔류화 라벨들(residualizing labels)"이라고 한다. 하나의 방법에서, 방사성요오드는 비-대사가능한(non-metabolizable) 탄수화물들에 부착되고, 나중에 일차 활성화된 후 항체들과 접합된다. 이러한 접근은 락티톨티라민(lactitoltyramine; LT) 및디락티톨티라민(dilactitoltyramine)(Strobel et al., Arch. Biochem. Biophys 1985; 240: 635-45) 및 타이로신 셀로바이오스(tyrosine cellobiose)(Ali et al., Cancer Research 1990; 50: 783s-88s)에 의해 실증된다. 다른 접근에서, 피리딘-기저 부위(pyridine-based moiety)인 "SIPC"가 이용되었다(Reist et al., Cancer Research 1996; 56: 4970-4977). 또한 모든 D 아미노산들과 복합 염기성 아미노산들(multiple basic amino acids)을 포함하는 펜타펩타이드들(pentapeptides)이 사용된다(Foulon et al., Cancer Research 2000; 60: 4453-4460). 또 다른 접근에서, D-아미노산들을 포함하는 DTPA-첨부 방사성요오드화된 펩타이드들이 잔류화 라벨로서 성공적으로 사용된다(Govindan et al., Bioconjugate Chemistry 1999; 10: 231-240; Stein et al., Cancer Research 2003; 63: 111-118).
이전 문단에서 주어진 참고자료에서 설명된 방사성요오드화된 저분자량 물질이 MAbs에 접합되는(이하, '방사성요오드화된 접합체'라고 한다) 경우, 보다 잘 제거되기 위하여 비접합된 물질을 필요로하는 부가적인 조건이 존재한다. 이는 정제의 부가적 컬럼 방법을 언제나 요구한다. 탄수화물법은 전체 수율 및 특이 활성을 낮게 하고, 과정의 말단에서 정제의 컬럼 방법을 포함한다. 상기 Reist et al. 및 Foulon et al.의 방법은 하나의 방사성요오드화 단계 및 다른 하나의 항체 접합 단계, 두 가지 컬럼 정제 단계들을 포함한다. 상기 Stein et al.에 더 기재된 상기 Govindan et al.의 방법은 보다 높은 전체 수율들 및 특이 활성들을 가지는 과정의 말단에서 하나의 컬럼 정제를 포함한다.
컬럼 방법들은 일반적으로 성가시고, 특히 임상 단계 준비를 위한 많은 I-131의 mCi를 취급하는 경우 방사선이 사람에게 노출되는 부가적인 결점 및 비극적인 컬럼 실패들을 가진다. 아이오도젠-기저 방사성요오드화에서, 산화물질이 수-불용성이고 방사성라벨된 물질을 간단히 주입하거나 또는 여과하여 제거된다. 이러한 사실들에서, 제거되어야하는 다른 물질은 비결합된 방사성요오드뿐이다. 요오드 대 인산염 또는 히드록시 이온이 강한 음이온-교환 수지에 결합하는 비교적 높은 친화력으로 인해, 비결합된 요오드는 음이온-교환 수지를 사용하여 제거가능하다(Weadock et al., J Nuclear Medicine 1990; 31: 508-511; Behr TM et al. Nuklearmedizin 2002; 41: 71-79). 음이온 교환 수지와 조합하여 통상적으로 사용가능한 "IODO-BEADS"® 와 같이 고정된 클로라민-T 산화물질의 사용을 생각할 수 있다. 특정 단순화가 상기 조합에 의한 직접적인 방사성요오드화된 항체들의 정제에서 완성되는 반면에, 방사성요오드화된 접합체들의 사용은 아직 비접합된 저분자량 부위들의 제거를 요구한다. 이러한 정제가 일반적으로 컬럼 방법에 의하여 많이 완수되는 경우, 그 방법들의 부수적인 결점들이 임상 적용을 위한 방사성라벨된 준비의 많은 mCi 정제에서 실질적인 문제들이 존재한다.
