Abstract
본 발명은 암 특이적 핵산 분자 전달용 조성물 및 이를 이용한 항암 치료용 조성물에 관한 것이다. 본 발명은 새로운 RS 융합 펩타이드를 이용하여 전달하고자 하는 핵산 분자와 나노복합체를 형성하여 암세포에 특이적으로 전달 효율도 극대화할 수 있는 유전자 전달 시스템으로 유용하게 이용될 수 있다. 본 발명은 암을 포함한 난치병들을 특이적으로 치료하는 새로운 유전자 치료제로 적용될 수 있다.
Description
본 발명은 유방암 세포에 특이적인 세포 투과성 펩타이드 (Cell Penetrating Peptide SP82)와 폴리 아르기닌(Polyarginine)의 RS 융합 펩타이드를 유효성분으로 포함하는 핵산 분자 전달용 조성물에 관한 것이다.
유전자 치료는 유전물질을 세포 내부로 전달하여 특정 유전자의 발현을 제어하는 기술로, 흔히 비정상적인 유전자 발현을 억제하거나 정상 단백질을 발현하는데 사용된다. 최근에는 이러한 유전자 치료 기술로 RNA interference 기술인 small interference RNA (siRNA)가 주목받고 있다. siRNA는 약 21 염기 쌍 정도의 짧은 RNA 가닥으로 목적하는 mRNA와의 상보성 결합을 이용해 특정 유전자의 발현을 조절한다. 그 기작은 세포질에서 RNA induced silencing complex(RISC)와 복합체를 형성하여 siRNA의 상보성으로 목적 mRNA와 결합하고 RISC의 절단 능력으로 절단을 유도함으로써 단백질의 번역과정이 일어나지 못하도록 억제하여 발현을 조절한다. 따라서 이러한 방법을 통해 유전질환뿐만 아니라 암, 바이러스 등 다양한 질환들을 초기 단계에서부터 치료하려는 연구들이 진행되고 있다. 하지만 siRNA는 핵산으로 인산기로 인해 음전하를 가지기 때문에 세포 내부로의 투과가 어렵다는 단점을 가지고 있다.
따라서 이러한 문제를 극복하기 위해 siRNA의 구조를 변형시켜 안정성을 높이거나 다양한 전달체를 이용하여 전달하는 방법이 있다. siRNA의 자체 구조를 변형시키는 방법은 당, 인산, 염기를 변형시켜 생체 내에서 안정성을 높이는 방법이다. 이러한 방법을 통해 siRNA의 혈청 내에서 지속력을 높이는 것으로 확인되었지만, 그에 따라 RNAi 효율은 크게 떨어져 사용에 어려움이 존재한다. 다음으로 전달체를 이용한 siRNA 전달에는 바이러스 기반의 플라스미드 siRNA 전달과 비바이러스 기반의 siRNA전달이 있다. 바이러스 기반의 전달체는 높은 효율로 유전물질을 세포내로 전달하는 장점을 가지고 있지만, 환자에게 다양한 합병증을 야기하고 면역 반응을 일으킬 시에 RNAi 효율을 낮추는 단점을 가지고 있다. 이와 반대로 비바이러스성 전달체는 바이러스성 전달체에 비해 낮은 전달 효율을 가지고 있지만 안정적이고 높은 생체적합성을 가지고 있다. 비바이러스성 전달체의 예시로는 리포좀, 폴리머 등이 존재한다. 하지만 각각은 혈청에서의 낮은 안정성, 낮은 특이성, 낮은 전달 효율, 낮은 생체적합성 등의 이유로 사용이 제한적이다.
그에 비하여 펩타이드 기반의 전달체는 cell penetrating peptide(CPP)로 높은 생체 적합성과 서열에 따라서 특이성이나 핵산과의 상호 작용, 엔도좀으로부터의 탈출 기능 등의 다양한 특성을 가질 수 있다는 장점을 가지고 있다.
