본 발명은 흰점박이꽃무지(Protaetia brevitarsis)유충으로부터 분리된 프로태티아마이신의 α-helix영역의 9개 잔기서열을 바탕으로 설계된 서열번호 1,2,3,4로 기재된 항생 펩타이드 9Pbw 1,2,3,4와 그의 용도 및 작용기작에 관한 것이다. 본 발명의 새로운 항균 펩타이드 4종 중 9Pbw1,2,4는 탁월한 항균 활성을 나타내고 세포 독성이 없으므로 인체에 안전한 항생제로 유용하게 사용될 수 있다.

본 발명은 인체에 항생제로서 유용하게 사용될 수 있는 펩타이드에 관한 것으로 더욱 상세하게는 딱정벌레목에 속하는 흰점박이꽃무지(Protaetia brevitarsis)유충으로부터 분리된 프로태티아마이신의 helix영역의 잔기서열을 기반으로 하여 설계된 유도체에 관한 것이다.
페니실린 이후 수많은 종류의 항생제가 개발되어 생체에 외부침입의 세균을 퇴치를 위하여 사용되어 왔다. 그러나 최근에 들어서 이들 항생제에 내성을 가지는 균주들이 등장하여 큰 문제로 여겨진다. 생명에 위협을 가할 수 있는 엔테로코쿠스 패칼리스 (Enterococcus faecalis), 마이코박테리움 투버쿨로시스 (Mycobacterium tuberculosis) 및 슈도모나스 애루기노사 (Pseudomonas aeruginosa) 등의 세균 종들은 지금까지 알려진 모든 항생제에 대한 저항력을 키워 왔다 (Stuart B. Levy, Scientific American, 46-53, 1998). 페니실린, 세파로스포린등과 같은 종래의 화학항생제는 미생물의 세포벽 또는 단백질의 합성저해에 의하여 항생작용을 나타낸다. 이들 내성균주를 퇴치할 수 있는 새로운 작용 메카니즘을 가지는 항생제의 개발이 시급한 실정이며 종래의 화학 항생제와 상이한 항생기전을 나타내는 항생펩타이드들은 새로운 개념의 차세대 항생제로서 주목을 받고 있다 (Zasloff M. 넓은 의미로서 면역반응으로 불리어지는 항원항체 반응은 인간을 포함한 무척추동물에 있어서 생체방어의 주된 역할을 해 왔다. 그러나 곤충은 항원 항체반응과 같은 후천적 면역기구와는 다른 선천적 면역기구를 강화하는 것으로 자신을 보호하고 있다. 이와 같이 선천적 면역은 곤충뿐만 아니라 사람을 포함한 척추동물에 있어서도 초기감염에 대해 중요한 방어계로서 역할을 하고 있다. 곤충의 선천적 면역기구의 아주 중요한 수단은 다양한 미생물에 대해 살균, 불활성화작용을 가진 물질을 획득한다는 것이다. 그 중에서도 항균성 펩타이드를 중심으로한 항미생물작용을 가진 펩타이드나 단백질은 우리 인간을 포함한 가축등의 질병 방제에 대해서 이용 가능성 높다. 이러한 곤충유래 항병원성 단백질은 인간 및 가축의 질병에 대한 가치뿐만 아니라 각종 병원균의 감염에 의해 야기되는 농작물의 피해 방지 및 치료를 위해서도 매우 유용하게 사용될 수 있을 것이다.
항생 펩타이드는 구조에 따라 세 개의 그룹으로 나눌 수 있다. 첫 번째는 시스테인이 풍부한 (cysteine-rich) β-쉬트(sheet) 펩타이드이고, 두 번째는 α-나선형 (helical)의 양친매성 펩타이드 분자이며, 세 번째는 프롤린이나 트립토판등의 특정 아미노산이 풍부한 (proline-rich, Trp-rich) 펩타이드이다 (Mayasaki et al., Int. J. Antimicrob. Agents, 9, 269-280, 1998; Andreu, D.; Rivas, L. Biopolymers 1998, 47, 415-433). 이들 항균 펩타이드들은 숙주 방어 및 선천적 면역계에 있어서 중요한 역할을 담당하는 것으로 알려져 있다. 또한, 상기 항균 펩타이드들은 아미노산 서열에 따라 다양한 구조를 갖는데, 이들 구조 중 가장 흔한 것은 곤충에서 발견된 항균 펩타이드인 멜리틴 (melittin)이나 세크로핀 (cecropin)과 같이 시스테인 (cysteine) 잔기가 없고 양친화성 α-나선형을 형성하는 구조이다.
