Abstract
Description
例えば、特表2008−525454号公報(特許文献2)には、抗体を製造するように遺伝子改変された植物細胞を、懸濁培養により増殖させ、抗体を生産するシステムが提案されている。しかしながら、植物では分化した組織や器官で特異的に有用物質の生産及び蓄積を行うことが知られている。二次代謝産物でいえば、例えばオウレンのベルべリンは根茎に、ムラサキのシコニンは根に特異的に蓄積されるし、タンパク質の例では、種子に貯蔵タンパク質として蓄積されること等が知られている。従って、脱分化した培養細胞を用いた場合は、植物組織の場合よりも、有用タンパク質の生産効率において劣る問題がある。
本発明の好ましい態様によれば、この方法は、液体培養条件下にて行われる。
アグロバクテリウム法では、目的遺伝子を含む発現可能な核酸構築物を含むアグロバクテリウム菌を、植物に接種する。接種に使用するアグロバクテリウム菌の調製方法は以下の通りである。
本発明の方法は、植物体の液体培養を継続的に実施できるような環境で行うことが望ましい。
したがって、植物体の液体培地液体無菌もしくはそれに準じる環境であることが好ましい。
本発明においてはさらに、得られた植物体から、目的とする有用タンパク質を、増殖させた植物体もしくは液体培地から抽出し、または分離する工程をさらに含むことができる。すなわち、増殖させた植物体もしくは液体培地から目的タンパク質を含む画分を取得し、目的タンパク質を適当な方法により分離・精製することが好ましい。なお、目的タンパク質をコードする目的遺伝子を含む核酸構築物には、例えば、精製のためのタグ配列や植物細胞外への分泌のためのシグナル配列を含んでもよい。
本発明の方法は、容器内において実施する。容器内において実施すると、無菌もしくはそれに準じる環境を保持し易い。すなわち、ここで培養容器とは、室内に培養槽を配置したような培養室自体や水耕栽培可能な温室自体などを含まない意味で使用される。好ましくは、無菌化した培養容器内で実施するのがよく、より好ましくは、フィルター等で無菌化した空気等ガスの導入部と排出部を有する容器内での実施が望ましい。
本発明の方法は、好ましくは、袋型培養槽内において行うのがよい。
好ましい袋型培養槽(培養袋)およびそのような培養槽を使用した大量生産設備の例を図12および図13にそれぞれ例示する。
植物体培養法による成長速度の比較を行うため、プラントボックスにて固体培養法と液体培養法でのジャガイモ植物体の成長量の比較を行った。
植物としては、ばれいしょ(Solanum tuberosum L.)の品種「サッシー」の組織培養苗を用いた。
ここで、カナマイシン耐性遺伝子の配列を特異的に増幅するプライマーとして、TAAAGCACGAGGAAGCGGT(配列番号1)、及びGCACAACAGACAATCGGCT(配列番号2)を用いた。
ジャガイモのin vitro培養条件は以下の通りである。
培養容器はプラントボックス(株式会社三商より入手)を使用した。得られた形質転換植物体は、固体培地にて頂芽を用いて継代した。固体培地は、MS培地(スクロース3%を添加し、pH5.8)に寒天0.8%を加えたものを使用した。
有用タンパク質であるβ−グルクロニダーゼの生産量を示す指標として、β−グルクロニダーゼ(GUS)活性を以下のように測定した。
結果は下記の通りであった。
次に、異なる液体培地量での成長量の比較を行った。
用いた植物材料、有用タンパク質の生産量検証方法および液体培地組成は実施例1と同様であった。
袋型培養槽(8L容)に、プラントボックスで固体培地を用いて約2か月培養した約2gの植物体を、1Lの液体培地に入れたもの(1L区)と、4Lの液体培地に入れたもの(4L区)で、GUSを発現する遺伝子組換えジャガイモの成長を40日間培養後に比較した。
結果は下記の通りであった。
40日間の培養後の新鮮重量を比較すると、1L区では139g、4L区では576gとなり、その収穫量は4.1倍であった(図2)。この際、1L区も4L区もいずれも良好に成長し、1L区では主に液体培地の液面より上部(気相部)への成長が、4L区では主に液体培地内(液相部)への成長が認められた。
結果は下記の通りであった。
液体培地に常時浸漬していた液相部は気相部よりも、総タンパク質あたりのGUS活性が高かった。すなわち、葉では気相部で315.8μmol/min/mg proteinに対し、液相部が700.8μmol/min/mg proteinであり、液相部で2.2倍高い値となった(図4)。
この結果から、有用タンパク質生産には液体に浸漬させた状態での培養が適していることが示された。
上記実施例3で確認された液体浸漬培養時の葉でのGUS活性値と、同じ組換え植物を土耕栽培した場合と葉比較した。ジャガイモの温室での土耕栽培条件は以下の通りであった。
得られた形質転換植物体の頂芽を固体培地にて継代し、14週経過したin vitro苗を園芸用土(コンパル高級園芸用土、スミリン農産工業株式会社)に馴化し、4週間温室にて育成した。
結果は下記の通りであった。
液体浸漬培養の葉と比較して、土耕栽培の葉では総タンパク質あたりのGUS活性が123.9μmol/min/mg proteinに低下した(図6)。このことからも今回達成された液体浸漬培養による有用タンパク質生産は産業上有用な方法であることが確認できた。
リーフレタスにおける植物体培養法による有用タンパク質生産量の比較を行うため、固体培養法と液体浸漬培養法での比較を行った。
植物としては、リーフレタス(Lactuca sativa)の品種「グリーンウェーブ」の組織培養苗を用いた。
ここで、ハイグロマイシン耐性遺伝子の配列を特異的に増幅するプライマーとして、CGGAAGTGCTTGACATTGG(配列番号3)、及びAGAAGAAGATGTTGGCGACC(配列番号4)を用いた。
リーフレタスのin vitro培養条件は以下の通りである。
培養容器はプラントボックス(株式会社三商より入手)を使用した。得られた形質転換
植物体は、固体培地にて頂芽を用いて継代した。固体培地は、MS培地(スクロース3%
を添加し、pH5.8)に寒天0.8%を加えたものを使用した。
液体培地の場合には、MS培地(スクロース3%及びBA0.02ppm、ジベレリン0.001ppmを添加し、pH5.8)を使用した。
有用タンパク質であるβ−グルクロニダーゼの生産量を示す指標として、β−グルクロ
ニダーゼ(GUS)活性を以下のように測定した。
有用タンパク質であるトランスフェリンの生産量を以下のように測定した。
結果は下記の通りであった。
12.9gであり、液体培養法の方が21.5倍の増加量となった。このデータから、植物体の成長には個体培養法よりも液体培養法の方が適していることが示された(図7)。

Claims (10)
- 容器内において、有用タンパク質の生産が可能な植物体の少なくとも一部を、液体培地に浸漬させたまま培養し、植物体を増殖させることを含む、植物による有用タンパク質の生産方法。
- 植物体が、有用タンパク質の生産が可能なように改変された植物体である、請求項1に記載の方法。
- 植物体が、安定な遺伝子組換え植物である、請求項1または2に記載の方法。
- 生産される有用タンパク質が、医療用タンパク質または産業用タンパク質である、請求項1〜3のいずれか一項に記載の方法。
- タンパク質が構成的発現プロモーターを用い発現されるものである、請求項1〜4のいずれか一項に記載の方法。
- 増殖させた植物体から、有用タンパク質を抽出または分離する工程をさらに含む、請求項1〜5のいずれか一項に記載の方法。