본 발명의 목적은, 펩타이드에 양전하를 부여하여 정전기적 상호작용을 통해 핵산과 나노복합체를 형성하면 유전자 치료 물질인 siRNA의 단점인 친수성이며 음전하를 갖는 특징을 펩타이드 서열 디자인을 통해 siRNA의 세포 내부로의 전달 효율을 증가시킬 수 있는 암 특이적 핵산 분자 전달용 조성물 및 그 용도를 제공하는 것이다.
또한, 유방암에 특이성을 보이는 homing peptide를 이용하면 일반적으로 목적하는 세포의 표면에만 발현되고 일반 세포에는 발현되지 않는 수용체 혹은 마커 등을 표적으로 할 수 있다. 대부분의 homing peptide는 파지 디스플레이를 통해 그 서열을 발견하며, 동물 실험에서도 종양 부위에만 들어가는 것으로 알려져 있다.
따라서, 물질들을 homing peptide와 연결하면, 세포 내부로 전달이 가능하여 여러 인자나 약물 등을 연결하여 특이적으로 전달하여 기타 독성을 최소화 하여 암의 진행 상황의 확인이나 치료 등에 활용이 가능하다.
본 발명에서는 목적 물질을 세포 내부로 높은 효율로 전달할 뿐만 아니라 유방암 세포에 특이적으로 전달할 수 있는 기술을 개발하기 위해서 노력하였다. 본 발명의 목적을 성취하기 위해 세포 투과력이 좋은 것으로 밝혀진 폴리아르기닌 11개 서열과 무작위 서열을 이용하여 파지 디스플레이를 통해 유방암 특이적인 세포 투과를 야기하는 펩타이드로 알려진 SP82를 융합한 새로운 융합 펩타이드를 디자인하였다.
이를 통해 핵산과 나노복합체를 형성해 EPR(Enhanced Permeability and Retention Effect) 효과를 이용해 세포 내부로의 전달과 유방암 특이적으로 물질을 전달하는 펩타이드인 SP82를 이용하여 높은 특이성과 효율로 세포 내부로 siRNA를 전달할 것으로 기대한다.
본 발명의 목적은 세포로의 핵산 분자 전달용 조성물을 제공하는 데 있다. 본 발명의 다른 목적은 암 치료용 조성물을 제공하는 데 있다.
본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명, 청구범위 및 도면에 의해 보다 명확하게 된다.
상기 과제를 해결하기 위하여, 본 발명자들은 특정 타입의 암세포에 대해 특이적이고, 높은 효율로 핵산 분자를 전달할 수 있는 유전자 전달 시스템 (gene delivery system)을 개발하기 위하여 연구 노력하였다. 이를 위하여, SP82 펩타이드와 폴리아르기닌 펩타이드를 조합하여 목적 세포에 특이성과 투과성이 상승함을 발견함으로써, 본 발명을 완성하게 되었다.
본 발명의 일 양태에 따르면, 본 발명은 암 세포 진입(Cell Permeating, SP82)펩타이드 및 폴리아르기닌(polyarginine) 펩타이드를 포함하는 세포로의 핵산 분자 전달용 조성물을 제공한다.
구체적으로, 본 발명의 일 실시형태에 따르면, 서열목록 제1 서열: QLEFYTGLAKLI을 갖는 암 세포 진입(Cell Permeating, SP82)펩타이드 및 서열목록 제2 서열: RRRRRRRRRRR (R11)을 갖는 폴리아르기닌(polyarginine) 펩타이드를 포함하는 암 특이적 핵산 분자 전달용 조성물을 제공한다.
본 발명자들은 높은 효율과 특이성을 가지고 핵산 분자를 목적 세포 내로 전달할 수 있는 우수한 유전자 전달 시스템(gene delivery system)을 개발하기 위하여 예의 연구 노력하였다. 그 결과, SP82 펩타이드와 폴리아르기닌 펩타이드의 조합을 이용하여 전달하고자 하는 핵산 분자와 나노복합체를 형성할 경우 세포막에 대한 투과성이 크게 개선될 뿐만 아니라 특이성도 향상되어 목적 세포로의 핵산 분자 전달 효율이 현저하게 상승함을 발견하였다.