디에틸아미노에틸-셀룰로오스 음이온-교환기의 컬럼을 통과하고 물의 몇몇의 물 분획으로 용리하여 비라벨된 DOTA-펩타이드로부터 방사선금속-킬레이트 DOTA-펩타이드를 정제하는 컬럼 방법이 Li et al.(Bioconjugate Chemistry 1994; 5: 101-104)에 기재되어 있다. 이 경우, 킬레이터 분획에서 음전하를 띠는 비라벨된 금속이 음이온-교환 컬럼에 의해 잔류하는 반면, 방사선금속-킬레이트 DOTA 펩타이드는 중성 전하를 띠고, 따라서 컬럼으로부터 용리된다. 이는 방사선금속-킬레이트 이관능기(bifunctional) 물질을 정제하고 이어서 항체들과 접합되며, 생성물이 크기-배제 컬럼 방법에 의하여 정제되기 위하여 필연적이다. 따라서, 상기 Li et al.의 과정은 다중 단계 접근 및 두 가지 컬럼-기저 정제 단계를 포함한다. 게다가, 상기 컬럼-기저 방법들은 표적 제제들에 접합하여 저분자량 부위들의 큰 규모 방사성요오드화에 적용하는 경우 비현실적일 것이다. 후자의 경우, 방사성 요오드의 많은 mCi를 포함하는 보다 간단한 정제 방법을 필요로 한다.
도 1은 조(정제되지 않은) 생성물(I-131-IMP-R4-hMN-14)의 크기-배재(SEC) HPLC를 보여준다. 메인 피크 ∼10분 보존은 라벨된 hMN-14에서 기인하는 반면에, 피크들 근처의 14분 및 18분은 각각 비접합된 I-131-IMPR4 및 I-131을 나타낸다.
도 2는 정제된 I-131-IMPR4-hMN-14의 SEC HPLC를 보여주고, 비접합된 I-131-IMPR4 및 I-131이 음이온 교환에 의하여 완전히 제거된 것을 입증한다.
도 3은 정제된 생성물과 암태아성 항원(carcinoembryonic antigen; CEA) 복합의 혼합물의 SEC-HPLC를 보여주고, 이 HPLC는 생성물의 면역반응성이 라벨된 항체로 인하여 항체-항원 복합체의 보다 높은 MW 물질로 피크가 완전히 이동한 것에 의해 설명되는 것과 같이 완전히 유지됨을 보여준다.
본 발명에서는 방사성요오드화된 아미노폴리카르복실레이트-첨부 펩타이드들을 표적 제제들에 공유 결합하여 제조하는 방사성요오드화된 접합체들의 정제 방법을 제공하여 상기의 문제들을 해결한다.
본 발명은 다양한 방사성요오드화된 아미노폴리카르복실레이트-첨부 부위들 및 표적 제제들로부터 유도된 방사성요오드화된 접합체를 위한 정제 시스템의 필요성을 신청한다. 항체들과 같은 표적 제제들의 직접적인 방사성요오드화와 관련된 당업계에 이미 알려진 종래 기술과는 대조적으로, 본 발명은 타이로신-함유 이관능기 펩타이드와 같은 저분자량 물질의 방사성요오드화를 포함하고, 방사성요오드화된 부위와 항체들을 접합하는 과정에 관한 것이다. 이와 같이, 본 발명은 비결합된 방사성요오드 및 비접합된 방사성요오드화된 부위 모두를 정제하여 제거하는 필요성을 신청한다.

용어정의

표적 제제 : "표적 제제"는 표적 세포에 선택적으로 결합하는 능력을 가지는 분자 부위이다. 표적 제제의 예로는 종양 또는 감염성 병변에 의해 발현되는 항원 또는 수용체를 표적할 수 있는 단백질 분자; 종양 또는 병변에 있는 특이적 수용체를 표적할 수 있는 저분자량 부위; 또는 종양 세포들 또는 병변들에 표적되는 합성 뉴클레오타이드들을 포함한다. 바람직한 표적 제제들은 항체들 및 펩타이드들을 포함한다.

아이오도젠 방법 : 이 용어는 산화물질로서 아이오도젠을 사용하는 것을 포함하는 방사성요오드화 과정을 말한다. 아이오도젠(1,3,4,6-테트라클로로-3α,6α-디페닐글리콜우릴(diphenylglycoluril))은 상기에서 언급한 Wilbur에서 거론된 방사성요오드의 용액과 접촉하여 활성 방사성요오드화 종을 생성하는 수-불용성 물질이다.
클로라민 -T 방법 : 이 용어는 상기에서 언급한 Wilbur에서 거론된 방사성요오드의 용액과 혼합하여 활성 방사성요오드화 종을 생성하는 수용성 산화물질로서 클로라민-T(소듐 N-클로로톨루엔설폰아미드)을 사용하는 것을 말한다.
