Abstract
본 발명은 하기 일반식(Ⅰ)로 표현되는 에틸아민 유도체 및 이의 약학적으로 허용되는 산 부가염을 제공한다:
일반식(I)
상기 식에서,
R1은 수소, 수산기, 저급 알콕시기 또는 할로겐이고,
R2는 탄소수 2 내지 5의 알킬기이고,
R3은 수소, 탄소수 2 내지 5의 알킬기, 탄소수 2 내지 5의 알킬카르보닐기, 탄소수 6 내지 10의 아릴기 또는 탄소수 7 내지 11의 아릴알킬기를 의미하며,
A고리는 적어도 하나 이상이 벤젠 고리이고, 다른 한쪽이 헤테로 원자를 함유할 수 있는 포화 또는 불포화 고리의 5 내지 6원자로 이루어진 비시클릭 화합물을 의미하며, 단, A고리가 인돌 및 1,3-벤조디옥솔인 경우, R2와 R3가 동시에 탄소수 2가 되지 않고, 또한 R3이 수소인 경우, A고리는 인돌 및 벤조[b]티오펜 이외의 환상 화합물이며, R2는 탄소수 3 내지 5의 알킬기이다. 이러한 화합물은, 카테콜아민 증강 작용과 CAE 효과에 대한 높은 선택성을 지니며, 향정신제, 항우울제, 항 파킨슨병 약 및 항 알츠하이머병 약으로 유용하다.
Description
매우 많은 에틸아민 유도체가 간행물에 기재되어 있다. 예를 들어, 유럽 특허 제 110781호에는 6-(2-아미노에틸) 벤족사졸리논 유도체가 항불안제, 심부전 치료제로서 기재되어 있고, 프랑스 특허 제 2035749호에는 아미노알킬 벤족사지논 유도체가 중추신경계 장해 치료약으로서 유용하다고 기재되어 있다. 또한, 일본 특허 공고공보 평성 6년(1994년)-99420호에는 본 발명의 화합물과 유사한 알킬아민 화합물이 향정신제로서 기재되어 있다. 그러나, 이러한 것들은 중추신경계에 대한 카테콜아민 치환형 방출 촉진 효과를 갖기 때문에, 시냅스 소포 등의 저장 부위로부터의 과도한 카테콜아민 방출이 일어나기 쉽고, 각성제와 비슷한 신경 독성이나 행동 이상(과다 복용시에 나타나는 흥분 작용: 조건 회피 반응 측정시에 나타나는 시그널간 반응의 증가) 등의 부작용이 지적되고 있다. 또한, 과도한 카테콜아민의 방출을 촉진시키는 약제의 연속 투여는, 카테콜아민 수용체의 감소를 유도하여 환자의 약제에 대한 반응성을 저하시키게 되어, 결과적으로 충분한 치료 효과를 얻을 수 없게 된다. 이것은 장기간의 연속적인 치료가 필요하거나 요구되고 있는 파킨슨병이나 알츠하이머병에 대한 약제로서 불만족스러운 것이다.
한편, 국제 공개 88/2254호에 페네틸아민 유도체가 향정신제로서 개시되어 있다. 이 페네틸아민 유도체는, 상기 카테콜아민 치환형 방출 촉진 작용과는 다른 메카니즘(mechanism)에 의거한 카테콜아민 작동 시스템의 활성 증강 효과(막 전위 의존성 엑소사이토시스의 증강에 의한 카테콜아민 방출 촉진 효과: (CAE(Catecholaminergic Activity Enhancing)효과, [라이프·싸이언스(Life Sciences) 58권 945∼952페이지(1996)]을 갖고 있다. 하지만, 그렇다고 해도 또한 행동 이상의 지표인 조건 회피 반응 측정시에 나타나는 시그널간 반응을 증가시키는 작용이 해결되지 않았기 때문에, 더욱이 CAE효과에 대한 높은 선택성을 지닌 약제의 개발이 요망되고 있었다.
따라서, 중추신경에 대한 부작용이 적은, 향정신제, 항우울제, 항 파킨슨병 약 및 항 알츠하이머병 약 등의 약제로서 유용한 화합물의 개발이 요망되고 있어, 이러한 요망에 대응할 수 있는 것이 본 발명의 목적이다.
본 발명은, 향정신제, 항우울제(antidepressant), 항 파킨슨병 약 및 항 알츠하이머병 약으로서 유망한 새로운 에틸 아민 유도체를 제공하는 것이다.
도 1은, 전기 자극에 의한 쥐의 적출 뇌간으로부터의 노르아드레날린 유리량에 대한, 화합물 번호 3, 20, 25 및 27의 영향을 나타내는 도면이다.

본 발명자들은, 상기 페네틸아민 유도체 등의 중추 신경계에 대한 신경 독성이나 행동 이상은, 각성제인 암페타민이나 메탐페타민과의 구조상의 유사성에 기인하거나 그것이 관여하고 있다고 생각하여, 본 발명의 목적을 달성하기 위해 예의 검토를 거듭해 온 결과, 각성제와는 구조를 달리 하는 어느 종류의 특정한 새로운 에틸아민 유도체 및 그 산 부가염이, 카테콜아민 증강 효과와 CAE효과에 대한 높은 선택성을 지님을 발견하여, 중추신경에 대한 신경 독성이나 행동 이상 등의 부작용이 적은 향정신제, 항우울제, 항 파킨슨병 약 및 항 알츠하이머병 약을 제공하는 것을 목적으로 하는 본 발명을 완성시키기에 이르렀다.
본 발명에 있어서는, 하기 일반식(Ⅰ)로 표현되는 새로운 에틸아민 유도체 및 이의 산 부가염, 및 상기 화합물을 유효 성분으로서 함유하는 향정신제, 항우울제, 항 파킨슨병 약 또는 항 알츠하이머병 약을 제공할 수 있다:

여기에서, 산 부가염으로서는, 약학적으로 허용되는 것이 바람직하며, 염산, 황산, 브롬화수소산, 질산, 메탄술폰산 등과 같이 무기산과의 염, 또는 글루콘산, 타르타르산, 말레산, 푸마르산, 숙신산, 말산(malic acid), 시트르산, 만델산 등의 유기산과의 염을 구체예로서 들 수 있다.
일반식 (Ⅰ)에서 말하는 저급 알콕시기는, 탄소수 1 내지 4의 알콕시기로서, 예를 들어 메톡시기, 에톡시기, n-프로폭시기, 이소프로폭시기, n-부톡시기, 이소부톡시기 및 3차-부톡시기 등을 들 수 있다.
A고리로서의 구체예로서는, 나프탈렌, 인돌, 1,3-벤조디옥솔, 1,4-벤조디옥산, 인단, 인덴, 벤조푸란, 벤조[d]티오펜, 2,3-디히드로벤조푸란, 디히드로나프탈렌 또는 테트랄린 등을 들 수 있고, 치환은 임의의 위치이어도 상관없다.
일반식(Ⅰ)의 특히 바람직한 화합물은, 1-(2-나프틸)-2-프로필아미노펜탄, 1-(인돌-3-일)-2-프로필아미노펜탄, 1-(벤조푸란-2-일)-2-프로필아미노펜탄, 1-(1,3-벤조디옥솔-5-일)-2-프로필아미노펜탄, 1-(1,3-벤조디옥솔-4-일)-2-프로필아미노펜탄, 1-(인돌-6-일)-2-프로필아미노펜탄, 1-(인돌-4-일)-2-프로필아미노펜탄, 1-(벤조[b]티오펜-2-일)-2-프로필아미노펜탄 및 1-(벤조[b]티오펜-3-일)-2-프로필아미노펜탄 등을 들 수 있다.
본 발명의 화합물은 전부 문헌에 기재되어 있지 않은 새로운 화합물로서, 예를 들어 하기의 어느 하나의 방법에 의해 유리하게 제조할 수 있다.
[루트 1]
하기 일반식(Ⅱ)로 표현되는 니트로 화합물을 테트라히드록푸란(THF)과 같은 불활성 용매 중에서, 수소화 리튬 알루미늄과 같은 환원제의 존재 하에 실온 내지 환류 온도의 범위에서 1 내지 200 시간 반응시켜서, 하기 일반식(Ⅲ)로 표현되는 아민 화합물을 얻는다:

이 공정에서 얻어지는 아민 화합물 중에서, A고리가 인돌 및 벤조[b]티오펜 이외의 비시클릭 화합물이며, 또한 R2가 탄소수 3 내지 5의 알킬기인 화합물은 본 발명의 새로운 목적 화합물의 일부를 이루고, 또 다른 일반식(Ⅲ)과 함께 본 발명의 또 다른 새로운 화합물 제조의 중간체 화합물로서 사용할 수 있다.
따라서, 본 발명에서는 상기 일반식(Ⅲ)으로 표현되는 화합물이, 목적하는 바에 따라 하기 일반식(Ⅳ)로 표현되는 산할로겐화물과 불활성 용매 중에서 염기성 축합제의 존재 하에, 실온 내지 환류 온도의 범위에서 반응하여, 바람직하게는 THF, 클로로포름, 4염화 탄소 등의 불활성 용매 중에서 트리에틸아민 또는 피리딘과 같은 축합제의 존재 하에서, 실온 내지 환류 온도의 범위에서 1 내지 48시간 반응하여 하기 일반식(Ⅴ)로 표현되는 화합물로 수득된다:

따라서, 본 발명에서는 상기 일반식(Ⅴ)으로 표현되는 화합물이 다시 목적하는 바에 따라, 비활성 용매 중에서, 환원제의 존재하에, 실온 내지 환류 온도의 범위에서 반응 처리되며, 바람직하게는 THF, 에테르와 같은 불활성 용매 중에서 수소화 리튬 알루미늄, 수소화 알루미늄과 같은 환원제의 존재 하에서, 실온 내지 환류 온도의 범위에서 1 내지 48시간 반응해서, 하기 일반식(Ⅰ)로 표현되는 새로운 에틸아민 유도체로 수득되며, 목적하는 바에 따라 이의 약학적으로 허용되는 염으로 유도된다:

