Abstract
Description
各種ワクチンの中でも、生ワクチンは最も有効の高いものの一つであるが、一般に、新興ウイルスの弱毒性ワクチンの開発には非常に長い期間を必要とすることが知られている。これまでに、本発明者らが開発した、狂犬病ウイルス用又はリンダペスト用の組換えワクシニアウイルス(RVV)は、野外試験等において、優れた感染発症予防効果を発揮することが実証されている(非特許文献2)。また、重症急性呼吸器症候群(SARS)に対しても、その病原体として知られるSARS-CoVのcDNAを有する組換えワクシニアウイルスの作製に成功しており、優れた予防効果と再投与可能な製剤であることが確認されている(非特許文献3)。更に、H5N1 HPAIVに対して、H5N1 HPAIVのHAのcDNAを有する組換えワクシニアウイルスの作製にも成功しており、優れた予防効果を示す製剤であることが確認されている(非特許文献4)。
(1)ワクシニアウイルスDIs株のゲノム中に、発現プロモーターと、インフルエンザウイルス由来タンパク質をコードするDNAの全部若しくは一部とを含む組換えワクシニアウイルス、又は不活化したインフルエンザウイルスを含む、インフルエンザに対する予防又は治療用医薬。
(2)インフルエンザに対する予防又は治療が、活性化されたCD4陽性細胞及び/又は活性化されたCD8陽性細胞の作用によるものである(1)に記載の医薬。
(3)インフルエンザウイルス由来タンパク質に対する抗体をさらに含む、(1)又は(2)に記載の医薬。
(4)抗体が結合抗体である(3)に記載の医薬。
(5)インフルエンザウイルス由来タンパク質が、H5N1高病原性鳥インフルエンザウイルス由来ヘマグルチニンタンパク質である(1)〜(4)のいずれか1項に記載の医薬。
(6)以下の(a)〜(c)のいずれかの成分を含む、インフルエンザに対する予防又は治療用医薬。
(a) 活性化されたCD4陽性細胞、CD4陽性細胞の活性化剤、及びCD4陽性細胞とCD4陽性細胞の活性化剤との組み合わせからなる群から選ばれる少なくとも1つの成分
(b) 活性化されたCD8陽性細胞、CD8陽性細胞の活性化剤、及びCD8陽性細胞とCD8陽性細胞の活性化剤との組み合わせからなる群から選ばれる少なくとも1つの成分
(c) 上記(a)及び(b)の成分の組み合わせの成分
(7)ワクシニアウイルスDIs株のゲノム中に、発現プロモーターと、インフルエンザウイルス由来タンパク質をコードするDNAの全部若しくは一部とを含む組換えワクシニアウイルス、又は不活化したインフルエンザウイルスをさらに含む、(6)に記載の医薬。
(8)インフルエンザウイルス由来タンパク質に対する抗体をさらに含む、(6)又は(7)に記載の医薬。
(9)抗体が結合抗体である(8)に記載の医薬。
(10)インフルエンザウイルス由来タンパク質が、H5N1高病原性鳥インフルエンザウイルス由来ヘマグルチニンタンパク質である(7)〜(9)のいずれか1項に記載の医薬。
(11)ワクシニアウイルスDIs株のゲノム中に、発現プロモーターと、インフルエンザウイルス由来タンパク質をコードするDNAの全部若しくは一部とを含む組換えワクシニアウイルス、又は不活化したインフルエンザウイルスを含む、インフルエンザに対する予防又は治療用キット。
(12)インフルエンザに対する予防又は治療が、活性化されたCD4陽性細胞及び/又は活性化されたCD8陽性細胞の作用によるものである(11)に記載のキット。
(13)インフルエンザウイルス由来タンパク質に対する抗体をさらに含む、(11)又は(12)に記載のキット。
(14)抗体が結合抗体である(13)に記載のキット。
(15)インフルエンザウイルス由来タンパク質が、H5N1高病原性鳥インフルエンザウイルス由来ヘマグルチニンタンパク質である(11)〜(14)のいずれか1項に記載のキット。