결합 부위 : 결합 부위는 말레이미드(maleimide), 클로로아세트아미드(chloroacetamide), 브로모아세트아미드(bromoacetamide), 요오드아세트아미드(iodoacetamide), 비닐술폰(vinylsulfone), N-히드록시숙신이미드 에스테르(N-hydroxysuccinimide ester), N-히드록시술포숙신이미드 에스테르(N-hydroxysulfosuccinimide ester), 아미데이트 에스테르(amidate ester), 이소시아네이트(isocyanate) 또는 이소티오시아네이트(isothiocyanate)를 포함하는 군에서 선택되고, 표적 제제와 공유 결합을 생성할 수 있는 관능기이다. 결합 부위는 동종이관능기(homobifunctional) 및 이종이관능기(heterobifunctional) 교차 결합 분자들로 사용하여 아미노카르복실레이트-첨부 펩타이드에 도입된다. 교차 결합기로서 동종이관능기 및 이종이관능기 반응물의 사용은 당업계에 잘 알려져 있다(Wong, S.S., 1991; Chemistry of protein conjugation and cross-linking; CRC Press, Boca Raton, Fl; pp 1-48).
놀랍게도, 본 발명자들은 음이온-교환 수지와의 간단한 접촉이 비접합된 저분자량 부위를 효과적으로 제거함을 발견하였다. 정제된 생성물은 음이온-교환 수지로부터 여과하여 분리된다. 접합체로부터 수지를 여과하고, 반응하지 않은 요오드 및 요오드화된 아미노폴리카르복실레이트-첨부 펩타이드를 수지로 제거하기 위하여, 미립자 물질을 포착할 수 있는 어떠한 여과 장치가 적용될 수 있다. 0.22㎛ 필터 장치가 가장 바람직하다. 음이온-교환 수지와 교반하여 비접합된 저분자량 물질을 제거하기 위한 방법은 DTPA와 같이 본 발명의 아미노폴리카르복실레이트 부위들을 이용하여 특히 잘 수행된다.
간단한 원-포트 정제에 의한 비접합된 폴리카르복실레이트 부위들의 효과적인 제거는 뜻밖에 발견되었다. 음이온 교환 수지에 대한 반대이온의 선택성(selectivity)에 기초하여, 음이온 교환 수지와의 단기간 접촉이 폴리카르복실레이트와 같이 비접합된 저분자량 부위들을 효과적으로 제거할 수 있음을 예상할 수 없었다. 예를 들면, 수산화물이 음이온 교환 수지에 약하게 결합하는 능력을 가진다고 여겨지고, 아세트산염 기가 수산화물보다 더 최소한으로만 결합한다. 아세트산염 기는 아미노폴리카르복실레이트가 질소 원자에서 아세트산 잔기들을 포함하는 경우 실제 아미노폴리카르복실레이트와 관계가 있고, 이들 아세트산 잔기들은 pH 6 이상의 완충액에서 아세트산으로 완전히 이온화된다. 예를 들면, 제조업자의 제품 안내서(AG®1 및 AG2 Strong Anion Exchange Resin Instruction Manual, BioRad Corporation)에 기초하는 요오드의 선택성은 AG1® 음이온-교환 수지상의 수산화물(1에서와 같이 임의로 유지됨)의 175배인 반면에, 아세트산염의 선택성은 수산화물 보다 3.2배만이 높았다. 더욱이, 상기 수지는 아세트산염의 선택성보다 약간 높은 5(수산화물에 대한 1과 비교하여)의 상대적 선택성을 가지는 인산염 형태로 사용된다. 음이온 교환 수지에 결합하기 위한 요오드의 상대적으로 높은 선택성은 이온-교환 과정에 의한 요오드의 성공적인 제거로만 예측된다. 그러나, 음이온 교환 수지에 결합하는 아세트산염 대 인산염 이온의 낮은 선택성에도 불구하고, 본 발명자들은 이온 교환법이 음이온-교환 수지 결합 선택성에 기초하여 예측될 수 없는 아미노폴리카르복실레이트를 포함하는 비접합된 저분자량 물질을 제거하는데 효과적으로 작용한다는 것을 발견하였다. 어떠한 원리에 의해 결합되기를 바라지 않는데도, 본 발명자들은 이러한 개선된 선택성이 저분자량 부위의 아미노폴리카르복실레이트 하부구조(substructure)에 의해 나타나는 것과 같이 동일한 분자에서 다중 아세트산염 기의 존재에서 기인한다고 믿는다. 따라서, 저분자량 물질 내에서 아미노폴리카르복실레이트 부위들의 통합에 의해 비접합된 방사성요오드화된 물질을 음이온-교환 수지의 현탁액과 몇 분동안 간단히 교반하고 생성물을 여과하여 효과적으로 제거될 수 있음을 발견하였다. 본 발명의 간단한 과정은 정제 컬럼 방법을 완전히 피하게 되는 상술한 Li et al.에서 조작상 독특하고 편리하다.
