- Phenylacetyl chloride (8.06 g) in dichloromethane (20 ml) was added dropwise to a solution of o-phenylenediamine (5.63 g) in dichloromethane (100 ml) and triethylamine (6.3 g) at 0° C. The mixture was stirred for 18 h and the suspension washed with water and brine. The solution was dried (Na 2 SO 4 ) and evaporated. The solid residue was purified by flash column chromatography (CH 2 Cl 2 /ether (3:1)) to give the title compound as colourless solid (1.32 g).
- Example 12 A solution of example 12 (200mg) in methanol (5 ml) was treated at RT with a solution of lithium hydroxide (51 mg) in water (5 ml). The mixture was stirred at RT for 4 h, then concentrated in vacuo. - Compounds of general formula (I) were evaluated in a HOSP.1 rat mammary carcinoma model of cancer (S.Eccles et al (1995), Cancer Research, in press). This model consists of the intravenous inoculation of female CBH/cbi rats with 2 ⁇ 10 4 tumour cells into the jugular vein. Compounds of general formula (I) are administered by the oral route as a suspension in 1% methyl cellulose or intraperitoneally as a suspension in phosphate buffered saline in 0.01% Tween-20. At the conclusion of the experiment (4-5 weeks) the animals were killed, the lungs were removed and individual tumours counted after hours fixation in Methacarn.