(16)以下の(a)〜(c)のいずれかの成分を含む、インフルエンザに対する予防又は治療用キット。
(a)活性化されたCD4陽性細胞、CD4陽性細胞の活性化剤、及びCD4陽性細胞とCD4陽性細胞の活性化剤との組み合わせからなる群から選ばれる少なくとも1つの成分
(b) 活性化されたCD8陽性細胞、CD8陽性細胞の活性化剤、及びCD8陽性細胞とCD8陽性細胞の活性化剤との組み合わせからなる群から選ばれる少なくとも1つの成分

本発明の範囲はこれらの説明に拘束されることはなく、以下の例示以外についても、本発明の趣旨を損なわない範囲で適宜変更し実施し得る。なお、本明細書において引用した特許文献、非特許文献その他の刊行物は、参照として本明細書に組み込まれるものとする。
現行のH5N1 HPAIVに対するプレパンデミックワクチンは、リバースジェネティクス技術を用いて、マルチベーシック部位を欠損させた H5N1 HAを持つインフルエンザウイルスを作出し、ホルマリンにより不活化させた全粒子ワクチンである。効果的な免疫応答を誘導する為には、3週間間隔で2回の接種が必要である事や接種ワクチンとは異なるクレードのウイルス株に対して、予防効果が顕著に減弱する。
他方、H5N1 HPAIV用ワクチン(rDIs/mCl2.2)接種個体では、H5N1 HPAIVに対する中和抗体が誘導されずとも良好な予防効果を発揮する事が判明した(図3、図4)。
そこで本発明においては、中和抗体に依存しない防御効果の解明を目指した。
更に、感染3日後から6日までのウイルス排除には、CD8陽性細胞が重要な役割を果たす事が判明した(図8)。更に、CD4陽性細胞及びCD8陽性細胞の両方を枯渇する事で、ワクチン接種個体においても、感染10日後にも関わらずウイルスを排除できず、著しく体重が減少し、死亡し始めた(図9)。
また、インフルエンザウイルスHAタンパク質に対して活性化されたCD4陽性細胞及び/又は活性化されたCD8陽性細胞は、急激な流行を示すインフルエンザウイルスに対して、安全かつ有効な予防薬又は治療薬となることが分かった。
本発明の一つの態様では、活性化されたCD4陽性細胞、CD4陽性細胞の活性化剤、又はCD4陽性細胞の活性化剤とCD4陽性細胞との組み合わせをインフルエンザの予防又は治療用医薬(予防又は治療用医薬組成物)として使用する。
また本発明の一つの態様では、活性化されたCD8陽性細胞、CD8陽性細胞の活性化剤、又はCD8陽性細胞とCD8陽性細胞の活性化剤との組み合わせを、インフルエンザに対する予防薬又は治療用医薬(予防又は治療用医薬組成物)として使用する。
以下、CD4陽性細胞若しくはCD8陽性細胞、又はCD4陽性細胞及びCD8陽性細胞の両者を、「CD4/CD8陽性細胞」ともいう。
CD4陽性細胞は、ヘルパーT細胞(Th1、Th2又はTh17に分類される)及び制御性T細胞である。CD4陽性細胞が持つT細胞受容体(TCR)はマクロファージやB細胞が持つクラスIIのMHC分子と結合する。ヘルパーT細胞はマクロファージなどからMHCクラスII分子を通じて、抗原提示を受け、B細胞や細胞傷害性T細胞を活性化する。他方、制御性T細胞は、免疫応答の終結に関与する。
不活化したインフルエンザウイルスとは、ワクチン製造用のインフルエンザウイルスを発育鶏卵に接種して増殖させ、漿尿液から精製・濃縮したウイルスをエーテルで部分分解し、更にホルマリンで不活化したものを意味する。但し本発明においては、ウイルス粒子そのものを不活化したものも使用することができる。
採取された細胞がCD4/CD8陽性細胞であることの確認は、フローサイトメトリー法により行うことができる。
CD4陽性細胞の活性化剤としては、インターロイキン(IL)-2、IL-4、CD86、OX40リガンド、抗原であるHAタンパク質やその一部などが挙げられる。CD4陽性細胞を上記活性化剤とともに培養することにより活性化させる。