음이온 교환 수지
이들 실험들을 위하여, 스티렌 디비닐벤젠 공중합체 격자에 부착되는 4급 암모늄기를 포함하는 통상적으로 사용가능한 강한 염기성 음이온-교환 수지를 사용하였다. 이러한 수지는 1000Da의 분자량 배제 한계를 가지는 포어 크기 및 100-200(150-75㎛ 입자 직경)의 배지 메쉬(mesh) 크기에 대응하는 8% 교차-결합을 가진다.
그러나, 본 발명은 상기에 언급된 특정 음이온-교환 수지 특성에만 한정되는 것이 아니며, 상기에 기재된 강한 염기성 음이온-교환 수지 및 통상적으로 사용가능한 디에틸아미노에틸 셀룰로오스와 같은 약한 염기성 음이온-교환 수지를 포함한다. 또한, 사용된 수지에서 교차-결합의 한도는 한정되지 않는다. 강한 염기성 음이온 수지의 특정한 예에서, 교차-결합은 2%(2700Da의 분자량 배제 한계를 가지는 포어 크기)에서 12%(400Da의 분자량 배제 한계를 가지는 포어 크기)까지 변화할 수 있고, 수지의 입자 크기는 850-38㎛ 입자 직경 범위에 대응하는 20-400 메쉬의 범위가 될 수 있다.
접합체들
본 발명은 방사성활성이 생체 내에서 잔류화되도록 라벨링 항체들을 위하여 사용되는 비대사가능한(nonmetabolizable) 및 방사성요오드화된 펩타이드들의 사용에 관한 것이다. 이들 특정하게 설계된 펩타이드들은 2-40 아미노산을 포함하고, 바람직하게는 2-5 아미노산을 포함하며, D-아미노산들을 포함하는 것이 바람직하다. 다른 구현예에서, 이관능기 요오드화가능한 아미노폴리카르복실레이트는 아미노폴리카르복실레이트에 대한 티라민 기 및 항체 결합 기를 부착하여 유도된다. 프로테아제에 민감한 결합은 이들 구조들을 포함하지 않는다. 선택적으로, 아미노폴리-카르복실레이트, 항체-결합 유닛으로 치환된 골격은 대응하는 이무수물들(dianhydrides)로 전환된 다음 D-타이로신과 반응하여 두 개의 D-타이로신 잔기들을 포함하는 존재물을 획득한다. 이관능기 아미노폴리카르복실레이트와 D-타이로신 사이의 아미드 결합이 프로테아제에 의해 인지되지 않기 때문에, 이들은 잔류화 요오드 라벨들의 다른 버젼을 구성한다.

항체들
본 발명의 바람직한 구현예에서, MAbs, 다가 항체들 및 다특이성 항체들과 같은 항체들은 표적 세포들에서 높은 레벨로 발현되고 정상 조직뿐만 아니라 골수 세포 및 부갑상선, 비장 및 자궁내막과 같은 전위 조직과 같이 제거되는 조직들 및 특정 정상 세포들과 결합되는 항원들 및 빠르게 내재화되는 항체들에 대비하여 질병 세포들에서만 또는 우세하게 발현되는 지표들 또는 종양 결합 항원들을 인지하거나 또는 결합하는데 사용된다. "다가" 항체에 의하여 항체가 동일한 또는 다른 구조를 가지는 하나 이상의 항원에 동시에 결합될 수 있다. "다특이성" 항체에 의하여 환자 항체는 다른 구조를 가지는 둘 이상의 항원들에 동시에 결합될 수 있다. 예를 들면, 두 개의 다른 특이성들을 가지는 항체는 동시에 두 가지의 구조적으로 다른 표적들에 결합할 수 있으므로 다가 및 다특이성으로 간주될 수 있다. 또 한편으로는, 동일 표적에 결합하는 둘 이상의 특이적 팔들(arms)을 가지지만 다른 특이성이 없는 항체는 다가이지만 다특이성은 아니다. 다양한 다특이성 및/또는 다가 항체들은 분자 공학(molecular engineering)을 사용하여 제조될 수 있다. 예를 들면, 양특이성 융합 단백질은 하나의 항원에 대한 단일 결합 부위를 가지는 scFv와 두번째 항원에 대한 단일 결합 부위를 가지는 Fab 단편으로 이루어진 일가가 될 수 있다. 또한 양특이성 항체는 하나의 항원에 대한 두 개의 결합 부위들을 가지는 IgG와 두번째 항원에 대한 두 개의 결합 부위들을 가지는 두 개의 scFv로 이루어진 이가가 될 수 있다.

