Abstract
본 발명은 피부 스트레스 방지 및 치료용 조성물의 제조에서 벤조디아제핀 말초 수용체 (PBR)와 결합하는 물질의 용도에 관한 것이다. 본 발명은 또한 상기 물질을 포함하는 조성물에 관한 것이다. 상기 조성물은 화장품용 또는 제약용, 특히 피부학에서 국소 용도일 수 있다.
Description
본 발명은 벤조디아제핀 말초 수용체에 대한 리간드를 함유하는 국소 용도용 조성물에 관한 것이다.
본 발명은 피부 스트레스 예방 또는 치료용 조성물의 제조에서, 벤조디아제핀 말초 수용체 (PBR)에 특이적으로 결합하는 물질의 용도에 관한 것이다.
본 발명은 또한 이러한 물질을 함유하는 조성물에 관한 것이다. 이러한 조성물들은 화장품용 또는 제약용일 수 있고, 특히 국소 피부학용 조성물일 수 있다.
용어 "피부 스트레스"는 손상을 야기하는 약품에 상관없이 특히 표피에 손상을 야기할 수 있는 다양한 경우를 의미한다. 이러한 약품은 신체의 내부 및(또는) 외부에, 예를 들어 화학 또는 자유 라디칼제, 또는 다르게는 자외선 복사선과 같은 외용제일 수 있다.

조성물에 함유된, "물질"이라고도 언급되는 PBR 리간드는 비펩티드 화합물, 펩티드, 동물 또는 식물 기원의 세포 추출물 또는 조직 추출물, 또는 미생물의 발효, 예를 들어 박테리아 또는 진균류의 발효에 의해 얻어진 생성물일 수 있다.
많은 PBR 리간드가 문헌에 개시되어 있다. 언급될 수 있는 예는 Ro 5-4864 또는 클로로디아제팜, Ro 5-2807 또는 디아제팜 및 PK 11195를 포함하고, 문헌 [Peripheral Benzodiazepine Receptors, Ch. III, J. J. Bourguignon, Ed. E. Giesen-Crouse, Academic Press.]를 참조한다.
PBR은 말초 조직 미토콘드리아의 외막에 위치한 18-kd 단백질이다. 이는 5 개의 막통과 도메인 및 시토솔을 향한 카르복시-말단 부분으로 구성된다. 스테로이드형성 (steroidogenesis)의 조절, 헴의 생합성, 세포 분화 및 성장, 미토콘드리아 호흡의 통제와 같은 몇몇 작용은 조직의 특징에 따르는 PBR에 의한 것으로 고려되고 있다 (문헌 [Krueger KE, Biochimica and Biophysica Acta 1995, 1241, 453-470]). 그의 정확한 작용이 완전히 밝혀지지는 않았지만, 몇몇 최근 실험 데이타는 PBR이 세포예정사 방법의 조절 및 자유 라디칼로부터의 보호에서 중요한 역할을 수행할 수 있다는 것을 제안하고 있다.

본 발명의 내용에서, 조혈성 기원의 세포에서 PBR이 자유 라디칼로부터 보호하는 역할을 직접 관찰하여 PBR 밀도와 자유 라디칼로부터의 보호와의 밀접한 상호관계를 입증하였다. 또한, 동일한 연구에서 PBR이 이러한 수용체가 부족한 세포로 형질감염되는 것은 산화된 종에 의해 야기되는 손상으로부터 보호한다는 것을 입증하였다 (문헌 [Carayon P. et al., Blood 1996, 87, 3170-3178)].
몇몇 문헌의 데이타는 PBR이 세포소멸 방법을 조절하고, 자유 라디칼에 의해 야기된 손상으로부터 세포를 보호하는 중요한 역할을 수행할 수 있다는 것을 제안하고 있다.
