Abstract
본 발명은 하기 화학식 I의 화합물 또는 화학식 I의 카복실산의 약학적으로 허용가능한 염의 신규한 레티노이드 길항물질에 관한 것이다:

점선은 선택적으로, 점선이 존재하는 경우, R1은 저급 알킬이고, R2는 수소이며, 점선이 존재하지 않는 경우, R1및 R2는 함께 합쳐져서 메틸렌이 되어 시스-치환된 사이클로프로필 고리를 형성하고; R3는 하이드록시 또는 저급 알콕시이고; R4는 알킬 또는 알콕시이고; R5및 R6는 독립적으로 C4-12 알킬기, 또는 4급 탄소원자를 통해 페닐 고리에 연결된 일환 또는 다환의 C5-12 탄화수소기이다.

Description
레티노이드 수용체 작용 활성을 갖는 레티노이드는 피부 질환 및 종양 질환의 치료를 위한 모델 시스템에서 뿐만 아니라 보조 T 세포 제 1 형(Th1)으로 매개되는 면역계 질환의 치료를 위한 모델에서도 활성을 나타내는 것으로 알려져 있다. 레티노이드 수용체 작용 활성을 갖는 레티노이드는 진행성 다발성 관절염[Brinckerhoff 등, Science221, 756-758 (1983)] 및 실험적 알러지성 뇌척수염[Massacesi 등, J.Clin.Invest.88, 1331-1337 (1991); Racke 등, J.Immunol.154, 450-458 (1995)], 즉 각각 동물 모델의 류마티스성 관절염 및 다발성경화증의 치료에 활성을 갖는다. 이들 두 질환은 Th-1으로 매개되는 면역계 질환에 속하는 것으로 생각된다.
실험적으로, 레티노이드 수용체 길항 활성을 갖는 레티노이드(레티노이드 길항물질)는 세포 증식의 억제, 세포 분화의 유도, 세포소멸의 유도 및 혈관형성의 억제와 같이 레티노이드 수용체 작용 활성을 갖는 레티노이드(레티노이드 작용물질)의 많은 성질을 방해하는데 효과적이다[Bollag 등, Int.J.Cancer70, 470-472 (1997)]. 또한 레티노이드 길항물질은 레티노이드 작용물질의 독성 부작용, 즉 비타민 A 과다증후군 및 기형발생의 징후 및 증상을 억제한다[Standeven 등, Toxicol. 레티노이드 길항물질은 알러지성 기관지 천식의 치료에 효과적이다. 천식과 관련된 염증의 특징은 활성화된 호산구, 기도의 감수성 증가(과잉반응), 부종, 점액질 과다분비 및 기침의 존재에 있다. 염증 과정은 Th2 유형 세포의 생성 및 활성에 의해 매개된다. 레티노이드 길항물질의 Th1 유형 반응의 촉진 및 이로 인한 Th2 유형 반응의 억제능은, 알러지성 폐 염증/천식에 있어서 이들 화합물의 효능의 기초를 이루는 메카니즘으로 생각된다. 레티노이드 길항물질은 Th1 유형 세포에 작용하여 알러지성 폐 염증/천식의 징후 및 증상을 억제한다[Gavett 등, J.exp.Med. 182, 1527-1536 (1995); Kips 등, Am.J.Respir.Crit.Care Med.153, 535-539 (1996)]. 이들은 항원/알러지항원(예; 난백알부민)에 감작되고 항원투여된 동물에 있어 활성을 나타낸다. 전신 투어 또는 에어로졸에 의해 국소 투여되는 레티노이드 길항물질은 비특이성 자극물에 대한 기도의 과잉반응 뿐만 아니라 기관지 수축, 기도 부종 및 점액 과다분비, 기도 염증, 기관지-폐포 조직내 및 기관지-폐포 세정시의 호산구 및 호중구의 축적을 감소시키거나, 억제하거나 또는 역전시키는데 효과적이다.

본 발명의 세 번째 태양에 있어서, 레티노이드 길항물질, 이들의 약학적으로 허용가능한 염 및 이들의 약학적으로 허용가능한 가수분해성 에스테르의 투여는 골다공증을 앓는 환자를 치료하는데 효과적인 것으로 밝혀졌다.
그러므로, 이러한 태양에서 본 발명은 골다공증의 치료를 위한 약물을 제조하기 위한 레티노이드 길항물질, 이들의 약학적으로 허용가능한 염 및 이들의 약학적으로 허용가능한 가수분해성 에스테르의 용도에 관한 것이다.

본원에 사용된 용어 "골다공증"은 일차 뿐만 아니라 이차 골다공증을 포함하며, 낮은 골질량 및 골조직의 미세구조상의 악화로 인해 골 약화 및 이로 인한 골절 위험의 증가를 특징으로 하는 질환에 관한 것이다.

