Abstract

본 발명은 새로운 종류의 펩티딜 유도체, 그의 제조 방법 및 의약품에서의 이들의 용도에 관한 것이다.
금속단백질가수분해효소(matalloproteinases), (인간 섬유아 세포) 콜라게나아제(collagenase), 젤라티나아제(gelatinase) 및 종양 사멸 요소(tumour necrosis factor)(TNF) 및 이들의 작용 방식, 및 또한 이들의 억제제 및 이들의 임상적인 효과가 WO-A-9611209에 기재되어 있고, 그 내용은 참조를 위해 본 명세서에 포함시켰다.
정상 조직에서, 세포 결합 조직 합성(cellular connective tissue synthesis)은 세포외부의 매트릭스 감성(matrix degradation)에 의해 상쇄되고, 두 개의 반대 효과가 동적 평형에서 존재한다. 매트릭스 감성은 잔류(resident) 결합조직 세포 및 침범한 염증성 세포로부터 방출된 단백질가수분해효소의 작용에 의한 것이고, 부분적으로 적어도 4개의 금속단백질가수분해효소의 기(group)의 활성에 기인한다. 이들은 콜라게나아제(간질(間質)의 콜라게나아제, MMP-1; PMN 콜라게나아제, MMP-8, 콜라게나아제-3, MMP-13), 젤라티나아제(젤라티나아제 A, MMP-2, 72kDa-젤라티나아제, 유형 Ⅳ의 콜라게나아제; 젤라티나아제 B, MMP-9, 92kDa-젤라티나아제, 유형Ⅳ의 콜라게나아제) 스트로멜리신(stromelysin)(단백질글리카나아제(proteoglycanase), MMP-3, 스트로멜리신-1, 트란신(transin); 스트로멜리신-2, MMP-10; 스트로멜리신-3, MMP-11) 및 막 유형 매트릭스 금속단백질가수분해효소(MT-1, MMP-14; MT-2, MMP-15; MT-3, MMP-16 및 MT-4, MMP-17) 이다. 보통, 이화(異化)(catabolic) 효소는 이들의 합성 또는 분비의 농도 및 또한 이들 세포외부에서의 활성의 농도에서 철저하게 조정되고, 후자는 금속단백질분해효소와 불활성 착물을 만드는 TIMP(금속단백질가수분해제의 조직 억제제) 등의 특정한 억제제 및 α2-매크로글로불린(α2-macroglobulins)등의 보다 일반적인 단백질 가수분해 효소의 작용을 통해 조정된다.
금속단백질가수분해효소가 세포외부의 매트릭스의 흡수에 촉매작용을 미치는 것에 의하여 가속되고, 방치된 결합조직의 붕괴는 류머티양 관절염, 골관절염, 폐혈성 관절염, 각막 궤양, 표피 궤양 또는 위궤양; 종양 전이 또는 침입; 치주 질병, 단백뇨, 아테로마성 동맥경화성반 파열(atherosclerotic plaque rupture)과 관련된 관상동맥 혈전 및 뼈질병 등의 많은 병리학적 증상(condition)의 특징이다. 본 명세서에서 청구된 억제제는 영구적인 신체장애로 이끌 수 있는 외상성의 상처에 따른 병리학적 스쿠앨래(squaelae)를 방지하는데 유용하다. 상기 화합물들은 또한 배란이나 착상을 방지하여 산아 제한하는 수단으로써 유용성을 가질 수 있다. 이러한 질병의 발생은 금속단백질가수분해효소의 억제제를 투여하는 것에 의해 유익한 방식으로 수정되어질 수 있을 것으로 예상되고, 다양한 화합물이 상기 목적을 위해 제안되어왔다[for a general review see R C Wahl, et al Ann.Rep, Med. Chem.25:175-184, Academic Press Inc., San Diego(1990)].
금속단백질가수분해효소를 억제하는 다수의 작은 펩티드 화합물들이 기재되어있다. 아마도 이들 중 가장 주목할 만한 종류의 것은 안기오텐신 전환 효소(angiotensin converting enzyme)(ACE)와 관련된 것이고, 이러한 약품은 데카펩티드 안기오텐신Ⅰ이 효능 있는 승압 물질(pressor substance)인 안기오텐신Ⅱ로 전환되는 것을 막는다. 이러한 유형의 화합물은 EP-A-0012401에 기재되어있다. 또한, 메르캅토아미드 펩티딜 유도체(mercaptoamide peptidyl derivatives)는 시험관에서 및 생체 내에서 ACE 억제제 활성을 보여왔다(H N Weller et al(1984), Biochem Biophys.Res.Comm., 125(1):82-89).
TNF는 시토킨(cytokine)이고 이것은 초기에 세포와 관련된 28kD의 전구체로서 생성되었다. 이것은 활성이 있는 17kD의 형태로 방출되고(D-M Jue et al,(1990) Biochemistry, 39:8371-8377), 이것은 생체 내 다수의 해로운 효과를 중재할 수 있다. TNF를 동물 또는 인간에게 투여했을 때, 이것은 급성 감염이나 쇼크 상태 동안 볼 수 있는 증상과 유사한 염증, 열, 심장혈관 효과(cardiovascular effects), 출혈, 응고 및 급성 상 반응(acute phase response)을 일으킨다. 만성적인 투여는 또한 악액질(惡液質)(cachexia) 또는 식욕 감퇴를 일으킬 수 있다. 과량의 TNF의 축적은 치명적일 수 있다.
동물 모델 연구에서 특정한 항체를 이용하여 TNF의 효과를 차단하는 것은 급성 감염, 마비 상태, 이식세포 대 숙주 반응(graft versus host reaction) 및 자가면역 질병에서 유익할 수 있다는 상당한 증거가 있다. TNF는 또한 일부 골수종 또는 림프종에 대한 오토크린(autocrine) 성장 인자이고 이러한 종양을 가진 환자에 있어서 정상적 혈액 생성을 억제할 수 있다.
따라서, TNF의 생성 또는 작용을 방지하는 것은 많은 염증성, 전염성, 면역성 또는 악성 질병에 대한 효능 있는 치료법이 될 수 있다고 예상된다. 이들은, 폐혈성 마비, 혈액동역학적 마비 및 폐혈증 증후군(Mathison et al(1988) J.Clin. Invest.81: 1925-1937; Miethke et al(1992), J.Exp.Med.175:91-98), 허혈 재관류(再灌流) 후 손상(post ischaemic reperfusion injury), 말라리아(Grau et al (1989)), Immunol.Rev. 112:49-70); 미코박테리아성 감염(mycobacterial infection)(Barnes et al(1992) Infect.Imm.60:1441-6), 수막염, 건선, 울혈성 심부전증, 섬유조직 질병, 악액질, 이식거부 반응, 암, 자가면역 질병, 류머티양 관절염, 다중 경화증, 방선 손상(radiation damage), OKT3 또는 CAMPATH-1 등의 면역억제 단클론항체의 투여에 따르는 독성 및 하이퍼산화 치조 손상을 포함하지만, 이것으로 제한되지 않는다.
현재의 임상적인 반-TNF 치료법은 덱사메타손(dexamethasone) 등의 코르티코스테로이드(corticosteroids)의 이용, 및 시토킨 유전인자 전사의 비특정 억제제인 씨클로스포린-A(cycloporine-A) 또는 FK506의 사용과 관련된다. 펜톡시필린(pentoxyfilline) 등의 포스포디에스터라아제(phosphodiesterase) 억제제는 TNF 유전인자 전사의 더욱 특정한 억제제임이 알려져왔다.(Endres S.(1991) Immunol. 72:56- 60, Schandene et al(1992), Immunol. 76:30-34, Alegre ML, et al(1991); Transplantation 52:674-679, Bianco et al(1991) Blood 78:1205-1221). 탈리도미드(thalidomide)는 또한 백혈구에 의한 TNF의 생성을 막는 다는 것이 알려졌다(Sampajo et al(1991), J.Exp.Med.173:699-703). 실험 설정에서, 반-TNF 단클론항체, 가용성의 TNF 수용체 및 가용성 TNF 수용체/면역접착체(immunoadhesins)는 TNF 작용의 효과를 특히 억제함이 알려졌다(Bagby et al(1991) J.Infect.Dis.163: 83- 88, Charpentier et al(1991) Pressemed. 20:2009-2011, Silva et al(1990) J. 최근에 TNF의 효과가 두 개의 펩티드, TNFα 및 TNFβ에 의해 중재됨이 알려졌다. 비록 이들 펩티드가 단지 서로 30% 정도 상동하지만, 이들은 동일한 수용체를 활성화시키고 즉각적으로 인접한 유전인자에 의해 코드 된다. 따라서, 본 명세서에서 사용된 바와 같이, 종양 사멸 인자 또는 TNF 라는 용어는 종양 사멸 인자 α 및 TNFα와 높은 정도의 순차적 서열의 상동을 갖는 또는 TNFα에 대해 실질적으로 유사한 생리학적 효과를 갖는 펩티드, 예를 들면 TNFβ를 의미한다.