현재까지 밝혀진 곤충유래 항균 단백질로서는 쉬파리과(Sarcophaga peregrina)에서 발견된 sacotoxin Ⅲ(Baba et all., J. Biochem, 102, 1, 69-74, 1987), 노랑초파리(Drosophila melanogaster)에서 발견된 diptericin(Wicker et al., J. Chem., 274, 9320-9326, 1999), 남방장수풍뎅이(Oryctes rhinoceros)에서 발견된 Rhinocerosin, Scarabaecin(Yang et al., Eur. J. Biochem, 255, 734738, 1998 ; Tomie et al., Biochemical and Biophysical Research Communications, 307, 2, 261-266, 2003), 매미아과의 Cicada flammata에서 발견된 cicadin(Wang and Ng TB, Peptides, 23, 7~11, 2002), 큰우단하늘소(Acalolepta luxuriosa)로부터 발견된 Cecropin(Saito et al., Comparative Biochemistry and Physiology Part B: Biochemistry and Molecular Biology, 3, 142, 317-323, 2005), 담배 거세미나방(Spodoptera litura)으로부터 발견된 Moricin analogue(Oizumi et al., Biochim. Biophys. Acta, 1752, 83~92, 2005)등이 있다.
본 발명은 상기의 문제점을 해결하고, 상기의 필요성에 의하여 안출된 것으로서 본 발명의 목적은 인체무독성인 항균 펩타이드를 제공하는 것이다.
상기의 목적을 달성하기 위하여 본 발명은
서열번호 1 내지 서열번호 4로 기재된 아미노산 서열을 가지는 펩타이드로 이루어진 군으로부터 선택된 하나의 펩타이드를 제공한다.
또한 본 발명은 상기 본 발명의 펩타이드를 유효성분으로 포함하는 약학적 조성물을 제공한다.
본 발명의 일 구체예에 있어서, 상기 조성물은 인간의 적혈구 세포 또는 상피세포에 대하여 독성을 가지지 않는 것을 특징으로 한다.
또한 본 발명은 본 발명의 펩타이드를 유효성분으로 포함하는 미생물에 대한 항균용 조성물을 제공한다.
본 발명의 바람직한 실시예에 있어서 상기 펩타이드는 서열번호 1, 서열번호 2 또는 서열번호 4에 기재된 아미노산 서열을 가지는 펩타이드인 것이 바람직하나 이들 서열에 하나 이상의 치환, 결손 등의 변이를 통하여 본 발명의 항균특성을 나타내는 모든 변이 펩타이드들도 본 발명의 펩타이드에 포함된다.
본 발명의 일 구체예에 있어서, 상기 미생물은 그람양성균, 그람 음성균 또 는 진균인 것이 바람직하나 이에 한정되지 아니하며, 바람직하게는 상기 그람양성균은 B. subtilis, S. aureus, 또는 E. faecalis, 그람 음성균은 E. coli, P. aeruginosa, 또는 K. pneumoniae, 진균은 Candida Albicans인 것을 포함하나 이에 한정되지 아니한다.
또한 본 발명의 일 구체예에 있어서 상기 항균용 조성물은 인간의 적혈구 세포 또는 상피세포에 대하여 독성을 가지지 않는 것을 특징으로 한다.
이하 본 발명을 설명한다.
본 발명자들은 면역유도시킨 흰점박이꽃무지의 유충으로부터 면역화 유도후 cDNA 유전자 은행을 제작하여 기 보고된 항균 관련 유전자와 상동성이 높은 항진균 펩타이드 유전자를 선발하고, 프로태티아마이신 항균펩타이드의 아미노산 서열을 밝힌바 있다(대한민국 특허출원번호: 10-2008-0109281). 본 발명에서는 프로태티아마이신 항균펩타이드 helix영역의 아미노산 서열에 기반하여 높은 항생활성을 가져 특허 출원된 (대한민국 특허출원번호: 10-2008-0109281) 9Pbm6의 잔기서열에 기반하여 펩타이드 N말단과 C말단에 양이온성 아미노산을 치환하고 중간영역에 세포막과의 상호작용을 증대시킬수 있도록 소수성 잔기를 치환한 양쪽 친매성을 증대시킨 펩타이드를 설계하여 인간의 세포에는 무독하고 높은 항균활성을 가지는 박테리아 선택적 유도체를 설계하고 활성을 측정하여 인체의 항균치료제로 응용될 수 있음을 확인하므로서 본 발명을 완성하였다.