원료 화합물인 일반식(Ⅶ)으로 표현되는 화합물은, 저널 오브 메디시널 케미스트리(Journal of Medicinal Chemistry ) 29권 2009∼2015페이지(1986)에 기재된 합성 방법에 의해 합성할 수 있다.
본 발명의 화합물 및 그 산 부가염은, CAE효과의 결과로서, 조절적으로 카테콜아민 방출을 촉진시켜, 항 파킨슨병 약, 항우울제, 항정신제 및 항 알츠하이머병 약 등으로서 유효하며 부작용이 적은 의약으로서 사용할 수 있다.
즉, 본 발명의 화합물은, 종래의 항 파킨슨병 약이나 항우울제 등에서 보여지는 모노아민 옥시다아제 저해 작용, 카테콜아민 섭취 저해 작용, 치환형 방출 촉진 작용 또는 신경세포의 각종 수용체와의 친화성 등을 갖고 있지 않지만, 카테콜아민 고갈제인 테트라베나진을 투여하여 학습 능력을 저하시킨 쥐의 학습 능력을 유의적(significantly)으로 회복시켰다. 본 발명의 화합물의 작용은 활동 전위와 전달 물질 방출에 관련된 자극에 따른 증강(막 의존성의 엑소사이토시스의 증강)에 의해 생리적으로 조절된 카테콜아민 방출 촉진 효과인 CAE효과에 기인하는 것으로, CAE효과에 대한 높은 선택성은, 하기에 기재한 약리 실험, 특히 실시예 30 내지 실시예 32에 의해, 본 발명의 화합물에 카테콜아민 치환형 방출(release) 촉진 작용, 모노아민 옥시다아제 저해 작용 및 각종 신경세포의 수용체와의 친화 작용을 갖지 않는 것으로 확인되었다. 더욱이, 개구리 심장의 동방 섬유의 지연형 내향 칼슘 전류(slow inward Ca2+currents)를 상승시키고, 또한 쥐의 뇌간을 크렙스(Krebs) 용액에 담가, 정지 전위시 및 전기 자극시의 카테콜아민 방출량을 측정한 경우, 정지시에는 방출량을 변화시키지 않고, 자극시에만 유의적으로 증가시키는 것도 본 발명의 CAE효과를 지지하였다.
CAE효과는, 막 전위 의존성의 엑소사이토시스의 증강에 따른 카테콜아민 방출 촉진 효과로서, 활동 전위에 응답하여 효력을 발생하는 생리적인 작용 메카니즘에 따르기 때문에, 종래의 모노아민 옥사다아제 저해약, 카테콜아민 섭취 저해약, 암페타민 등의 각성제 또는 페네틸아민 등에 나타나는 카테콜아민 치환형 방출 촉진 작용약과는 전혀 상이하며, 지나친 카테콜아민의 방출, 고농도로의 급격한 상승 및 카테콜아민 신경말단의 아민 고갈을 유도하지 않는 특이한 작용 스펙트럼을 가지며, 그렇기 때문에 과도한 카테콜아민의 존재에 기인하는 카테콜아민 수용체의 감소나 장해도 발생하기 힘들다.
따라서, 본 발명의 화합물은, 행동량 이상 증가(흥분 작용), 중추신경에 대한 신경 독성 등의 부작용 및 환자의 응답성의 저하가 적고, 항우울제, 항정신제, 항 파킨슨병 약 및 항 알츠하이머병 약 등으로서, 안전성이 높고, 뛰어난 효과를 가지며, 또한 장기적인 연속 투여에 있어서도 그 효과의 유지를 기대할 수 있는 유용한 화합물이다.
본 발명의 화합물 및 이의 약학적으로 허용되는 산 부가염을 상기 의약으로서 이용할 경우, 통상 담체(carrier), 비히클(vehicle), 희석제, 용해 보조제 등과 혼합하여, 알약, 가루약, 과립제, 캅셀제, 시럽제, 주사제 등의 형태로 경구적 또는 비경구적으로 투여될 수 있다. 투여량은 환자의 증상, 연령, 체중 등에 따라 변할 수 있는데, 통상 성인의 하루당 경구 투여량은 0.1 내지 100mg의 범위이며, 1회 또는 몇 회에 걸쳐서 투여할 수 있다.
이하에, 본 발명의 화합물에 대해 구체적으로 설명하겠는데, 본 발명이 이에 한정되지 않음은 물론이다.
실시예 1
1-(2-나프틸)-2-아미노펜탄 염산염(화합물 번호 1)의 제조
수소화 리튬 알루미늄 2.42g(63.8mmol)을 무수 THF 30ml에 현탁시키고, 얼음 냉각 하에서 1-(2-나프틸)-2-니트로-1-펜텐 7.70g(31.9mmol)의 무수 THF 40ml용액을 1시간에 걸쳐 적하시켜, 실온에서 하룻밤 동안 교반하였다. 반응 후, 물을 가해서 과잉 수소화 리튬 알루미늄을 분해하여 여과시킨 후, 여과액을 농축하여, 에테르 100ml를 가해 0.5N염산 50ml로 3회 추출하였다. 얻어진 물 층을 28% 암모니아수를 이용하여 알칼리성으로 만든 후, 에테르 100ml로 3회 추출하였다. 에테르 층을 무수 황산 나트륨으로 건조한 다음 농축시켜, 플래시 컬럼 크로마토그래피(flash column chromatography)에 의해 클로로포름/메탄올(10:1)로 용출시키고, 담황색 기름 형태의 물질로서 1-(2-나프틸)-2-아미노펜탄 5.36g(79%)을 얻었다. 상기 기름 형태의 물질을 무수 에테르에 용해시켜, 냉각 하에서 포화 염화수소 에테르 용액을 가해서 염산염으로 변환시켰다.

실시예 2
N-[2-(1-(2-나프틸)펜틸]프로피온아미드(화합물 번호 2)의 제조
실시예 1에서 얻은 1-(2-나프틸)-2-아미노펜탄 5.36g(25.1mmol) 및, 트리에틸아민 3.81g(37.7mmol)을 무수 THF 40ml에 용해하고, 0℃로 냉각시켰다. 이 용액에 프로피온산 염화물 4.64g(50.1mmol)의 무수 THF 10ml 용액을 적하시켜, 실온에서 24시간 교반하였다. 반응 후 농축하고, 잔류물을 에테르에 용해하여, 5% 염산, 5% 탄산나트륨 및 포화 식염수의 순서로 세정하였다. 무수 황산나트륨으로 건조한 후 농축하여, 플래시 컬럼 크로마토그래피에 의해 클로로포름/헥산(5:2)으로 용출시킨 후, 에테르-헥산으로부터 재결정하여, 백색 결정으로서의 N-[2-(1-(2-나프틸))펜틸] 프로피온아미드 5.48g(81%)을 얻었다.