CD8陽性細胞の活性化剤としては、IL-2、IL-12、IFN-g、CD86、CD137 リガンド、OX40リガンド、抗原であるHAタンパク質やその一部などが挙げられる。CD8陽性細胞を上記活性化剤とともに培養することにより活性化させる。
他方、本発明において、CD4/CD8陽性細胞活性化剤を被検者(健常者又は患者)に投与すると、投与された被検者の体内において、被検者由来のCD4/CD8陽性細胞が活性化される。体内におけるCD4/CD8陽性細胞の活性化の仕組みは、IL-2、IL-4、CD86、OX40リガンド、抗原であるHAタンパク質やその一部などのCD4陽性細胞活性化剤が投与されると、CD40L 、CD44及びCD69などの表面分子発現が亢進し、その結果、CD4陽性細胞が活性化される。また、IL-2、IL-12、IFN-g、CD86、CD137 リガンド、OX40リガンド、抗原であるHAタンパク質やその一部などのCD8陽性細胞活性化剤が投与されると、CD44やCD69などの表面分子発現が亢進し、その結果、CD8陽性細胞が活性化される。
本発明における別の態様として、インフルエンザウイルス由来タンパク質に対する抗体を、本発明の組換えワクシニアウイルス、又は不活化したインフルエンザウイルスとともに、インフルエンザの予防又は治療用医薬として使用する。
また本発明における別の態様として、インフルエンザウイルス由来タンパク質に対する抗体は、CD4/CD8陽性細胞の活性化剤及び/又は活性化されたCD4/CD8陽性細胞とともに、インフルエンザの予防又は治療用医薬として使用する。
また、上記インフルエンザウイルス由来タンパク質に対する抗体又はその抗原結合断片を、インフルエンザの予防又は治療用医薬として使用する。
「中和抗体」とは、インフルエンザウイルス由来タンパク質(例えばH5N1ウイルスのHAタンパク質)を抗原とする抗体であり、後述のDIsワクチン接種後の個体から産生される抗体である。中和抗体の中和活性とは、ウイルスが標的細胞への接着及び/又は感染を阻害する機能を意味する。すなわち、中和抗体の機能は、インフルエンザウイルス由来タンパク質のどの部分に結合するかは問わず、当該タンパク質に結合することにより、ウイルスのタンパク質が標的細胞表面にある受容体に接着し感染することを阻害する機能を意味する。
さらに、本発明の別の態様において、本発明の抗体は、表1に記載のアクセッション番号に示す塩基配列と高ストリンジェントな条件下でハイブリダイズする塩基配列を含むポリヌクレオチドによりコードされる変異型ポリペプチドを認識するものであってもよい。
本発明の中和抗体は、上記の通り、DIsワクチン接種後の個体から産生される抗体である。
DIsワクチンをそれ自体で、あるいは担体、希釈剤と共に非ヒト哺乳動物、例えばウサギ、イヌ、モルモット、マウス、ラット、ヤギ、ウマ、ヒツジ等に投与することにより免疫する。免疫方法は周知である。
血清中の抗体の中和価の測定は、マイクロ中和抗体測定法等によって行うことができる。中和価が十分上昇したことを確認した後、全採血し、通常行われる方法により抗体を分離精製する。分離精製は、硫安塩析法、イオン交換クロマトグラフィー、ゲル濾過クロマトグラフィー、アフィニティークロマトグラフィー等の公知の方法を適宜選択して、又はこれらを組み合わせることにより、精製することができる。
本発明の結合抗体は、インフルエンザウイルス由来タンパク質(例えばH5N1ウイルスのHAタンパク質)に対する抗体であるから、当該タンパク質又はその部分ペプチドを抗原として用いて、通常のポリクローナル抗体又はモノクローナル抗体を作製する方法と同様にして作製することができる。
(1)ポリクローナル抗体の作製
インフルエンザウイルス由来タンパク質又は部分ペプチドをそれ自体で、あるいは担体、希釈剤と共に非ヒト哺乳動物、例えばウサギ、イヌ、モルモット、マウス、ラット、ヤギ等に投与することにより免疫する。抗原の動物1匹当たりの投与量、アジュバントを用いるときの投与量、アジュバンドの種類、免疫部位、免疫の間隔等は周知である。