최근 계통발생 연구는 PBR이 항산화성 작용에 관련된 세포소멸 조절자로서 작용한다는 새로운 견해를 강화하고 있다. 그 이유는 PBR과 광합성 박테리아인 로도박터 스페로이드즈 (Rhodobacter sphaeroides)의 단백질 CrtK간에 유의한 유사성이 존재하기 때문이다. 감광성 산소 검출자로서 작용하는 이러한 박테리아성 단백질은 산소 압력 및 빛의 강도에서의 환경 변화에 응답하여 광합성에 관련된 유전자의 발현을 조절한다. PBR과 CrtK를 비교하여 동일성이 35 %이고 이십억년전에 계통발생학상 분기된 이러한 두개의 단백질간에 서열이 보존된 것을 밝혀내었다. 이러한 상동성은 박테리아에서 CrtK의 고도 분화 및 보존 작용과 유사한 것으로 보이는 PBR의 고도 분화 및 보존 작용을 제안한다. 특정하게는, 로도박터 CrtK 돌연변이체로 형질감염된 포유류 PBR이 CrtK의 산소-검출 작용을 보완한다는 것이 최근입증되었다. 따라서, 이러한 연구는 산소-의존 신호의 형질도입에서 PBR의 주된 역할을 제안하고 있다 (문헌 [Yeliseev AA., et al., Proc. Natl. Acad. Sci. 1997, 94, 5101-5106]).
그러나, 현재까지 피부 PBR 수용체에 대해 특이적인 리간드로서 정확히 표시된 물질이 없는데, 이는 PBR 수용체에 특이적으로 결합하는 국소적으로 활성인 물질이 문헌에 개시되어 있지 않은 이유이기도 하다.
본 발명의 내용에서, 이제 PBR이 각질세포, 랑게르한스 (Langerhans) 세포, 모낭 및 피부 혈관계의 내피세포로 구성된 다양한 세포 분획물내의 피부에서 풍부하게 발현된다는 것을 보여주고 있다. 피부에서, PBR의 발현은 기저층으로부터 각질층으로 증가되는 구배를 따른다. 외부 스트레스에 가장 많이 노출되어 있는 표피의 분화된 세포에 유리한 이러한 주목할만한 유기조직은 표피의 가장 취약한 지역을 보호하기 위해 의심할 여지없이 근본 생리적으로 중요하다. 공초점 현미경에 의해 수행된 세포하 연구는, 예상한 바와 같이 미토콘드리아에서 PBR과 Bcl2와의 공동국재화를 나타내고 있다. 피부 표본에 대한 조직학적 연구는 PBR의 놀라운 분포를 밝혀내었다 (도 1 및 2).
특정하게는, 이러한 표피중 수용체의 발현은 기저층으로부터 각질 세포의 가장 분화된 층으로 증가하는 밀도 구배를 따른다. 밀도에 관해서 가장 바깥쪽의 것, 즉 표피의 가장 많이 노출된 세포에 유리한 이러한 고도로 조직된 공간 분포는 피부 중 PBR이 자외선 복사선에 대한 노출에 의해 발생된 자유 라디칼에 대한 천연 보호 시스템을 나타낼 수 있다고 가정하게 한다. 또한, 항-세포소멸 단백질 Bcl2의 분포가 엄격한 역구배를 따른다는 관찰은 가장 분화된 세포를 보호하는 PBR의 보충적인 역할을 제안한다.
보다 특정적으로 표피에서 PBR의 보호 역할을 제안하는 이러한 세트의 데이타는 천연 또는 합성 리간드를 발견하도록 하였고, 이는 이들의 PBR과의 상호작용이 화학 또는 라유-라디칼제에 의해, 또는 다르게는 UV에 대한 노출에 의해 유도된 다양한 피부 스트레스 상태에서 유익할 수 있다는 것을 보여주었다.
따라서, 본 발명의 한 면에 따라 본 발명은 피부 스트레스에서 PBR, Ro 5-4864에 대해 특이성이 있는 리간드의 용도에 관한 것이다. 이러한 리간드는 PBR 작용제이다.
본 발명의 또다른 면에 따르고, 이러한 관찰을 기초로 하여, 천연 PBR 리간드를 찾기위한 스크리닝을 수행하고, 수용체와 상호작용할 수 있는 몇몇 분획물을 단리할 수 있었다. 이어서, 이러한 천연 리간드의 잠재적인 보호성 효과를 피부 스트레스를 유도하는 다양한 실험, 특히 UV 조사에 의해 유도된 피부 홍반의 실험으로 평가하였다. 라디칼-제거 특성 및 피부 재생 능력을 또한 연구하였다.
생화학 및 약물학적 실험을 사용하여 다양한 피부 스트레스 상태에서 본 물질의 활성 및 장점을 입증하였다.