콜라게나아제, 스트로멜리신 및 젤라티나아제 등의 결합조직 붕괴에 관여하는 금속단백질가수분해효소의 작용을 억제하는 성질을 가진 화합물을 실험실 및 생체 내 모두에서 TNF의 방출을 억제하는 것이 알려져왔다(AJH Gearing et al(1994), Nature, 370:555-557;GM McGeehan et al(1994), Nature, 370:558-561; WO 93/ 20047). 이들 보고된 억제제 모두는 히드록삼산 아연 결합기를 포함한다.
본 기술의 숙련가들에 의해 이해되는 바와 같이, 인간 섬유 아세포 콜라게나아제, 스트로멜리신 및 젤라티나아제 사이의 상당한 부분의 상동은 하나의 효소를 억제하는 화합물이 다른 모두를 어느 정도 억제할 수 있다는 가능성을 이끌어 낸다.
콜라게나아제를 억제하는 화합물은 특허 받은 미국 특허 제 4,511,504호(1985. 4.16) 및 특허 받은 미국 특허 제 4,568,666호(1986. 2. 4)에 기재된 것을 포함한다. 스트로멜리신(프로테오글리카나아제(proteoglycanase))을 억제하는 것으로 청구된 것과 관련된 구조의 화합물은 특허 받은 미국 특허 제 4,771,037호(1988. 9.13)에 기재되어있다.
스트로멜리신 및 콜라게나아제 억제제는 폐결핵성 관절염과 관련된 관절 연골부 손상을 방지하는데 유용성을 가진다고 믿어진다. 관절의 박테리아성 감염은 감염 병원체 제거에 대한 요구를 지속하게 하는 염증성 반응을 유발할 수 있고, 이것은 구조 성분의 영구적인 손상을 초래한다. 박테리아성 병원체(bacterial agent)는 단백질 분해 활성을 관절 반응을 나타내는 동물 모델에 사용하여 왔다(See J.P. 스트로멜리신, 콜라게나아제 및 젤라티나아제 등의 분비된 단백질가수분해효소는 전이 종양의 침입 동안에 세포 이동과 관련된 과정에서 중요한 역할을 한다. 실제로, 매트릭스 금속단백질가수분해효소가 일정한 전이 종양 세포주에서 과다하게 스며 나온다는 것이 또한 증거이다. 이와 관련하여, 효소는 밑에 있는 기저막(basement membranes)을 통과하고 종양 세포를 일차 종양 형성 장소로부터 나오게 하며 순환하도록 한다. 종양 세포는 혈관 벽에 부착 된 후, 이것은 동일한 금속단백질가수분해효소를 이용하여 밑에 있는 기저막을 통과하고, 다른 조직을 통과하고, 그것에 의해 종양 전이된다. 이러한 과정의 억제는 전이를 막을 수 있고 현재의 화학요법 또는 및/또는 방사선 요법의 이용하여 현재의 치료의 효율을 향상시킨다.
이들 억제제는 치은점 등의 치주 질병을 제어하는데 유용하다. 콜라게나아제 및 스트로멜리신 활성 모두는 치은을 일으키는 것으로부터 나온 섬유 아세포로 부터 단리 되었다(Uitto et al(1981), J.Periodontal Res., 16:417-424). 효소 농도는 치은 질병의 심한 정도와 상관관계가 있다; C.M. Overall et al(1987), J.Periodontal Res., 22:81-88).
단백질분해과정은 또한 알카리 화상(alkali burns)에 따른 각막 궤양에서 관찰된다(S.I.Brown et al(1989), Arch.Opthalmol., 81:370-373). 메르캅토 함유 펩티드는 알카리 화상을 입은 토끼의 각막으로부터 단리된 콜라게나아제를 억제한다(F.R.Burns et al(1989), Invest.Opthalmol, 30:1569-1575). 알카리 화상을 입은 눈 및 시트르산 나트륨 또는 아스코르브산 나트륨 및/또는 항미생물과 배합된 금속내 단백질가수분해효소의 억제에 의해 감염된 결과로서 각막 궤양을 일으키는 눈의 치료는 각막 감성(degradation)의 진행을 막는데 효과적일 수 있다.
스트로멜리신은 신장의 신사구체(glomerular) 기저막의 구조 성분의 감성과 관련되고, 플라스마(plasma) 단백질의 소변으로 이동하는 것을 제한하는 것이 주된 역할이다(W.H.Baricos et al(1989), Biochem.J., 254:609-612). 신사구체 질병인 단백뇨는 GBM 대 플라스마 단백질의 증가된 투과율에 의해 야기되는 질병으로 소변 중 과량의 단백질이 있는 것이다. 증가된 GBM 투과율의 근본적 원인은 알려져있지 않으나, 스트로멜리신을 포함하는 단백질가수분해효소가 신사구체 질병에 중요한 역할을 하는 것 같다. 이러한 효소의 억제는 신장 기능부진과 관련된 단백뇨를 완화시킬 수 있을 것이다.