본 발명에 따르면, 하기 실시예에서 보는 바와 같이 SP82 펩타이드와 폴리아르기닌 펩타이드를 융합하여 사용한 경우 핵산 분자의 전달의 효율에 있어 유의한 상승적 효과(synergistic effect)를 나타낸다.
SP82 펩타이드는 유방암 세포에 대한 투과성을 개선하기 위해 사용되는 12개의 아미노산으로 이루어진 합성 폴리펩타이드로서, 거대분자의 국소적 전달에 주로 사용된다. 한편, 폴리아르기닌은 다수의 아르기닌(Arg) 잔기가 연속적으로 연결된 양이온성 합성 펩타이드로서, 음이온을 띄는 핵산 분자의 전하를 중화시켜 지질 이중막으로 구성된 세포막의 투과성을 향상시킨다.
본 발명의 구체적인 구현예에 따르면, 본 발명에서 사용되는 폴리아르기닌 펩타이드는 11개의 아르기닌 잔기로 구성된다.
본 발명의 구체적인 구현예에 따르면, 상기 SP82 펩타이드는 서열목록 제1 서열의 아미노산 서열을 포함한다.
본 발명의 SP82 펩타이드 및 폴리아르기닌 펩타이드는 분리된 각각의 분자로서 목적 핵산분자를 포함하는 시료에 동시에 또는 임의의 순서로 순차적으로 첨가될 수도 있고, 또는 하나의 융합된(fused) 형태로 이용될 수도 있다. 융합된 형태로 이용되는 경우, 상기 SP82 펩타이드 및 상기 폴리아르기닌 펩타이드가 직접 공유 또는 비공유 결합을 할 수도 있고, 또는 두 펩타이드가 링커로 연결될 수도 있다. 두 펩타이드가 링커로 연결될 경우, 상기 링커는 서열목록 제3서열을 포함하는 폴리펩타이드이다.
상기 암 세포 진입(Cell Permeating, SP82)펩타이드 및 폴리아르기닌(polyarginine) 펩타이드는 융합하여 RS 융합 펩타이드를 형성하는 것을 특징으로 한다.
상기 RS 융합 펩타이드는 상기 SP82 펩타이드와 상기 폴리아르기닌(polyarginine) 펩타이드가 서열목록 제3 서열: GGGG을 갖는 링커에 의해 서로 연결된 형태인 것을 특징으로 한다.
상기 RS 융합 펩타이드는 비극성 아미노산(A, G, I, L), 전하를 띠지 않는 극성 아미노산(Q, Y, T), 양전하를 가지는 아미노산(K, R)으로 이루어져 있는 것을 특징으로 한다.
상기 RS 융합 펩타이드는 염기성 부분이 전체의 44±11%, 산성 부분이 전체의 4±1%, 소수성 부분이 전체의 22±6.5% 및 중성 부분이 전체의 30±7.5%로 차지하는 것을 특징으로 한다.
상기 RS 융합 펩타이드는 서열 상동성이 반드시 일치하는 것만을 한정하는 것은 아니며, 예를 들어 펩타이드 서열 상동성이 75% 이상인 경우라면, 유사한 서열로서 본 발명의 RS 융합 펩타이드에 포함되는 것으로 인정할 수 있다.
본 발명의 다른 실시형태에 따르면, 암 세포 진입(Cell Permeating, SP82)펩타이드 및 폴리아르기닌(polyarginine) 펩타이드가 융합하여 형성된 RS 융합 펩타이드에 핵산 분자가 결합된 암 특이적 핵산 분자 전달용 나노 복합체를 제공한다.