血清中の抗体価の測定も周知であり、ELISA、EIA、RIA等によって行うことができる。抗体価が十分上昇したことを確認した後、全採血し、通常行われる方法により抗体を分離精製することができる。分離精製は、硫安塩析法、イオン交換クロマトグラフィー、ゲル濾過クロマトグラフィー、アフィニティークロマトグラフィー等の公知の方法を適宜選択して、又はこれらを組み合わせることにより、精製することができる。
モノクローナル抗体の作製法も周知である。例えば、抗体産生細胞の採取、ハイブリドーマを得るための抗体産生細胞とミエローマ細胞との細胞融合、ハイブリドーマの選別及びクローニング、モノクローナル抗体の採取などは、当業者に周知の方法により行うことができる。
さらに、本発明において、抗体のエピトープ(抗原決定基)は、抗原であるインフルエンザウイルス由来タンパク質の少なくとも一部であればよく限定はされない。本発明の抗体には、当該抗体が結合する部位(例えばエピトープ)に結合する抗体、例えば、本発明のハイブリドーマが産生する抗体が結合する部位に結合する抗体が含まれる。
本発明の抗体の好ましい態様の一つとして、遺伝子組換え抗体が挙げられる。遺伝子組換え抗体としては、限定はされないが、例えば、キメラ抗体、ヒト型化抗体及びヒト化抗体等が挙げられる。
キメラ抗体(すなわちヒト型キメラ抗体)は、マウス由来抗体の可変領域をヒト由来の定常領域に連結(接合)した抗体であり(Proc. Natl. Acad. Sci. U.S.A. 81, 6851-6855, (1984) 等を参照)、キメラを作製する場合は、そのように連結した抗体が得られるよう、遺伝子組換え技術によって容易に構築できる。
本発明で使用されるインフルエンザウイルス由来タンパク質に対する抗体の断片としては、例えば、Fab、Fab'、F(ab')2、Fv、diabody(dibodies)、dsFv、scFv(single chain Fv)などが挙げられる。上記抗体断片は、本発明の抗体を目的に応じて各種タンパク質分解酵素で切断することにより得ることができる。
H5N1 HPAIV用ワクチンの作製方法は公知であり(第5884100号特許)、当該公知方法に従って作製することができる。作製方法の概要を以下に示す。
例えば、下記表1に示すように、GenBankアクセッション番号に、H5N1 HPAIVの各クレード由来のHAタンパク質をコードする遺伝子、あるいはH5N1 HPAIVの各クレードのHAタンパク質をコードするDNAの一部を欠損させた遺伝子が登録されている。
不活化ワクチンの製造法は公知である。前記の通り、ワクチン製造用のインフルエンザウイルスを発育鶏卵に接種して増殖させ、漿尿液から精製・濃縮したウイルスをエーテルで部分分解し、更にホルマリンで不活化する。
本発明は、(i)DIs株由来の上記組換えワクシニアウイルス若しくは不活化ウイルス、又は(ii)当該組換えワクシニアウイルス若しくは不活化ウイルスとインフルエンザウイルス由来タンパク質に対する抗体との組み合わせを含む、インフルエンザの予防又は治療用医薬を提供する。

(b) CD4/CD8陽性細胞の活性化剤
(c) CD4陽性細胞とCD4陽性細胞の活性化剤との組み合わせ
(d) CD8陽性細胞とCD8陽性細胞の活性化剤との組み合わせ
さらに本発明は、上記医薬又は上記医薬を患者(被検者)に投与することを含む、インフルエンザの予防及び治療方法、インフルエンザの予防及び治療のための上記医薬の使用、上記新型インフルエンザの予防薬及び治療薬を製造するための上記医薬の使用等も提供する。
さらに、本発明の医薬組成物に含まれる抗体、CD4/CD8陽性細胞、CD4/CD8陽性細胞の活性化剤、又はこれらの組合せは、上記組換えワクシニアウイルス又は不活化ウイルスと併用することも可能である。
