Claims (9)
- 서열번호: 1로 표시되는 아미노산 서열을 가지며, 하기 이화학적 성질을 가지는 폴리포스페이트:AMP 포스포트랜스퍼라제.(A) 작용: 하기 2개의 반응을 촉매한다.NMP + PolyP(n)→ NDP + PolyP(n-1)dNMP + PolyP(n) → dNDP + PolyP(n-1)(식중, NMP는 뉴클레오시드 모노포스페이트, NDP는 뉴클레오시드 디포스페이트, dNMP는 데옥시뉴클레오시드 모노포스페이트, dNDP는 데옥시뉴클레오시드 디포스페이트, n은 폴리포스페이트의 중합도를 나타내는 것으로, 100이하의 정수이다.)(B) 기질 특이성: AMP, GMP, IMP, dAMP, dGMP에 특이적이며, CMP, UMP, dCMP, TMP에도 작용한다.(C) 분자량: 약 55∼56Kd (킬로달톤)이다.(D) 비활성: 효소단백 1 mg당 70단위(유니트) 이상이다.다만, l 단위(유니트)란 37℃에서 1분간에 1μmo1e의 ADP를 생성하는 활성을 나타내며, 이하의 조건으로 측정한다.<측정조건>20mM 염화마그네슘, 10mM AMP 및 폴리포스페이트(무기 인산으로서 30mM)을 함유하는 50mM 트리스 염산 완충액(pH 7.8)에 효소표품을 첨가하고, 37℃에서 보온함으로써 반응을 행하고, 100℃, 1분간 열처리하여 반응을 정지시키고, 고속 액체 크로마토그래피(HPLC)를 이용하여 반응액중의 ADP를 정량한다.

- 뉴클레오시드 모노포스페이트로부터 뉴클레오시드 디포스페이트를 효소적으로 제조하는 방법에서, 효소로서 제 1 항 기재의 폴리포스페이트:AMP 포스포트랜스퍼라제를 사용하고, 포스페이트 공여체로서 폴리포스페이트를 사용하는 뉴클레오시드 디포스페이트의 제조법.
- AMP로부터 ATP를 효소적으로 제조하는 방법에서, 효소로서 제 l 항 기재의 폴리포스페이트:AMP 포스포트랜스퍼라제와 아데닐레이트 키나아제의 2종의 효소를 사용하고, 포스페이트 공여체로서 폴리포스페이트를 사용하는 ATP의 제조법
- AMP, 폴리포스페이트, 폴리포스페이트:AMP 포스포트랜스퍼라제 및 아데닐레이트 키나아제로 이루어진 ATP의 생성/재생계에 있어서, 폴리포스페이트:AMP 포스포트랜스퍼라제로서 제 1 항 기재의 효소를 사용하는 ATP의 생성/재생계.
- ATP를 소비하는 효소 반응을 이용한 화합물의 제조법에 있어서, 생성한 AMP를 폴리포스페이트, 폴리포스페이트:AMP 포스포트랜스퍼라제 및 아데닐레이트 키나아제로 이루어진 ATP의 재생계를 이용하여 재생할 때, 폴리포스페이트:AMP 포스포트랜스퍼라제로서 제 1 항 기재의 효소를 사용하는, AMP로부터 ATP를 재생하면서 당해 효소반응을 행함을 특징으로 하는 당해 화합물의 제조법.