레티노이드 길항물질이 상기 언급된 재흡수 자극 화합물에 의해 유도된 골 흡수를 억제한다는 것이 입증되었다. 이것은 비포스포네이트와 같은 골다공증의 임상적 예방 및 치료에 유용한 제제를 사용할 수 있는 것으로 예견되는 광범위하게 사용되는 신생 마우스의 두개골(머리뼈) 모델에서 증명되었다. 실험은 하기 실시예 7에서 기술한다.
본 발명의 네 번째 태양은 피부 및 점막의 전암성 병변, 또는 피부, 점막, 두부 및 목, 폐, 위, 결장, 유방, 난소, 경관 및 전립선의 고형 종양과 같은 전암성 및 종양성 질환의 치료에 사용하고, 이러한 질환의 치료용 약제를 제조하기 위한, 선택적인 레틴산 수용체(RAR) 길항활성, 레티노이드 X 수용체(RXR) 길항 활성, 또는 혼합된 RAR-RXR 길항 활성을 갖는 레티노이드를 포함하는 레티노이드 길항물질의 용도에 관한 것이다(하기 실시예 8 참조).
상기 언급한 질환의 치료를 위하여, 활성 화합물, 즉 레티노이드 길항물질, 이들의 약학적으로 허용가능한 염 및 이들의 약학적으로 허용가능한 가수분해성 에스테르를 전신적으로 또는 국소적으로 투여한다. 바람직하게는, 상기 화합물을 상기 활성 화합물 및 상기 활성 화합물과 상용가능한 약학적으로 허용가능한 담체 또는 희석제를 함유하는 조성물로서 투여한다. 이러한 조성물을 제조함에 있어서, 임의의 통상의 약학적으로 허용가능한 담체를 사용할 수 있다. 약물을 경구 투여하는 경우, 일정한 간격을 두고 식사시간에 편리하게 투여하거나 또는 하루에 일회 투여하는게 일반적이다. 상기 화합물의 효과적인 투여량은 경구 투여 또는 국소 투여시 부작용이 전혀 없거나 약간 있는 정도로 정해져있다. 그러므로, 활성 화합물의 경구 또는 국소 투여가 일반적으로 바람직하다. 피부, 입, 코, 인두, 후두, 기관지 등의 질환을 치료함에 있어서, 경구 투여와 함께 국소 투여하는 것이 또한 유리할 수 있다.
상기 질환들을 치료함에 있어서, 레티노이드 길항물질은 경구 투여시 비타민 A 과잉증의 독성 증후에 속하는 역효과, 예를 들어 점액피부계, 근골격계, 신경계 징후 및 트랜스아미나제, 트리글리세라이드 및 콜레스테롤의 상승을 나타내지 않거나 약간 나타낸다. 또한, 전체-트랜스 레틴산(트레티노인), 13-시스 레틴산(이소트레티노인), 에트레티네이트 및 아시트레틴과 같이 피부 질환 및 종양 질환의 치료에 임상적으로 유용한 수용체 작용활성을 갖는 레티노이드에 비해 이들은 기형발생을 일으키지 않거나 약간 일으킨다.
보조 T세포 제 2 형으로 매개되는 면역계 질환을 치료함에 있어서, 레티노이드 길항물질, 이들의 약학적으로 허용가능한 염 및 이들의 약학적으로 허용가능한 가수분해성 에스테르는 단독으로 사용되거나 또는 기타 수단들과 혼합하여 사용될수 있는데, 예를 들면, 국소 또는 전신성 코르티코스테로이드, 항히스타민제 및 기관지 확장 제제와 같은 기타 약학적으로 허용가능한 활성 물질과 함께 혼합하여 사용할 수 있다. 기타 물질과 혼합하여 사용하는 경우, 레티노이드 길항물질 및 상기 기타 물질을 따로 투여하거나 또는 효과량을 하나의 약학 조성물로 혼입하여 투여할 수 있다.
골다공증의 치료에 있어서, 레티노이드 길항물질, 이들의 약학적으로 허용가능한 염 및 이들의 약학적으로 허용가능한 가수분해성 에스테르를 단독으로 사용하거나 또는 기타 수단과 혼합하여 사용할 수 있는데, 예를 들면, 칼슘, 비타민 D 유도체, 에스트로겐, 단백동화제, 칼시토닌 또는 비포스포네이트와 같은 기타 약학적 활성 물질과 혼합하여 사용할 수 있다. 기타 물질들과 혼합하여 사용하는 경우, 레티노이드 길항물질 및 상기 기타 물질을 따로 투여하거나 또는 효과량을 하나의 약학 조성물로 혼입하여 투여할 수 있다.
본 발명에 따라, 레티노이드 길항물질은 그의 약학적으로 하용가능한 가수분해성 에스테르의 형태로 투여될 수 있다. 임의의 약학적으로 허용가능한 가수분해성 에스테르를 본 발명의 조성물 및 방법에서 사용할 수 있다. 이중 바람직한 에스테르는 벤질 잔기가 치환되지 않거나 또는 저급 알킬, 할로, 니트로, 티오 또는 치환된 티오로 치환된 벤질 에스테르와 같은 방향족 에스테르; 또는 에틸, t-부틸, 사이클로펜틸, 사이클로헥실 또는 사이클로헵틸 에스테르, 또는 9-플루오레닐메틸 에스테르와 같은 저급 알킬 에스테르이다.
상기 언급한 레티노이드 길항물질, 이들의 염 및 이들의 에스테르는 특히 약학적으로 허용가능한 경구 또는 국소 방식으로 사용된다. 이들 약학적 조성물은 상기 활성 화합물 및 이들과 혼합된 상용가능한 약학적으로 허용가능한 담체 물질을 포함한다. 임의의 통상적인 담체 물질을 사용할 수 있다. 담체 물질은 경구 투여에 적합한 유기 또는 무기 불활성 담체 물질일 수 있다. 적합한 담체는 물, 젤라틴, 아라비아 고무, 락토오즈, 전분, 마그네슘 스테아레이트, 활석, 식물성 오일, 폴리알킬렌-글리콜, 바셀린 등을 포함한다. 더욱이, 약학적 활성 제제는 기타 약학적 활성제를 포함할 수 있다. 또한, 향신제, 방부제, 안정화제, 유화제, 완충제와 같은 첨가물을 약학적 배합의 허용된 방법에 따라 첨가할 수 있다.