PBR을 사용하여 수행된 시험들은 PBR이 세포소멸 공정을 조절하고, 다양한 유독한 스트레스 상태로부터 피부 세포를 보호하는데 잠재적으로 관계하는 것을 보여주기 위한 것이었다.
<실시예 1>
PBR의 피부 국재화의 면역조직학적 연구
웨스턴 블롯 분석에 의해 특이성 항-PBR 항체 Ac 8D7 (항-PBR 마우스 mAb, 동종형 IgG1, 문헌 [Dussossoy et al., Cytometry, 1996, 24:39-48])을 사용하여, 6개의 상이한 사람 각질 세포계 및 정상 사람 피부계에서 PBR이 풍부히 존재하는 것을 입증할 수 있었다 (도 1). 세포하 수준에서, 공초점 현미경에 의해 분석을 수행하여 각질 세포의 미토콘드리아 수준에서 공동국재화를 확인하였다 (도 2).
정상 사람 표피 표본에서 동일한 항체를 사용하여 수행한 면역조직학적 연구에 의해 PBR의 발현이 기저층에서 각질층으로 증가하기 때문에 매우 특수한 유기 조직임을 밝혀냈다. 따라서 이러한 수용체는 가장 분화되고, 각질층 바로 아래에 위치하는 각질 세포에 풍부하게 존재하였다 (도 3).
<실시예 2>
결합 및 특이성 연구
기준 리간드 [3H]-PK11195의 치환에 의해 각질 세포계 A-431 (사람 유표피 암종 (ATCC, CRL-1555))에서 결합 연구를 수행하였다. 스케차드 분석은 단일 결합 자리, 세포당 약 470000 수용체의 밀도 및 리간드의 높은 친화도를 나타내었다 (KD = 1.5 nM) (도 4). 기준 말초 리간드 (PK 11195) 치환의 효능이 하기 리간드에 의해 감소된다는 것을 보여주는 약물학적 연구에 의해 각질 세포에 의해 생성된 말초 수용체에 결합하는 특이성을 확인하였다: Ro 5-4864 = (IC50≒ 25 nM) > 디아제팜(IC50≒ 100 nM) >>> 클로나제팜 (3200 nM에서 비활성). 이는 마지막 화합물이 벤조디아제핀에 대한 중앙 수용체의 리간드이고, 디아제팜은 혼합되었고, Ro 5-4864는 PBR에 대해 특이성이라는 것을 상기시켜 주었다 (도 5).
<실시예 3>
산화된 라디칼에 대한 보호에서 PBR의 관련성
두가지 유형의 실험이 도 6에 기재되어 있다. 첫번째는 PBR 발현 수준에 관련된 산화된 라디칼의 독성에 저항하는 이들의 능력에 관한 림프구양 또는 골수양 기원의 상이한 계의 비교로 이루어진다. 이 결과는 세포당 PBR 자리의 수 및 H2O2에 의해 유도된 독성에 대한 내성간의 매우 강한 상호관계를 나타내었다. 또한, 이번에는 Bcl2의 발현 수준, 산화성 손상으로부터 세포를 보호한다고 공지된 단백질을 고려할 때 유사한 상호관계가 있었다. Bcl2 및 PBR이 미토콘드리아의 외부막에 위치한 단백질이라는 사실과 함께 이러한 데이타는 이들이 세포 보호에서 공동의 작용을 가질 수 있다는 것을 제안하고 있다. 흥미롭게도, PBR의 발현이 기저층으로부터 각질층의 범위까지 증가하는 밀도 구배를 따름에도 불구하고, 문헌의 데이타는 Bcl2 발현에 대한 역현상을 나타내고 있는데, 이는 각질 세포의 분화동안에 PBR이 Bcl2를 대신하여 자유 라디칼로부터 보호하는 작용을 한다는 것을 제안하였다.
두번째 실험에서는, H2O2의 존재하에서 동종 PBR- 세포와 비교하여 PBR+ 형질감염 줄캣 (Jurkat) 세포의 더 양호한 생육성의 논증으로부터 자유 라디칼의 독성으로부터의 보호에서 PBR의 가능한 역할이 강화되었다.
