Claims (21)
- a) 미성숙한 수상돌기 세포(DC)를 제공하는 단계;b) 하나 이상의 성숙 자극제가 있는 성숙 혼합물을 함유하고 있는 배양 배지에서 상기 미성숙 DC 를 배양하여 성숙한 DC 를 얻는 단계;c) 상기 성숙한 DC 를 어떠한 이종의 면역혈청도 함유되어 있지 않은 동결 배지에서 동결시키는 단계를 포함하는 즉시 사용가능한 동결된 성숙한 수상돌기 세포의 제조 방법.
- 제 1 항에 있어서, 상기 DC 동결 전 또는 동결된 DC 를 해동시킨 후, 상기 DC 에 항원 또는 항원-항체 컴플렉스를 로딩시키는 방법.
- 제 1 항 또는 제 2 항에 있어서,(i) 상기 미성숙한 DC 가 CD14+ 단핵 세포(단구 세포) 또는 CD34+ 세포로 부터 제조되고 ; 및/또는(ii) 상기 미성숙한 DC 가 혈액으로부터 직접 분리되고 ; 및/또는(iii) 상기 미성숙한 DC 또는 이의 전구 세포가 류카프레세이트(leucapheresates)로부터 얻어지고 ; 및/또는(iv) 상기 미성숙한 DC 또는 이의 전구 세포가 가공되지 않은 혈액 또는 골수로부터 얻어지는 방법.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 상기 성숙 자극제가 IL-1β와 같은 IL-1, IL-6, TNF-α, PGE2와 같은 프로스타글란딘, IFN-α, MPL(모노포스포릴리피드 A) 및 리포테이코산과 같은 리포다당류 및 기타 박테리아성 세포 생성물, 포스포릴콜린, 칼슘 이오노포레스, PMA 와 같은 포르볼 에스테르, 열-충격 단백질, ATP 와 같은 뉴클레오티드, 리포펩티드, Toll-유사 수용체에 대한 인공 리간드, 폴리-I:C 와 같은 이중 나선 RNA, 면역자극성 DNA 서열, CD40 리간드 등에서 선택되고, 상기 배양 배지 또는 상기 성숙 혼합물은 특히 IL-1β, IL-6, PGE2및 TNF-α를 함유하거나 또는 상기 성숙 혼합물은 단핵 세포-조건 배지(MCM) 또는 PGE2가 보충된 MCM 인 방법.
- 제 4 항에 있어서, 상기 미성숙 DC 가 0.1 내지 100 ng/ml 의 IL-1β, 0.1 내지 100 ng/ml 의 IL-6, 0.1 내지 10 ㎍/ml 의 PGE2및 0.1 내지 100 ng/ml 의 TNF-α의 존재 하에서 배양되는 방법.
- 제 4 항 또는 제 5 항에 있어서, 성숙을 위하여 DC 에 첨가된 활성 물질 IL-1β, IL-6, PGE2및 TNFα가 각 활성 물질의 정제된 조제물로부터 얻어지는 것을 특징으로 하는 방법.
- 제 1 항 내지 제 6 항 중 어느 한 항에 있어서,(i) 1 시간 이상, 바람직하게는 6 시간 이상 동안 성숙되고 ; 및/또는(ii) 성숙되는 동안, 배양 배지의 성숙 혼합물이 교체되고 및/또는, 추가의 성숙 자극제가 배양 배지에 첨가되고 ; 및/또는(iii) 면역/성숙 조절제, 특히 IL-10, 푸마르산 및 이의 에스테르, 미코페놀레이트 모페틸 및 비타민 D3 에서 선택된 조절제가 배양 배지에 첨가되는 방법.
