Claims (13)
- 키토산 중의 트로포미오신을, 키토산을 유기산 수용액에 용해한 상태로, 면역측정법(immunoassay)으로 측정하는 것을 특징으로 하는 트로포미오신 측정방법.
- 제 1 항에 있어서, 상기 면역측정법이, ELISA 방법인 측정방법.
- 제 2 항에 있어서, 상기 ELISA 방법이, 1차 항체로서 적어도 1종의 항-갑각류 트로포미오신 항체와, 적어도 1종의 표식 항체를 사용하는 샌드위치 방법인 측정방법.
- 제 3 항에 있어서, 상기 표식 항체가, 효소 표식 항-갑각류 트로포미오신 폴리크로날 항체 또는 효소 표식 항-면역글로불린 항체인 측정방법.
- 제 4 항에 있어서, 상기 갑각류가, 작은 새우류이며, 효소가, 페록시다아제, 알칼리 포스파타아제 및 β-갈락토시다아제로 이루어지는 군으로부터 선택되는 적어도 1종인 측정방법.
- 제 3 항에 있어서, 상기 항-갑각류 트로포미오신 항체가, 측정용 용기의 내벽에 코팅되어 있는 측정방법.
- 제 4 항에 있어서, 효소가, 페록시다아제이며, 그 기질로서 o-페닐렌디아민, 2,2'-아지노비스(3-에틸-벤즈티아졸린술폰산) 디암모늄염 및 테트라메틸벤지딘으로부터 선택되는 적어도 1종을 사용하는 측정방법.
- 제 7 항에 있어서, 트로포미오신의 측정을, 측정계의 청색을 측정하여 실시하는 측정방법.
- 제 8 항에 있어서, 트로포미오신의 측정을, 청색을 나타내는 측정계에 황산 또는 인산을 첨가하여, 측정계를 황색으로 바꾸어, 상기 황색을 측정하여 실시하는 측정방법.
- 제 1 항에 있어서, 상기 유기산이, 초산(acetic acid), 유산(lactic acid) 및 피롤리돈 카르본산(pyrrolidonecarboxylic acid)으로 이루어지는 군으로부터 선택되는 적어도 1종인 측정방법.
- 제 1 항에 있어서, 상기 유기산 수용액의 유기산 농도가, 0.1중량% 이상 2중량% 미만인 측정방법.
- 제 1 항에 있어서, 상기 키토산은, 그 불용해 성분이, 이하의 측정방법:(1) 키토산을 105℃에서 3시간 건조하는 단계,(2) 건조 전후의 중량비로부터 키토산 순도를 계산하는 단계,(3) 건조 전의 키토산을 1중량%의 초산 수용액에 키토산 순분(A그램) 농도 0.5중량%로 용해하는 단계,(4) 상기 수용액을, 일정 중량으로 건조된 G3 유리 필터(B그램)로 여과하는 단계,(5) 상기 필터상의 여과찌꺼기를 증류수로 세정하는 단계,(6) 여과찌꺼기를 포함한 필터를 105℃에서 3시간 건조하여, 중량(C그램)을 측정하는 단계, 및(7) 식[(C-B)/A×100]에 의해 키토산 중의 불용해 성분(중량%)을 계산하는 단계,에 의해 1.0중량% 이하의 키토산인 측정방법.