<실시예 4>
활성제의 항-세포소멸성 (anti-apoptotic) 활성은 10 % 소태아혈청을 보충한 DMEM (Dubelco's Mode Eagle Medium) 중 35 mm 페트리 접시 (5 x 105세포/웰)에 접종시키고, 합류점까지 증식시킨 사람 각질세포 및 섬유아세포 (ATCC)에서 측정하였다. 이어서 이러한 배양 배지를 제거하고, 세포를 헹구고, 0.1 % 소태아혈청을 식염수의 존재하에 첨가하였다. 연구하고자 하는 물질의 농도를 증가시켰다. 24 시간 후, 세포소멸을 ELISA (효소 면역 측정법) 분석 키트로 측정하였다.
각질 세포에 16 시간 동안 250 J/m2의 선량으로 유형 B의 자외선 복사선 (UVB)을 쬐었다 (문헌 [J. Invest. Dermatol. 1995., 104: 922-927]). PBR 리간드 Ro 5-4864의 존재하에서, 조사에 의해 유도된 세포 손상 방법은 리간드 농도 범위 10 nM 내지 10 ㎛에서 방지된다는 것을 보여주었다.
<실시예 5>
흰색 기니아 피그의 피부에 적용함으로써 리간드의 광보호성 효과를 평가하였다.
피부 국소 경로를 사용하여 사람에서의 사용 조건을 재현하였다.
프랑스 76410 셀레옹 세인트 오빈 레 엘보 소재의 챨스 리버 프랑스로부터의 할리 기니아 피그를 사용하였다.
처리 시작 24 시간 전, 동물을 면도하고 우측 및 좌측 후 측복부의 털을 뽑았다.
1차 처리 바로 전에 동물에 복사선을 조사하였다. 에너지를 체크한 후, 각 복사선 조사를 4000 mJ/cm2의 선량의 UVB 스펙트럼으로 우측 및 좌측 측복부에서 수행하였다.
조사 직후, 및 조사 2 시간 및 5 시간 후에 동물의 우측 측복부를 0.2 ml의 리간드 용액으로 처리하였다.
크세논 고압 증기 램프 (IDEM 300)로 조사하였다.
처리전, 및 조사 5 시간 및 24 시간 후에 국소 반응을 해석하였다.

발효에 의해 생성되는 PBR 수용체에 대한 천연 리간드의 예는 그들의 활성과 함께 하기에 기재하였다.
고체 또는 액체 배지에서 수행된 미생물 추출물에서 스크리닝을 수행하여 미생물 (미생물 진균류 및 박테리아)의 3 가지 균주를 선택할 수 있었다.
PBR 수용체와의 상호작용을 측정하기 위한 실험에서, 양호한 활성을 갖는 배양 추출물의 유의한 양을 제조하기 위한 조건을 최적화하기 위해 수행된 다양한 연구 후 선택된 3 가지 균주는 참조 번호 SRL 4988, SRL 5168 및 SRL 5189이었다.
상기 3 가지 균주를 파스퇴르 연구소의 CNCM에 1999년 8월 27자로 각각 일련 번호 제I-2305번, 제I-2306번 및 제I-2307번으로 기탁하였다.
토양 샘플로부터 단리되고 노카르디아 (Nocardia) 종으로 분류된 균주 SRL 4988은 2주간 28 ℃의 ISP2 배지에서 배양한 후 결정된 음성 광의존균, 유기영양균, 증온균, 음성 호염균과 같은 생태학-생리학적 특성을 갖는다. 이는 부동적이고, 개방된 비-환생체 균사이다.
토양 샘플로부터 단리되고 스트렙토미세스 (Streptomyces) 종으로 분류된 균주 SRL 5186은 2주간 28 ℃의 ISP2 배지에서 배양한 후 결정된 음성 광의존균, 유기영양균, 증온균, 음성 호염균과 같은 생태학-생리학적 특성을 갖는다. 이는 부동적이고, 적응성 있는 (flexible) 이환생체 균사이다.
악티노시네마 (actinosinnema) 종으로 분류된 균주 SRL 5189는 2주간 28 ℃의 ISP2 배지에서 배양한 후 결정된 음성 광의존균, 유기영양균, 증온균, 음성 호염균과 같은 생태학-생리학적 특성을 갖는다. 이는 부동적이고, 적응성 있는 일환생체 균사이다.