상기 핵산 분자는 gDNA 및 cDNA를 포함하는 DNA 분자와 mRNA 및 siRNA를 포함하는 RNA 분자 중 어느 하나 이상인 것을 특징으로 하며, 특히 siRNA 인 것을 특징으로 한다.
상기 RS 융합 펩타이드의 아민 그룹(N)과 상기 핵산 분자의 인산 그룹(P)의 비율(N/P ratio)는 5:1 이상인 것을 특징으로 한다. 구체적으로, 상기 RS 융합 펩타이드의 아민 그룹(N)과 상기 핵산 분자의 인산 그룹(P)의 비율(N/P ratio)는 5:1, 7.5:1, 10:1 혹은 그 이상인 것을 특징으로 한다.
본 발명의 다른 실시형태는 상기 암 특이적 핵산 분자 전달용 조성물을 포함하는 암 및 염증 치료용 조성물을 제공한다.
본 발명의 또 다른 실시형태는 상기 특이적 핵산 분자 전달용 나노 복합체를 세포와 접촉시키는 단계를 포함하는 암 특이적 핵산 분자 전달용 나노 복합체를 세포에 전달하는 방법을 제공한다.
본 발명의 특징 및 이점을 요약하면 다음과 같다:
(a) 본 발명은 세포로의 핵산 분자 전달용 조성물 및 이를 이용한 유방암 치료용 조성물을 제공한다.
(b) 본 발명은 SP82 펩타이드 및 폴리아르기닌 펩타이드의 조합을 이용하여 전달하고자 하는 핵산 분자와 나노복합체를 형성함으로써 세포막에 대한 투과성이 현저하게 개선되어, 효율적인 유전자 전달 시스템으로 유용하게 이용될 수 있다.
(c) 본 발명의 조성물은 특히 siRNA의 세포 내 투과 효율을 증진시킬 뿐 아니라 siRNA를 특정 세포에 전달하여 특정 질병 세포만을 타겟팅하여 전달할 수 있다.

본 발명의 구체적인 구현예에 따르면, 상기 핵산 분자는 목적 유전자의 발현 및 발현 억제용 핵산 분자이다. 본 명세서에서 용어 "발현"은 목적 유전자의 발현을 의미하며, "발현의 억제"는 목적 유전자의 활성 또는 발현의 저하를 야기시키는 것을 의미하며, 이에 의해 목적 유전자의 활성 또는 발현이 탐지 불가능해지거나 무의미한 수준으로 존재하게 되는 경우 뿐 아니라, 타겟 유전자의 생물학적 기능이 유의하게 저하될 수 있을 정도로 활성 또는 발현을 저하시키는 것을 의미한다. 본 발명의 발현 및 발현 억제용 핵산 분자는 구체적으로는 목적 유전자의 서열에 상보적인 핵산분자로서, 예를 들어 mRNA, shRNA, siRNA, miRNA, gRNA(guideRNA) 및 안티센스 올리고뉴클레오타이드를 포함하나, 이에 제한되지 않고 당업계에 공지된 모든 유전자의 발현 및 발현 억제수단인 핵산분자가 이용될 수 있다.구체적으로는, 본 발명에서 사용되는 발현 억제용 핵산 분자는 siRNA이다.
본 명세서에서 용어 "siRNA(small interfering RNA)"는 인 비보 상에서 단일 가닥 21개로 구성된 뉴클레오타이드로서, RNA 간섭을 통해 타겟 유전자의 발현을 억제하기 위한 RNA 서열을 의미한다.

본 명세서에서 용어 "gRNA(guideRNA)"는 타겟 유전자를 인식하여 핵산분해효소(nuclease)를 유도함으로써 인식된 분위를 특이적으로 절단하는 유전자 편집 시스템에 사용되는 RNA 분자를 의미한다. 이러한 유전자 편집 시스템에는 대표적으로 CRISPR(Clustered Regularly Interspaced Short Palindromic Repeats) 시스템이 있다.