본 발명의 다른 바람직한 구현예에서, 항체들은 빠르게 내재화된 후 세포에 의해 순환하는 면역접합체의 지속적인 흡수 및 침착을 가능하여 세포 표면에서 재발현되고, 처리되며 존재한다. 가장 바람직한 항체/항원 쌍의 예로는 LL1, 항-CD74 mAb(불변 사슬, Ⅱ종 특이적 샤프론, Ii)이다. CD74 항원은 B 세포 림프종, 특정 T 세포 림프종, 흑색종 및 특정 다른 암들에서 높게 발현된다(Ong et al., Immunology 1999; 98: 296-302).
항-CD74 항체들로 처리되는 질병의 예로는 비호지킨스 림프종, 흑색종 및 다발성 골수종을 포함하나, 이에만 한정되는 것은 아니다. 표적 세포의 표면에서 짧은 시간 동안의 CD74 항원의 지속적인 발현에 이은 항원의 내재화 및 항원의 재발현은 "페이로드(payload)"와 같이 운반하는 어떠한 치료 부위와 함께 내재화되어 LL1 항체를 표적할 수 있다. 이는 상기 세포들 내부에 축적되는 LL1-치료 면역접합체의 높은 치료 농도를 허용한다. 내재화된 LL1 면역접합체들은 리소좀들 및 엔도좀들(endosomes)을 통하여 순환되고, 따라서 잔류화 방사성 부위는 표적 세포들 안에 잔존한다.
바람직한 구현예에서, 뮤린 및 키메라 버젼의 항체들도 사용될 수 있으나, 인간 질병을 치료하는데 사용되는 인간 또는 인간화된(cdr-이식된) 버젼의 항체들이 사용될 수 있다. 수의사가 이용하는 경우, 교차-종의 IgG가 유용할 수 있으나, 동일 종의 IgG가 가장 유효한 벡터가 될 수 있다. 운반제와 같은 동일 종의 IgG 분자들은 면역 반응들을 최소화하는데 가장 바람직하다. 이는 특히 반복 치료를 고려하는 경우에서 중요하다. 인간에 대하여, 인간 또는 인간화된 IgG 항체는 환자로부터 항-IgG 면역 반응을 보다 적게 초래할 수 있다. 내재화 항원을 표적하는 hLL1 및 hLL2와 같은 항체들은 표적 세포에 결합한 후 빠르게 내재화되며, 운반되어지는 방사성요오드화된 항체 접합체가 세포로 빠르게 내재화되는 것을 의미한다.
MAb는 뮤린, 키메라, 인간화된 또는 인간 항체들이며, 온전한, 단편 또는 이들의 다양한 제작 버젼들이 될 수 있다. 특히 바람직한 구현예에서, MAb는 티올기들을 가지는 유도된 또는 이중황-환원된 것일 수 있다.

로 라벨링하기 위한 간편화된 원-포트(one-pot) 제조 및 정제 방법
이 과정은 방사성요오드화되고 이중황-환원 항-CEA MAb, hMN-14에 접합되는 저분자량 물질로서 IMP-R4를 사용하여 이하 설명한다. IMP-R4는 MCC-Lys(MCC)-Lys((1-(p-CSNH)벤질)DTPA)-D-Tyr-D-Lys((1-(p-CSNH)벤질)DTPA)-OH 구조를 가지고, 여기에서 MCC는 4-(N-말레이미도메틸)-사이클로헥산-1-카르보닐이다. IMP-R4는 2000년 10월 26일자로 출원된 미국특허 출원 제09/696,740호의 주제의 일부이다.