돼지 중의 울혈성 심기능부전증(Congestive Heart Failure)(CHF)의 모델이 최근에 제시되었는데, CHF 동안에 심장의 형태학적인 구조의 변화가 눈에 띄었다. 세포외부의 매트릭스의 변화에 의해 심실의 확장 및 박막화(wall thinning)는 심장근세포(cardiomycytes)와 소량인 전체 콜라겐 사이의 콜라겐 결합을 거의 없게 만든다. 이러한 예에서 보다 약해진 수축력으로 인해 심실 작용이 비능률적으로 된다. 매트릭스 금속단백질분해효소의 특정한 억제제는 세포외부 매트릭스을 안정화시키는데 중요한 역할을 하고 따라서 CHF의 치료 및/또는 방지에 중요하다고 믿어진다.
콜라게나아제 및 스트로멜리신 등의 매트릭스 금속단백질분해효소는 또한 인체 가용성 CD23의 형성을 억제시킨다는 것이 최근에 제시되었다(WO 96/0240). CD23은 B 및 T 림포세포, 대식세포, NK 세포, 란저한스 세포(Langerhans cells), 단세포(monocytes), 호산구 및 혈소판을 포함하는 여러 가지 성숙 세포의 표면에서 발현되는(expressed) 45kDa의 유형Ⅱ의 필수 단백질(integral protein)이다(Delespesse et al(1991), Adv.Immunology, 49:149; Grangette et al(1989), J., Immunol, 143:3580). IgE 조절과 모두 관련되어 있는 여러 활성이 사람 안에 있는 가용성 CD23에 있다고 여겨져 왔다. 특별한 활성이 다음을 포함한다:
ⅰ) 항원 태위(antigen presentation)
ⅱ) IgE 매개된 호산구 세포독성
ⅲ) 림프절(lymph nodes) 또는 비장(spleen)으로 향하는 B 세포

따라서, 전체 과량의 가용성 CD23의 생성은 외인성 천식, 비염, 알레르기성 결막염, 습진, 아토피성 피부염 및 아나필락시(anaphylaxis) 등의 알레르기성 질병의 특징인 IgE의 과량생성을 의미한다(Sutton et al(1993), Nature, 336:421). 가용성 CD23의 증가된 농도는 또한 만성 B 림포세포 백혈병을 가진 환자의 혈청에서 관찰되고(Safarti et al(1998), Blood, 71:94), 류머티양 관절염을 가진 환자의 활액 흐름에서 관찰된다(Chomarat et al (1993), Arthritis and Rheumatism, 36:234).
최근의 보고는 MMP계의 새로운 효소가 또한 예를 들어 셀렉틴(selectin) 등의 L-셀렉틴의 유착 분자(adhesive molecule)의 발산(shedding)을 중재한다. 이러한 가용성 유착 분자는 암, 자가면역을 포함하는 다수의 질병 및 염증성 반응과 관련된다. 일단 절단된 후 셀렉틴은 특별한 리간드(ligand)와 결합하고 이것은 그들의 생물학적 활성을 설명한다고 제안되어왔다. 따라서, 리간드가 셀렉틴과 결합하는 것을 간섭하거나 방지하는 약은 상기의 여러 가지 질병을 치료하기 위해 유용한 약물이다. 그러므로, 본 발명의 추가적 목적은 어떤 유착 분자의 발산을 억제하는 화학물을 제안하고 따라서 암, 자가면역 질병 또는 염증성 질병 예를 들어 염증성 장 질병 및 다중 경화의 치료 또는 예방을 위한 약물의 제조를 제안한다.
스트로멜리신 및 콜라게나아제의 특정한 억제제는 산아 제한제로서 유용하다고 믿어진다. 스트로멜리신 및 콜라게나아제를 포함하는 금속단백질가수분해효소의 발현은 미수정 난자나 접합자(zygotes)의 추가적인 분할 단계에서 관찰되고 태아 발육의 낭포 단계에서 및 내배엽 분화와 함께 증가되는 것이 입증되었다(C.A.Brenner et al(1989), Genes & Develop., 3:843-59). 종양 침입과 유사하게, 낭포는 이식동안 자궁벽의 세포외부 매트릭스를 통과하기 위하여 금속단백질가수분해효소를 압출한다. 이들의 초기 발육 과정 동안 스트로멜리신 및 콜라게나아제의 억제는 아마 자궁내의 정상적인 배의 발육(embryonic development) 및/또는 착상을 막을 것이다. 이러한 조정은 산아 제한의 새로운 방식이다. 덧붙여서 콜라게나아제가 배란 과정에 중요하다는 것은 분명하다. 이러한 예에서, 소포의 정점 영역을 통해 콜라겐으로 피복한 것은 난자가 빠져나가기 위해 통과되어야만 한다. 콜라게나아제는 이러한 과정에서 검출되었고 억제제는 배란을 방지하는데 효과적인 것으로 나타났다(J.F.Woessner et al(1989), Steroids, 54:491-499). 배란 동안 스트로멜리신에 대한 역할이 있다(C.K.L. 콜라겐분해성 및 스트로멜리신 활성은 또한 부영양 표피수포증에서 관찰되었다(A.Kronberger et al(1982), J.Invest.Dermatol., 79:208-211; D.Sawamura et al(1991), Biochem.Biophys.Res. Commun., 184:1003-8). 금속단백질분해효소의 억제는 피부 결합 성분의 빠른 파괴를 제한한다.
구조 성분을 이루는 세포외부 매트릭스에 덧붙여, 스트로멜리신은 생체 내 실험에서 억제제 α1-단백질분해효소 억제제를 포함하는 다른 기질을 감성할 수 있고 따라서 엘라스티아제(elastase)(P.G.Winyard et al(1991), FEBS Letts, 279,1: 91-94) 등의 다른 단백질분해 효소의 활성에 영향을 미칠 수 있다. 매트릭스 금속단백질가수분해효소의 억제는 내생성 억제제의 반단백질가수분해효소(antiprot- einase)의 활성을 높일 수 있다.
최근의 문헌으로 MMP계의 여러 가지 새로운 효소가 동정(同定)되었고(identified), 이 중 일부가 질병에서 중요할 수도 있다는 것이 증명되었다. 흉부암세포에 유일한 효소인 콜라게나아제 3은 흉부암에서 유용성을 가질 수 있고(JMP Freije et al(1994), J.Biol.Chem., 269(24):16766-16773), 반면에 MT-MMPs, MMP계의 다른 성분들은 젤라티나아제 A의 활성화에 중요한 효소인 것으로 나타났다(H Sato et al(1994), Nature, 370:61-65). 젤라티나아제 A는 (상기에 명시된) 종양의 성장과 전이에 중요한 효소이다.
b-아밀로이드 전구 단백질(b-Amyloid Precursor Protein)(APP)의 감성은 알츠하이머 병(Alzheimers Disease)(AD)에서 발견되는 노인성반(senile plaques)의 주요한 구성 성분인 아밀로이드반(amyloid plaques)을 발생시킨다. 두 개의 최근 문헌에서는 APP를 아밀로이드반으로 분해하는 금속단백질분해효소를 확인했다(CR Abraham et al(1994), Biochemistry, 33:192-199; G Huber et al(1994), Biochem. Biophys.Res.Comm, 201(1):45-53).
본 발명의 숙련가들에 의해 이해되는 바와 같이, 이러한 새로운 효소 및 다른 MMP사이의 상동관계의 중요한 부분은 하나의 효소를 억제하는 화합물이 어느 정도 이들 새로운 효소를 억제할 수 있는 가능성으로 이끈다. 그러므로, 본 발명에 따른 억제제는 이러한 새로운 효소와 관련하여 상기 질병에 유용하게 쓰일 수 있다.
따라서, 본 발명의 추가적인 목적은 TNF 방출을 억제하는 것 외에 또한 MMPs의 작용을 억제하는 화합물을 제공하는 것이고, 그러므로 TNF 및/또는 MMPs로 중재된 증상을 겪는 환자의 치료에 쓰일 수 있다.
따라서, 본 발명의 추가적인 목표는 상기된 바와 같이 가용성 CD23의 과생성과 관련된 알레르기성 및 자가면역 질병 등의 병의 치료 또는 예방을 위한 약물의 제조를 위해 인체 가용성 CD23의 형성을 억제하는 화합물, 그의 염, 용매 화합물 및 수화물을 제공하는 것이다.
본 발명은 식(I)의 새로운 메르캅토알킬펩티딜 화합물(mercaptoalkyl- peptidyl compound)에 관한 것이고 상기 화합물은 퇴행성 질병(degenerative diseases)(상기되어 있는 것과 WO-A-9611209에 기재되어있다)을 포함한 금속단백질분해효소 및/또는 TNFα 매개 질병에 대한 유용한 억제제이다.
본 발명의 첫 번째 측면은 일반식(I)의 화합물, 및 이의 염, 용매화물, 및 수화물을 제공하는 것이다:

본 명세서의 식에서, ~라인은 R- 및 S - 배열의 가능성을 나타내는 잠재적 비대칭 중심에서 사용되고, <라인 및 ......라인은 비대칭 중심에서 유일한 배열을 나타내기 위해 사용된다.

일반식(Ⅰ)의 화합물은 당업계에 공지된 어떤 적절한 방법 및/또는 본 발명의 일부를 형성하는 하기 방법에 의해 제조될 수 있다.
본 발명의 두 번째 측면에 따르면, 상기에 정의된 바와 같이 일반식(Ⅰ)의 화합물을 제조하기 위한 방법을 제공한다. 식(Ⅰ)의 특별한 입체이성질체가 필요한 경우, 본 명세서에 기재된 합성 방법은 적합한 호모키랄(homochiral) 출발 물질로 사용되고/사용되거나 이성질체는 종래의 분리 기술(예를 들어 HPLC)을 이용하여 혼합물로부터 분해될 수 있다.
본 발명에 따른 화합물은 하기 방법에 의해 제조될 수 있다. 아래에 기재된 식에서, R1, R2, R3, R4, R5, R6, R7, R8, R9, R10, R11, R12, R13, R14, R15, R16, R17, A, B, C, R, S, W, X, Y 및 Z는 따로 표시한 것을 제외하고는 상기에 정의한 바와 같다. 아래에 기재된 여러 가지 화합물에 존재하는 적절한 위치에서 유지되는 것이 바람직한 아미노, 수산화 또는 카르복시기 등의 작용기는 어떤 반응을 시작하기 전에 보호되는 것이 필요하다. 이러한 예에서, 보호기의 제거는 특별한 반응에서 최종 단계이다. 이러한 역할을 위한 적절한 보호기는 본 기술의 숙련가들에게 명백하다. 보다 상세한 설명을 위해서는 "Protective Group in Organic Synthesis", Wiley Interscience, T W Greene, PGM Wutts 참조.
일반식(Ⅰ)의 화합물을 제조하기 위한 방법은 식 R7S-CHR8-CO-NR16-CHR1-CO-NR2-CHR3-X(Ⅱ)(R7은 적절한 보호기(예를 들어 3차-부틸, 트리틸, 벤조일 또는 아세트산염)를 나타낸다)의 화합물의 보호기 제거(예를 들어 가수분해)로 이루어진다.
식(Ⅰ)의 특별한 입체이성질체가 필요한 경우, HPLC(High Performance Liquid Chromatography) 등의 종래의 분리 기술에 의해 얻어질 수 있음이 이해될 수 있다. 그러나, 바람직한 경우는 적합한 호모키랄 출발 물질을 결합 반응(coupling reaction)에 사용하여 식(Ⅰ)의 특별한 입체이성질체를 생산하는 것이다.
일반식(Ⅱ)의 중간 생성물은 식 R7S-CHR8-COOH(Ⅲ)(R7 및 R8은 상기에서 정의된 바와 같거나, 또는 이것의 활성 있는 유도체이다)의 산을 식 R16NH-CHR1-CO-NR2- CHR3-X(Ⅳ)의 아민과 결합시켜 제조한다. 일반식 (III)의 산 활성 유도체는 산 염화물과 같은 산 무수화물 또는 산 할로겐화물 등을 포함한다.

