Abstract
Description
(1)ピロロキノリンキノン及び/又はその塩と、
非還元性糖と、
ゲル化剤と、
を含有するゼリー。
(2)ピロロキノリンキノン及び/又はその塩を0.004〜0.05重量%、
非還元性糖を2〜20重量%、
ゲル化剤を0.2〜5重量%、
含有する、(1)に記載のゼリー。
(3)ピロロキノリンキノンの塩がピロロキノリンキノンジナトリウム塩である(1)記載のゼリー。
(4)非還元性糖がソルビトール、キシリトール又はショ糖である(1)から(3)のいずれかに記載のゼリー。
(5)ゲル化剤が寒天又はゼラチンである、(1)から(4)のいずれかに記載のゼリー。
(6)ホモジナイズされている、(1)から(5)いずれかに記載のゼリー。
(7)ピロロキノリンキノン及び/又はその塩と、非還元性糖と、ゲル化剤と、を配合する工程を含む、(1)〜(6)のいずれかに記載のゼリーを製造する方法。
(8)有効成分として非還元性糖を含む、ピロロキノリンキノン又はその塩を含有するゼリーの安定化剤。
(9)有効成分として非還元性糖を含む、ピロロキノリンキノン又はその塩を含有するゼリーの着色抑制剤。
ピロロキリンキノンジナトリウム 0.02重量% ソルビトール10重量% 寒天2重量%、水残余、からなる組成の試料(総重量50g)を調製し、これをオートクレーブ(121℃)で15分加熱した。加熱した溶液を混合することで均一にした。得られた溶液を冷蔵庫(4℃)で冷やし、1日保存することでゼリー食品が得られた。得られたゼリーはピロロキノリンキノンに起因する赤色を呈しており、その色は全体的に均一であった。
各ゼリー1gを水10mlと混合し、ポリプロピレン製15mL遠心分離容器に入れ、室温で30分振った。この溶液を液体クロマトグラフィー用の溶離液で10倍に希釈した。これを0.2μmフィルターで濾過した。ピロロキノリンキノン含有量について、高速液体クロマトグラフィー(HPLC)を用い以下の条件で分析した。
HPLC条件:259nm, 40℃、YMC−Pack ODS−A 150mm, 4.6mm, 30mM 酢酸−70mM酢酸アンモニウム、装置LC2010(株式会社島津製作所製)
その結果、添加したピロロキリンキノンジナトリウム含有量はゼリー中で100%維持されていた。
異性化糖は王子コーンスターチ株式会社製王子の異性化糖HC(果糖分55%以上)を使用した。ピロロキリンキノンジナトリウム0.02重量% 異性化糖10重量% 寒天2重量%、水残余、からなる組成の試料(総重量50g)を調製し、これをオートクレーブ(121℃)で15分加熱して均一にした。得られた溶液を冷蔵庫(4℃)で冷やした。1日経過しても、溶液はゼリー状に固まっておらず、当初赤色だった混合物の色が黄色に変色していた。また、HPLC分析により、ゼリー中のピロロキリンキノンジナトリウム含有量は添加量との比較で59%に減少していた。
実施例1の試料の組成中、ピロロキノリンキノンジナトリウムの含有量のみを変えたゼリー食品を実施例1と同様の手法により調製した(実施例2〜5)。同様に、比較例1の試料の組成中、ピロロキノリンキノンジナトリウムの含有量のみを変えたゼリー食品を比較例1と同様の手法により調製した(比較例2)。また、比較例1の異性化糖に代えて、グルコース(比較例3)と果糖(比較例4)を添加したゼリー食品も調製した。調製した各ゼリー食品5gを口に入れ30秒保持し、その後、5分後に舌全体を見て舌におけるピロロキノリンキノンの着色のしやすさを観察した。
実施例6、7、参考例1〜3
ゲル化剤の種類を変更してゼリーの形成を検討した。ピロロキリンキノンジナトリウム 0.02重量% ソルビトール10重量% ゲル化剤2重量% 水残余、からなる組成の試料(総重量50g)をオートクレーブ(121℃)で15分加熱して均一にした。冷蔵庫(4℃)で冷やし、1日後、ゼリーの形成を観察した。その結果を表2に示す。
国際公開第2012/020767号(上掲)の実施例13に基づき、ピロロキノリンキノンの塩と分散媒を用いてピロロキノリンキノン自体のゲルを調製した。まず、PQQ粉末0.1gを15mlのプラスチック製遠心分離用容器に加え、水10ml加え、室温で振って混合した(約25℃)。これを冷蔵庫で冷却して4℃になった後、さらに振って混合した。一晩、冷蔵庫に保存すると全体が均一なゲル状に変化した。続いて、寒天0.01gを水10mlに加え、電子レンジで溶解したものを約40℃まで冷却し、上記のピロロキノリンキノン1%ゲルと1mlずつ混合した。混合物を冷蔵庫で一晩冷却したところ、ゲル化しており、光学顕微鏡で繊維が確認できた。得られたゼリーを口に入れ30秒保持したところ、ゼリーの色が舌に着色した。
ホモジナイザーを用い、実施例1で作製したゼリーを室温で均一の大きさに壊れるように砕いた。固まっていたゼリーが砕かれて流動性のあるクラッシュゼリーになった。
ピロロキノリンキノンジナトリウムの濃度を変えて舌への着色を調べた。ソルビトール10重量%、寒天2%に固定した点を除き、ゼリーの調製は上述のとおり行った。
ピロロキノリンキノンジナトリウム0.04重量%、ソルビトール10重量%の水溶液を口に30秒含み着色を調べた結果、舌に着色が見られた。この結果から、驚くべきことに、ピロロキノリンキノンの着色抑制には非還元性糖をゼリーに配合する必要があることが分かった。

- ピロロキノリンキノン及び/又はその塩と、
非還元性糖と、
ゲル化剤と、
を含有するゼリー。 - ピロロキノリンキノン及び/又はその塩を0.004〜0.05重量%、
非還元性糖を2〜20重量%、
ゲル化剤を0.2〜5重量%、
含有する、請求項1に記載のゼリー。 - ピロロキノリンキノンの塩がピロロキノリンキノンジナトリウム塩である、請求項1又は,2に記載のゼリー。
- 非還元性糖がソルビトール、キシリトール又はショ糖である、請求項1から3のいずれか1項に記載のゼリー。
- ゲル化剤が寒天又はゼラチンである、請求項1から4のいずれか1項に記載のゼリー。
- ホモジナイズされている、請求項1から5のいずれか1項に記載のゼリー。
- ピロロキノリンキノン及び/又はその塩と、非還元性糖と、ゲル化剤とを配合する工程を含む、請求項1〜6のいずれか1項に記載のゼリーを製造する方法。
- 有効成分として非還元性糖を含む、ピロロキノリンキノン又はその塩を含有するゼリーの安定化剤。
- 有効成分として非還元性糖を含む、ピロロキノリンキノン又はその塩を含有するゼリーの着色抑制剤。