약학적 제제는 임의의 통상적인 형태로 제조될 수 있으며, 특히 다음과 같은 형태를 포함한다: (a) 정제, 캡슐(예, 경질 또는 연질의 젤라틴 캡슐), 환제, 향낭(sachet), 분말, 과립 등과 같은 경구 투여용 고형 형태; (b) 용액, 현탁액, 연고, 크림, 겔, 미세분말, 스프레이, 에어로졸 등과 같은 국소 투여용 제제. 약학적 제제는 멸균될 수 있고/있거나 방부제, 안정화제, 습윤제, 유화제, 삼투압을 변화시키기 위한 염 및/또는 완충제와 같은 아쥬반트를 포함할 수 있다.
피부 또는 점막에 국소 투여하기 위하여, 상기 언급한 유도체를 연고, 팅크제, 크림, 겔, 용액, 로션, 스프레이; 흡입용 에어로졸 및 건조 분말, 현탁액, 샴푸, 두발용 비누, 향수 등으로 제조하는 것이 바람직하다. 사실상, 임의의 통상적 조성물을 본 발명에 사용할 수 있다. 본 발명의 제제를 포함하는 조성물을 적용하는 바람직한 방법에는 연고, 겔, 크림, 로션, 스프레이; 흡입을 위한 에어로졸 또는 건조 분말의 형태가 있다. 피부로의 국소 투여를 위한 약학적 제제는 상기 언급한 활성 성분을 비독성이고, 치료요법적으로 불활성이며, 고형 또는 액상의 이러한 제제의 제조에 흔히 사용되는 담체와 함께 혼합하여 제조할 수 있다. 이들 제제는 조성물의 총 중량을 기준으로 일반적으로 0.01 내지 5.0 중량%, 바람직하게는 0.1 내지 1.0 중량%의 활성 성분을 포함한다.
상기 기술한 국소용 제제를 제조함에 있어서, 국소 제제의 약학적 배합 기술분야에서 통상적인 방부제, 농후화제, 향수 등과 같은 첨가제를 사용할 수 있다.
또한, 상기 언급한 활성제를 함유하는 국소 제제에 통상의 산화방지제 또는 통상의 산화방지제의 혼합물을 혼입할 수 있다. 이들 제제에 사용될 수 있는 통상의 산화방지제에는 N-메틸-α-토코페롤아민, 토코페롤, 부틸화된 하이드록시아니솔, 부틸화된 하이드록시톨루엔, 에톡시퀸 등이 포함된다. 본 발명에 따라 사용되는 활성제를 함유하는 크림을 기본으로 한 약학적 배합물은 지방산 알콜, 반고형 석유계 탄화수소, 에틸렌 글리콜 및 유화제를 포함하는 수성 에멀젼으로 구성된다.
본 발명에 따른 활성제를 포함하는 연고 배합물은 활성 물질의 용매 분산물과 반고형의 석유계 탄화수소의 혼합물을 포함한다. 본 발명에 사용하기 위한 활성 성분을 포함하는 크림 조성물은 습윤제, 점도 안정화제 및 물의 수성상, 지방산 알콜, 반고형 석유계 탄화수소 및 유화제의 유성상, 및 수성 안정화제-완충 용액내에 분산된 활성제를 포함하는 상으로부터 형성되는 에멀젼을 포함하는 것이 바람직하다. 안정화제는 국소 제제에 첨가될 수 있다. 통상의 안정화제를 본 발명에 사용할 수 있다. 유성상에서, 지방산 알콜 성분은 안정화제로서 기능한다. 이들 지방산 알콜 성분은 14 개 이상의 탄소원자를 함유하는 긴 사슬의 포화 지방산을 환원시키므로써 유도할 수 있다. 또한, 두발용 국소 제제에 일반적으로 사용되는 통상의 향수 및 로션을 본 발명에 사용할 수 있다. 더욱이, 필요하다면, 통상의 유화제를 본 발명의 국소 제제에 사용할 수 있다.
알러지성 비염 및 알러지성 기관지 천식의 국소 치료를 위해, 경비용 및 흡입용 에어로졸을 사용한다. 이러한 에어로졸을 위한 배합물은 문헌[Drugs and Pharmaceutical Science, Marcel Dekker, New York, 1996, Vol.72, pp 547-574]에 기술되어 있다. 또한, 활성 화합물은 건조 분말 흡입에 의해 전달될 수 있다. 이러한 배합물 및 장치는 문헌[Pharmaceutical Technology, June 1997, pp 117-125]에 기술되어 있다.
바람직한 경구 투여물의 형태는 정제, 환제, 향낭, 또는 경질 또는 연질 젤라틴, 메틸셀룰로즈 또는 소화관내에서 용이하게 용해되는 기타 적합한 물질로 된 캡슐을 포함한다. 각 정제, 환제, 향낭 또는 캡슐은 바람직하게는 약 5 내지 약 200 mg, 더욱 바람직하게는 약 20 내지 약 100 mg의 활성 성분을 포함할 수 있다. 본 발명에 따라 의도된 경구 투여량은 환자의 필요에 따라 주치의에 의해 결정되기 때문에 달라질 수 있다. 그러나, 일일 투여량은 일반적으로 0.05 내지 20 mg/환자체중 kg, 바람직하게는 0.1 내지 7 mg/환자체중 kg, 가장 바람직하게는 약 0.3 내지 약 1.5 mg/환자체중 kg으로 사용된다. 이러한 투여량은 환자의 필요사항에 따라 주치의가 정하는 임의의 투여 스케줄에 따라 투여될 수 있다.
치료 투여량은 통상적으로 투여 경로, 환자의 연령, 체중 및 질환 상태에 따라 달라진다. 적합한 투여형은 당해 기술 분야에 공지되어 있으며, 공지된 방법으로 용이하게 얻을 수 있다. 본 발명의 범주에 특히 적합하거나 또는 상기 기술내용에 따라 용이하게 조절할 수 있는 로션, 겔, 크림, 스프레이; 흡입용 에어로졸 및 건조분말, 경질 또는 연질 젤라틴 캡슐, 정제 및 향낭의 배합물이 당해 기술분야에 있다.
상기 개시한 바와 같은 레티노이드 길항물질의 약리학적 활성은 하기 실시예 1 내지 8에 나타낸 것과 같은 다양한 시험 모델에서 설명될 수 있다. 실시예 1 내지 8에서 하기의 화합물을 사용하였다:

실시예 2
털없는 래트에서 전체-트랜스 레틴산에 의해 야기되는 피부 자극에 미치는 억제 효과
매일 일회씩, 일주일에 5일씩(월요일부터 금요일까지), 연속 4주동안 실험 동물의 피부 위를 아세톤/에탄올(1:1)내의 시험 화합물의 혼합 용액으로 처리하였다(0.025 ml/2cm2).
실험을 시작한지 약 24시간 후부터 동물의 각 시험 부위에서 홍반 및 부종의 징후를 매일 월요일부터 금요일까지 관찰하였다. 유도된 모든 피부 반응의 강도를 하기 등급 기준에 따라 데이터 시이트에 기록하였다.
0= 피부 반응 없음
1= 약간의 피부 반응(약간 내지 뚜렷한 홍반)

실시예 3
다양한 레티노이드의 시험관내 기형발생 활성에 미치는 억제 효과
13일된 래트 태아의 앞다리 아 및 뒷다리 아를 37℃에서 0.1% 트립신 및 0.1% EDTA를 함유하는 칼슘-마그네슘 없는 균형잡힌 염 용액중에서 분리하였다. 세포 밀도는 10% NU-혈청을 함유하는 CMRL 배지중에서 2x107다리 아 세포/ml로 조절하였다. 24-웰 접시의 각 웰의 중심에 20㎕의 세포 현탁액을 불연속적인 방울로서 분배하므로써 준비하였다. 배양물을 적절한 배양 배지로 잠기게 하기 전에 37℃에서 1 내지 2 시간동안 세포를 붙어있도록 하였다. 이어서 시험 물질을 디메틸설폭사이드에 용해시키고 배양 1일째 동일한 시간에 첨가하였다. 동량의용매(0.4%)를 대조 배양물에 첨가하였다. 배양 7일 후, 간엽세포의 연골세포(연골 생성)로의 분화 등급을 측정하기 위하여 프로테오글리칸을 알시안청으로 착색하였다. 결합된 염료를 4M의 구아니딘 하이드로클로라이드로 추출하고 600 nm에서의 흡광도를 분광광도법으로 측정하였다.
도 3은 다양한 농도의 화합물 A와 일정한 농도의 전체-트랜스 레틴산(화합물 C)의 상호작용을 보여준다. C의 농도는 실험동안 1x10-6mol/l로 일정하게 유지하였다. 화합물 A의 농도는 1x10-9mol/l에서 2x10-6mol/l로 증가시켰다. 화합물 A의 농도가 증가함에 따라 간엽세포의 분화가 약 5%에서 약 30%로 증가하였다.
도 4는 다양한 농도의 화합물 A(1x10-9mol/l 내지 2x10-6mol/l)와 화합물 D의 상호작용을 보여주며, 화합물 D의 농도는 배양시키는 동안 1x10-6mol/l로 일정하게 유지하였다. 화합물 A의 농도가 증가함에 따라 간엽세포의 분화가 약 30%에서 약 70%로 증가하였다.
도 5는 다양한 농도의 화합물 A(1x10-9mol/l 내지 2x10-6mol/l)와 화합물 E의 상호작용을 보여주며, 화합물 E의 농도는 배양시키는 동안 5x10-7mol/l로 일정하게 유지하였다. 화합물 A의 농도가 증가함에 따라 간엽세포의 분화가 약 5%에서 약 80%로 증가하였다.
도 6은 다양한 농도의 화합물 A(1x10-9mol/l 내지 2x10-6mol/l)와 화합물 F의 반응을 보여주며, 화합물 F의 농도는 배양시키는 동안 1x10-8mol/l로 일정하게 유지하였다. 화합물 A의 농도가 증가함에 따라 간엽세포의 분화가 약 5%에서 약 75%로 증가하였다.
도 7은 다양한 농도의 화합물 A(1x10-9mol/l 내지 2x10-6mol/l)와 화합물 G의 반응을 보여주며, 화합물 G의 농도는 배양시키는 동안 1x10-6mol/l로 일정하게 유지하였다. 화합물 A의 농도가 증가함에 따라 간엽세포의 분화가 약 5%에서 약 110%로 증가하였다.
실시예 4
레티노이드 길항물질에 의한 IL-12 생성 유도의 시험관내 분석시험
THP-1 세포를 아메리카 조직 배양물 컬렉션(American Tissue Culture Collection)으로부터 구입하고 완전 배지에서 배양하였다. IL-12 생성을 분석하기 위하여, 1.25x106세포/ml의 THF-1 세포를 에스.아우레우스 코완 균주(S.aureusCowan strain)(SAC) 및 인간 재조합 인터페론-γ(huIFN-γ)(1000 U/ml)로 자극하였다[Ma 등, J.Exp. Med,183, 147-157 (1996)].
이와는 다르게, 37℃에서 16시간동안 0.5x106인간 말초혈 단핵세포(PBMC)(48 웰 플레이트에 1 ml의 배양물)를 huIFN-γ(1000 U/ml)로 프라임(prime)시키고, 이어서 SAC(1/1000)으로 자극하였다. 48 시간 후에 상청액을 수집하고 분석시험 전까지 -20℃에서 얼렸다[Panina-Bordignon 등,J.Clin.Invest.100, 1513-1519 (1997)].
IL-12 생성은 코팅 완충액중의 2.5 ㎍/ml의 20C2 항체(래트 항 인간 IL-12 이질이량체 p40-p35), 및 기술한 바와 같은 분석 완충액중의 250 ng/ml의 퍼옥시다제-결합된 4D6항체(래트 항 인간 IL-12)를 사용하여 특정 효소 연결 면역 수착제 분석법(enzyme linked immuno sorbant assay)(ELISA)으로 측정하였다[Zhang 등, J.Clin.Invest.93, 1733-1739 (1994)]. 분석 완충액으로 희석된 표준(재조합 인간 IL-12, 800 pg/ml 내지 6 pg/ml) 및 샘플(100㎕)을 이중 웰에 첨가하였다. 흡광도는 450 내지 650 nm에서 읽었다. 샘플의 IL-12 미지 농도는 상응하는 표준 곡선으로부터 읽고 상응하는 희석 인자를 곱하였다. 최대 IL-12 생성은 200 내지 400 pg/ml로 다양했다.
동결건조된 레티노이드 길항물질을 황색광 하에서 DMSO 내에서 희석시켜, 얼음 위에서 2 mM의 농도를 나타내게 하였다. 완전 RPMI 배지내에서 일련의 희석물(1μM 내지 1 pM)을 제조하였다. 각 희석물의 10㎕를 1 ml의 배양물에 첨가하였다.
실험 결과는 시험된 레티노이드 길항물질이 IL-12 생성에 영향을 미치는 것으로 나타났다. 특히, 시험된 레티노이드 길항물질은 활성화된 인간 단구에 의한 IL-12 생성을 자극한다. 표 2 및 표 3 참조한다. 레티노이드 길항물질은 특히 활성화된 단구에 의한 IL-12 생성을 증가시킨다.

