Abstract
Description
도 2는 복숭아껍질에서 분리된 효모들의 pH에 따른 성장곡선을 나타낸 것이다.
도 3은 팜슬러지에서 분리된 효모들의 pH에 따른 성장곡선을 나타낸 것이다.
도 4는 팜슬러지 퇴비에서 분리된 효모들의 pH에 따른 성장곡선을 나타낸 것이다.
도 5는 0, 4, 5 및 6% 젖산이 포함된 YPD 액체배지에서의 성장곡선을 나타낸 것이다.
도 6은 30, 37, 42 및 50℃에서 배양하여 온도에 따른 성장곡선을 나타낸 것이다.
도 7은 신규 효모 2-2, NG1 및 NG7 균주의 글리세롤이 포함된 배지에서의 성장 여부를 나타낸 것이다.
도 8은 신규 효모 2-2, NG1 및 NG7 균주와 효모 사카로마이세스 세레비지에(SC)와 클루베로마이세스 막시아누스(KM)의 pH에 따른 세포성장 및 에탄올 생성능을 비교한 그래프이다.

도 12는 NG7 균주의 URA3 유전자 1카피를 불활성화 시키기 위한 모식도이며 형질전환체의 유전자 도입을 확인하기 위한 PCR 반응 결과를 나타낸 것이다.
도 13은 NG7 균주의 2 번째 URA3 유전자를 불활성화 시키기 위한 모식도이며 형질전환체의 유전자 결실을 확인하기 위한 PCR 반응 결과를 나타낸 것이다.
도 14는 URA3 선별마커를 이용하여 D-LDH 유전자를 PDC1 유전자 위치에 삽입 후 각 번호에 해당하는 프라이머를 이용하여 PCR 반응을 통해 확인한 결과를 나타낸 것이다.
도 15는 PDC1 유전자가 모두다 결실된 NG7Δpdc1/D-LDH 균주를 중화제 사용 없이 배양하여 포도당 소비량, 젖산 생산능 및 에탄올 생산능을 확인한 그래프를 나타낸 것이다.
도 16는 PDC1 유전자가 모두다 결실된 NG7Δpdc1/D-LDH 균주의 배양시 배양기 rpm이 젖산 생산에 미치는 영향을 확인한 그래프이다.
도 17는 NG7Δpdc1/D-LDH 균주의 초기 접종량을 증가시켰을 경우 배양기 rpm이 젖산 생산에 미치는 영향을 확인한 그래프이다.
도 18은 NG7Δpdc1/D-LDH 균주를 pH 중화제(CaCO3)를 농도별(0~3%)로 첨가하여 배양하면서 젖산 생산을 확인한 그래프이다.
도 19는 NG7Δpdc1/D-LDH 균주를 pH 중화제(CaCO3)를 3%와 5%를 사용하여 배양하면서 포도당 소모량과 젖산 생산량을 확인한 그래프이다.

- 내산성 및 내열성을 가지는 피키아 쿠드리압즈비(Pichia kudriavzevii) NG7 균주로서,
상기 균주는 기탁번호 KCTC18297P로 기탁된 것인, 균주.
- 삭제
- 제1항에 있어서,
상기 균주는 오로티딘 5인산 탈탄산효소(Orotidine 5'-phosphate decarboxylase, URA3)의 활성이 추가로 불활성화된 것인, 균주.
- 제1항에 있어서,
상기 균주는 피루베이트 디카르복실라제(Pyruvate Decarboxylase)의 활성이 내재적 활성보다 약화되고 젖산 탈수소 효소(Lactate Dehydrogenase) 활성이 추가로 도입된 것인, 균주.
- 제4항에 있어서,
상기 균주는 기탁번호 KCTC18300P인, 균주.
- 제1항에 있어서, 상기 균주는 탄소원으로 글리세롤을 사용할 수 있는, 균주.
- 제3항에 있어서,
상기 균주는 피루베이트 디카르복실라제 (Pyruvate Decarboxylase)의 활성이 불활성화되고 젖산 탈수소 효소(Lactate Dehydrogenase) 활성이 추가로 도입된 것인, 균주.