일반식(Ⅱ)의 화합물은 식 R8R18C-CO-NR16-CHR1-CO-NR2-CHR3-X(Ⅶ)(R18은 적절한 이탈기(leaving group)(예를 들면 브롬 등의 할로겐, 또는 메탄술폰산염 등의 알킬술폰산 에스테르 등)를 나타낸다)와 식 R7SH(Ⅷ)의 티올을 WO90/05719에 예시된 본 기술의 숙련가에게 알려진 표준 조건을 이용한 친핵 치환(nucleophilic substitution)에 의해 제조된다.
식(Ⅷ)의 티올은 본 기술의 숙련가들에게 알려진 상업적으로 이용 가능한 출발 물질로부터 얻을 수 있다. 일반식 (Ⅶ)의 많은 티올이 또한 상업적으로 유용하다.
식(Ⅶ)의 화합물은 식 R18R3CH-COOH(Ⅸ)(R18 및 R8은 상기에 정의되었거나 이들을 적절하게 보호한 것이다), 식(Ⅳ)의 아민 또는 이들의 활성 유도체를 식(Ⅱ)의 화합물의 제조를 위해 기재된 유사한 결합 조건을 사용한 결합에 의해 제조될 수 있다.
식(Ⅲ) 및 (Ⅸ)에 표시된 구조의 카르복시산은 키랄 또는 라세미 형태로 얻어질 수 있다. 많은 이들 유도체들이 본 기술의 숙련가들에게 알려진 방식을 이용하여 상업적으로 이용 가능한 출발 물질로부터 쉽게 얻어질 수 있다(See WO90/05719).
식(Ⅱ)의 중간 생성물은 R7S-CHR8-CO-NR16-CHR1-COOH(Ⅹ)(R1, R7 및 R8은 상기에 정의된 바와 같거나, 또는 이들의 활성 유도체)와 식(Ⅵ)의 아민을 상기의 절차를 이용하여 결합하는 것에 의해 제조하였다.
일반식(Ⅹ)의 산은 식(Ⅲ)의 산 또는 이들의 활성 유도체를 식(Ⅴ)의 아민, 또는 나중에 보호기가 제거될 수 있는 적절하게 보호된 유도체와의 결합에 의해 차례로 제조될 수 있다.
식(Ⅹ)의 산의 활성 유도체는 예를 들면 상기에 약술한 산 염화물 또는 산 무수화물 또는 산 할로겐화물의 산 무수물을 포함한다.
식(Ⅰ)의 화합물은 식(Ⅰ)의 다른 화합물로의 상호전환(interconversion)에 의해 제조될 수 있다. 따라서, 예를 들면 R1이 C1-6알킬기인 식(Ⅰ)의 화합물은 R1이 C2-6알케닐기인 식(Ⅰ)의 화합물의 수소화(알콜-예를 들면 에탄올의 적절한 용매 중에의 탄소상의 팔라듐을 이용한다)에 의해 제조된다. 추가적인 예는 R7이 R10CO인 식(Ⅰ)의 화합물을 식(Ⅰ)의 R7이 H인 화합물과 아실화(acylation)(염소화된 용매-예를 들면 디클로로메탄 등의 적절한 용매 중에 트리에틸아민 등의 염기의 존재시, 적절한 염화 산 R10COCl을 이용하였다) 시켜 제조될 수 있다.
Ⅹ가 헤테로아릴인 화합물은 PCT/GB96/01137에 기재된 절차에 따라 제조될 수 있다.
얻어진 최종 생성물 또는 중간 생성물의 어떤 혼합물은 성분의 물리-화학적 차이에 기초하여 공지된 방법, 예를 들면 색층분리법, 증류, 분별 결정에 의해 또는 만약 적절하거나 가능한 환경 하에 있다면 염의 생성에 의해 순수한 최종 생성물 또는 중간 생성물로 분리될 수 있다.
본 발명에 따른 화합물은 실험실 조건에서 스트로멜리신, 콜라게나아제 및 젤라티나아제에 대한 활성을 억제한다. 본 발명에 따른 화합물은 실험실 조건에서 TNFα의 방출을 억제한다. 화합물의 활성 및 선택률은 적합한 효소 억제 실험, 예를 들면 하기 실시예 A 및 WO-A-9611209에 기재된 실험에 의해 결정될 수 있다. 동일한 문헌에서 본 발명의 화합물을 실험하기 위한 다른 적합한 실험(실시예 B 내지 G)이 제시되어있다.
본 발명은 또한 상기한 바와 같이 매트릭스 금속단백질가수분해효소 및/또는 TNFα에 기인하는 환자(사람 및/또는 유제품, 고기 또는 모피 산업 또는 애완용으로 양육되는 포유 동물)의 치료 방식에 관한 것이고, 더욱 상세하게는 활성 성분인 식(Ⅰ)의 매트릭스 금속단백질분해효소 억제제의 투여와 관련된 치료 방식에 관한 것이다.
따라서, 식(Ⅰ)의 화합물은 골관절염 및 류머티양 관절염의 치료, 및 어떤 전이 종양 세포주에서 발견되는 이러한 금속단백질가수분해효소의 과잉발현(over-exprssion)으로부터 유발되는 질병 및 징후에서 다른 것들 사이에 사용될 수 있다.
상기에서, 식(Ⅰ)의 화합물은 TNFα 및 MMPs의 억제제와 같은 활성이 있기 때문에 인간 또는 수의학에서 유용하다. 따라서 또 다른 측면에서,
본 발명은
포유류에 상기 식(Ⅰ)의 화합물의 효과적인 양의 투여 또는 제약학적으로 허용 가능한 이들 염의 투여로 이루어진, 포유류, 특히 인간에 있어서 TNFα 및/또는 MMPs에 의해 중재된 질병 또는 증상의 치료 처리(management)(치료 또는 예방(prophylaxis)을 의미한다) 방식과;
인간의 또는 수의학 약품, 특히 TNFα 및/또는 MMPs에 의해 중재된 질병 또는 통증의 처리(치료 또는 예방을 의미한다)에 이용하기 위한 식(Ⅰ)의 화합물과;
TNFα 및/또는 MMPs에 의해 중재된 질병 또는 통증의 처리(치료 또는 예방을 의미한다)를 위한 제제의 제조에서 식(Ⅰ)의 화합물의 용도와 관련된다.
상기에서 언급된 질병 또는 증상은 염증성 질병, 자가면역 질병 암, 심장혈관 질병, 류머티양 관절염, 골관절염, 골다공증, 신경변성, 알츠하이머병, 아테로마성 동맥경화증, 울혈성 심부전증, 발작, 혈관염, 크론 질병, 궤양성 대장염, 다중 경화, 치주염, 치은염을 포함하는 조직 파열 질병 및 골 흡수(bone resorption), 히드록시출혈(hydroxy-haemorrhage), 응고, 급성 상 반응, 악액질 및 식욕부진, 급성 감염, HIV 감염, 열, 마비 상태, 이식세포 대 숙주 반응, 피부병, 외과 상처 치료(surgical wound healing), 건선, 아토피성 피부염, 표피박탈 수포(epidermolysis bullosa), 종양 성장, 맥관 형성 및 이차 변이에 의한 침입, 안과 질병, 망막증, 각막 궤양, 재융상(再融像) 손상(reperfusion injury), 편두통, 수막염, 천식, 비염, 알르레기성 결막염, 습진 및 아낙필락시 등의 상기 조직 파열과 관련된 질병을 포함한다.
류머티양 관절염, 골관절염 및 어떤 전이 종양 세포주에서 발견되는 매트릭스 금속단백질가수분해효소의 과압출로부터 초래된 질병 또는 징후 및 매트릭스 금속단백질가수분해효소 및 증가된 TNFα에 의해 중재된 다른 질병의 치료를 위해, 식(Ⅰ)의 화합물은 경구적으로, 국소적으로, 비경구적으로 투여되거나, 흡입 분사 또는 비독성의 제약학적으로 허용 가능한 담체, 보조액 및 부형제(賦形劑)를 포함하는 투약 단위 형태로 직장으로 투여할 수 있다. 본 명세서에서 이용된 바와 같이 비경구적 투약은 피하 주사, 정맥 주사, 근육내, 흉골내부 주사 또는 주입 기술을 포함한다. 쥐, 토끼, 말, 소, 양, 개, 고양이 등의 온혈 동물의 치료 외에도, 본 발명의 화합물은 인간의 치료에 효과적이다.