실시예 5
레티노이드 길항물질에 의한 인간 고유의 T 세포의 보조T세포 2(Th2)으로의 분화억제에 관한 실험관내 분석
코드 혈(cord blood)로부터의 고유의 T세포를 분리하고 문헌[Panina-Bordignon 등, J.Clin.Invest.100, 1513-1519 (1997)]에 기술한 바와 같이 처리하였다. 간단하게, 단핵세포로부터 유도한 코드혈을 항-CD45RA 및 항-CD-4 단클론성 항체와 함께 배양하였다. 20분간의 배양 후에, 세포를 세척하고, 고트 항-마우스 Ig로 피복된 자기구슬과 함께 배양하였다. 양성 세포를 분리하고, 1mM의 화합물 H 또는 화합물 B의 존재 또는 비존재하에서 자가 접착성 세포인 PHA 및 IL-4와 함께 5일동안 24웰 플레이트내에서 1 x 106세포/ml로 접종하였다. 10일 후에, 세포를 수집하고, PMA(50ng/ml) 및 이오노마이신(1㎍/ml)으로 4시간동안 재자극하였다. 마지막 2시간동안 브레펠딘 A(Brefeldin A)를 첨가하였다. 이어서, 세포를 4%의 파라포름알데히드로 고정시키고 사포닌으로 투과성을 갖도록 하였다.고정된 세포를 FITC-항 IFNγ 및 PE-항-IL-4mAbs로 착색시키고 세포형광측정 분석법(cytofluorimetric analysis)으로 분석하였다.