실시예 3
1-(2-나프틸)-2-프로필아미노펜탄 염산염(화합물 번호 3)의 제조
수소화 리튬 알루미늄 2.70g(71.1mmol)을 무수 에테르 30ml에 현탁시키고, 냉각 하에서 무수 염화 알루미늄 2.71g(20.3mmol)의 무수 에테르 용액 20ml를 적하시켜서 수소화 알루미늄의 무수 에테르 용액을 제조하였다. 이것에 실시예 2에서 얻은 N-[2-(1-(2-나프틸))펜틸]프로피온아미드 5.48g(20.3mmol)의 무수 에테르 용액 40ml를 냉각 하에서 적하시키고, 0℃에서 잠시 교반한 후, 실온에서 다시 하룻밤 교반하였다. 반응 후, 냉각 하에서 물을 조금씩 가하고, 5N 수산화나트륨을 가해서 알칼리성으로 여과하였다. 여과액을 농축시킨 후, 에테르 100ml를 가해 0.5N 염산 70ml으로 4회 추출하였다. 더욱이, 물 층(water phase)을 28% 암모니아수로 알칼리성으로 만든 다음, 에테르 100ml로 3회 추출하고, 무수 황산나트륨으로 건조한 다음 농축시켜, 플래시 컬럼 크로마토그래피에 의해 클로로포름/메탄올(30:1)로 용출시키고, 담황색 기름 형태의 물질로서 1-(2-나프틸)-2-프로필아미노펜탄을 2.40g(46%)를 얻었다. 얻어진 기름 형태의 물질을 실시예 1과 같은 방법에 의해 염산염으로 변환시켰다.

실시예 3에서 얻은 1-(2-나프틸)-2-프로필아미노펜탄 0.7g(2.7mmol)에 말레산 0.15g(1.3mmol)의 에탄올 2ml 용액을 첨가하였다. 약 5℃에서 방치한 후, 얻어진 석출 결정을 여과시켜서 1-(2-나프틸)-2-프로필아미노펜탄 말레산염 0.33g(32%)를 얻었다.

1-(1,3-벤조디옥솔-5-일)-2-부틸아미노펜탄 염산염(화합물 번호 29)의 제조
아르곤 분위기 하에서 1-(1,3-벤조디옥솔-5-일)펜탄-2-온 2.0g(9.70mmol), n-부틸아민 0.71g(9.7mmol)의 1,2-디클로로에탄 34ml 용액에, 수소화 트리아세톡시 붕소나트륨 2.88g(13.6mmol) 및 아세트산 0.58g(9.7mmol)을 가해, 실온에서 4일간 교반하였다. 반응액에 1N 수산화나트륨을 가해 알칼리성으로 만들고, 물 50ml를 가해서 에테르 70ml로 3회 추출하여, 포화 식염수로 세정하였다. 무수 황산나트륨으로 건조한 후, 농축하여 컬럼 크로마토그래피(용출액: 클로로포름)에 의해 담황색 기름형태 물질로서 얻었다. 이것을 실시예 1과 같은 방법으로 염산염으로 변환시켜, 1-(1,3-벤조디옥솔-5-일)-2-부틸아미노펜탄 염산염 1.42(4.7mmol)을 얻었다(수율 48%).