활성 성분을 포함하는 제약학적 조성물은 경구 이용에 적절한 형태, 예를 들면 정제, 구내정, 약용 드롭스(lozenges), 수성 또는 유성 현탁, 분사 분말 또는 과립, 유제, 경질 또는 연질 캡슐, 또는 시럽 또는 엘릭서제(elixirs)의 형태로 있을 수 있다. 경구 사용을 위한 조성물은 제약학적 조성물의 제조를 위한 기술에서 공지된 어떤 방법에 따라서 제조될 수 있고 이러한 조성물은 제약학적으로 우아하고 맛있는 조제를 위해 감미제, 조미제, 착색제 및 보존제등으로 이루어진 군으로부터 선택된 하나 이상의 제제를 포함할 수 있다. 정제는 정제 생산에 적합한 비독성의 제약학적으로 허용 가능한 부형제(excipients)와의 혼합물에서 활성 성분을 포함한다. 이러한 부형제는 예를 들어 탄산 칼슘, 탄산 나트륨, 락토오스, 인산 칼슘 또는 인산 나트륨 등의 불활성의 희석제와; 과립제 또는 분해제, 예를 들어 옥수수 전분, 또는 알긴산(alginic acid)과; 결합제(bonding agent), 예를 들어 전분, 젤라틴 또는 아카시아(acasia) 및 윤활제, 예를 들면 스테아르산 마그네슘, 스테아르산(stearic acid) 또는 활석등 이다. 상기 정제는 코팅되지 않을 수 있고 위장도(gastrointestinal tract)에서의 분해 및 흡수를 지연시키는 공지 기술로 코팅 될 수 있고 이것에 의해 보다 긴 기간을 통해 유지 작용된다. 예를 들어, 글리세릴 모노스테아르산염 또는 글리세릴 디스테아르산염 등의 시간 지연 물질이 이용될 수 있다. 이들을 또한 방출을 제어하기 위한 삼투 치료 정제를 만들기 위해 US특허 제 4,256,108; 4,166,452; 및 제 4,265,874호에 기재된 기술로 코팅하였다.
경구사용을 위한 제제는 또한 활성 성분 중 불활성 고체 희석제, 예를 들면 탄산 칼슘, 인산 칼슘 또는 카오린(kaolin)과 혼합된 경질 젤라틴 캡슐 또는 활성 성분 중 물 및 기름 중간 생성물, 예를 들면 땅콩 기름, 액체 파라핀 또는 올리브 기름과 혼합된 연질의 젤라틴 캡슐로서 존재한다.
수성 현탁액은 수성 현탁액의 제조에 적절한 부형제와 혼합한 활성 물질을 포함한다. 이러한 부형제는 현탁제, 예를 들면, 나트륨 카르복시메틸셀룰로스, 메틸셀룰로스, 히드록시프로필메틸셀룰로스, 나트륨 알긴산 폴리비닐-피롤리돈, 트라가칸스 고무(gum tragacanth) 및 아카시아 고무(gum acasia); 분산 또는 습윤제는 천연적으로 생기는 인지질(phosphatide) 예를 들면 레시틴(lecithin), 또는 지방산, 예를 들면 폴리옥시에틸렌 스테아르산염 및 알킬렌 산화물의 응축 생성물, 또는 긴 사슬의 지방족 알콜, 예를 들어 헵타데카에틸렌옥시세탄올과 에틸렌 산화물의 응축 생성물 또는 지방산으로부터 나온 에스테르 에틸렌 산화물 및 지방산으로부터 나온 부분적 에스테르와 이러한 폴리옥시에틸렌의 응축 산물 및 헥시톨 무수화물, 예를 들어 폴리옥시에틸렌 소르비탄 모노올레산염(polyoxy ethylene sorbitan monooleata)이 될 수 있다. 수성 현탁액은 또한 하나 이상의 방부제, 예를 들면 에틸 또는 n-프로필, p-히드록시벤조산염, 하나 이상의 착색제, 하나 이상의 조미제, 및 자당(sucrose) 또는 사카린(saccharin) 등의 하나 이상의 감미제를 포함한다.
유성 현탁액은 식물 기름, 예를 들면 아라치스 기름(arachis oil), 올리브 기름, 참기름 또는 코코넛 기름 중 또는 액체 파라핀 등의 광물 기름 중의 활성 성분을 현탁하여 형성될 수 있다. 이러한 유성 현탁액은 농후제(thickening agent), 예를 들면 밀납, 경질 파라핀 및 세틸 알콜을 포함할 수 있다. 상기 감미제 및 조미제가 맛있는 경구용 제제를 제조하기 위해 첨가된다. 이러한 조성물은 아스코르브산 등의 항산화제(anti-oxidant)의 첨가에 의해 보존될 수 있다.
물을 첨가하는 것으로 수성 현탁액을 제조하기 위한 적절한 분산 분말 및 과립은 분산 또는 습윤제, 현탁제 및 하나 이상의 방부제와의 혼합물 중 활성 성분을 제공한다. 적절한 분산 또는 습윤제 및 현탁제는 예시화되어 있고, 예를 들어 감미, 조미 및 착색제가 또한 존재할 수 있다.
본 발명의 제약학적인 조성물은 또한 물 중 오일 유탁액(oil-in-water emulsions)의 형태로 있다. 유층(oily phase)은 채소 기름, 예를 들면 올리브 기름, 아라치스 기름 또는 광물 기름, 예를 들면 액체 파라핀 또는 이것의 화합물일 수 있다. 적절한 유제는 자연적으로 나오는 고무, 예를 들면, 아카시아 고무 또는 트라가칸스 고무 일 수 있고, 자연적으로 나오는 인지질, 예를 들면 콩, 레시틴(lecithin), 및 지방산으로부터 나온 에스테르 또는 부분적인 에스테르 및 헥시톨 무수화물, 예를 들면 소르비탄 모노올레산염 및 에틸렌 산화물, 예를 들면 폴리옥시에틸렌 소르비탄 모노옥살산염과 이들의 부분적 에스테르의 응축 생성물이 있을 수 있다. 상기 유탁액는 조미 및 감미제를 포함한다.
시럽 및 엘리서제는 또한 감미제, 예를 들면 글리세롤, 프로필렌 글리세롤, 소르비톨 또는 자당을 이용하여 형성된다. 이러한 시럽 및 엘리서제 또한 진통제, 방부제 및 조미 및 착색제를 포함한다. 제약학적 조성물은 무균의 주입 가능한 수성의 또는 유성의 현탁액의 형태로 있을 수 있다. 이러한 현탁액은 상기된 적절한 분산 또는 습윤제 및 상기의 유화제를 이용하는 공지 기술에 따라 만들어졌다. 무균의 주입 가능한 제조물은 또한 예를 들면 1,3-부탄 디올중의 용액으로서 비독성의 비경구적으로 허용 가능한 희석제 또는 용매중의 무균의 주입 가능한 용액 또는 현탁액 중에 있을 수 있다. 허용 가능한 부형제 사이에서 사용 가능한 용매는 물, 링거 용액(Ringer's solution) 및 염화 나트륨 등장(isotonic) 용액이다. 덧붙여, 무균의 비휘발성 기름은 용매 또는 현탁 매개체로서 종래에 사용되었다. 이러한 목적을 위하여 어떤 비자극(bland) 비휘발성 기름은 합성의 모노- 또는 디글리세리드를 포함하여 사용될 수 있다. 추가적으로, 올레산 등의 지방산은 주입물의 제조에서 이용함을 알 수 있다.
식(Ⅰ)의 화합물은 또한 약의 직장(rectal) 투여를 위한 좌약의 형태로 투여될 수 있다. 이러한 조성물은 약을 적절한 비자극성의 부형제와 혼합하여 제조될 수 있고, 이러한 부형제는 상온에서는 고체지만 직장 온도(rectal temperature)에서는 액체이고 따라서 직장에서 융융되어 약이 방출되도록 할 것이다. 이러한 물질은 코코넛 버터와 폴리에틸렌 글리세롤류이다.
국소 사용을 위하여, 식(Ⅰ)의 화합물을 포함하는 크림, 연고(ointments), 젤리, 용액 또는 현탁액 등을 사용한다.(이러한 적용의 목적을 위하여, 국소 적용은 구강세척제 및 청정제 사용을 포함할 것이다.)
매일 체중 킬로그램 당 약 0.05 내지 140㎎의 정상적인 복용량의 한계는 상기 증상의 치료에 유용하다(매일 환자당 약 2.5㎎ 내지 7gms). 예를 들어, 염증은 매일 체중 킬로그램당 화합물 약 0.01 내지 50㎎의 투여에 의해 효과적으로 치료될 수 있다(매일 환자당 약 0.5㎎ 내지 약 3.5gms).
일회 복용 형태를 생산하기 위한 담체 물질과 배합될 수 있는 활성 성분의 양은 치료받는 숙주 및 투여되는 특별한 투여 양식에 따라 다양해질 것이다. 예를 들어, 인간의 경구 투여를 위한 형태는 전체 조성물의 약 5 내지 약 95%로 다양할 수 있다. 복용 단위 형태는 일반적으로 활성 구성성분의 약 1 내지 약 500㎎을 포함한다.
그러나, 어떤 특별한 환자에 대한 복용 농도는 사용된 특정한 화합물의 활성, 나이, 체중, 일반적인 건강 상태, 성별, 투여 약의 소화 시간, 투여 경로, 배출 속도, 약의 조성물 및 치료가 진행되는 동안 특정한 질병의 정도를 포함하는 여러 가지 인자들에 의존하여 이해될 수 있다.
다음의 실시예는 식(Ⅰ)의 화합물의 제조를 나타내기 위한 것이고, 청구항에 설명되어있는 바와 같이 본 발명은 이로 인해 제한되는 것은 아니다.