이하 본 발명을 단계에 따라 보다 구체적으로 설명하고자 한다.
본 발명에서는 4종의 항생펩타이드를 합성하여 6종의 박테리아에 대한 항생 활성을 검정하였다. 그 결과 본 발명에 의한 항균 펩타이드의 유도체를 처리시 공시한 6종의 균들의 생장이 억제되었다. 9Pbw1-4 네 펩타이드 모두 대조 펩타이드인 9Pbm6보다 높은 항균활성을 나타냈다. 특히 9Pbw4 항생 펩타이드는 MIC분석 방법을 통해서 E.coli와 S. aureus, B. subtilis 에 대해 2μM/ml의 농도 이하에서도 균의 활성이 억제됨을 확인 할 수 있었다. 한편 항균 활성이 있는 유도체에 대해 세포 독성을 나타내는지를 확인하기 위하여 적혈구 파괴능을 조사한 결과, 양성 대조군으로 사용한 벌독인 멜리틴에 비해 9Pbw1-4 네 개 펩타이드가 현저히 낮은 활성을 가졌다. 특히 9Pbw1,2,4 세 개의 펩타이드는 거의 적혈구를 파괴하지 않는 것을 알 수 있었다.
또한 비록 구체적인 실시예로 제시되지는 않았지만, 항진균 작용이 있는 본 발명에 의한 항균 펩타이드 유도체를 유효성분으로 함유하는 항균제용 약학적 조성물이 가능함이 당업자에게 당연할 것이다.

상기 표는 펩타이드의 아미노산 서열을 나타낸다.
표 1은 기존 발명된 9Pbm6 잔기서열을 기반으로 개발된 본 발명의 서열번호 9Pbw1-4로 기재되는 된 고기능성 펩타이드 유도체 4종의 잔기서열과 비교펩타이드의 잔기서열이다.
우선, 본 발명의 항균 펩타이드 9Pbw1-4가 상기 용도로 사용될 수 있는지 확인하기 위하여 본 발명자들은 본 발명에서 합성한 항균 펩타이드및 대조군 펩타이드에 대하여 그람-음성균 (3종류의 균주) 및 그람-양성균 (3종류의 균주)에 대하여 항균 활성을 측정하였다. 항균 활성은 균체가 분열되지 않는 펩타이드의 최소성장 억제농도 (minimal inhibitory concentration: MIC), 이하 “MIC”라 약칭함)를 측정하여 관찰하였다. 그람 양성균으로써 Bacillus subtilis, Staphylococcus aureus, E. faecalis를, 그람 음성균으로써 Escherichia coli, Psedomonas aeruginosa, K. pneumoniae를 그 대상으로 하였다.
9Pbw1-4 항생펩타이드들은 모두 대조군인 9Pbm6보다 월등한 항생활성을 나타냈다. 특히9Pbw4는 대조군 펩타이드인 멜리틴과 Bacillus subtilis, Staphylococcus aureus 균에 대해 비슷한 높은 활성을 보였으며 대조군 펩타이드인 9Pbm6 보다 평균 4배정도 강한 항균활성을 나타내었다.(표 2 참조).
본 발명의 항균 펩타이드 9Pbw1-4의 숙주세포에 대한 독성을 조사한 결과 대조군인 멜리틴은 독성이 높은 반면 9Pbw 유도체들은 9Pbw3를 제외하고는 고농도에서도 용혈활성을 나타내지 않았다 (도 1 참조).
또한 도 2는 형광염료(fluorescent dye) 칼세인(calcein)을 포획시킨 인간의 적혈구를 모방하는 중성지질인 EYPC/CH (1:1, w/w)리포좀에 대해 본 발명의 펩타이드 9Pbw1-4에 의한 % 염료방출(dye leakage)을 펩타이드 농도에 따라 나타낸 것이다. 멜리틴은 아주 저 농도에서도 100% 염료방출을 나타내 독성이 높으며, 발명펩타이드 중 9Pbw1,2,4가 인간의 세포막을 모방하는 중성지질에서는 약한 염료방출을 나타내서 인간의 세포에는 독성이 없음을 알 수 있다.