본 명세서에서 용어 "발현시키다"는 대상체가 외래(exogenous) 유전자를 발현하게 하거나 또는 내인성(endogenous) 유전자의 자연적 발현량을 증가시키기 위해 유전자 전달체를 이용하여 인위적으로 이를 도입함으로써 유전자가 대상체 세포 내에서 염색체외 인자로서 또는 염색체 통합 완성에 의해 복제 가능하게 되는 것을 의미한다. 따라서, 용어 "발현"은 "형질전환(transformation)", "형질감염(transfection)" 또는 "형질도입(transduction)"과 동일한 의미이다.
본 명세서에서, 용어 "유전자 전달 시스템(gene delivery system)"은 유전자를 세포 내로 운반하는 모든 수단을 의미하며, 유전자 전달은 유전자의 세포내 침투(transduction)와 동일한 의미를 가진다. 조직 수준에서, 상기 용어 유전자 전달은 유전자의 확산(spread)과 동일한 의미를 가진다. 따라서, 본 발명의 유전자 전달 시스템은 유전자 침투 시스템 및 유전자 확산 시스템으로 기재될 수 있다.
본 명세서에서 용어 "치료"는 (a) 질환, 질병 또는 증상의 발전의 억제; (b) 질환, 질병 또는 증상의 경감; 또는 (c) 질환, 질병 또는 증상을 제거하는 것을 의미한다. 본 발명의 조성물이 대상체에 투여되면 타겟 유전자의 발현 억제를 통해 유방암 세포의 증식 및 성장을 억제하여 유방암을 치료하는 역할을 한다. 따라서, 본 발명의 조성물은 그 자체로 염증 또는 자가면역 질환의 치료 조성물이 될 수도 있고, 혹은 다른 약리성분과 함께 투여되어 상기 질환에 대한 치료 보조제로 적용될 수도 있다. 이에, 본 명세서에서 용어 "치료" 또는 "치료제"는 "치료 보조" 또는 "치료 보조제"의 의미를 포함한다.
본 명세서에서 용어 "투여" 또는 "투여하다"는 본 발명의 조성물의 치료적 유효량을 대상체에 직접적으로 투여함으로써 대상체의 체내에서 동일한 양이 형성되도록 하는 것을 말한다.
본 발명에서 용어 "치료적 유효량"은 본 발명의 조성물을 투여하고자 하는 개체에게 치료적 또는 예방적 효과를 제공하기에 충분한 정도로 함유된 조성물의 함량을 의미하며, 이에 "예방적 유효량"을 포함하는 의미이다.
본 명세서에서 용어 "대상체"는 제한없이 인간, 마우스, 래트, 기니아 피그, 개, 고양이, 말, 소, 돼지, 원숭이, 침팬지, 비비 또는 붉은털 원숭이를 포함한다. 구체적으로는, 본 발명의 대상체는 인간이다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시 예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실험방법
siRNA/RS 융합 펩타이드 나노복합체 형성
대조군 siRNA(Genolution, Seoul, Korea), 실험군 siRNA (GenePharma, Shanghai, China), 유방암을 사멸시키는 siRNA (Genolution, Seoul, Korea)를 합성하여 nuclease-free water(Integrated DNA Technologies Inc., IA, USA)에 1 μM가 되도록 녹였다. RS 융합 펩타이드(GL Biochem Ltd., Shanghai, China)인 SP82-R11(RS): RRRRRRRRRRRGGGGQLEFYTGLAKLI는 95% 이상의 순도를 가지도록 합성하여 3차 증류수에 2μM이 되도록 녹였다.
본 발명의 일 실시형태에 따르면, 서열목록 제1 서열: QLEFYTGLAKLI을 갖는 암 세포 진입(Cell Permeating, SP82)펩타이드 및 서열목록 제2 서열: RRRRRRRRRRR (R11)을 갖는 폴리아르기닌(polyarginine) 펩타이드를 포함하는 암 특이적 핵산 분자 전달용 조성물을 제공한다.