ECD(1.47g, 7.71mmol)을 3℃에서 테트라히드로퓨란(40㎖) 중 [(2R-브로모-5-프탈리미도]펜타노산 용액(see WO-A-9611209, intermediate 117, 2.40g, 7.35mmol) 및 N-히드록시벤조트리아졸(1.04g, 7.71mmol) 용액에 첨가하였다. 상기 용액에 L-류실-L-삼차-류신 N-메틸아미드(see WO-A-9611209, intermediate 116, 1.89g, 7.35mmol)를 첨가하고, 이 혼합물을 서서히 RT까지 가열하였고, 밤새도록 교반하였다. 상기 혼합물을 아세트산 에틸 및 물 사이에 분배하였고 수층을 아세트산 에틸로 추출하였다. 배합된 유기층을 1N HCl, 포화 중탄산 나트륨 용액 및 브린(brine)으로 세척하고, 건조하고(MgSO4) 진공에서 기화하여 추가적인 정제 없이 다음 단계로 이용될 수 있는 흰 거품의 형태로 표제 물질(3.93g, 95%)을 얻었다.

칼륨 삼차-부톡시드(677㎎, 6.03mmol)를 3℃에서 디메틸포름아미드(70㎖) 중 트리페닐메틸메르캅탄(1.66g, 6.03mmol)의 교반되고 있는 용액에 첨가하였다. 20분 동안 교반 한 후에 중간 생성물 1(3.25g, 5,75mmol)을 첨가하여 상기 혼합물을 천천히 RT까지 가열하고 밤새도록 교반하였다. 이어서 혼합물에 물(200㎖)을 쏟아 붓고 결과 침전물을 여과해서 수집하고 진공에서 기화하여 엷은 노란색 고체로서 표제 화합물(3.12g, 71%)을 얻었다.

40% 메틸아민 수용액(10㎖, 116mmol)을 RT에서 메탄올(20㎖) 중 중간 생성물 2(1.13g, 1.48mmol) 용액에 첨가하였다. 결과 현탁액을 RT에서 밤새도록 교반하였다. 상기 용액을 진공에서 기화시키고, 잔여물을 아세트산 에틸에 용해시켜 물 및 브린으로 세척하고, 건조하고(MgSO4) 진공에서 기화하여 오렌지색 거품의 표제 화합물(850㎎, 91%)을 얻었다.

EDC(65㎎, 0.34mmol)을 3℃에서 테트라히드로퓨란(15㎖) 중 중간 생성물 3(200㎎, 0.32mmol), N,N-디메틸글리신(33㎎, 0.32mmol) 및 N-히드록시벤조트리아졸(46㎎, 0.34mmol)의 교반되고 있는 용액에 첨가하였다. 혼합물을 RT까지 서서히 가열하고 밤새도록 교반하였다. 상기 혼합물을 아세트산 에틸 및 물 사이에 분배하고 수용액 층을 아세트산 에틸로 추출하였다. 배합된 유기 추출물을 포화 중탄산 나트륨 용액 및 브린으로 세척하고, 건조하고(MgSO4) 진공에서 기화하여 하얀색 고체로서 표제 화합물(163㎎, 0.23mmol, 71%)을 얻었다.

붕소 트리플루오리드 에테르산염(2㎖)을 RT에서 디클로로메탄(15㎖) 및 헥산 (50㎖) 중의 5-브로모부티르산(8.3g, 50mmol) 및 삼차-부틸 2,2,2-트리클로로아세티미산염(10.5g, 50mmol)의 혼합물에 첨가하였다. 혼합물을 RT에서 추가적으로 18시간 동안 교반하였고, 이어서 중탄산 나트륨을 첨가하여 가라앉혔다. 상기 혼합물은 셀라이트(Celite)를 통해 여과하고 여액을 진공에서 기화하여 무색의 기름으로서 표제 화합물(7.2g, 64%)을 얻었다.

수소화 나트륨(60%, 1.3g, 32mmol)을 0℃에서 디메틸포름아미드(10㎖) 중 1,5,5,-트리메틸히단토인(4.0g, 28.2mmol) 용액에 첨가하였고 이 혼합물을 질소 하에서 30분간 교반하였다. 이어서 중간 생성물 7(7.1g) 용액을 첨가하고, 결과 혼합물을 RT에서 밤새도록 교반한 다음 물(100㎖)을 쏟아 부은 다음 삼차-부틸 메틸 에테르(100㎖)로 추출하였다. 유기층을 물 및 브린으로 세척하고, 건조하고(MgSO4) 진공에서 기화하여 무색 기름으로서 표제 화합물(3.7g, 46%)을 얻었다.

트리플루오로아세트산(10㎖)을 RT에서 디클로로메탄(10㎖) 중 중간 생성물 7 (3.6g) 용액에 첨가하였고 상기 용액을 18시간 동안 교반하였다. 결과 용액을 진공에서 기화하고 잔여 트리플루오로아세트산을 톨루엔(3×50㎖)과 공비 혼합하여 무색의 점액질의 기름으로서 표제 화합물을 얻었고 이 화합물을 직접 다음 단계에서 이용하였다.

콜라게나아제 억제 활성
콜라게나아제의 억제제로서 작용하는 일반식(Ⅰ)의 화합물의 효능이 Cawston 및 Barrett(Anal. 스트로멜리신 억제 활성
스트로멜리신의 억제제로서 작용하는 일반식(Ⅰ)의 화합물의 효능이 Nagase et al(Methods in Enzymology Vol 254, 1994)의 절차에 의해 결정되었고 그것에 의해 실험필의 0.1mM 억제제 용액 또는 이들의 희석제를 스트로멜리신 및 3H 트랜스페린(transferrin)(10mM CaCl2, 150M NaCl, 0.05% 브리즈 35 및 0.02% NaN3을 포함하는 pH6.7인 50mM 트리스로 완충하였다)과 함께 37℃에서 16 시간 동안 배양하였다. 상기 트랜스페린을 3H 아이오도아세트산(iodoacetic acid)으로 카르복시메틸화하였다. 1mM 억제제 또는 이것의 희석제의 존재시, 스트로멜리신 활동도를 억제제 없는 제어에서의 활성과 비교하였고 그 결과를 스트로멜리신의 50% 억제를 나타내는 억제제 농도(IC50)로서 보고했다.