전기 자극에 의한 활동 전위 상태에서의 쥐 적출 뇌간으로부터 생체 아민 유리 작용:
라이프 싸이언스 58권 2101∼2114페이지(1996)에 기재된 방법을 이용하여, 체중 280∼350g의 수컷 쥐의 후두부를 강타하여 기절시켜, 뇌간(평균 중량 약 800mg)을 적출하여, 95% O2, 5% Co2로 통기시킨 37℃의 크렙스 용액에 30분간 담가 놓고, 그 후 20㎕의3H-노르아드레날린(아마샴:Amersham, 30∼50Ci/mmol 1-[7, 8-3H] 노르아드레날린)을 조직 표본(preparation)에 첨가하여, 45분간 방치하고3H-노르아드레날린을 섭취시켰다. 크렙스 용액의 조성(mmol/l)은, Na+137.4, K+5.9, Ca2+2.5, Mg2+1.2, Cl-120.2, H2PO4 -1.2, HCO3 -25.0, SO4 2-1.2, 글루코오스 11.5, 아스코르브산 0.3, Na2EDTA 0.03 및 파르길린 12로 하며, 표식 물질의 섭취 기간 중에, MAO 활성을 저해시키기 위해 파르길린(12mmol)을 크렙스 용액에 가했다.
3H-노르아드레날린을 섭취하게 한 후, 뇌간을 5ml의 크렙스 용액(37℃)이 들어간 오간 배스(organ bath) 안에 고정시켰다. 다음에 뇌간을 0.03mmol의 코카인을 함유하는 산소로 포화된 크렙스 용액으로 8ml/분의 유속으로 세정하였다. 100분 후에, 관류(perfusion) 속도를 4ml/분으로 감소시키고, 더욱이 0.05mmol의 코오티코스테론(Corticosterone)을 함유하는 것으로 변경하여, 코카인과 코오티코스테론의 존재 하에서(이 상태에서는3H-노르아드레날린의 86%는 대사되지 않으며, 신경 안이나 신경 바깥에 흡수되지 않음) 실험을 했다. 관류액의 분별(fractionation)은 3분마다 이루어지고, 3분 동안에 방출되는3H-노르아드레날린 양이 5ml인 아쿠아세이프 300플러스(진서 : Zinsser)에 관류액을 1ml 가해 신틸레이션 카운터(베크만:Beckmann, LS-900)로 측정되었다. 또한, 세로토닌에 대해서도 동일하게 측정하였다. 전기 자극(3Hz, 1ms, 60V)은 3분 동안 인가되었다. 실험을 시작할 때 3회 분별분의 휴지 기간을 최초의 전기 자극에 앞서 두었다. 그 후, 7회 분별의 휴지 기간을 자극과 자극 사이에 두었다. 본 발명의 화합물은, 0.5, 1, 2.5, 5㎍/ml가 되도록 관류액 중에 용해시켰다. 본 발명의 화합물을 첨가한 관류액은, 전기 자극 전에 3분간 관류되었다.
도 1 및 도 2의 결과에 나타난 바와 같이, 본 발명의 물질은, 신경 세포가 전기 자극을 받았을 경우에 있어서, 노르아드레날린 및 세로토닌의 유리를 촉진시키고, 엑소사이토시스를 증강시키는 것이 확인되었다.
실시예 27
셔틀 박스에 있어서의 쥐의 조건 회피 반응에 대한 작용:
라이프 싸이언스 58권 817∼827(1996)에 기재된 방법을 이용해, 테트라베나진 처법에 의해 학습 능력이 저하된 암컷과 수컷 쥐(200∼220g)의 셔틀 박스에 있어서의 조건 회피 반응(Conditioned Avoidance Reflex:CARs)에 대한 작용을 검토하였다.
장치는, 사이코파머콜로지아(Psychophamacologia) 10권 1∼5페이지(1995)에 기재된 고안에 따라서 조립되며, 체중 200∼220mg의 암수컷 쥐를 이용하여, 조건 자극(Conditioned Stimulus:CS, 빛과 부저음)을 받아, 칸막이를 빠져 나오도록 훈련시켰다. 만일 실패하면, 무조건 자극(Unconditioned Stimulus:US)인 푸트 쇼크(footshock)(1mA)를 주었다. 쥐가 US에 대해, 5초 이내에 반응하지 않으면 도피 실패(Escape Failures:EFs)로 간주했다. 또한, 조건과 상관없는 행동량을 시그널간 반응(Intersignal Reactions:IRs)으로 간주했다. 쥐에게는 1일 100시행, 5일간 훈련하고, 각 시행은 15초 간격으로 행했다. 테트라베나진은 생리 식염액에 용해시키고, 시험의 1시간 전에 1mg/kg의 용량으로 피하 투여하여, CARs, EFs 및 IRs가 자동적으로 측정되었다. 본 발명의 화합물은 생리 식염액에 용해되어, 테트라베나진과 함께 1, 2, 2.5 또는 5mg/kg의 용량으로 피하 투여했다. 통계적인 유의성은 일원 배치 분산분석에 의해 해석하였다.
그 결과의 유의성에 대해 표 6에 나타내고, 더욱 특징적인 결과를 도 3 내지 도 6에 나타내었다. 본 발명의 화합물은 테트라베나진 투여에 의해 유발된 학습 능력의 저하를 유의적으로 개선시키고, 우울증 및 인지 기능을 개선하는 것이 확인되었다. 또, 도피 실패(EFs)를 유의적으로 저하시키기 때문에 우울증이나 파킨슨병에서 보여지는 움츠림(freezing)을 개선하는 것도 인정되었다. 그리고, 국제 공개 88/2254호의 대표적인 화합물 1-페닐-2-프로필아미노펜탄(PPAP) 등과는 달리, 무조건 자극시의 행동 이상(흥분 작용)을 나타내는 IRs반응을 유의적으로 상승시키지는 않았다.