실시예 6
알러지항원으로 유도된 기도 염증 및 과잉반응증의 쥐 모델
0 일 및 14일 째 C57BL/6 마우스(생후 8 내지 9 주)에 0.2 ml의 멸균 염수내의 10㎍ OA + 1 mg Al(OH)3(겔 현탁액)을 복강내 주사하므로써 상기 마우스를 난백알부민(OA)에 매우 민감해지도록 하였다. 21 일째, 상기 마우스를 18분동안 5.0% OA 에어로졸로 항원투여(challenge)하였다. 상기 에어로졸은 드 빌비스 울트라-넵 90 (De Vilbiss Ultra-Neb 90) 초음파 분무기에 의해 생성하였으며, 상기 분무기의 출구는 상기 동물이 들어있는 작은 플렉시글래스 상자로 연결되어 있다. 3일동안 매일, 48 시간, 24 시간, 및 OA 항원투여 직전에 상기 마우스에 RXR 길항 화합물 B(복강내로 10 및 30 mg/kg)를 투여하였다. 동물은 21일째에 사용하였다.
기도 염증성 세포 축적: 24일째, OA 에어로졸로 항원투여한 후 3일 후, 상기 동물을 우레탄(2.4g/kg)으로 마취시키고 23 게이지 카테터로 기관절개하였다. Ca++및 Mg++이 없는 멸균된 행크 균형 염 용액의 분취량(2x1 ml)으로 폐를 세정하였다. 세정액을 30 초후에 부드럽게 흡인하여 회수하고 각 동물에 대해 모아 두었다. 이어서 5℃에서 샘플을 2000 rpm으로 15 분동안 원심분리하였다. 0.5 ml의 증류수를 사용하여 생성된 펠릿으로부터 적혈구를 용해해내고, 펠릿에 남아있는 세포는 5 ml의 HBSS로 재구성하였다. 샘플을 5℃, 2000rpm에서 15분동안 이차 원심분리하였다. 생성된 펠릿을 1 ml의 HBSS에 분산시켰다. 세포 현탁액의 분취액으로부터 혈구계를 사용하여 총 세포 수를 계산하였다. 세포학적 연구용 준비를 위해, 상기세포를 개질된 라이트 스테인(Wright's stain)으로 착색된 세포원심분리된 슬라이드상에 고정시켰다. 세포를 분류하기 위하여 표준의 형태학적 기준을 사용하여 300 개 이상의 세포 상에 차등적인 계수를 수행하였다.
실험 결과는 시험된 레티노이드 길항물질이 알로젠으로 유도된 기도 염증성 세포의 축적을 억제하는 것으로 나타났다(표 5).

기도 과잉반응증
24일째, OA 에어로졸로 항원투여된지 3일 후에, 동물들을 펜토바비탈 나트륨(100mg/kg, 복강내)로 마취시키고 기관절개하였다(PE-190). 정맥내 약물 전달을 위해 경정맥내에 실라스틱 튜브(sylastic tubing)로 캐뉼라 삽입하였다. 동물을 내장형 호흡속도묘사기가 설치된 전신 혈량계 내에 눕히고, 기계적으로 환기시키고(Vf=150/분, Vt=0.3ml; 모델683, Havard Apparatus, S.Natic, MA) 즉시 이어서 판큐로늄 브로마이드(0.1 mg/kg, 정맥내)로 처리하였다. 미분 압력 변환기(Validyne DP 103-08, Northridge, CA)를 사용하여 호흡량 신호를 통합하므로써 주기적인 부피를 얻었다. 미분 압력 변환기(Validyne DP 45-30, Northridge, CA)를 사용하여 기관내 압력과 흉강내 압력(늑간 공간에 삽입된 캐뉼라로부터 얻음) 사이의 차이로서 폐통과(transpulmonary) 압력을 측정하였다. 메타콜린 투여량의 증가(30, 100, 300, 1000 ㎍/kg, 정맥내)에 대한 폐 저항성의 변화(cm H2O/ml/s)를 폐통과 압력, 주기적 부피, 및 호흡량 측정치로부터 변조장치 신호처리 시스템(Malvern, PA)을 이용하여 계산하였다.
실험의 결과는 레티노이드 길항물질이 알러지성 기도염증을 예방하거나 또는 반전시킬 수 있고, 알러지성 기관지 천식과 같이 알러지성 기도 질환에 전형적인, 항원으로 유도되는 기관지 수축을 억제할 수 있는 것으로 나타났다.

골 흡수에 미치는 레티노이드 길항물질의 효과의 실례
골 흡수 분석
일련의 제제는 골 흡수를 유도하거나 자극하는 것으로 알려져 있다. 본 분석실험에서는 골 흡수 유도체의 골 흡수 활성을 억제하거나 방해할 수 있는 레티노이드 길항물질의 능력을 시험하였다.
재료 및 방법
골 흡수는 조직 배양 시스템내에서 신생마우스의 두개골(머리뼈)로부터 상청액 배지로의 칼슘 방출의 양을 재므로서 측정하였다. 두개골은 시간을 조절하여 w짝짓기를 시킨 스위스 알비노 마우스의 생후 4일된 마우스(체중 4 내지 4.5 kg)로부터 준비하였다. 전두부 및 두정부 두개골 부분을 입체현미경 하에서 절개하고, 중앙 봉합부를 따라 이등분하였다. 기체와 배지의 경계부에서 골을 지지하기 위한 스테인레스 스틸 격자판을 갖는 6-웰 배양 접시에 모든 동물의 두개골 절반을 무작위로 분배하였다. 1 mg/ml의 BSA(소혈청 알부민, SIGMA)가 보충되는 특정 배합에 따른 BGJ 배지(Bioconcept, 스위스)에서 상기 조직을 배양시켰다. 두개골을 37℃, 5% CO2및 95% 공기로 구성된 가습 대기하에서 배양하였다. 24시간동안의 예비배양 후에, 이들을 1.7 ml의 신선한 배지 및 시험 물질을 포함하는 새로운 접시에 옮겨놓았다. 골 흡수 유도체 및 레티노이드 길항물질을 단일 제제로서 또는 혼합물로서 시험하였다. 배양은 72시간동안 계속되었다.
배양 상청액내의 칼슘 농도는 각 배양 기간 직후에 측정하였다. 메틸티몰 블루를 함유하는 키트(Biomerieux)를 사용하여 안정한 유리 칼슘을 25㎕ 샘플에서 분광광도법으로 측정하였다[E.M.Gindler와 J.D.King, Am.J.Clin.Pathol, 1972, 58, 376-382]. 재흡수 반응은 3일간 처리하는 동안 절반의 두개골당 방출되는 칼슘의 ㎍으로 표시하였다.
두개골내에 남아있는 칼슘을 측정하기 위한 추가의 실험을 수행하였다. 칼슘을 5% 트리클로로아세트산(TCA)으로 추출하고, NaOH로 중화시킨 후 50㎕ 샘플에서 측정하였다. 또한, 세포독성을 배제하기 위하여 골조직 생존력을 측정하였다. 이것은 비색분석 MTT 테트라졸륨 시험[T.Mosmann, J.Immunol.Methods 1983,65, 55-63]에 의해 정량화하였다.
이들 시험에서 레티노이드 길항물질 H 및 B를 사용하였다.

