캠토테신(CPT, 1, 도 1)은 현저한 항종양 효과를 갖는 천연 발생 알칼로이드이다1-3. 그의 항종양 활성은, 비가역적 약물-효소-DNA 삼원복합체를 안정화시키고 Topo I에 의해 유도된 단일 가닥 DNA 절단의 재결합을 방지함으로써 DNA 토포이소머라제 I (Topo I)의 촉매 사이클을 방해하는 능력에 기인하는 것으로 생각되었다.4,5 지난 수십년에 걸쳐 집중적인 합성 의약품 화학 노력은, 난소암, 소세포 폐암 및 대장암을 치료하기 위해 현재 임상적으로 이용되는 토포테칸(2) 및 이리노테칸(3)을 포함한 강력한 1-유도체를 낳았다. 또한, 몇 가지 유도체, 예를 들면 기마테칸(4), CKD-602(5), 및 BNP-1350(6)은 전임상 또는 임상 개발의 다양한 단계에 있다.6-8 임상적으로 사용되는 1-유도체는 유망한 부류의 항종양제로 남아있지만, 이들의 치료 용도는, 독성 문제 및 난수용성에 기인한 전달 문제는 물론, 열린(opened) 카르복실레이트의 혈청 알부민에의 우선적 결합에 기인한 활성 락톤 형태의 불안정성에 의해, 심각하게 방해되었다.9 ,10

실험 부분
화학 일반 정보. N-Boc-아미노산 및 TFA는 GL 바이오켐(샹하이) 컴패니로부터 구입하였다. DIPC 및 DMAP는 시그마 케미칼 컴패니(중국)로부터 구입하였다. 다른 시약 및 용제는 상업적 공급원으로부터 구입하였으며 또한 받은 대로 사용하였다. 출발 1는 중국 약용식물 씨. 아쿠미나타(C. acuminata)로부터 분리하고 사용되기 전에 정제하였다(> 98% 순도). 분석용 박층 크로마토그래피 (TLC) 및 분취용 박층 크로마토그래피(PTLC)는 실리카 겔 60 GF254 (Qingdao Haiyang Chemical Co., Ltd.)을 사용하여 실리카 겔 플레이트로 수행하였다. 융점은 코플러 융점 장치로 얻고 보정하지 않았다. 화합물 9a -9l에 대한 일반적 합성 절차. 트리에틸아민 (1.2 mmol)을 CH2Cl2 (35 mL)중 TFA 염 8 (0.5 mmol)의 현탁액에 서서히 첨가하고, 이 혼합물을 투명한 용액이 얻어질 때까지 10분 동안 교반하였다. N2 분위기 하에, 알킨 (0.5 mmol), p-톨루엔설포닐 아지드(0.6 mmol), 및 CuI (0.05 mmol)를 첨가하였다. 반응 혼합물을 실온에서 2 내지 6시간 교반하였다. 이 원고에서 사용된 인간 종양 세포주는 A-549 (폐 암종), MDA-MB-231 (삼중음성 유방암), DU-145 (호르몬-비감수성 전립선암), KB (비인두의 표피암으로부터 최초로 분리됨), KBvin (빈크리스틴-내성 KB 서브라인), 및 HCT116 (결장직장 선암)이었다. 이들 세포주는 와이.-씨.첸(Y.-C. 형태학적 관찰. 배양 세포의 형태학적 변화는 위상차 현미경으로 관찰하였으며, 디지탈 카메라(일본 니콘)로 촬영하였다.
아폽토시스 평가. 아폽토시스는 아넥신 V-FITC/프로피듐 아이오다이드 이중 염색 키트(BD Biosciences)로 검출하였다. A-549 세포는 24 시간 또는 48 시간 동안 9a 로 처리하였다. 세포는 트립신 처리법으로 회수하고 빙냉 PBS로 세척하였다. 세포를 어두운데서 실온에서 15분 동안 아넥신 V-FITC 및 프로피듐 아이오다이드로 표지하였다. 표지된 세포를 FACSCalibur 유세포 분석기(Becton Dickinson)로 분석하였다.