라이프 싸이언스 58권 2101∼2114페이지(1996)에 기재된 2중 자당 격리배열 전위 고정법의 변법(A modified version of the voltage clamp method on a double sucrose gap arrangement)을 이용하였다. 직경이 약 100㎛이고, 길이가 3∼4mm인 섬유를 라나(Rana) 식용 개구리의 심장 동방 영역으로부터 쌍안 현미경을 사용해서 제조하였다. 섬유를 냉각시킨 환경(18±0.5℃)에서 실험용 접시에 얹어 놓고, 섬유의 길이를 따라 0.1mm의 경계에 4개의 와세린(vaseline) 벽을 설치해 둠으로써 5개의 구획(compartment)을 작성하였다. 중앙의 구획은 시험 용액(본 발명의 화합물을 Na+가 첨가되지 않은 링게르액(염화콜린 함유)으로 0.5, 1, 2, 4 또는 8㎍/ml가 되도록 용해시킨 것)으로 관류하고, 2개의 바깥측 구획은 등삼투압의(isotonic) KCI용액으로 탈분극시켰다. 중앙의 구획에 인접한 나머지 2개의 구획은, 바깥측의 구획으로부터 중앙의 구획을 전기적으로 분리하기 위해, 등삼투압의 자당 용액으로 관류하였다. 중앙과 바깥측의 2개의 구획은 비분극 Hg/Hg2Cl2전극에 의해 사령 전위(command potential)를 막에 부하시켰다. 적분형 피드백 증강기가 막 전위를 일정하게 유지시키고, 막 전류와 동등한 보정 전류로 조정하였다. 지연형 내향 칼슘 전류가 Na+를 첨가하지 않은 링게르액(염화콜린 함유)으로 시험 구획을 관류함으로써 측정되었다.
그 결과, 도 7에 도시된 바와 같이, 지연형 내향 칼슘 전류를 증강시키고, 막 의존성 엑소사이토시스의 증강 작용을 갖는 것이 확인되었다. 이는, 본 발명물의 CAE효과를 뒷받침해 주는 것이다.
실시예 29
뇌 조직으로부터 방출되는 생체 아민의 측정:
라이프 싸이언스 56권 611∼620페이지(1995)에 기재된 방법에 따라, 위스터 계열의 쥐의 머리를 자른 후, 곧바로 뇌 조직(선조체(striatum), 흑질(substantia nigra), 후결절(tuberculm olfactorium), 청반핵(locus coeruleus) 및 봉선핵(raphe)을 적출하여, 95% O2, 5% CO2로 버블링한 37℃의 크렙스 용액에 담가 놓았다. 크렙스 용액의 조성(mmol/단위)은 NaCl 111, KCl 4.7, CaCl22.5, MgSO41.64, NaHCO325, KH2PO41.2, 글루코오스 12, 아스코르브산 및 200mg/l Na2EDTA로 하였다. 선조체, 흑질 및 후결절은 4등분, 청반핵 및 봉선핵은 8등분으로 해서, 항온조에 담갔다. 목적의 조직을 20분간 인큐베이션(incubation)시킨 후, 크렙스 용액을 교체하여, 본 발명의 화합물을 용해한 크렙스 용액에 조직을 20분간 담그고, 이 시간 내에 방출된 모노아민 양을 측정하였다. 본 발명의 화합물 및 대조 물질 PPAP는, 생리 식염액에 용해시켜, 뇌 적출 30분 전에 피하 투여되며, 노르아드레날린 및 도파민을 측정하기 위해, 저널 오브 파머콜로지 앤드 실험 요법(Journal of Parmacology and Experimental Therapeutics) 138권 360∼372페이지(1962)에 기재된 방법에 의해, 시료를 60mg의 알루미나를 함유한 마이크로 컬럼(BDH 케미칼스:BDH Chemicals Ltd.)으로 정제하고, 세로토닌을 측정하기 위해 시료 15mg의 세파덱스(Sephadex) G-10 마이크로컬럼(파머시아: Pharmacia)으로 정제하였다. 도파민, 노르아드레날린, 세로토닌은 전기 화학 검출기(워터즈: Waters 460 ECD)가 부착된 고속 액체 크로마토그래피(Waters 746)에 의해 측정하였다. 분리 컬럼은 워터즈·리졸브(Waters Resolve, 5μ Spherical C183.9×150mm)를 사용하여, 이동상(mobile phase)으로는 6% 아세토니트릴에 200mg/1 옥탄술폰산 나트륨, 18mg/l Na2EDTA를 첨가한 pH5.2의 트리에틸아민-인산 완충액을 사용하였다. 각각의 아민의 방출량은 20분 동안 방출된 nmol/g(조직)로 나타내었다. 평균값의 차이는 Student's t-테스트에 의해 검정되고, 유의 레벨은 p<0.05로 하였다.
그 결과, 표 7에 도시된 바와 같이, 본 발명 물질은, 선조체, 흑질 및 후결절에 있어서 도파민의 유리를 유의적으로 상승시키고, 청반핵의 노르아드레날린 및 봉선핵의 세로토닌에 있어서도 도파민의 유리를 상승시켰다.