제조업자로부터 획득한 I-131 요오드화 나트륨을 그 부피의 7배량인 pH 7.4의 0.5M 인산 나트륨으로 완충처리하고, pH 7.4의 40mM 인산 나트륨 1.4mL을 부가적으로 사용하여 교반 바(stir bar)를 가지고 IMP-R4(I-131의 0.13μmol/100mCi)를 포함하는 아이오도젠-코팅된 바이알(vial)로 이전하였다. 상기 용액을 8-15분간 교반하고 결합하지 않은 방서성요오드는 과잉의 4-히드록시페닐아세트산으로 억제하였다. 방사성요오드화된 IMP-R4는 30분간 이중황-환원 hMN-14(한 실험에서 IgG-SH 대 IMP-R4 비율 :0.6)에 접합시키고 사용되지 않은 티올기들은 테트라티오네이트(tetrathionate) 나트륨으로 캡을 씌웠다. 최종적으로, 인산염에 형태에 용해한 음이온-교환 수지 AG 1X8®(100-200 메쉬)의 20% w/v 현탁액 2-3mL을 첨가하고 5분간 교반하였다. 그런 다음 방사성라벨된 물질을 MilliFil®GS 0.22㎛ 필터 유닛(Millipore Corporation)을 사용하여 멸균한 격막-밀봉 용기로 여과하였다. 인간 혈청 알부민은 1-2.5% 최종 농도로 생성물에 선택적으로 첨가한다.
일반적인 회복율들은 하기와 같다. I-131의 57.4mCi에서 출발하여 최종 생성물 40mCi(69.9%)를 포함한 반면, I-131의 123mCi를 포함하는 하나의 주행에서 최종 생성물은 63mCi(51%)를 포함하였다. I-131의 40-123mCi를 사용하는 방사성라벨들(n=9)에서, 전체 결합은 HPLC 분석에 의해 61-75% 범위였던 반면, 회복율들은 50-70% 범위였다. CEA와 복합되고 HPLC에 의해 분석하여 평가되는 면역반응들은 95% 이상이었다. 최종 생성물들은 HPLC 분석에 의해 순도 95% 이상이었고, 응집이 5% 이하이며, 5-6mCi/mg 범위에서 특이 활성을 가졌다.

완충액
내에서 혈청-면역성 검사에 대한 안정성
실시예 1(총 방사성의 62.9mCi)의 정제된 생성물을 0.9mCi/mL의 농도 및 항체 농도 0.2mg/mL에서 2.5% HSA를 포함하는 인산 완충액에 넣고 20시간 동안 상온에서 방치한 다음 크기-배제 HPLC로 분석하였다. 예시적인 인산 완충액은 pH가 6.0-7.5 사이로 조절된 0.1M 인산 나트륨 수용액이다. 이는 생성물이 실제로 변화하지 않았음을 보여주었다. 20배 몰의 과잉의 CEA 항원과의 복합체화는 면역반응성의 보존을 나타내었다.
두번째 실험에서, 상기 생성물은 인간 혈청으로 33.3배 희석하고 37℃에서 20시간동안 항온배양하였다. 이 시점에서 분석은 라벨의 사소한 손실을 보였고 상기 물질은 아직 면역반응성의 보존을 나타내는 CEA와의 복합체를 이루고 있었다.

I-131 요오드화 나트륨의 완충작용을 pH6의 0.3M 인산 나트륨의 그 7배의 부피량으로 수행한 후 pH 6의 0.03M 인산 나트륨 1.4mL을 사용하여 아이오도젠 바이알로 완충된 I-131을 이전하도록 실시예 1의 과정을 변형하여 수행하였다. 방사성요오드화 이후, 이중황-환원 hMN-14에 접합하고 AG® 1X8 음이온 교환 수지로 교반하여 정제하여 I-131 요오드화 나트륨의 56.5mCi로부터 출발하여 정제된 I-131-IMP-R4-hMN-14의 39mCi(69%) 회복율을 획득하였다. 다른 실험에서, 동일한 과정을 거쳐 정제된 생성물(63.1%)의 69.3mCi가 I-131 요오드화 나트륨의 109.8mCi로부터 획득되었다. 다시 이들 두 가지 실험들에서 면역반응성의 완전한 보존이 CEA와의 복합체화를 이용하여 입증되었다.

Claims (50)
- (A) (i) 결합되지 않은 방사성요오드 (ii) 표적 제제에 접합되지 않는 방사성요도드화된 아미노폴리카르복실레이트-첨부 펩타이드 및 (iii)표적 제제와 접합되는 방사성요도드화된 아미노폴리카르복실레이트-첨부 펩타이드를 포함하는 용액을 제공하는 단계;(B) 상기 용액을 음이온-교환 수지에 접촉시키는 단계; 및(C) 음이온-교환 수지 입자들의 트래핑(trapping)하는 여과 능력을 통하여 상기 음이온-교환 수지와 용액을 함께 통과시켜 방사성요오드화된 아미노폴리카르복실레이트-첨부 펩타이드와 표적 제제의 정제된 접합체를 획득하는 단계;를 포함하는 방사성요오드화된 아미노폴리카르복실레이트-첨부 펩타이드와 표적 제제의 접합체를 제조 및 정제하는 방법.