상기 암 세포 진입(Cell Permeating, SP82)펩타이드는 하기의 서열목록 제1 서열을 가진다.
제1 서열: QLEFYTGLAKLI
상기 폴리아르기닌(polyarginine) 펩타이드는 11개의 아르기닌 잔기로 구성된다.
구체적으로, 상기 폴리아르기닌(polyarginine) 펩타이드는 하기의 서열목록 제2 서열을 가진다.
제2 서열: RRRRRRRRRRR (R11)
상기 암 세포 진입(Cell Permeating, SP82)펩타이드 및 폴리아르기닌(polyarginine) 펩타이드는 융합하여 RS 융합 펩타이드를 형성하는 것을 특징으로 한다.
본 발명에서는 두 가지의 기능성을 가지는 펩타이드 서열 사이에 링커 서열(GGGG)을 추가하여 디자인하였다.
상기 RS 융합 펩타이드는 상기 SP82 펩타이드와 상기 폴리아르기닌(polyarginine) 펩타이드가 서열목록 제3 서열: (Gly Gly Gly Gly)을 갖는 링커에 의해 서로 연결된 형태인 것을 특징으로 한다.
상기 링커는 하기의 서열목록 제3 서열을 가진다.
제3 서열: GGGG
RS 융합 펩타이드의 구조는 크게 SP82 펩타이드 및 폴리아르기닌 펩타이드 사이에 링커 서열로 구성되며, RS 융합 펩타이드는 암세포에 특이적 전달 기능을 폴리아르기닌은 양전하를 띄어 음전하인 핵산과 정전기적인 상호작용을 하여 자가조립 기능을 가진다. 반면에 링커 서열은 두 펩타이드가 서로 영향을 주고 받지 않으며 활동할 수 있는 중간다리 역할을 한다. RS 융합 펩타이드의 아미노산 조성을 분석하면 RS 융합 펩타이드는 비극성 아미노산(A, G, I, L), 전하를 띠지 않는 극성 아미노산(Q, Y, T), 양전하를 가지는 아미노산(K, R)으로 이루어져 있으며 염기성 부분이 전체의 44%, 산성 부분이 4%, 소수성 부분이 22% 및 중성 부분이 30% 내외이기 때문에 siRNA와의 정전기적 상호작용이 가능하다. 구체적으로, 상기 RS 융합 펩타이드는 염기성 부분이 전체의 44±11%, 산성 부분이 전체의 4±1%, 소수성 부분이 전체의 22±6.5% 및 중성 부분이 전체의 30±7.5%로 차지하는 것을 특징으로 한다. siRNA/RS 융합 펩타이드의 자가 조립 나노복합체 형성을 위하여, 펩타이드의 아민 그룹(N)과 RNA의 인산 그룹(P)의 비율을 계산한 후 적절한 비율에 따라 상온에서 30분 동안 배양하였다.
겔 지연 분석
대조군 siRNA와 RS 융합 펩타이드의 N/P 비율을 0.005:1, 0.01:1, 0.025:1, 0.05:1, 0.1:1, 0.25:1, 0.5:1, 1:1까지 계산하여 각각 상온에서 30분 동안 배양하였다. 1% 아가로스 겔 (w/v)을 10,000X TopRed Nucleic Acid Gel Stain (GenomicBase, Seoul, Korea)이 들어있는 1X TAE 완충액에서 제조하고 각 샘플에 6X 로딩 dye (Biofact)를 첨가한 후 겔에 로딩하였다. Mupid-2plus 전기영동 시스템(Optima Inc., Tokyo, Japan)을 이용하여 35분 동안 전기영동을 수행한 후 결과를 겔 기록 시스템 LSG 1000(iNtRON biotechnology, Seongnam, Korea)으로 확인하였다.