상기의 결과로부터, 본원 발명의 서열번호 1,2,4로 기재되는 9Pbw1,2,4펩타이드는 탁월한 항균 활성을 가지며 용혈활성이나 독성이 낮아서 높은 세균선택성을 가지는 고기능성 항균펩타이드임을 확인할 수 있었다.
또한 본 발명의 약학 조성물은 약학적으로 허용되는 부형제, 담체, 희석제 등을 추가로 포함할 수 있다.
본 발명에서 사용가능한 담체로는 리포좀, 다당, 폴리락트산, 폴리글리콜산, 중합체성아미노산, 및 아미노산 공중합체와 같이 천천히 대사되는 거대분자를 들 수 있다. 예를 들면, 하이드로클로라이드, 하이드로브로마이드, 포스페이트 및 설페이트와 같이 무기산의 염; 아세테이트, 프로피오네이트, 말로네이트 및 벤조에이트와 같은 유기산의 염과 같은 약제학적으로 허용가능한 염; 물, 염수, 글리세롤 및 에탄올과 같은 액체, 및 수화제, 유화제 또는 pH 완충 물질과 같은 보조적 물질을 사용할 수 있다.
약제학적으로 허용가능한 담체에 관해서는 문헌 [Remingtion's Pharmaceutical Sciences, Mack Publishing Company, 1991]에 기재되어 있다.
또한, 상기 조성물은 약학적 분야에서 통상의 방법에 따라 환자의 신체 내 투여에 적합한 단위투여형의 제제, 바람직하게는 펩타이드 의약품의 투여에 유용한 제제 형태로 제형화시켜 당업계에서 통상적으로 사용하는 투여방법을 이용하여 경구, 또는 피부, 정맥내, 근육내, 동맥내, 골수내, 수막강내, 심실내, 폐, 경피, 피하, 복강내, 비강내, 소화관내, 국소, 설하, 질내 또는 직장 경로를 포함하는 비경구투여 경로에 의하여 투여될 수 있으나, 이들에 한정되는 것은 아니다.
즉, 본원 발명의 펩타이드는 실제로 비경구의 여러 가지 제형으로 투여될 수 있는데, 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 비경구투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함된다. 비수성용제, 현탁용제로는 프로필렌글리콜(Propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용 될 수 있다.
또한, 상기 펩타이드는 생리식염수 또는 유기용매와 같이 약제로 허용된 여러 전달체(carrier)와 혼합하여 사용될 수 있고, 안정성이나 흡수성을 증가시키기 위하여 글루코스, 수크로스 또는 덱스트란과 같은 카보하이드레이트, 아스코르브 산(ascorbic acid) 또는 글루타치온과 같은 항산화제(antioxidants), 킬레이팅 물질(chelating agents), 저분자 단백질 또는 다른 안정화제(stabilizers)들이 약제로 사용될 수 있다.
상기 펩타이드의 유효용량은 1 일당 0.01 내지 20 ㎎/㎏체중이고, 바람직하기에는 0.1 내지 10 ㎎/㎏ 이며, 하루 1 내지 수회 투여될 수 있다.
본 발명의 약학적 조성물에서 본 발명의 펩타이드 및 그의 유도체의 총 유효량은 거환(bolus) 형태 혹은 상대적으로 짧은 기간 동안 확산(infusion) 등에 의해 단일 투여량(single dose)으로 환자에게 투여될 수 있으며, 다중 투여량(multiple dose)이 장기간 투여되는 분할 치료 방법(fractionated treatment protocol)에 의해 투여될 수 있다. 상기 펩타이드의 농도는 약의 투여 경로 및 치료 횟수뿐만 아니라 환자의 나이 및 건강상태 등 다양한 요인들을 고려하여 환자의 유효 투여량이 결정되는 것이므로 이러한 점을 고려할 때, 이 분야의 통상적인 지식을 가진 자라면 상기 펩타이드의 약학적 조성물로서의 특정한 용도에 따른 적절한 유효 투여량을 결정할 수 있을 것이다.
상기에서 살펴본 바와 같이, 본 발명은 흰점박이꽃무지 유충으로부터 분리된 펩타이드의 α-helix 영역의 잔기서열을 기반으로 설계된 서열번호 1-4로 기재되는 4종의 항생펩타이드 서열을 제공한다. 이들은 그람 양성균 및 그람 음성균 모두에서 탁월한 항균작용을 나타내고, 특히 9Pbw1, 2, 또는 4 항균펩타이드는 항균활성이 탁월함과 동시에 세포독성을 전혀 가지지 않는 박테리아 선택성이 탁월한 항생 펩타이드이다. 본 발명에 의해 제공되는 항생 펩타이드는 세포독성을 나타내지 않으므로 인체에 안전한 항생제로 유용하게 사용될 수 있다.