- 제 1항에 있어서, 상기 (A)의 용액은아미노폴리카르복실레이트-첨부 펩타이드를 방사성요오드화하여 방사성요오드화된 펩타이드를 생성하는 단계; 및방사성요오드화된 아미노폴리카르복실레이트-첨부 펩타이드를 표적 제제에 접합시켜 방사성요오드화된 아미노폴리카르복실레이트-첨부 펩타이드의 접합체를 생성하는 단계;에 의하여 제조됨을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 펩타이드의 방사성요오드화는 방사성 요오드화 나트륨으로부터 펩타이드에 결합가능한 방사성요오드를 생성하는 산화물질과 함께 수행됨을 특징으로 하는 방법.
- 제 3항에 있어서, 상기 산화물질은 아이오도젠(iodogen) 또는 클로라민-T(chloramine-T)임을 특징으로 하는 방법.
- 제 4항에 있어서, 상기 산화물질은 클로라민-T이고, 방사성요오드화는 클로라민-T의 고정된 불용성 비드 형태의 존재하에서 수행됨을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 필터는 0.1-0.3㎛의 포어(pore) 직경을 가짐을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 음이온-교환 수지는 폴리머 격자 상의 4급 암모늄 관능기로 이루어지고, 상기 폴리머 상의 과잉 교차-결합은 2-12% 임을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 음이온-교환 수지는 폴리머 격자 상에 사차 아민 관능기로 이루어짐을 특징으로 하는 방법.
- 제 7항 또는 제 8항에 있어서, 상기 수지의 입자 크기는 20-400 메쉬(850-38㎛ 입자 직경) 임을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 접합체의 정제된 용액은 음이온 교환 수지에 노출되기 전에 용액내에 원래부터 존재하던 결합된 비접합된 방사성요오드 및 비접합된 방사성요오드화된 펩타이드의 10% 미만을 포함함을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 용액과 함께 수지를 교반하는 단계를 더 포함함을 특징으로 하는 방법.
- 제 1항에 있어서, 음이온 교환 수지를 포함하는 컬럼을 통하여 용액을 통과시키는 단계를 더 포함함을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 아미노폴리카르복실레이트-첨부 펩타이드가 하기와 같이 이루어진 군으로부터 선택된 것임을 특징으로 하는 방법:X-Gly-D-Tyr-D-Lys((1-(p-CSNH)벤질)DTPA)-OH;X-D-Ala-D-Tyr-D-Tyr-D-Lys(DTPA);[X-D-Ala-D-Tyr-D-Tyr-D-Lys(½DTPA)]2;X-Lys(X)-Lys((1-(p-CSNH)벤질)DTPA)-D-Tyr-D-Tyr-D-Lys((1-(p-CSNH)벤질)DTPA)-OH;X-Lys(X)-Lys((1-(p-CSNH)벤질)DTPA)-D-Tyr-D-Lys((1-(p-CSNH)벤질)DTPA)-OH;X-Asp-D-Tyr-D-Lys((1-(p-CSNH)벤질)DTPA)-OH;X-Lys(X)-Asp-D-Tyr-D-Lys((1-(p-CSNH)벤질)DTPA)-OH;X-Asp-D-Tyr-D-Lys((1-(p-CSNH)벤질)DTPA)-OH; 및X-Lys(X)-Asp-D-Tyr-D-Lys((1-(p-CSNH)벤질)DTPA)-OH,상기에서, X는 표적 제제와 공유 결합을 생성할 수 있는 결합 부위를 포함하는 교차-결합기임.
- 제 1항 또는 13항에 있어서, 상기 표적 제제는 말레이미드, 클로로아세트아미드, 브로모아세트아미드, 요오드아세트아미드, 비닐설폰, N-히드록시숙신이미드 에스테르, N-히드록시설포숙신이미드 에스테르, 아미데이트(amidate) 에스테르, 이소시아네이트 또는 이소티오시아네이트를 포함하는 결합 부위에 의해 아미노폴리카르복실레이트-첨부 펩타이드에 접합됨을 특징으로 하는 방법.
- 제 14항에 있어서, 상기 말레이미드 결합 부위는 4-(N-말레이미도메틸)-사이클로헥산-카르보닐 부위임을 특징으로 하는 방법.