따라서, 이러한 균주 및 이들의 증식성 돌연변이체는 본 발명의 또다른 주제를 구성한다.
영양 한천 배지에서 배양하고 수회 후속 계대 배양하여 풍부하고 순수한 배양액을 제조한 후, 스톡 균주의 저장 뱃치 0에 이어, 제1 및 제2 접종 뱃치를 제조하였다.
이를 수행하기 위하여, 포자 현탁액을 페트리 접시의 영양 한천 배지중 배양액 및 흡수 배지로부터 제조하였다. 이러한 배지는 냉동 저장 동안 포자의 양호한 생육성을 보존하기 위해 부동제를 함유한다.
얻어진 포자 현탁액을 -80 ℃에서 저장되는 저온튜브 (cryotube)로 분배하였는데, 이러한 튜브는 뱃치 0을 구성한다
뱃치 0으로부터의 튜브를 사용한 것을 제외하고는 동일한 프로토콜에 따라, 제1 접종 뱃치를 제조하였다. 다음으로, 다시 동일한 프로토콜에 따라, 제1 접종 저온튜브로부터 제2 접종 뱃치를 제조하였다. 접종 뱃치 0, 1 및 2의 제조는 균주의 오래-지속되는 이용성을 유지하고, 이에 따라 목적 활성을 유지한다. PBR 수용체의 천연 리간드를 얻기 위한 이러한 3개의 균주를 배양하는 것은 보통의 호기성 배양 방법을 사용하여 유사한 방법으로, 예를 들어 임의 부피의 발효기중 액체 배지를 pH 및 통기의 인-라인 (in-line) 모니터링에 의해 수행할 수 있다.
<실시예 A SRL 4988>
균주 SRL 4988용 삼각 플라스크중에서의 배양 실시예로서, 제2 접종 튜브를 사용하여 하기 조성에 따라 악티노미세트 포자형성을 촉진하는 배지로 제조된 페트리 접시에 접종하였다.

5 ml의 포자 현탁액을 사용하여, 선반이 있는 진탕기상 28 ℃의 온실 또는 자율 배양기에 접종된 전배양물을 구성하는 50 ml의 동일한 배지를 함유하는 살균 250 ml 플라스크를 접종하였고, 각 시스템의 회전 속도는 210 rpm이었다.
2일 동안 진탕한 후, 전배양 플라스크를 사용하여 조성이 하기와 같은 배양 배지 (100 ml)을 함유하는 500 ml 삼각 플라스크당 5 ml의 전배양 배지의 속도로 실제 배양 플라스크에 접종하였다.

따라서, 이러한 특정 경우에, 5 개의 전배양 플라스크를 사용하여 500 ml 삼각 플라스크당 100 ml의 배양 배지로 40 개의 배양 플라스크에 접종한 후 210 rpm으로 설정된 회전식 진탕기상 28 ℃의 온실에서 6 일 동안 진탕 배양하여 4 l의 박테리아성 현탁액을 얻었다.
4 l의 발효 브로스 (broth)를 4 ℃의 온도에서 13500 rpm의 방법 (즉, 사용한 회전자로 27500 x g)으로 수회 원심분리하여 바이오매스, 즉, 사용한 영양배지로부터의 물로 주로 구성되고, 용액으로 나머지 영양배지 성분 및 또한 이들 성장의 다양한 상 동안에 박테리아성 세포에 의해 생성되고 분비된 대사물을 함유하는, 배양 상청액으로부터 세포를 합한 펠렛을 분리하였다.
이어서 바이오매스 및 상청액을 -20 ℃에서 냉동하였다.
<PBR 수용체의 천연 리간드의 추출>
4 l의 해동된 상청액을 10 l 비이커에 배치하였다. 400 g의 앰버라이트 (Amberlite) XAD 16 폴리스티렌-디비닐벤젠 수지 (롬 앤드 하스 (Rohm & Haas))를 용액에 첨가하였다. 상청액을 패달 축이 장착된 모터를 사용하여 진탕하고, 15 시간 동안 20 rpm에서 회전시켰다. 이어서 용액을 여과하고, 여액을 제거하고, 배수된 수지를 1 l의 메탄올에 용해하였다. 이 혼합물을 1 시간 동안 부드럽게 교반하였다. 수지를 다시 여과하고, 동일한 방법으로 1 l의 메탄올로 재처리하였다. 3번째 실시 동안에, 수지를 이번에는 1 l의 아세톤으로 재처리하였다. 이어서 배수된 수지를 제거하고 3 l의 합한 유기 용매를 진공하 회전식 증발기 중에서 증발시켜 건조하였다.