세포 주기 분석. A-549 세포는 빙냉 70% EtOH로 고정시킨 다음 프로피듐 아이오다이드 염색을 하였다. 샘플을 세포주기 결정을 위해 유세포 분석기로 분석하였다. 세포 주기의 각 기(phase)에 있는 집단은 배수성(ploidy)에 기초하여 계산하고 (서브-G1기로서 < 2N; G1기로서 2N; S기로서 2N 내지 4N; G2/M기로서 4N), 스튜던트 t-검정으로 평가하였다 (P < 0.01).
웨스턴 블롯 분석. 세포는 프로테이나제 억제제 및 포스파타제 억제제를 함유하는 PBS 중에서 회수하여 초음파 처리하였다. 전체 세포 용해액을 SDS-PAGE로 분리하고 Immobilon P 막(EMD Millipore)으로 옮겼다. 막을 1차 항체로 배양한 다음 호오스래디쉬 퍼옥시다아제(HRP)-접합된 이차 항체 (EMD Millipore)로 표지하였다. 화학발광(Chemilluminence) 기질 키트(EMD Millipore)를 막 결합 HRP의 검출을 위해 사용하고, 발광 영상 분석기 LAS4000 (일본 후지 포토 필름사)로 가시화하였다.
항체. 카스파제-3, 카스파제-8, 카스파제-9, PARP, 포스포-ATM (Ser1981), ATM, 포스포-ATR (Ser428), 포스포-Chk1 (Ser345), 포스포-Chk2 (Thr68), Chk2, 포스포-H2AX (Ser139), 및 포스포-p53 (Ser15)에 대한 항체는 셀 시그날링 테크노놀지(Cell Signaling Technology)로부터 구입하였다. ATM, ATR, Chk1, 및 PUMA에 대한 항체는 산타 크루즈 바이오테크놀로지(Santa Cruz Biotechnology)로부터 입수하였다. Topo I, Topo IIα, Topo IIβ, p53, FADD, BAX, Bcl-xL, 및 Bcl-2에 대한 항체들은 비디 바이오사이언스(BD Biosciences)로부터 입수하였다. β-액틴에 대한 항체는 이엠디 밀리포어(EMD Millipore)로부터 구입하였다.
무세포 시스템에서 토포이소머라아제 I 활성 분석. 1 유닛의 재조합 인간 토포이소머라아제 I 효소 (TopoGEN)를 최종 용적 20 μL 반응 완충액 (10 mM 트리스-HCl, pH 7.9, 1 mM EDTA, 150 mM NaCl, 0.1% BSA, 0.1 mM 스페르미딘, 5% 글리세롤) 중 비히클, 9a, 1, 3, 또는 SN-38과 함께 37℃에서 20분 동안 미리 배양한 다음, 250 ng의 초나선 플라스미드 DNA와 함께 20분 동안 배양하였다. 초나선, 이완 또는 절개된 DNA를 1 x TAE (트리스-아세테이트-EDTA)완충액 중의 1% 아가로오스 겔로 분리하였다. 에티듐 브로마이드 염색된 아가로오스 겔을 Gel Doc XR (Bio-Rad)를 사용하여 촬영하였다.
토포이소머라아제 I 활성 분석. Topo I 활성 시험은 제조업자의 지시에 따라 분석 키트(TopoGEN)를 사용하여 수행하였다. 9a-처리된 A-549 세포로부터의 핵 추출물을 37℃에서 초나선형 DNA (Topo I에 대해) 또는 연쇄형(catenated) kDNA (Topo II에 대해)와 함께 30분 동안 배양하였다. 반응 혼합물은 1 x TAE 완충액 중의 1% 아가로오스 겔로 분리하였다. 겔은 에티듐 브로마이드로 염색하고 Gel Doc XR (Bio-Rad)을 사용하여 촬영하였다.