이하, 본 발명을 실시예에 의해 상세히 설명한다.
단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.
<실시 예 1>펩타이드들의 합성 및 분리정제
서열번호 1-5로 기재되는 항균 펩타이드들은 Fmoc (9-fluorenyl -methoxycarbonyl)-chemistry를 이용한 고상 펩타이드 합성법(solid phase peptide synthesis) (Merrifield, R. B. Science 232: 341-347)에 의하여 제조하였다. 각 Fmoc-아미노산 및 Fmoc-Nlys(Boc)-OH의 coupling에 의한 펩타이드 chain의 연장은 Coupling시약인 N-hydroxybenzotriazole(HOBt) 및 dicyclohexycarbodiimide (DCC)를 사용하였다. 각 펩타이드의 아미노 말단의 Fmoc-아미노산인 Fmoc-Val-OH을 coupling 시킨 후, 20% piperidine/N-methyl pyrolidone (NMP)용액으로 Fmoc기를 제거하고 NMP 및 dichoromethane (DCM)으로 여러 번 씻어준 다음 질소 가스로 말렸다. 여기에 TFA (trifluoroacetic acid)-phenol-thioanisole-ethanedithiol-H2O (82.5: 5: 5: 2.5: 5, vol./vol.) 용액을 가하고 실온에서 2시간 반응시켜 보호기의 제거 및 수지(resin)으로 부터 펩타이드를 분리시킨 다음, diethylether로 펩타이드를 침전화 시켰다. 이렇게 하여 얻은 crude 펩타이드는 정제형 역상 칼럼 (Vydac C18, 300Å, 15μm, 2.2 cm × 25 cm)이 부착된 0.05% TFA가 포함된 CH3CN (acetonitrile) gradient로 하는 크로마토그래피(HPLC)로 정제하였다. 정제를 완료한 순수한 펩타이드는 MALDI 질량 분석법(Hill, et al. Rapid Commun. Mass Spectrometry, 5, 395, 1991)을 이용하여 얻은 분자량이 아미노산 서열로부터 계산하여 얻은 분자량과 일치하므로 정확한 아미노산 서열을 가지는 펩타이드가 합성되었음을 확인하였다.
<실험예: 1> 항균활성의 측정
서열번호 1-5로 표기되는 펩타이드의 항균활성을 측정하기 위하여 다음과 같이 실시하였다. 그람-양성균(Gram-negative bacteria)로서는 한국생명공학연구원의 Korean Collection for Type Cultures(KCTC)로부터 분양받은 그람 양성균으로써 Bacillus subtilis(KCTC 3068), Staphylococcus aureus(KCTC 1621), E. faecalis (KCTC 2011)를, 그람 음성균으로써 Escherichia coli (KCTC 1682]), Psedomonas aeruginosa (KCTC 1637), K. pneumoniae(KCTC 2242)를 그 대상으로 하였다. 세균수가 ml 당 2×106 colony-forming units (CFUs)가 되도록 1% peptone으로 희석하고 50 ㎕씩 96-웰 마이크로적정 플레이트(microtiter plate)에 분주한 후, 1% peptone으로 희석한 펩타이드 용액 (2:1 단계적으로 희석된 용액)을 각 웰에 50 ㎕씩 첨가한다. 플레이트를 37℃에서 18시간 동안 배양 한 후 ELISA판독기 (Bio-Tek Instruments)로 620nm에서 각 웰의 흡광도를 측정하여 펩타이드의 최소성장억제농도 (MIC: minimal inhibitory concentration)를 결정한다. 대조군의 펩타이드로서 항균 펩타이드 멜리틴과 9Pbm6를 사용하였다. 그 결과 9Pbw1-4 네 개의 펩타이드모두 대조군인 9Pbm6보다 높은 항균활성을 가져 성공적으로 펩타이드 설계가 이루어졌음을 알 수 있었다. 또한 9Pbw4는 모든 균에 대해서 대조군인 9Pbm6 항생펩타이드보다 월등히 높은 항균활성을 나타냈고 S. aureus, B. subtilis에 대해서는 melittin과 같은 높은 활성을 나타냈다.