다양한 제제에 의해 유도되는 골 흡수는 절반의 두개골로부터 상청액 배지로 방출되는 칼슘의 양을 측정하므로써 알 수 있었다. 칼슘 방출은 1.7 ml의 배지내에서 절반의 두개골 당 ㎍으로 나타냈다. 두개골로부터 배지로의 방출뿐만 아니라 배지로부터 두개골로의 흡수도 또한 측정할 수 있다. 골 흡수 유도체의 활성을 억제하는 레티노이드 길항물질의 효능은 억제율%로 표시한다. 골 흡수 유도체 단독에 의해 방출된 칼슘과 골 흡수 유도체 및 레티노이드 길항물질과의 혼합물에 의해 방출된 양 사이의 차이값으로부터 계산하였다. 레티노이드 길항물질 단독으로는 칼슘 방출과 섭취를 모두 유도할 수는 없다. 비히클 대조 시험에서의 기초 재흡수율은 5% 또는 그 미만이었다.
결과
실험의 결과(표 6 내지 11)는 레티노이드 길항물질 H 및 B가 6개의 상이한제제에 의해 유도되는 골 흡수를 방해함을 나타낸다. 후자는 포유동물 및 인간에서 골다공증 및 골절의 원인인 골 흡수에 기여하는 것으로 생각된다. 표 6 내지 11로부터 알 수 있는 바와 같이, 6가지의 골 흡수 제제 모두가 투여량에 의존적인 방식으로 두개골로부터 상청액 배지로의 칼슘 방출을 유도하였다. 레티노이드 길항물질은 이들 6가지 골 흡수 제제에 의해 유도되는 칼슘 방출을 억제할 수 있었다.

실시예 8
피부의 전암성 병변(다중 광화학선작용 케라토스)의 치료에서 화합물 B의 국소 적용의 효과
임상적 시도에서, 1%의 화합물 B를 함유하는 크림을 폐쇄적 드레싱을 하지 않고 하루에 두 번씩 병변 부위에 적용하였다. 결과는 표 12에 나타냈다. 표 12에서 볼 수 있는 바와 같이, 화합물 B를 국소적으로 투여하는 경우 피부의 전암성 병변의 치료에 효과적이었으며, 다른 레티노이드에 비해 병변 부위 및 주변 피부의 자극을 유발함이 없이 잘 받아들여졌다.

화학식 XVIII의 화합물은 신규하며 또한 본 발명의 목적이기도 하다. 화학식 XVIII의 화합물은 하기 반응식 1 및 반응식 2에 기술된 바대로 제조할 수 있다:

0℃에서 100ml의 디메틸포름아미드(DMF)중의 13g의 2-브로모-4,6-디-t-부틸페놀 용액을 100ml의 DMF중의 2.2g의 나트륨 하이드라이드(광유내 50%) 현탁액에적가하였다. 반응 혼합물을 수소가 더 이상 발생하지 않을 때가지 실온에서 교반하고, 다시 0℃로 냉각시키고, 50ml의 DMF 내의 22g 의 에틸 요오다이드 용액으로 처리하였다. 실온에서 약 1 시간동안 교반시킨 후, 반응 혼합물을 얼음물에 부어넣고, 에틸아세테이트로 추출하고, 세척하고(H2O), 건조시키고(Na2SO4), 증발시켜 13.5 g의 1-브로모-2-에톡시-3,5-디-t-부틸-벤젠을 백색 결정으로 얻었다. 융점: 55 내지 56℃.