Acta, 258:566-576, 1972)의 절차를 이용하여 결정되었고 그것에 의해 실험 필의 1mM 억제제 용액 또는 이들의 희석제를 젤라틴 및 열변성된 3H 및 14C-아세틸화 콜라겐(5mM CaCl2, 0.05% 브리즈(Brij) 35 및 0.02% NaN3을 포함하는 pH7.6인 50mM 트리스로 완충하였다)과 함께 37℃에서 16시간 동안 배양하였다. 3H 또는 14C 젤라틴을 Cawton 및 Murphy(Methods in Ezymology, 80:711, 1981)의 방식에 따라 60℃에서 30분 동안 배양하여 제조된 3H 또는 14C-콜라겐을 변성시켜 제조하였다. 소화되지 않은 젤라틴을 트리클로로아세트산의 첨가 및 원심분리에 의해 침전시켰다. 1mM 억제제 또는 이것의 희석제의 존재시, 젤라티나아제 활성을 억제제 없는 제어에서의 활성과 비교하였고 그 결과를 젤라티나아제 50% 억제를 나타내는 억제제 농도(IC50)로서 보고했다(IC50).
실시예 D
MMP 억제 활성-형광 분석(Fluorimetric Assay)
콜라게나아제-1(MMP-1), 콜라게나아제-2(MMP-8), 젤라티나아제-A(MMP-2), 젤라티나아제-B(MMP-9) 및 스트로멜리신-1(MMP-3)의 억제제로서 작용하는 일반식(Ⅰ)의 화합물의 효능이 하기 절차에 의해 결정되었다:
억제제를 0.02% b-메르캅토에탄올을 포함한 디메틸설폭시드 중에 용해하였고 일련의 희석제를 제조하였다. 활성화된 효소를 pH 7.4인 50mM 트리스, 5mM CaCl2, 0.02% NaN3 및 브리즈 35를 포함한 분석 완충 용액 중에서 억제제가 존재하는 상태와 존재하지 않은 상태에서 배양하였다. 시료를 형광분석 기질(Mca-Pro-Leu-Dpa- Ala-Arg-NH2)을 최종 농도 10mM 까지 첨가하기 전에 37℃에서 15분 동안 예비배양하였다(preincubated). 상기 분석물을 37℃에서 90분 동안 배양한 다음 lex(355nm) 및 lem(460nm)에서 플루오로스캔 Ⅱ를 읽었다.
효소 활성을 콜라게나아제 활성을 억제제 없는 제어에서의 활성과 비교하였고 그 결과를 억제제의 농도가 스트로멜리신 50% 억제를 나타내는 억제제 농도(IC50)로서 보고했다.
실시예 E
TNFα 생성의 억제
TNFα 생성 억제제로서 작용하는 일반식(Ⅰ)의 화합물의 효능이 하기 절차를 이용하여 결정되었다. 실험 필한 1mM 억제제 용액 또는 이들의 희석제를 37℃에서 5% CO2의 대기에서 RPMI 1640 배지 중 현탁된 THP-1 세포(인간 단세포) 및 세포밀도 1×106/㎖에서의 20μM β-메르캅토에탄올을 사용하여 배양하였고 5㎍/㎖ LPS 최종 농도로 자극했다. 18시간 후 위에 뜨는 물질을 상업적으로 이용 가능한 ELISA 킷(R & D System)을 이용하여 TNFα의 농도를 분석하였다.
0.1mM의 억제제 또는 이것의 희석제의 존재시 활성을 억제제 없는 제어에서의 활성과 비교하였고 그 결과를 억제제의 농도가 TNFα생성의 50% 억제를 나타내는 억제제 농도로서 보고했다.
실시예 F
매질 관절염(adjuvant arthritic) 쥐 모델
일반식(Ⅰ)의 화합물을 B.B.Newbould(Br.J.Pharmacol, 21,127-136, 1963) 및 C.M.Pearson 및 F.D.Wood(Arthritis Rheum, 2, 440-459, 1959)에 의해 이용된 방식에 기초하여 쥐 중에서 매질 관절염 모델에서 평가하였다. 간단하게 수컷의 위스타 쥐(180 내지 200g)에 프론드 매질(Freund's adjuvant)을 꼬리 아래에 주사했다. 12일 후 반응 동물(responding animals)을 실험군으로 임의 추출하였다. 일반식(Ⅰ)의 화합물을 12일 내지 실험이 끝나는 22일 까지 1% 메틸 셀룰로스 중 현탁물로서 경구적으로 또는 0.2% 카르복시메틸셀룰로스 중 현탁물로서 복막조직내로 투여하였다. 뒷발의 용적을 12일 부터 매 2일 간격으로 측정하고 실험이 끝나는 날에 뒷다리를 X-선 촬영하였다. 12일 값을 통한 발용적의 퍼센트 증가로서 결과를 표현하였다.
실시예 G
마우스 난소 암종 이종이식편 모델
일반식(Ⅰ)의 화합물을 B.Davies et al(Cancer Research, 53, 2087-2091, 1993)에 기재된 방식에 기초하여 난소 암종 이종이식편 암 모델에서 평가하였다. 이 모델은 간단하게 암컷의 누/누 마우스에 1×109 OVCAR3-icr 세포를 복막의 동공으로 접종하는 것으로 이루어져 있다. 일반식 (Ⅰ)의 화합물을 1% 메틸 셀룰로스중 현탁물로서 경구적으로 또는 0.01% 트윈-20(Tween-20) 중 살린으로 완충된 인(phosphate) 중 현탁물로서 복막내로 투여하였다. 이 실험(4 내지 5주)의 결과로, 복막 세포의 수를 계수하였고 어떤 고형의 종양 침전물의 무게를 달았다. 일부 실험에서 종양 발육을 종양의 특정한 항체를 측정하여 관찰한다.
실시예 H
쥐 유방암 모델

- 매트릭스 금속단백질가수분해효소와 관련되거나 또는 TNFα 또는 L-셀렉틴 세다제(sheddase)에 의해 중재되는 증상의 치료 또는 예방용 인간이나 수의학적 약품의 제조를 위한 제1항 내지 제4항 중 어느 하나의 화합물로서, 상기 증상은 치주염 및 치은염으로부터 선택된 것인 화합물.
- 매트릭스 금속단백질가수분해효소와 관련되거나 또는 TNFα 또는 L-셀렉틴 세다제(sheddase)에 의해 중재되는 증상의 치료 또는 예방용 인간이나 수의학적 약품의 제조를 위한 제1항 내지 제4항 중 어느 하나의 화합물로서, 상기 증상은 비염, 알레르기성 결막염, 습진 및 아나필락시로부터 선택된 것인 화합물.
- 매트릭스 금속단백질가수분해효소와 관련되거나 또는 TNFα 또는 L-셀렉틴 세다제(sheddase)에 의해 중재되는 증상의 치료 또는 예방용 인간이나 수의학적 약품의 제조를 위한 제1항 내지 제4항 중 어느 하나의 화합물로서, 상기 증상은 리스토노시스, 울혈성 심부전증, 자궁내막증, 아테로마성 관절염 및 내경화증으로 부터 선택된 것인 화합물.