증발 잔류물 (17.7 g)을 50 ml의 메탄올 중에 슬러리화하고, 얻어진 현탁액을 3000 rpm에서 15 분 동안 원심분리하였는데, 얻어진 안정된 상청액은 배양 상청액 추출물을 구성하였다.
이러한 추출물을 PBR 수용체에 대한 결합 억제를 위해 희석하여 실험하여 1/200 희석에서 평가된 활성을 제공하였다 (50 % 억제).
2 l 비이커중 합한 바이오매스 (199 g)을 교반하면서 750 ml의 염화메틸렌 및 750 ml의 메탄올의 혼합물로 처리하였다. 실온에서 15 시간 동안 교반을 계속하였다. 이어서 현탁액을 여과하고, 얻어진 투명한 용액을 회전식 증발기에서 진공하에 농축하였다. 이어서, 증발 잔류물 (5.4 g)을 50 ml의 메탄올 중에 슬러리화하였는데, 이는 바이오매스 추출물을 구성하였다.
이 추출물을 PBR 수용체에 대한 결합 억제에 대해 실험하여, 1/2200에서 측정한 활성을 얻었다 (ID50= 2200-1).
<실시예 B SRL 5168>
동일한 개개의 프로토콜 및 배지, 즉
- 페트리 접시상 계대배양용 한천 배지,
- 액체 전배양 배지,
- 액체 생성 배지를 사용하여
5 % 접종된 100 ml의 생성 배지를 함유하는 14 x 500 ml 삼각 플라스크를 210 rpm로 회전하는 회전식 진탕기상 28 ℃의 온실에서 6 일 동안 배양하였다.
생성된 상청액 및 바이오매스를 원심분리하고, -20 ℃의 냉동실에서 하루 또는 이틀 동안 저장한 후, 해동하여 이 생성물을 추출하였다.
바이오매스 (54.9 g)를 비이커 중에서 교반하면서 250 ml의 디클로로메탄 및 250 ml의 메탄올의 혼합물로 10 시간 동안 처리하였다. 이어서 현탁액을 여과하고, 얻어진 투명한 용액을 회전식 증발기에서 농축하여 건조하였다.
건조 잔류물 (1.4 g)을 17.5 ml의 메탄올에 슬러리화하고, 얻어진 현탁액을 3000 rpm에서 15 분 동안 원심분리하였다. 수거된 원심분리 상청액은 바이오매스 추출물을 구성하였다.

동일한 개개의 프로토콜 및 배지, 즉
- 페트리 접시상 계대 배양용 한천 배지,
- 액체 전배양 배지,
- 액체 생성 배지를 사용하여
5 % 접종된 100 ml의 생성 배지를 함유하는 10 x 500 ml 삼각 플라스크를 210 rpm으로 회전하는 회전식 진탕기상 28 ℃의 온실중에서 8일 동안 배양하였다.
생성된 상청액 및 바이오매스를 원심분리하고, -20 ℃의 냉동실에서 하루 또는 이틀 동안 저장한 후, 해동하여 이 생성물을 추출하였다.
바이오매스 (69.5 g)를 비이커 중에서 교반하면서 150 ml의 디클로로메탄 및 150 ml의 메탄올의 혼합물로 10 시간 동안 처리하였다. 이어서 현탁액을 여과하고, 얻어진 투명한 용액을 회전식 증발기에서 농축하여 건조하였다. 건조 잔류물 (1.5 g)을 12.5 ml의 메탄올 중에 슬러리화하고, 얻어진 용액을 3000 rpm에서 15 분 동안 원심분리하였다. 수거된 원심분리 상청액은 바이오매스 추출물을 구성하였다. 본 발명에 따른 조성물에서, PBR에 결합하는 물질은 바람직하게 조성물 총 중량의 0.00001 중량% 내지 20 중량% 범위의 양으로, 및 특히 조성물 총 중량의 0.001 중량% 내지 10 중량% 범위의 양으로 사용된다.
