Description
Claims (27)
- (a) 하기 화학식 I에 의해 정의되는 하나 이상의 스캐폴드를 제공하는 단계<화학식 I>(상기 식에서, X1, X2 및 X3은 서로 독립적으로 sp2- 또는 sp3-혼성화 탄소 원자이고, X4는 친핵체이고, m은 1 또는 2의 정수임);(b) 상기 스캐폴드의 환식 구조를 유지하면서 스캐폴드의 친핵성 X4 기를 유도체화하여, 금속 킬레이트 관능기가 보호 또는 비보호된 형태인, 하나 이상의 여러자리 금속 킬레이트 친화성 리간드 아암을 각 스캐폴드 상에 제공하는 단계;(c) 하나 이상의 금속 킬레이트 친화성 리간드 아암을 스캐폴드에 추가하는 시약을 첨가하여, 유도체화된 스캐폴드의 카르보닐과 황 사이의 결합에서 개환 반응시키는 단계; 및 단계 (b)에서 금속 킬레이트 관능기가 보호된 경우,(d) 단계 (b)에서 제공된 리간드 아암(들)의 관능기를 탈보호하는 단계를 포함하는, 하나 이상의 여러자리 금속 킬레이트 친화성 리간드의 형성 방법.
- 삭제
- 제1항에 있어서, 화학식 I에서, X4가 -NH2인 방법.
- 제1항 또는 제3항에 있어서, 화학식 I에서, m이 1이고 스캐폴드가 호모시스테인 티오락톤인 방법.
- 제1항 또는 제3항에 있어서, 단계 (b)에서, 친전자성이고 따라서 화학식 I의 X4와 반응할 수 있는 한 부분 및 금속 킬레이트 친화성 리간드인 한 부분으로 이루어진 1종 이상의 유도체화제를 첨가함으로써 유도체화하는 방법.
- 제5항에 있어서, 2개의 상이하거나 동일한 금속 킬레이트 관능기를 포함하는 2종의 유도체화제를 첨가함으로써 유도체화하는 방법.
- 제5항에 있어서, 1종의 유도체화제가 할로겐화된 보호된 에스테르인 방법.
- 제7항에 있어서, 1종의 유도체화제가 브로모-아세트산 에틸 에스테르인 방법.
- 제1항 또는 제3항에 있어서, 단계 (c)에서, 개환 반응이 염기를 첨가함으로써 제공되는 가수분해인 방법.
- 제1항 또는 제3항에 있어서, 금속 킬레이트 관능기가 단계 (b)에서 보호되며, 이후에 단계 (c) 및 단계 (d)가 본질적으로 동시에 수행되는 방법.
- 제1항 또는 제3항에 있어서, 단계 (a) 및 (b)를 보다 사전에 수행하여 미리 유도체화된 스캐폴드를 제공하는 방법.
- 제1항 또는 제3항에 있어서, 단계 (d)로부터 얻어진 생성물이, 분리 매질을 제조하기 위해 그의 티올기를 통해 기재 기질에 커플링되는 방법.
- 제12항에 있어서, 티올기가 기재 기질의 알릴기에 커플링되는 방법.
- 제12항에 있어서, 기재 기질을 알릴화하여 반응성 기를 제공하는 단계를 더 포함하는 방법.
- 제12항에 있어서, 리간드의 티올기가 알릴 글리시딜 에테르 (AGE)의 알릴기를 통해 기재 기질에 커플링되는 방법.
- 제12항에 있어서, 기재 기질의 반응성 기를 활성화하는 단계를 더 포함하는 방법.
- 제16항에 있어서, 활성화가 브롬화에 의해 수행되는 방법.