실시예 30
쥐의 뇌 시냅토솜에 있어서의 모노아민 유리에 대한 작용:
시냅토솜은 이하와 같이 제조되었다. 8주된 수컷 위스터계열 쥐(닛뽄 S.L.C.)로부터 신선한 선조체(도파민 측정용) 또는, 전뇌(노르아드레날린 및 세로토닌 측정용)를 적출하였다. 이 조직에 10배 양의 얼음 냉각 0.32M 자당(pH 7.4)을 가하여, 모터 회전식 테프론제 막자(pestle)를 구비한 유리제 호모제나이저에 의해 균질화하였다. 그 후, 4℃에서 10분간, 900×g로 원심 분리하여, 얻어진 상청액을 다시 11500×g로 20분간 원심 분리하였다. 발생한 펠릿(pellet)을 얼음 냉각 완충액(ice-cold buffer)으로 재현탁시켰다. 시냅토솜 시료는 최종 농도 0.6mg 단백질/ml로 제조하였다. 또한, 단백질 정량은 저널 오브 바이올로지컬 케미스트리(Journal of Biological Chemistry) 193권 265∼275페이지(1951)에 기재된 방법에 준해서 행하며, 표준으로는 혈청 알부민을 이용했다. 시냅토솜으로부터 모노아민을 유리시키는 것은 저널 오브 파머콜로지 앤드 실험 요법 241권 27∼33페이지(1987)에 기재된 방법에 준해서 다음과 같은 방법으로 행했다. 0.6mg/ml의 시냅토솜 용액을 35℃, 95% O2, 5% CO2혼합 공기 하에서 5분간 예비인큐베이션(preincubation)시켰다. 다음에, 시냅토솜 용액에 25nM3H-모노아민을 첨가하고, 35℃에서 5분간 인큐베이션시켰다. 인큐베이션 후의 시냅토솜을 챔버에 넣고, 35℃에서 일정한 유속(0.7ml/min)으로 25분간 과융해 완충액(Superfusion buffer)으로 관류시킴으로써, 과잉 표지 화합물(labelled compound)을 제거하였다. 25분 후, 시냅토솜 용액 0.25ml로부터 유리되는3H-모노아민을 2분마다 20분간에 걸쳐 회수하였다. 더욱이, 본 발명의 화합물은 전부 다 최종 농도 10-5M에서 분별 5(10분∼)에서부터 시냅토솜에 노출되도록 과융해 완충액 중에 용해해서 첨가하였다. 회수한 과융해 완충액에 신틸레이션 칵테일 3.5ml(에멀션화제 신틸레이터 플러스TM[팩카드(Packard)]를 첨가하여, 신틸레이션 카운터를 이용하여 시냅토솜으로부터 유리된3H-양을 측정하였다.
피실험물질 첨가 전에 과융해 완충액을 관류시킬 때를 100%로 하고, 피실험 물질에 의한 시냅토솜으로부터의3H-모노아민의 유리 작용을 측정하며, 바탕값(blank)은 과융해 완충액으로부터 산출하였다.
또한, 사용한 과융해 완충액의 조성은, NaCl 115mM, CaCl22mM, MgSO41.5mM, KH2PO41.5mM, NaHCO325mM, D-글루코오스 10mM, 에틸렌 글리콜 비스-(아미노에틸 에테르)-N, N'-4아세트산(EGTA) 0.5mM 및 L-아스코르브산 0.1mM(pH 7.4)로 하였다.
그 결과를 도 8 및 도 9에 도시하였다. 본 발명의 화합물은, 시냅토솜에서의 티라민과 같은 작용을 나타내지 않을 뿐만 아니라, 동시에 첨가한 경우에는 티라민에 의한 효과와 길항 작용을 나타내었다. 이 시냅토솜에 있어서의 작용은, 본 발명의 화합물에는 카테콜아민 치환형의 카테콜아민 유리 효과가 없거나 그러한 효과를 억제시키는 것을 의미하며, 본 발명의 화합물의 CAE효과에 대한 높은 선택성을 나타내는 것이다.
실시예 31
모노아민 옥시다아제(MAO)에 대한 작용:
3.8% 시트르산이 첨가된 사람의 혈액을 950rpm, 10분간 원심 분리하여, 혈소판이 많은 혈장(PRP:platelet rch plasma)을 얻고, 나머지 혈구 부분을 3000rpm, 10분간 원심 분리하여, 혈소판이 부족한 혈장(PPP:platelet poor plasma)을 얻었다. PRP의 일부를 취해, 혈소판을 측정한 후, 이 PRP 및 PPP는, -20℃에서 동결한 후, MAO활성 측정시에 해동하고, 초음파에 의해 분리 현탁하여 혈소판 MAO의 조직 표본으로 하였다. 또한 사람 혈소판의 MAO는 거의 대부분이 B형인 것이 알려져 있다.
MAO-B의 측정은, 바이오케미컬 파머콜로지(Biochem. Pharmacol.) 제 14권 제 1684∼1685페이지(1965)의 방법에 준해서 행했다. 62.5mM 인산 완충액 880㎕에, 비교물 셀레길린(selegiline) 및 본 발명의 화합물을 62.5mM 인산 완충액으로 각 농도로 제조한 후 120㎕를 가해, 37℃에서 5분간 인큐베이션시켰다. 더욱이, PRP 또는 PPP 80㎕를 가해, 37℃에서 20분간 인큐베이션시켰다. 다음에 기질(kynuramine dihydrobromide 0.33mg/ml) 120㎕를 가하고, 37℃에서 30분간 인큐베이션시킨 후, 10% 트리클로로 아세트산 800㎕를 가해서 반응을 정지시키고, 3000rpm, 10분간 원심 분리하여, 상청액 1ml에 1N 수산화나트륨 2ml를 가해, Ex312nm 및 Em380nm으로 형광을 측정하였다. PRP는 62.5mM 인산 완충액으로 혈소판 수를 25만 cell/㎕로 제조하여, 측정시의 기준은 4-히드록시퀴놀린으로 하였다.
표 8의 결과에 나타나는 바와 같이, 비교물인 셀레길린은 10-7M의 농도에서 98.1%의 저해율을 나타내었으나, 본 발명의 화합물은 10-5M의 농도에 있어서도 저해율은 50% 이하였다. 이것은, 본 발명의 화합물에 MAO-B 저해 작용이 없음을 보여주는 것이며, 더욱이 본 발명물의 CAE효과에 대한 높은 선택성이 지지되었다.

실시예 32
신경 세포 수용체 α1, α2, D1, D2, 5-HT1 및 5-HT2에 대한 친화성:
수용체 용액의 제조는, 쥐의 머리를 자른 후, 즉시 뇌를 적출하여, 중량을 측정하였다. 그 중량의 10배 양의 얼음 냉각 0.32M 수크로오스 용액(pH 7.4) 안에서 균질화하여(모터 회전식 테프론제 막자를 구비한 유리제 호모제나이저), 균질화 용액을 만들어, 4℃, 900×g로 10분간 원심 분리하였다. 얻어진 상청액은 4℃, 22000×g로 20분간 원심 분리하고, 침전물에 5mM 인산 완충 용액(pH 7.4)을 가해 현탁시켰다. 이 현탁액을 37℃에서 30분간 인큐베이션시킨 후, 다시 한번 4℃, 22000×g로 20분간 원심 분리하고, 그 침전물에 5mM 인산 완충 용액(pH 8.0) 10ml를 가하여 현탁시켜, 수용체 용액으로 하였다.
리간드(ligand)에3H-프라조신(prazocin)을 사용하여, 튜브에 5mM 인산 완충 용액(pH 8.0)로 제조한 0.25nM3H-프라조신, 본 발명의 화합물 및 수용체 용액을 첨가하여 25℃에서 90분간 인큐베이션시켰다. 또한, 0.1μM 프라조신 존재하에서의3H-프라조신의 결합값을 특이하지 않은(비휘발성) 결합으로 하였다. 반응은, 유리 섬유 필터(pore size:0.5㎛, Whatman GF/C)로 수용체를 결합한3H-프라조신(결합형:B)과 유리형의3H-프라조신(유리형:F)을 B/F 분리장치(Brandel사 제품, USA)에 의해 분리하여, 결합형만을 회수하였다. 유리 섬유 필터에 흡착된 결합형은, 얼음 냉각 생리 식염액으로 세정하고, 유리 섬유 필터를 약병(vial)으로 옮겨서, 신틸레이션 칵테일 10ml를 첨가해서 방사능을 측정하여, α1수용체와 그 리간드와의 결합 저해율로부터, 본 발명의 화합물의 친화성을 구했다. 동일하게 조작해서, α2, D1, D2, 5-HT1및 5-HT2수용체에 대한 친화성을, 리간드 용액으로서 각각 0.7nM3H-라우울신(rauwolscine), 1.4nM3H-SCH-23390바운드, 2.0nM3H-스피페론(spiperone), 2nM3H-5-HT 및 0.5nM3H-케탄세린(ketanserin)을 사용하고, 여과기(displacer) 용액으로서 각각 1μM 요힘빈(yohimbine), 10μM(+)-부타클라몰(butaclamol), 10μM 할로페리돌(haloperidol), 10μM 5-HT 및 1μM 케탄세린(ketanserin)을 사용하여, 각각의 수용체에 있어서의 본 발명의 화합물의 친화성을 구하였다.
수용체와의 친화성이 높으면, 리간드와 수용체의 결합을 저해한다. 일반적으로는, 각각의 리간드와 수용체의 결합을 50% 저해하는 데 필요한 몰 농도가, 10-6M의 정도에서는 생리 활성을 발휘할 만큼 강하지 않은 화합물로 되어 있다. 본 발명의 화합물은, 표 9에 나타낸 바와 같이, 이러한 수용체에서는, 10-6M 내지 10-5M 이상의 농도를 필요로 하며, 생리 활성을 발휘하기 위해 필요한 수용체와의 친화성은 관찰되지 않았다.