<실험예: 2> 용혈활성의 측정
서열번호 1-5로 표기되는 펩타이드들에 대하여, 사람의 적혈구 용혈활성을 통하여 세포독성 여부를 판별하기 위하여 다음과 같이 실시하였다. 사람의 적혈구(human erythrocytes)를 PBS(phosphate-buffered saline: 35 mM 포스페이트 완충용액/150 mM NaCl의 혼합용액, pH 7.4)으로 희석한 후 원심력 1000g로 10분간 원심분리 하는 것에 의하여 3회 세척한다. PBS로 희석한 8.0% 적혈구 용액을 96-웰 마이크로적정 플레이트(microtiter plate)에 100㎕씩 로딩한 후, 펩타이드 용액 100㎕씩 섞어 준 후, 37℃에서 1 시간동안 배양한 후, 96-웰 마이크로적정 플레이트를 원심력 1000g로 5분간 원심분리 한다. 상층액 100㎕씩 취하여 다른 96-웰 마이크로적정 플레이트에 옮긴 후, 405 nm에서의 흡광도를 측정하였다. 또한, 대조군 펩타이드로, 멜리틴과 9Pbm6를 사용 하였다. 이때, 0.1% 트리톤 X-100로 처리하였을 경우의 값을 100% 용혈로 계산하고, 펩타이드의 용혈활성(세포파괴 정도)을 % hemolysis로서 수학식 1에 의하여 산출하였다. 여기서 A는 펩타이드 용액에서의 405 nm의 흡광도이고, B는 0.1% 트리톤 X-100에서의 405 nm의 흡광도이고, C는 PBS 용액에서의 405 nm의 흡광도를 나타낸다. 이 때 대조군의 펩타이드로서 강한 용혈활성을 나타내는 항균 펩타이드로서는 멜리틴를 사용하였다. 도 1에 나타낸 바와 같이 서열번호 1,2,4로 표기되는 발명 펩타이드 9Pbw1,2,4는 고농도에서도 전혀 용혈활성을 나타내지 않았다.
<실험예: 3> 박테리아세포막과 적혈구 세포막을 모방하는 인공지질막에 대한 파괴능 측정
서열번호 1-5로 표기되는 펩타이드들의 항균작용이 인간의 적혈구 세포막을 모방하는 중성지질에 대해 독성이 없음을 보이기 위해 다음과 같은 실험을 수행하였다. Fluorescent dye (형광염료)인 칼세인(calcein)을 포획시킨 인간의 적혈구 세포막을 모방하는 중성지질인 EYPC/CH (1:1, w/w) LUVs (large unilamellar vesicle)의 리포좀을 제작하여 다음의 형광염료방출실험(dye leakage assay)을 행하였다. 일정 구성비로 구성된 인지질을 10mM/ml이 되도록 클로로포름(CHCl3)에 녹 인 다음, rotary evaporator로 클로로포름을 제거하고, 하룻밤 동안 동결건조 시킨다. 건조된 지질필름(lipid films)을 트리스-완충용액 (10mM Tris-HCl/ 0.1mM EDTA /150 mM NaCl / pH 7.4) 1ml에 calcein을 70mM이 되도록 녹인다. 리포좀 용액을 액체질소로 얼림-녹임(frozen-thaw)을 10번(ten cycles) 반복하여 행하고 polycarbonate 필터 (pore size가 100 nm인 필터를 두장을 겹친것)로 장착된 LiposoFast extruder (Avestin, Inc. Canada)장치를 이용하여 10회 통과시켜서 칼세인을 포획한 LUVs를 제작한다. 다음은 Sephadex G-50 컬럼을 이용하여 LUVs에 포획되지 않은 free calcein을 제거 하고 calcein이 포획된 리포좀을 모은다. Spectrofluorimeter (Shimadzu RF 5301 PC spectrofluorimeter, Japan)의 excitation wavelength을 490nm에 emission wavelength를 520nm에 맞춘다. 트리스-완충용액 3ml을 quartz cuvette (1cm path length)에 넣고 여기에 적당량의 LUVs 용액을 넣고, 10% Triton-X100 20μl를 넣어 형광세기(fluorescence intensity) 가 1000을 넘지 않도록 LUVs의 양을 조절한다. 