각 추출물의 전자공여능은 DPPH(1,1-diphenyl-2-picryl hydrazyl)를 이용하여 시료의 radical 소거능을 측정하였다. 추출물을 각각 농도별로 용해시킨 시료액을 0.2 mM DPPH을 혼합하여 15분간 암실에서 반응 시킨후 517 nm에서 Microplate reader(Epoch, BioTek)로 흡광도를 측정하였다. DPPH radical 소거능은 시료 첨가군와 비첨가군를 비교하여 free radical 소거능을 백분율로 나타내었고, 양성대조군으로는 ascorbic acid(Sigma)를 사용하였다. [실험예 2 : 복합물의 피부 자극 정도 확인][Experimental Example 2: Confirmation of skin irritation level of complex]

본 실험에서는 상기 실시예 1에서 제조한 황금하귤추출물, 섬모시풀추출물, 환삼덩굴추출물과 상기 실시예 2에서 제조한 복합물의 피부 자극 정도를 확인하였다. 각 추출물과 복합물의 배합비에 따른 피부 자극도는 아래 표 4와 같으며, 20~30대의 성인 15명을 임의로 선발하여 각 추출물과 복합물로부터 자극이 느껴지는 정도를 1부터 10까지의 수치로 나타내었다.