나노복합체의 크기 및 제타 전위 측정
대조군 siRNA와 RS 융합 펩타이드의 N/P 비율을 5로 하여 상온에서 30분 동안 배양하였다. 최종 siRNA의 농도가 200 nM이 되도록 nuclease-free water를 첨가하여 희석한 후, 10 mL 주사기와 0.45 μm 주사기 필터를 이용하여 여과하고 30초 동안 볼텍싱하였다. Zetasizer Nano ZS(Malvern Instruments Ltd., Worcestershire, UK)를 이용하여 크기와 제타 전위를 측정하였다.
세포 배양
인간 삼중음성유방암 세포인 MDA-MB-231 세포를 T75 cell culture flasks (SPL Life Science, Korea)에 10% FBS와 1% 페니실린-스트렙토마이신(Life Technologies, CA, USA)이 포함된 8 mL의 DMEM(dulbecco's modified eagle's medium, Corning, MA, USA)에서 37℃, 5% CO2 표준 조건으로 CO2 incubator (CCL-170B-8-FD; ESCO, Singapore)에서 배양하였다. 계대 배양은 세포의 컨플루언시 80~90%로 3-4일 간격으로 수행하였다.
유세포 분석
MDA-MB-231, MCF-7, BT-474, RAW264.7 세포를 6-웰 플레이트에 2.0 x 106 세포/웰로 총 2 mL씩 씨딩한 후 37℃, 5% CO2에서 18시간 동안 배양하였다. 배지를 Opti-MEM(Gibco, USA)으로 교체한 후, 200nM의 FITC가 표지된 siRNA(GenePharma, Shanghai, China)와 RS 융합 펩타이드를 5:1의 N/P 비율로, R11 펩타이드를 20:1의 N/P 비율로 상온에서 30분 동안 배양하여 형성된 나노복합체를 세포에 처리하고 표준 조건에서 4시간 동안 배양하였다. Dubercco's phosphate buffered saline (DPBS)(sigma-aldrich)로 세척한 후 각 웰에 트립신-EDTA(Thermo Fisher Scientific, MA, USA)를 첨가하여 세포 부착성을 떨어뜨리고, 배지 2mL를 첨가하여 300 x g에서 5분 동안 원심분리하였다. 상층액을 제거하고 DPBS로 재부유한 후 동일한 조건에서 원심분리를 2번 반복하였다. 최종적으로 pellet을 차가운 PBS로 재부유하여 유세포 분석기 전용 튜브에 옮겨 유세포분석기 (CytoFLEX, Beckman, USA)를 이용하여 측정하였다.
세포 독성 확인
MDA-MB-231 세포를 동일한 배지와 조건에서 배양하였다. 충분히 자란 세포를 96-웰 플레이트에 5.0×10³세포/웰로 씨딩하여 37℃, 5% CO2에서 20시간 동안 배양하였다. RS 융합 펩타이드를 1, 2, 3, 4 μM의 농도로 희석하여 세포에 4시간 처리한 후 48시간 표준 조건에서 배양한 뒤, D-PlusTM CCK cell viability assay kit (Dongin biotech, Seoul, Korea)을 2시간 반응시킨 뒤 마이크로 플레이트 리더 (EPOCH2; BioTek, Winooski, VT, USA)를 이용하여 흡광도를 측정하였다. 그 후 RS 융합 펩타이드가 처리되지 않은 세포의 흡광도를 100%로 환산하여 비교하였다.
실험결과
siRNA/RS 융합 펩타이드 나노복합체 형성 및 특성 분석
siRNA와 RS 융합 펩타이드가 상호작용하여 형성한 나노복합체를 확인하고 특성을 분석하였다. RS 융합 펩타이드 없이 siRNA만 로딩하였을 때 RNA 밴드를 확인하였다. 이 위치를 기준으로 RS 융합 펩타이드가 N/P 비율 0.5:1로 첨가되었을 때까지는 거의 동일한 위치에 밴드가 나타났지만, 밴드가 약간 끌리며 밝기는 조금 감소한 것을 확인하였다. 그러나 RS 융합 펩타이드가 N/P 비율 3:1로 증가하였을 때부터는 RNA 밴드의 지연이 일어나 로딩 웰에 밴드가 관찰되었다 (도 1). 따라서 RS 융합 펩타이드는 3:1의 N/P 비율 이상부터 siRNA와 효과적으로 상호작용하여 나노복합체를 형성할 수 있음을 확인하였다.