- 제 14항에 있어서, 상기 말레이미드 결합 부위는 2-(N-말레이미도)아세틸 부위임을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 아미노폴리카르복실레이트는 EDTA, DTPA, 벤질-EDTA, 벤질-DTPA, 벤질-DOTA, TTHA(트리에틸렌테트라민헥사아세트산), NOTA 또는 벤질-NOTA임을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 펩타이드는 MCC-Lys(MCC)-Lys((1-(p-CSNH)벤질)DTPA)-D-Tyr-D-Lys((1-p-(CSNH)벤질)DTPA)-OH인 IMP-R4이고, 상기에서 MCC가 4-(N-말레이미도메틸)-사이클로헥산-카르보닐 부위임을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 표적 제제는 단클론 항체(MAb)임을 특징으로 하는 방법.
- 제 19항에 있어서, 상기 단클론 항체는 악성 질병과 결합되는 것임을 특징으로 하는 방법.
- 제 19항에 있어서, 상기 단클론 항체는 심장혈관 질병과 결합되는 것임을 특징으로 하는 방법.

- 제 19항에 있어서, 상기 단클론 항체는 알츠하이머 질병과 결합되는 것임을 특징으로 하는 방법.
- 제 19항에 있어서, 상기 단클론 항체는 감염 기관과 결합되는 것임을 특징으로 하는 방법.
- 제 19항에 있어서, 상기 단클론 항체는 내재화 항체임을 특징으로 하는 방법.
- 제 19항에 있어서, 상기 단클론 항체는 항-CEA MAb, MN-14; 항-EGP-1 MAb; 항-CD22 MAb; 항-CD20 MAb; 항-결장-특이 항원-p MAb; 항-CD74 MAb; 항-MUC1 MAb; 항-AFP MAb; 항전립선 특이막 항원; 또는 항-탄산탈수효소 Ⅸ MAb 임을 특징으로 하는 방법.
- 제 19항에 있어서, 상기 단클론 항체는 뮤린, 키메라, 인간화된 또는 인간 항체들이고, 온전한, 단편 또는 이들의 다양하게 제작된 버젼이 될 수 있음을 특징으로 하는 방법.
- 제 19항에 있어서, 상기 단클론 항체는 티올 기들을 가지는 유도된 또는 이중황-환원된 것임을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 방사성요오드는 I-123, I-124, I-125 또는 I-131임을 특징으로 하는 방법.
- 제 1항에 있어서, 표적 벡터에 의해 표적되는 상기 항원들은 HER/neu, CD19, CD20, VEGF, EGF 수용체, 알카라인 포스파타아제, 전립선 산 포스파타아제(prostatic acid phosphatase), 테나신(tenascin), 태반 성장 인자(P1GF), 인슐린류 성장 인자(ILGF) 및 갱글리오사이드(gangliosides)를 포함함을 특징으로 하는 방법.
- 제 19항에 있어서, 상기 단클론 항체는 심장혈관 병변, 아밀로이드 침적, 감염 기관들, 염증, 자가면역 질병들, 대체(displaced) 및 전이 정상 조직들, 유동성 암 또는 고형암을 표적할 수 있는 것임을 특징으로 하는 방법.
- 제 19항에 있어서, 상기 단클론 항체는 응고, 색전증, 동맥경화반(atherosclerotic plaques), 아밀로이드증(amyloidosis), 세균, 곰팡이, 리케차, 바이러스, 기생충, 류마티스 관절염, 전신성 홍반성 낭창, 다발성 경화증, 대체(displaced) 및 전이 부갑상선 조직, 대체(displaced) 및 전이 자궁내막 조직, 대체(displaced) 및 전이 비장 조직, 대체(displaced) 및 전이 흉선 조직, 백혈병, 림프종, 암종, 육종, 신경교종 또는 흑색종을 표적할 수 있는 것임을 특징으로 하는 방법.
- 제 1항에 있어서, 상기 표적 제제는 다가 항체 또는 다가, 다특이성 항체임을 특징으로 하는 방법.
- 제 35항에 있어서, 상기 다가 항체 또는 다가, 다특이성 항체는 악성 질병에 결합되는 것임을 특징으로 하는 방법.
- 제 35항에 있어서, 상기 다가 항체 또는 다가, 다특이성 항체는 심장혈관 질병에 결합되는 것임을 특징으로 하는 방법.
- 제 35항에 있어서, 상기 다가 항체 또는 다가, 다특이성 항체는 자가면역 질병에 결합되는 것임을 특징으로 하는 방법.
