Abstract
Description
도 2는 본 발명의 siRNA 전달용 재조합 단백질의 3차원 분자구조를 시뮬레이션한 결과를 나타낸 것이다.
도 3은 본 발명의 siRNA 전달용 재조합 단백질을 대장균 대량발현 시스템을 이용하여 발현 유도 하였을 때의 전기영동 결과를 나타낸 것이다.
도 4는 본 발명의 siRNA 전달용 재조합 단백질을 액체 크로마토그래피의 히스티딘 친화도 컬럼을 사용하여 정제한 결과를 보여주는 전기영동 결과를 나타낸 것이다.
도 5는 본 발명의 siRNA 전달용 재조합 단백질과 siRNA의 결합비율을 확인하기 위해서 siRNA 전달용 재조합 단백질의 농도에 따라 결합하는 siRNA의 양을 정량분석한 전기영동한 결과를 나타낸 것이다(siRNA 양을 고정하면서 siRNA 전달용 재조합 단백질의 양을 변화시킬 때 결합에 참여한 siRNA는 p19-YSA/siRNA 밴드로, 결합에 참여하지 못한 siRNA는 free siRNA 밴드로 나타난다).
도 6은 MTT 분석를 통하여 본 발명 p19-YSA/siRNA 결합체의 농도에 따른 세포 독성 및 생체적합성을 시험한 결과를 나타낸 것이다.
도 7은 p19-YSA/siRNA 결합체의 결합되어 있는 siRNA가 pH가 감소할수록 해리되는 것을 보여주는 전기영동 결과를 나타낸 것이다.
도 8은 10% FBS(fatal bovine serum) 조건 하에서 naked siRNA와 siRNA 전달용 재조합 단백질에 결합되어 있는 siRNA의 안정성을 비교한 결과를 나타낸 것이다.
도 9는 형광체 Cy5.5와 FITC로 각각 표지된 Cy5.5-p19-YSA/FITC-siRNA 결합체가 B16F10 암세포에 세포 투과되는 것을 보여주는 형광현미경 분석결과를 나타낸 것이다(DAPI 염색은 세포 핵의 위치를 나타내며 merge 이미지는 각각의 이미지가 중첩된 결과이다).
도 10은 CPZ, Amil 및 Filip으로 이루어진 군으로부터 선택된 1종 이상의 저해제를 처리하였을 때의 세포 투과 감소됨을 형광현미경을 통해서 보여주는 결과를 나타낸 것이다.
도 11은 100배 고농도의 YSA 펩타이드를 먼저 처리한 세포에 Cy5.5-p19-YSA/siRNA 결합체를 처리한 후의 형광현미경 및 flow cytometry를 나타낸 것이다.
도 12a 및 도 12b는 HeyA8, A549, MDA-MB231, SKOV3ip1, B16F10, HeLa, H9C2 및 HEK293 세포에 Cy5.5-p19-YSA/siRNA 결합체를 처리한 후의 flow cytometry를 나타낸 것이다.
도 13은 p19-YSA/siRNA 결합체를 형광단백질 RFP가 발현되는 암세포 (RFP/B16F10)에 처리한 후의 형광현미경 분석 결과(그림 A), 이의 정량분석 결과 (그림 B), 시간에 따른 유전자 발현 억제 효과를 보여는 결과(그림 C)를 나타낸 것이다(Control: RFG 유전자가 발현되는 B16F10 세포, p19-YSA/siRNA: siRNA 전달용 재조합 단백질을 이용하여 siRNA를 세포에 주입한 경우, p19-YSA/sc siRNAN: RFP 유전자에 매칭하지 않는 scramble siRNA를 siRNA 전달용 재조합 단백질에 봉입하여 세포에 주입한 경우, LF/siRNA: 기존 siRNA 전달체인 리포펙타민을 사용한 경우, p19-YSA: siRNA 없이 siRNA 전달용 재조합 단백질만을 주입한 경우).
도 14는 도 13에서 RFP 유전자의 mRNA가 siRNA에 의하여 분해되었음을 RT-PCR을 통하여 확인한 전기영동 결과 및 정량분석 결과를 나타낸 것이다 (Control: RFG 유전자가 발현되는 B16F10 세포, p19-YSA/siRNA: siRNA 전달용 재조합 단백질을 이용하여 siRNA를 세포에 주입한 경우, p19-YSA/sc siRNAN: RFP 유전자에 매칭하지 않는 scramble siRNA를 siRNA 전달용 재조합 단백질에 봉입하여 세포에 주입한 경우, LF/siRNA: 기존 siRNA 전달체인 리포펙타민을 사용한 경우).
도 15은 도 13에서 RFP 유전자의 mRNA가 siRNA에 의하여 분해되어 RFP 단백질의 발현이 감소되었음을 RFP 항체를 이용하여 보여주는 western blot 결과를 나타낸 것이다 (Control: RFG 유전자가 발현되는 B16F10 세포, p19-YSA/siRNA: siRNA 전달용 재조합 단백질을 이용하여 siRNA를 세포에 주입한 경우, p19-YSA/sc siRNAN: RFP 유전자에 매칭하지 않는 scramble siRNA를 siRNA 전달용 재조합 단백질에 봉입하여 세포에 주입한 경우, LF/siRNA: 기존 siRNA 전달체인 리포펙타민을 사용한 경우).
Claims (14)
- siRNA 결합 단백질 및 표적지향성 펩타이드를 포함하며,
상기 siRNA 결합 단백질은 p19 RNA 결합 단백질(p19 RNA binding protein)로서,
p19 RNA 결합 단백질(p19 RNA binding protein)의 C 말단에 서열번호 3의 아미노산 서열을 포함하는 표적지향성 펩타이드가 결합된 것을 특징으로 하는,
siRNA 전달용 재조합 단백질. - 삭제