siRNA와 RS 융합 펩타이드 나노복합체의 크기와 제타 전위를 DLS(dynamic light scattering) 분석으로 측정하였다. 나노복합체는 N/P 비율 5:1에서 278 nm의 평균 크기를 나타내었다. 또한, 모든 비율에서 평균 PdI 값이 0.25 정도로 매우 균일한 크기를 가지는 것을 확인하였다. 이를 통해 5:1의 N/P ratio에서는 상호작용이 충분하여 응축되고 균일한 나노복합체가 형성된 것임을 알 수 있으며 이는 겔 지연 분석 결과와 유사한 양상을 나타내었다. 또한, 5:1의 N/P ratio에서는 siRNA의 음전하를 모두 상쇄시켜 약한 양전하를 나타내었다(도 2). 따라서 이후 실험에서는 RS 융합 펩타이드의 비율을 5:1로 고정하여 실험을 진행하였다.
siRNA/펩타이드 나노복합체의 세포 내 투과 효율 평가
정량적인 세포 투과율을 확인하기 위하여 유세포 분석기를 사용하여 앞의 실험과 동일한 조건에서 형광세기를 비교하였다. MDA-MB-231, MCF-7, BT-474, SK-BR-3 세포에서 RS 융합 펩타이드가 없는 siRNA에 비하여 나노복합체의 경우 형광 세기가 증가한 세포들이 측정되어 뚜렷한 이동(shift)이 90.03, 81.54, 55.12, 32.5%로, 33.45%로 분석된 일반세포에(RAW264.7) 비해 특이성을 나타내었다(도 3a, 3b, 3c, 3d, 3e). 이는 RS 융합 펩타이드가 일반세포에 비해 특정 암세포에 특이적으로 전달된다는 결과를 확인할 수 있었다. 또한 양성 대조군인 R11에 비하여 핵산 전달 효율이 우수한 결과를 확인할 수 있었다. 또한, FITC가 표지된 siRNA가 세포 내부로 전달된 비율을 정량화하여 그래프로 나타내었을 때, RS 펩타이드가 R11 펩타이드보다 크게 향상된 siRNA 전달효율을 보여준다. 유방암 세포 중 전달 효율이 높은 순으로 MDA-MB-231, BT-474, MCF-7이었고, SK-BR-3와 일반세포인 RAW264.7은 차이를 보이지 않았다. 본 측정 결과 특정 타입의 유방암 세포에 특이성을 확인할 수 있었다 (도 4).
RS 융합 펩타이드의 세포 독성 평가
RS 융합 펩타이드가 세포에 미치는 독성의 영향을 HDF-n, MDA-MB-231 세포에서 확인하였다(도 5a, 5b). RS 융합 펩타이드의 농도가 증가하여도 흡광도 수준은 대조군과 비교하여 두 세포에서 모두 통계적으로 유효한 차이가 확인되지 않았다. 앞선 세포 실험에서 사용한 RS 융합 펩타이드 농도는 이 범위 안에 포함되기 때문에 사용 범위 내에서 세포 독성 없이 물질 전달이 가능한 것으로 확인하였다.
이상, 본 발명의 일 실시예에 대하여 설명하였으나, 해당 기술 분야에서 통상의 지식을 가진 자라면 특허청구범위에 기재된 본 발명의 사상으로부터 벗어나지 않는 범위 내에서, 구성 요소의 부가, 변경, 삭제 또는 추가 등에 의해 본 발명을 다양하게 수정 및 변경시킬 수 있을 것이며, 이 또한 본 발명의 권리범위 내에 포함된다고 할 것이다.