併用の態様は特に限定されるものではなく、本発明の医薬と本発明の組換えワクシニアウイルス又は不活化ウイルスとを同時に投与することもでき、また、一方を投与後、一定時間経過後に他方を投与する方法により生体に導入することもできる。
投与量は、有効成分の種類、投与経路、投与対象、患者の年齢、体重、性別、症状その他の条件により適宜選択される。
活性化されたCD4/CD8陽性細胞:静脈内
CD4/CD8陽性細胞の活性化剤:経口、静脈内
一方で、感染1日後から3日後までのウイルス感染防御には、CD4陽性細胞が必須であることが、CD4陽性細胞の枯渇実験の結果から明らかとなった。更に、感染3日後から6日までのウイルス感染防御にはCD8陽性細胞が必要であることが判明した。
本発明のキットには、RVV(rDIs)若しくは不活化ウイルス、インフルエンザウイルス由来タンパク質に対する抗体、又はこれらの組合せを含めることができる。
また、本発明のキットには、 以下の(a)〜(d)のいずれか、又はこれらの組合せが含まれる。
(a) 活性化されたCD4/CD8陽性細胞
(b) CD4/CD8陽性細胞の活性化剤
(c) CD4陽性細胞とCD4陽性細胞の活性化剤との組み合わせ
(d) CD8陽性細胞とCD8陽性細胞の活性化剤との組み合わせ
本発明のキットは、H5N1高病原性鳥インフルエンザに対する予防用又は治療用に使用される。
以下に、実施例を挙げて本発明をより具体的に説明する。これらの実施例は説明のためのものであり、本発明の範囲を限定するものではない。
1) インフルエンザウイルスヘマグルチニンタンパク質遺伝子を導入したDIs株由来組換えワクシニアウイルスの作製
ワクシニアウイルスプロモーターmH5配列の下流に人工合成したH5N1 HPAIVの抗原であるヘマグルチニンタンパク質(HA)遺伝子を連結させ、相同組換え用プラスミドベクターへ挿入した(図1)。この際、H5N1亜型HA配列を挿入する場合には、制限酵素部位としてHind III部位を使用した。予めDIs株を感染させておいたCEFへ作製した相同組換え用プラスミドベクター(制限酵素により一箇所切断し、線状化した。)を遺伝子導入することによって、DIs株の遺伝子欠損部位の隣接領域にて相同組換えを起こさせてワクシニアウイルスのゲノム中にmH5プロモーター及びインフルエンザウイルスHAタンパク質遺伝子を挿入した組換えワクシニアウイルス(RVV)、具体的には、rDIs/mCl1、rDIs/mCl2.1、rDIs/mCl2.2、及びrDIs/mCl2.3の各種RVVを作出した。インフルエンザウイルス由来のHAタンパク質は、モルモット赤血球に対して凝集反応を示すが、ワクシニアウイルス由来のHAタンパク質はモルモット赤血球に対して凝集反応を起こさない。従って、上記組換えウイルスをCEFに感染させ、これにより形成するプラークへのモルモット赤血球の凝集反応を指標にして、RVVのスクリーニングを行った。目的の組換えウイルスは、赤血球凝集活性を持つレッドプラークを形成するものを選別した。
組換えワクシニアウイルス(rDIs/mCl2.2)を1x107 PFUでマウスの背部皮内へ接種した。接種から4週間後にH5N1 HPAIVの攻撃感染を行い、連日体重測定を行い、生存率を求めた。更に、1、3、6、9日後に採血と肺組織サンプル採取を行い、肺組織中ウイルス量の測定を行った。
採取した血清を用いて、マウスへの感染に使用したH5N1 HPAIVと同一のウイルス株を用いて、血清中に含まれる中和価を測定した。
H5N1 HPAIVへの結合抗体を移入したマウスへH5N1 HPAIVを攻撃感染し、感染1日後と6日後に肺組織サンプルを採取して、肺組織中ウイルス価を測定した。