본 발명에 따른 조성물은 국소 적용에서 보통 사용되는 임의의 제공 형태일 수 있다.
본 발명에 다른 조성물중 다양한 성분의 양은 당해 분야에 통상적으로 사용되는 양 및 이들의 제공 형태에 적절한 양이다.
국소 적용을 위해, 본 발명의 조성물은 피부와 상화성인 매질을 포함한다. 이러한 조성물은 특히 크림 또는 겔, 극소-에멀젼 또는 에어로졸 모양인 수성, 알코올성 또는 수성-알코올성 용액, 겔, 유중수 또는 수중유 에멀젼의 형태, 또는 다르게는 이온성 및 비이온성 지질을 함유하는 소포성 분산액의 형태일 수 있다. 이러한 제공 형태는 당해 분야의 통상의 방법에 따라 제조된다.
이러한 국소 적용을 위한 조성물은 특히 얼굴, 목, 손 또는 몸의 화장품용 또는 피부학용 보호 치료 또는 관리 조성물 (예, 데이 크림, 나이트 크림, 태양 차단 크림 또는 오일 또는 바디 밀크), 메이크업 조성물 (예, 화운데이션) 또는 인공 태닝 (taning) 조성물을 구성한다.
본 발명의 조성물이 에멀젼일 경우, 지방족 물질을 조성물 총 중량의 5 중량% 내지 80 중량%, 및 바람직하게 5 중량% 내지 50 중량% 범위의 비율로 함유한다. 에멀젼 형태의 조성물중 사용된 지방족 물질 및 유화제는 통상적으로 화장품용 또는 피부학용으로 사용되는 것으로부터 선택된다.
본 발명에 사용될 수 있는 지방족 물질로서, 광유 (바셀린), 식물성유 (카라이트 버터 (karite butter)의 액체 분획물) 및 그의 수소화 유도체, 동물성유, 합성유 (퍼히드로스쿠알렌), 실리콘유 (폴리디메틸실록산) 및 플루오로 오일을 언급할 수 있다. 또한 언급될 수 있는 다른 지방족 물질은 지방족 알코올 (세틸 알코올 또는 스테아릴 알코올), 지방산 (스테아르산) 및 왁스를 포함한다.
유화제는 조성물 총 중량의 0.3 중량% 내지 30 중량% 범위, 바람직하게 0.5 중량% 내지 30 중량%의 비율로 조성물에 존재할 수 있다.
공지된 방법에서, 본 발명의 화장품용 또는 피부학용 조성물은 또한 상응하는 분야에서 통상적인 부형제, 예를 들어 친수성 또는 친지질성 겔화제, 방부제, 항산화제, 용매, 방향제, 충전제, 스크리닝제 및 염료를 함유할 수 있다. 또한 이러한 조성물은 친수성 또는 친지질성 활성제를 함유할 수 있다. 이러한 다양한 부형제 또는 활성제의 양은 화장품 또는 피부학에서 통상적으로 사용되는 양, 예를 들어 조성물 총 중량의 0.01 중량% 내지 20 중량%로 사용된다. 이들의 특성에 따라, 이러한 부형제 또는 이러한 활성제들은 지방족 상, 수성상 및(또는) 지질 소포에 도입될 수 있다.
본 발명의 조성물이 함유할 수 있는 이러한 활성제 중, 특히 주름살 및 잔주름의 치료에 효과가 있는 활성제, 특히 각질용해 활성제를 언급할 수 있다. 용어 "각질용해"는 탈락, 박리 또는 스크럽 특성을 갖는 활성제, 또는 각질층을 연화시킬 수 있는 활성제를 의미한다.
주름살 및 잔주름의 치료에 효과가 있는 본 발명의 조성물이 함유할 수 있는 이러한 활성제중, 특히 히드록시산 및 레티노이드를 언급할 수 있다.
히드록시산은 예를 들어, 선형, 분지형 또는 고리형, 및 포화 또는 불포화일 수 있는 α-히드록시산 또는 β-히드록시산일 수 있다. 탄소쇄의 수소 원자도 또한 할로겐, 할로겐화, 알킬, 아실, 아실옥시, 알콕시카르보닐 또는 알콕시 라디칼 (탄소수 2 내지 18임)로 치환될 수 있다.