위의 조건이 확립되면 LUVs에 펩타이드를 투여한다. 여기서 100 % dye leakage는 0.01 % Triton-X100을 넣었을 때의 형광 세기로 표시한다. 여기서 EYPC 는 egg yolk 1,2-diacyl-sn-glycero-3-phosphocholine의 약어이다. CH는 콜레스테롤이다. 여기서 대조군의 펩타이드로서는 멜리틴과 서열번호 1의 9Pbm6를 사용하여 비교하였다. 도 2에 나타낸 바와 같이 대조 펩타이드인 멜리틴은 펩타이드 농도증가에 따라서 적혈구 세포막을 모방하는 중성지질인 PC/CH (1:1)리포좀에 대해 대조 펩타이드인 멜리틴은 낮은 농도에서도 거의 100 %에 가까운 칼세인 방출을 일으켰다. 그러나 발명 펩타이드 9Pbw1,2,4는 인간의 적혈구를 모방하는 PC/CH (10:1, w/w) 리포좀에서는 거의 칼세인 방출 (calcein leakage)이 일어나지 않아 대부분의 균에 대한 MIC이상인 8μM에서도 거의 5 % 미만의 극히 적은 칼세인 방출을 일으켜 인간의 세포에 독성이 없을것임을 보였다. 반면에 9Pbw3는 8μM에서 50%의 염료방출을 나타내 독성이 있음 알수 있었다. 따라서 9Pbw1,2,4는 높은 항균활성을 가짐과 동시에 박테리아세포 선택성이 높아 우수한 고기능성 항균펩타이드 후보물질이다.
도 1은 특허 출원(10-2008-0109281)된 9Pbm6로부터 항균활성이 증대되도록 설계된 9mer 펩타이드 9Pbw 1,2,3,4를 인간의 적혈구 세포에 처리한 후, 펩타이드 농도에 따른 펩타이드의 용혈활성을 % hemolysis로 나타낸 것이고, 여기서는 대조군 (용혈활성을 나타내는 펩타이드)으로 항균 펩타이드9Pbm6과 melittin을 사용하였다.
도 2는 본 발명의 펩타이드 9Pbw 1,2,3,4에 의한 형광염료(fluorescent dye) 칼세인(calcein)을 포획시킨 인간의 적혈구 세포막을 모방하는 중성지질 EYPC/CH (1:1. w/w)에 대한 % 염료방출(dye leakage)를 펩타이드 농도에 따라 나타낸 것이고, 여기서는 대조군 (세포막 표적의 작용기작을 나타내는 펩타이드)으로 항균 펩타이드 9Pbm6과 melittin을 사용하였다.

Claims (8)
- 서열번호 1 내지 서열번호 4로 기재된 아미노산 서열을 가지는 펩타이드로 이루어진 군으로부터 선택된 하나의 펩타이드.
- 제1항의 펩타이드를 유효 성분으로 포함하는 약학적 조성물.
- 제 2항에 있어서, 상기 조성물은 서열번호 1, 서열번호 2 및 서열번호 4에 기재된 아미노산 서열을 가지는 펩타이드로 구성된 군으로부터 선택된 펩타이드를 유효성분으로 포함하고, 인간의 적혈구 세포 또는 상피세포에 대하여 독성을 가지지 않는 것을 특징으로 하는 약학적 조성물.
- 제1항의 펩타이드를 유효성분으로 포함하는 미생물에 대한 항균용 조성물.
- 제 4항에 있어서, 상기 펩타이드는 서열번호 1, 서열번호 2 및 서열번호 4에 기재된 아미노산 서열을 가지는 펩타이드로 구성된 군으로부터 선택된 펩타이드인 것을 특징으로 하는 미생물에 대한 항균용 조성물.
- 제4항 또는 제5항에 있어서, 상기 미생물은 그람양성균, 그람 음성균 또는 진균인 것을 특징으로 하는 미생물에 대한 항균용 조성물.
- 제6항에 있어서, 상기 그람양성균은 B. subtilis, S. aureus, 또는 E. faecalis, 그람 음성균은 E. coli, P. aeruginosa, 또는 K. pneumoniae, 진균은 Candida Albicans인 것을 특징으로 하는 미생물에 대한 항균용 조성물.
- 제5항에 있어서 상기 항균용 조성물은 인간의 적혈구 세포 또는 상피세포에 대하여 독성을 가지지 않는 것을 특징으로 하는 미생물에 대한 항균용 조성물.