組換えワクシニアウイルス(rDIs/mCl2.2)を1x107 PFUでFcR gamma欠損マウスと野生型マウスの背部皮内へ接種した。接種から4週間後にH5N1 HPAIVの攻撃感染を行い、連日体重測定を行い、生存率を求めた。更に、1、3日後に肺組織サンプル採取を行い、肺組織中ウイルス量の測定を行った。
組換えワクシニアウイルス(rDIs/mCl2.2)を1x107 PFUでBALB/cマウスの背部皮内へ接種した。接種から4週間後にH5N1 HPAIVの攻撃感染を行った。攻撃感染に先立ち、感染2日前より、一日おきにCD4陽性細胞枯渇用抗体を静脈内投与した。連日体重測定を行い、生存率を求めた。更に、1、3、6日後に肺組織サンプル採取を行い、肺組織中ウイルス量の測定を行った。
組換えワクシニアウイルス(rDIs/mCl2.2)を1x107 PFUでBALB/cマウスの背部皮内へ接種した。接種から4週間後にH5N1 HPAIVの攻撃感染を行った。攻撃感染に先立ち、感染2日前より、一日おきにCD8陽性細胞枯渇用抗体、又はCD4陽性細胞及びCD8陽性細胞枯渇用両抗体を静脈内投与した。1、3、6日後に肺組織サンプル採取を行い、肺組織中ウイルス量の測定を行った。
組換えワクシニアウイルス(rDIs/mCl2.2)を1x107PFUでBALB/cマウスの背部皮内へ接種した。接種から4週間後にH5N1 HPAIVの攻撃感染を行った。攻撃感染に先立ち、感染2日前より、一日おきにCD4陽性細胞及びCD8陽性細胞枯渇用両抗体を静脈内投与した。連日体重測定を行い、生存率を求めた。
作製したRVV(rDIs/mCl2.2)単回接種マウスでは、H5N1 HPAIVによる攻撃感染後に一過性の体重減少を示したが、その後速やかに回復、100%の生存率を示した。一方、ワクチンを接種していないコントロール群では、急激な体重減少を示し、全匹死亡した(図2)。この時、コントロール群及びワクチン接種群の肺組織中ウイルス価を測定した結果、コントロール群では、感染9日後まで高いウイルス価を示したが、ワクチン接種群では、殆どの個体において、感染6日後までにウイルス価は、検出限界以下にまで低下していた(図3)。
そこで、ワクチン接種個体における防御作用機序を詳細に解析する目的で、ワクチン接種個体から回収した抗血清中に含まれるIgGを精製し、ナイーブマウスへ静脈内投与した後、H5N1 HPAIVの攻撃感染を行った。その結果、感染1日後の肺組織中ウイルス価は、ワクチン接種個体へのH5N1 HPAIV感染時と同様に、ワクチン非接種個体に比べて、1/5から1/10にまで低下していた(図5)。次に、抗原特異的IgGと結合するFcRを有する細胞群がこの感染初期のウイルス価の低下に関与するか否かを検討する目的で、FcR gamma欠損マウスにワクチン接種を行った後、H5N1 HPAIVを攻撃感染した。
インフルエンザウイルスHAタンパク質遺伝子を発現するDIs株由来RVVは、H5N1 HPAIV用ワクチン(rDIs/mCl2.2)としては、1x107 PFU量のRVVを単回接種することで、致死性のH5N1 HPAIV攻撃感染に対しても速やかな体重回復と100%の生存率を示した。よって、DIs株由来RVVは、H5N1 HPAIV用ワクチンとしても十分な効果を有するものである。通常、H5N1 HPAIV用ワクチン(rDIs/mCl2.2)接種個体では、H5N1 HPAIV感染6日後において、ウイルスは検出限界以下にまで排除される。このワクチン接種個体において、H5N1 HPAIV感染1日後までの感染防御には、結合抗体とFc gammaレセプターを有する細胞群がウイルス感染防御に重要である事が判明した。一方で、感染1日後から3日後までのウイルス感染防御には、CD4陽性細胞が必須であることが、CD4陽性細胞の枯渇実験の結果から明らかとなった。
Claims (20)
- ワクシニアウイルスDIs株のゲノム中に、発現プロモーターと、インフルエンザウイルス由来タンパク質をコードするDNAの全部若しくは一部とを含む組換えワクシニアウイルス、又は不活化したインフルエンザウイルスを含む、インフルエンザに対する予防又は治療用医薬。