- 전기한 항들 중 어느 한 항에 있어서, 상기 동결 배지가(i) 5 내지 25 % (v/v) 의 하나 이상의 동결보조제로서, 특히 DMSO, 글리세롤, 폴리비닐피롤리돈, 폴리에틸렌 글리콜, 알부민, 콜린 클로라이드, 아미노산, 메탄올, 아세트아미드, 글리세롤 모노아세테이트 및 무기염, 더욱 바람직하게는 DMSO 으로부터 선택되는 동결보조제 ; 및/또는(ii) 2 내지 30 % (w/v) 의 하나 이상의 폴리올 화합물로서, 특히 글루코스, 덱스트란, 수크로스, 에틸렌 글리콜, 에리트리톨, D-리비톨, D-만니톨, D-소르비톨, 이노시톨 및 D-락토스, 더욱 바람직하게는 글루코스로부터 선택되는 화합물 ; 및/또는(iii) 2 내지 90 % (w/v) 의 이종이 아닌 면역혈청 성분으로서, 특히 자원성면역혈청 또는 혈청 또는 동종이계 면역혈청 또는 혈청, 더욱 바람직하게는 인간 면역혈청 알부민, 자원성 면역혈청, 동종이계 인간 면역혈청 또는 풀 면역혈청을 함유하는 방법.
- 전기한 항들 중 어느 한 항에 있어서, 5 ×106내지 100 ×106세포/ml의 농도로 세포가 동결되는 것을 특징으로 하는 방법.
- 제 2 항 내지 제 9 항 중 어느 한 항에 있어서, 상기 DC 에 항원 또는 항원-항체 컴플렉스가 동결되기 전에 로딩되는 방법.
- 제 2 항 내지 제 10 항 중 어느 한 항에 있어서, DC 에 로딩된 상기 항원이 8 개 이상의 아미노산을 갖는 단백질 또는 이의 단편, 바람직하게는 배양 배지에 IL-1β, IL-6, PGE2및 TNFα가 함유되어 있을 때, 상기 단백질 또는 단편이 상기 배양 배지에 특히 0.01 내지 1000 μM 의 농도로 첨가되는 방법.
- 제 2 항 내지 제 10 항 중 어느 한 항에 있어서, 상기 DC 에 상기 항원을 코딩하는 DNA 또는 RNA 분자가 로딩되는 방법.
- 제 2 항 내지 제 10 항 중 어느 한 항에 있어서, 단백질 단편, 세포 또는 세포 단편 또는 핵산 서열이 상기 항원-항체 컴플렉스에서 항체로서 이용되고, 항체는 클래스 IgG 또는 IgE 중 하나인 방법.
- 제 1 항 내지 제 13 항 중 어느 한 항에 있어서, 상기 DC 가 동결되기 전 또는 재해동된 후 세포소멸을 억제할 수 있는 분자와 접촉되는데, 이 때 세포소멸을 억제할 수 있는 상기 분자는 바람직하게는 CD40 리간드, TRANCE 및 RANKL 로 이루어진 그룹으로부터 선택되는 방법.
- 제 1 항 내지 제 14 항 중 어느 한 항에 있어서, 동결된 DC 의 갯수를 기준으로 하여 75 % 이상, 바람직하게는 85 % 이상의 DC 가 DC 를 동결시키고 재해동 시킨 후에 생존하는 방법.
- 제 1 항 내지 제 15 항 중 어느 한 항에 있어서, 성숙한 동결저장된 DC 로부터 백신 제조에 적합한 방법.
- a) 미성숙한 DC 를 제공하는 방법;b) 상이한 성숙 자극제의 존재 하에서 미성숙한 DC 를 상이하게 배양하는 단계 ;c) 이후 시토킨이 존재하거나 존재하지 않는 배지에서 DC 배양하는데, 이 때 시토킨이 존재하지 않는 배양이 바람직한 단계 ;d) 시토킨이 존재하거나 존재하지 않는 상기 배지에서 배양한지 1 일이상이 지난 다음 생존 세포의 비율을 측정하는 단계; 및e) 가장 높은 생존율을 나타내는 성숙 자극제를 결정하는 단계를 포함하는, DC 의 동결에 유리한 조건을 찾는 방법.
- 제 17 항에 있어서, 시토킨이 존재하거나 존재하지 않은 채로 배양한지 2 일 이상, 바람직하게는 3 일 이상 지난 후에 생존 세포의 비율을 측정하는 방법.
- 항원이 로딩된 동결된 성숙한 수상돌기 세포.
- 제 19 항에 있어서, 제 1 항 내지 제 16 항 중 어느 한 항의 방법으로 얻을 수 있는 세포.
- 제 19 항 또는 제 20 항의 수상돌기 세포를 포함하는 백신.