이하, 안전성 시험의 결과를 나타낸다.
실시예 33
간 세포 초기 배양을 이용한 독성 실험:
쥐로부터 콜라게나아제 관류법에 의해 간 세포를 채취하여, 5×105세포/ml의 세포 현탁액을 제조하였다. 24시간 배양하여, 본 발명의 화합물이, 100 및 10μM을 함유하는 윌리엄스 E배지(GIBCO BRL)로 교환하였다. 교환후 24시간째에, 배지 안에 방출된 유산 탈수소 효소(LDH)를 측정하였다.
그 결과를 용매 대조(0.5% 생리 식염액 또는 0.5% DMSO)에 대한 백분율로 해서 표 10에 나타내었다. 시료 농도 100μM로서 처리했을 경우, 본 발명의 화합물은 PPAP와 같거나 또는 그 이하의 LDH의 유리를 나타내었으나, 10μM으로 했을 경우, PPAP에서만 유의적인 LDH의 유리가 관찰되었다.

실시예 34
1회 투여에 있어서의 생쥐의 급성 독성 시험:
생쥐에게 50mg/kg의 본 발명의 화합물(화합물 번호 3, 25, 27 및 28)을 1회 피하 투여한 후, 2주간 일반 증상을 관찰하였다. 그 결과, 모든 화합물이 PPAP의 LD50값인 50mg/kg에서는 세 가지 예 중에서 세 개 모두 생존하였다.
Claims (9)
- 하기 일반식(Ⅰ)로 표현되는 에틸아민 유도체 및 이의 약학적으로 허용되는 산 부가염:일반식(I)상기 식에서,R1은 수소, 수산기, 저급 알콕시기 또는 할로겐이고,R2는 탄소수 2 내지 5의 알킬기이고,R3은 수소, 탄소수 2 내지 5의 알킬기, 탄소수 2 내지 5의 알킬카르보닐기, 탄소수 6 내지 10의 아릴기 또는 탄소수 7 내지 11의 아릴알킬기를 의미하며,A고리는 적어도 하나 이상이 벤젠 고리이고, 다른 한쪽이 헤테로 원자를 함유할 수 있는 포화 또는 불포화 고리의 5 내지 6원자로 이루어진 비시클릭 화합물을 의미하며, 단, A고리가 인돌 및 1,3-벤조디옥솔인 경우, R2와 R3가 동시에 탄소수 2가 되지 않고, 또한 R3이 수소인 경우, A고리는 인돌 및 벤조[b]티오펜 이외의 환상 화합물이며, R2는 탄소수 3 내지 5의 알킬기이다.
- 제 1 항에 있어서, R3가 탄소수 2 내지 5인 알킬기, 탄소수 6 내지 10의 아릴기 또는 탄소수 7 내지 11의 아릴알킬기 중 어느 하나인 에틸아민 유도체 및 이의 약학적으로 허용되는 산 부가염.
- 제 1 항에 있어서, R1이 수소이고, R2및 R3가 프로필기인 에틸아민 유도체 및 이의 약학적으로 허용되는 산 부가염.
- 제 1 항 내지 제 3 항에 있어서, A고리가 나프탈렌, 인돌, 1,3-벤조디옥솔 중의 어느 하나인 에틸아민 유도체 및 이의 약학적으로 허용되는 산 부가염.
- 제 1 항에 있어서, 일반식(Ⅰ)로 표현되는 화합물이 1-(2-나프틸)-2-프로필아미노펜탄, 1-(6-메톡시나프탈렌-2-일)-2-프로필아미노펜탄, 1-(인돌-3-일)-2-프로필아미노펜탄, 1-(인돌-4-일)-2-프로필아미노펜탄, 1-(인돌-6-일)-2-프로필아미노펜탄, 1-(벤조푸란-2-일)-2-프로필아미노펜탄, 1-(1,3-벤조디옥솔-4-일)-2-프로필아미노펜탄 또는 1-(1,3-벤조디옥솔-5-일)-2-프로필아미노펜탄 중의 어느 하나인 에틸아민 유도체 및 이의 약학적으로 허용되는 산 부가염.
- 제 1 항 내지 제 5 항의 어느 한 항에 따른 화합물을 유효 성분으로 함유하는 향정신제.
- 제 1 항 내지 제 5 항의 어느 한 항에 따른 화합물을 유효 성분으로 함유하는 항우울제.