- インフルエンザに対する予防又は治療が、活性化されたCD4陽性細胞及び/又は活性化されたCD8陽性細胞の作用によるものである請求項1に記載の医薬。
- インフルエンザウイルス由来タンパク質に対する抗体をさらに含む、請求項1又は2に記載の医薬。
- 抗体が結合抗体である請求項3に記載の医薬。
- インフルエンザウイルス由来タンパク質が、H5N1高病原性鳥インフルエンザウイルス由来ヘマグルチニンタンパク質である請求項1〜4のいずれか1項に記載の医薬。
- 以下の(a)〜(c)のいずれかの成分を含む、インフルエンザに対する予防又は治療用医薬。
(a) 活性化されたCD4陽性細胞、CD4陽性細胞の活性化剤、及びCD4陽性細胞とCD4陽性細胞の活性化剤との組み合わせからなる群から選ばれる少なくとも1つの成分
(b) 活性化されたCD8陽性細胞、CD8陽性細胞の活性化剤、及びCD8陽性細胞とCD8陽性細胞の活性化剤との組み合わせからなる群から選ばれる少なくとも1つの成分
(c) 上記(a)及び(b)の成分の組み合わせの成分 - ワクシニアウイルスDIs株のゲノム中に、発現プロモーターと、インフルエンザウイルス由来タンパク質をコードするDNAの全部若しくは一部とを含む組換えワクシニアウイルス、又は不活化したインフルエンザウイルスをさらに含む、請求項6に記載の医薬。
- インフルエンザウイルス由来タンパク質に対する抗体をさらに含む、請求項6又は7に記載の医薬。
- 抗体が結合抗体である請求項8に記載の医薬。
- インフルエンザウイルス由来タンパク質が、H5N1高病原性鳥インフルエンザウイルス由来ヘマグルチニンタンパク質である請求項7〜9のいずれか1項に記載の医薬。
- ワクシニアウイルスDIs株のゲノム中に、発現プロモーターと、インフルエンザウイルス由来タンパク質をコードするDNAの全部若しくは一部とを含む組換えワクシニアウイルス、又は不活化したインフルエンザウイルスを含む、インフルエンザに対する予防又は治療用キット。
- インフルエンザに対する予防又は治療が、活性化されたCD4陽性細胞及び/又は活性化されたCD8陽性細胞の作用によるものである請求項11に記載のキット。
- インフルエンザウイルス由来タンパク質に対する抗体をさらに含む、請求項11又は12に記載のキット。
- 抗体が結合抗体である請求項13に記載のキット。
- インフルエンザウイルス由来タンパク質が、H5N1高病原性鳥インフルエンザウイルス由来ヘマグルチニンタンパク質である請求項11〜14のいずれか1項に記載のキット。
- 以下の(a)〜(c)のいずれかの成分を含む、インフルエンザに対する予防又は治療用キット。
(a)活性化されたCD4陽性細胞、CD4陽性細胞の活性化剤、及びCD4陽性細胞とCD4陽性細胞の活性化剤との組み合わせからなる群から選ばれる少なくとも1つの成分
(b) 活性化されたCD8陽性細胞、CD8陽性細胞の活性化剤、及びCD8陽性細胞とCD8陽性細胞の活性化剤との組み合わせからなる群から選ばれる少なくとも1つの成分
(c) 上記(a)及び(b)の成分の組み合わせの成分 - ワクシニアウイルスDIs株のゲノム中に、発現プロモーターと、インフルエンザウイルス由来タンパク質をコードするDNAの全部若しくは一部とを含む組換えワクシニアウイルス、又は不活化したインフルエンザウイルスをさらに含む、請求項16に記載のキット。
- インフルエンザウイルス由来タンパク質に対する抗体をさらに含む、請求項16又は17に記載のキット。
- 抗体が結合抗体である請求項18に記載のキット。
- インフルエンザウイルス由来タンパク質が、H5N1高病原性鳥インフルエンザウイルス由来ヘマグルチニンタンパク質である請求項17〜19のいずれか1項に記載のキット。