Claims (29)
- 하기 화학식 I의 화합물 또는 화학식 I의 카복실 산의 약학적으로 허용가능한 염:화학식 I상기식에서,점선은 선택적으로; 점선이 존재하는 경우, R1은 저급 알킬이고, R2는 수소이며, 점선이 존재하지 않는 경우, R1및 R2는 함께 합쳐져서 메틸렌이 되어 시스-치환된 사이클로프로필 고리를 형성하고;R3는 하이드록시 또는 저급 알콕시이고;R4는 알킬 또는 알콕시이고;R5및 R6는 독립적으로 C4-12 알킬기, 또는 4급 탄소원자를 통해 페닐 고리에 연결된 일환 또는 다환의 C5-12 탄화수소기이다.
- 제 1 항에 있어서,하기 화학식 Ia의 화합물 또는 화학식 Ia의 카복실산의 약학적으로 허용가능한 염:화학식 Ia상기식에서,R1은 저급 알킬이고;R3내지 R6는 제 1 항에서 정의한 바와 같다.
- 제 2 항에 있어서,(2E,4E,6Z)-7-[3,5-디-t-부틸-2-에톡시페닐]-3-메틸-2,4,6-옥타트리엔산인 화합물.
- 제 2 항에 있어서,(2E,4E,6Z)-7-[3,5-디-t-부틸-2-부틸옥시페닐]-3-메틸-2,4,6-옥타트리엔산인 화합물.
- 제 2 항에 있어서,(2E,4E,6Z)-7-[3,5-디-t-부틸-2-부틸옥시페닐]-3-메틸-2,4,6-옥타트리엔산 에틸 에스테르,(2E,4E,6Z)-7-[3,5-디-t-부틸-2-메톡시페닐]-3-메틸-2,4,6-옥타트리엔산,(2E,4E,6Z)-7-[3,5-디-t-부틸-2-헥실옥시페닐]-3-메틸-2,4,6-옥타트리엔산, 및(2E,4E,6Z)-7-[3,5-디-t-부틸-2-옥틸옥시페닐]-3-메틸-2,4,6-옥타트리엔산중에서 선택된 화합물.
- 제 1 항에 있어서,하기 화학식 Ib의 화합물 또는 화학식 Ib의 카복실산의 약학적으로 허용가능한 염:화학식 Ib상기식에서,R3내지 R6는 제 1 항에서 정의한 바와 같다.
- 제 6 항에 있어서,(2E,4E)-(1RS,2RS)-5-[2-(3,5-디-t-부틸-2-에톡시-페닐)-사이클로프로필]-3-메틸-펜타-2,4-디엔산 에틸 에스테르,(2E,4E)-(1RS,2RS)-5-[2-(3,5-디-t-부틸-2-에톡시-페닐)-사이클로프로필]-3-메틸-펜타-2,4-디엔산, 및(2E,4E)-(1RS,2RS)-5-[2-(3,5-디-t-부틸-2-부톡시-페닐)-사이클로프로필]-3-메틸-펜타-2,4-디엔산중에서 선택된 화합물.
- 하기 화학식 XVIII의 화합물:화학식 XVIII상기식에서,A는 포밀이고;R1, R2, 및 R4내지 R6는 제 1 항에서 정의한 바와 같다.
- 제 1 항의 화학식 I의 화합물 또는 화학식 I의 카복실산의 약학적으로 허용가능한 염 및 통상의 약학 담체 물질을 포함하는 약학 제제.
- 레티노이드 작용물질 치료를 받는 환자에게서 일어나는 역효과를 감소시키거나 또는 제거하기 위한 레티노이드 길항물질의 용도.
- 제 10 항에 있어서,상기 레티노이드 작용물질 치료가 여드름, 건선, 또는 전암성 질환 또는 악성 질환을 위한 것인 용도.
- 면역글로불린 E(IgE)로 매개되는 알러지성 질환과 같은 보조 T 세포 제 2 형(Th2)으로 매개되는 면역계 질환, 또는 IL-4 및 IL-5와 같은 Th2-관련 사이토카인에 의해 매개되는 질환을 치료하기 위한 레티노이드 길항물질의 용도.
- 제 12 항에 있어서,상기 질환이 아토피성 피부염, 알러지성 비염 또는 알러지성 기관지 천식인 용도.
- 골다공증을 치료하기 위한 레티노이드 길항물질의 용도.
- 제 14 항에 있어서,상기 골다공증이 폐경기 골다공증, 남성 및 여성의 노인성 골다공증, 또는 모든 유형의 이차 골다공증인 용도.
- 전암성 질환 또는 종양성 질환을 치료하기 위한 레티노이드 길항물질의 용도.
- 제 16 항에 있어서,상기 전암성 질환이 피부 및 점막의 전암성 병변이고, 상기 종양성 질환이 피부, 점막, 두부 및 목, 폐, 위, 결장, 유방, 난소, 경관 및 전립선의 고형 종양인 용도.
- 레티노이드 작용물질의 치료를 받는 환자에게서 일어나는 역효과를 감소시키거나 제거하기 위한 약제를 제조하는데 사용되는, 제 1 항에서 정의된 화학식 I의 화합물 또는 화학식 I의 카복실산의 약학적으로 허용가능한 염의 용도.
- 제 18 항에 있어서,상기 레티노이드 작용물질 치료가 여드름, 건선, 전암성 질환 또는 악성 질환을 위한 것인 용도.
- 면역글로불린 E(IgE)로 매개되는 알러지성 질환과 같은 보조 T 세포 제 2 형(Th2)으로 매개되는 면역계 질환, 또는 IL-4 및 IL-5와 같은 Th2-관련 사이토카인에 의해 매개되는 질환을 치료하기 위한 약제를 제조하는데 사용되는 레티노이드 길항물질의 용도.

- 제 24 항에 있어서,상기 전암성 질환이 피부 및 점막의 전암성 병변이고, 상기 종양성 질환이 피부, 점막, 두부 및 목, 폐, 위, 결장, 유방, 난소, 경관 및 전립선의 고형 종양인 용도.
- 제 10 항 내지 제 25 항중 어느 한 항에 있어서,상기 레틴산 길항물질이 제 1 항에서 정의한 바와 같은 화학식 I의 화합물인 용도.
- 제 26 항에 있어서,상기 화학식 I의 화합물이 제 6 항에서 정의된 화학식 Ic의 화합물인 용도.
- 제 27 항에 있어서,상기 화학식 Ic의 화합물이 (2E,4E)-(1RS,2RS)-5-[2-(3,5-디-t-부틸-2-부톡시-페닐)-사이클로프로필]-3-메틸-펜타-2,4-디엔산인 용도.
- 전술한 바와 같은 신규한 화합물, 조성물 및 용도.