이종이식 모델 항종양 분석. 5 내지 6주령 암컷 nu/nu 마우스 (대만, 국립 실험실동물센터)는 옆구리에 2 x106 인간 결장직장 선암종 HCT116 세포로 피하로 접종되었다. 이식된 종양 용적이 200 ㎣의 평균 용적에 도달한 때, 마우스는 4개의 그룹으로 무작위로 분리하였다 (n = 8). 처리 계획은 표 2에 나타낸다. 비히클 대조군 및 화합물9a를 7일 동안 하루에 한번(QD) 정맥내 5 또는 10 mg/kg로 투여한 다음 마지막까지 하루에 한번 복강내 투여하였다. 실험 대조군으로서, 100 mg/kg의 화합물 3은 마지막까지 일주일에 한번(QWK) 정맥내 투여하였다. 이식편의 길이(L) 및 폭 (W)은 마지막까지 3 내지 4일마다 측정하였으며, 또한 종양 용적은 LW 2 / 2로서 계산하였다. 결과는 스튜던트 t-검정에 의해 통계적으로 계산하였다. 이 연구는 국립 타이완 대학교(대만, 타이페이)의 동물실험관련 위원회 (Institutional Animal Care and Use Committee, IACUC)에 의해 승인되었으며 또한 기관 가이드라인에 따라 수행하였다.
생체내 독성의 병리학적 평가. 60 마리의 8-주령 수컷 BALB/c 마우스 (대만, 타이페이, 국립실험실 동물센터)를 사용하여 단일용량 독성을 평가하였다. 마우스는 6개의 그룹(n = 10)으로 무작위로 분리되어 제0일에 0 (비히클), 30, 100, 200 또는 300 mg/kg로 9a의 단일 복강내 주입을 받았다. 하나의 그룹은 정상 대조군으로서 미처리하였다. 체중은 15일 동안 3일마다 측정하였다. 실험기간의 마지막에, 모든 동물은 CO2로 안락사시키고 간, 폐, 신장 및 비장으로부터 조직을 칭량하였다(데이터 도시안함). 조직은 10% 포르말린으로 고정하고 파라핀 포매하였다. 두께 3-5㎛의 박편(section)을 병리조직학적 검사를 위해 준비하였다. 헤마토실린 및 에오신 (H&E) 염색된 파라핀 박편은 Shackelford et al.36에 기술된 가이드라인에 따라 병리조직학적으로 평가하고 증후성 병변을 등급화하였다. 병변의 정도는 다음과 같이 중증도에 따라 1 내지 5개로 등급화하였다: [현저하지 않음, 1 = 최소 (< 1%), 2 = 약간 (1-25%), 3 = 중간 (26-50%), 4 = 중 정도 중증(modelately severe) (51-75%), 5 = 중증/높음 (76-100%)]. 통계적으로 유의한 결과(P < 0.05)를 나타냈다. 이 연구는 중국의과대학 (대만 타이중)의 기관 동물실험 관련 위원회(IACUC)에 의해 승인되고 기관 가이드라인에 따라 수행하였다.

실시형태 II
화합물 10a-10t는 실시형태 I에 기술된 것과 유사한 절차를 사용하여 합성하고 확인하였으며, 이는 표 3에 나타낸다.
트리에틸아민 (1.2mmol)을 CH2Cl2 (35mL)중 다양한 TFA 염 8 (0.5mmol)의 현탁액에 서서히 첨가하고, 이 혼합물을 투명한 용액이 얻어질 때까지 10분 동안 교반하였다. N2 분위기하에, 알킨(0.5 mmol), 설포닐아지드 (0.6 mmol), 및 CuI (0.05 mmol)를 실온에서 이 반응혼합물에 첨가하였다.