사용될 수 있는 히드록시산은 특히, 글리콜산, 락트산, 말산, 타르타르산, 시트르산, 2-히드록시알카논산, 만델산, 살리실산 및 그의 알킬 유도체, 예를 들어 5-n-옥타노일살리실산, 5-n-도데카노일살리실산, 5-n-데카노일살리실산, 5-n-옥틸살리실산, 5-n-헵틸옥시살리실산, 또는 4-n-헵틸옥시살리실산 및 2-히드록시-3-메틸벤조산 또는 그의 알콕실화 유도체, 예를 들어 2-히드록시-3-메톡시벤조산이다.
레티노이드는 특히 레티노산 및 그의 유도체, 레티놀 (비타민 A) 및 그의 에스테르, 예를 들어 레티닐 팔미테이트, 레티닐 아세테이트 또는 레티닐 프로피오네이트 및 그의 염일 수 있다.
이러한 활성제는 특히 조성물 총 중량의 0.0001 중량% 내지 5 중량% 범위의 농도로 사용될 수 있다.
Claims (15)
- 피부 스트레스 치료에서 화장품용, 제약용 또는 피부학용 조성물의 제조를 위한, 벤조디아제핀 말초 수용체 또는 PBR에 결합하는 1종 이상의 물질의 용도.
- 주름을 감소시키거나, 일광 홍반을 감소시키거나, 자유 라디칼로부터 보호하기 위한 화장품용 및(또는) 피부학용 국소 조성물의 제조를 위한 PBR에 결합하는 1종 이상의 물질의 용도.
- 제1항 또는 제2항에 있어서, PBR에 결합하는 물질이 합성 분자, 천연 추출 물질 및 발효에 의해 얻은 물질로부터 선택된 PBR 작용제인 것을 특징으로 하는 용도.
- 제1항 내지 제3항 중 어느 한 항에 있어서, PBR에 결합하는 물질이 Ro 5-4864인 것을 특징으로 하는 용도.
- 제1항 내지 제3항 중 어느 한 항에 있어서, PBR에 결합하는 물질이 발효에 의해 얻은 물질인 것을 특징으로 하는 용도.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 화장품용 또는 피부학용 조성물에서 물질이 조성물의 총중량의 0.00001 중량% 내지 20 중량% 범위의 양으로 존재하는 것을 특징으로 하는 용도.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 화장품용 또는 피부학용 조성물에서 물질이 조성물의 총중량의 0.001 중량% 내지 10 중량% 범위의 양으로 존재하는 것을 특징으로 하는 용도.
- 제1항 내지 제7항 중 어느 한 항에 있어서, 조성물이 또한 히드록시산 및(또는) 레티노이드를 함유하는 것을 특징으로 하는 용도.
- 제8항에 있어서, 히드록시산이 선형, 분지형 또는 고리형, 및 포화 또는 불포화일 수 있는 α-히드록시산 및 β-히드록시산으로부터 선택되는 것을 특징으로 하는 용도.
- 제9항에 있어서, 레티노이드가 레티노산 및 그의 유도체, 및 레티놀 및 그의 에스테르를 포함하는 군으로부터 선택되는 것을 특징으로 하는 용도.
- PBR에 결합하는 물질을 활성 성분으로서 함유하는 것을 특징으로 하는 화장품용 및(또는) 피부학용 국소 조성물.
- 파스퇴르 연구소의 C.N.C.M.에 제I-2305번으로 기탁된 균주 노카르디아종 SRL 4988 및 그의 증식성 돌연변이체.
- 파스퇴르 연구소의 C.N.C.M.에 제I-2306번으로 기탁된 균주 스트렙토미세스종 SRL 5186 및 그의 증식성 돌연변이체.
- 파스퇴르 연구소의 C.N.C.M.에 제I-2307번으로 기탁된 균주 악티노시네마종 SRL 5189 및 그의 증식성 돌연변이체.
- 제12항 내지 제14항에 따른 균주를 발효시킴으로써 얻어진 물질을 활성 물질로서 함유하는 것을 특징으로 하는 화장품용 및(또는) 피부학용 국소 조성물.
