Abstract
Description
本発明(2)は、前記リン酸カルシウム微粒子の平均粒子径が、10〜500nmである、前記発明(1)の製造方法である。
本発明(3)は、前記組成物が、熱分解性粒子を含む、前記発明(1)又は(2)の製造方法である。
本発明(4)は、前記熱分解性粒子が、親水性ポリマーである、前記発明(3)の製造方法である。
本発明(5)は、前記熱分解性粒子が、(メタ)アクリル酸ポリマー又はその誘導体である、前記発明(3)又は(4)の製造方法である。
本発明(6)は、前記焼成リン酸カルシウム成形体が、ハイドロキシアパタイトからなる、前記発明(1)〜(5)のいずれか一つの製造方法である。
本発明(7)は、前記焼成リン酸カルシウム成形体が、前記焼成リン酸カルシウム成形体の全質量を基準として、ハイドロキシアパタイトを80質量%以上含有する、前記発明(6)の製造方法である。
<成分>
本発明に係る焼成リン酸カルシウム成形体は、リン酸カルシウムからなる。ここで、リン酸カルシウムとしては、ハイドロキシアパタイト、β−リン酸三カルシウム(TCP)、α−TCP及びこれらの混合体を挙げることができる。尚、水不溶性やタンパク吸着特性等が求められる用途にて本発明に係る焼成リン酸カルシウム成形体を用いる場合、リン酸カルシウムはハイドロキシアパタイトであることが好適である(例えば、焼成リン酸カルシウム成形体の全質量を基準とした場合、ハイドロキシアパタイトが、好適には80質量%以上、より好適には90質量%以上)。ここで、ハイドロキシアパタイトの含有量は、XRDにて測定可能である。
<非含有成分>
本発明に係る焼成リン酸カルシウム成形体は、後述のように酸化カルシウムを実質的に含有しない{XRDで測定した際、酸化カルシウムのピーク(2θ=37.34°)が出現せず}。
本発明に係る焼成リン酸カルシウム成形体は、多孔質であっても緻密体であってもよい。ここで、緻密体とは、気孔率が5%以下のものを指す(多孔質とは、気孔率が5%超のものを指す)。ここで、気孔率は、{(体積×構成成分の理論密度−実質量)/(体積×構成成分の理論密度)}×100(%)で算出された値である。尚、大きな表面積が求められる用途等においては、多孔質であることが好適である。ここで、多孔質である場合、好適な焼成リン酸カルシウム成形体は、微細な孔(好適には、平均孔径=10〜1000nm)と、大きな孔(好適には、平均孔径=50〜500μm)と、を有している。ここで、平均孔径は、走査型電子顕微鏡(SEM)にて撮影した画像により算出した。なお、微細な孔は、10,000倍で撮影した画像の視野中に存在するすべての孔の長径を計測し、平均した値を平均孔径とした。また、大きな孔は、30倍で撮影した画像の視野中に存在するすべての長径を計測し、平均した値を平均孔径とした。
次に、本発明に係る焼成リン酸カルシウム成形体の製造方法を詳述する。
{ポリエーテル変性のリン酸カルシウム微粒子}
本製造方法で使用される一原料は、ポリエーテル変性のリン酸カルシウム微粒子(リン酸カルシウム微粒子とポリエーテルとの複合体)であり、好適にはポリエーテル変性のリン酸カルシウムナノ粒子である。以下、好適例であるポリエーテル変性のリン酸カルシウムナノ粒子を使用した態様を詳述する。当該粒子は、リン酸カルシウムナノ粒子の表面に、ポリエーテルが化学結合したものである。尚、化学結合しているか否かは、リン酸カルシウムナノ粒子単独及びポリエーテルをTG−DTA解析やFT−IR解析にて、これら成分単独では出現しないピークが、ポリエーテル変性のリン酸カルシウムナノ粒子では出現することにより確認できる。以下、当該粒子の各構成成分と製造方法を説明する。
ポリエーテルは、主鎖に複数のエーテル結合を有する高分子であり、環状エーテル(例えばクラウンエーテル)の開環重合、グリコール(例えばポリエチレングリコール(PEG))の自己縮合、ジオール化合物の酸化重合などにより得られる。ポリエーテルの具体例としては、特に限定されないが、例えば、ポリエチレングリコール、ポリエチレンオキサイド、ポリプロピレングリコール、ポリプロピレンオキサイド、ペンタエチレングリコール、ポリテトラメチレンオキサイド、ポリフェニレンエーテル、ペンタエチレングリコールモノドデシルエーテル、デキストリンやシクロデキストリン等の多糖類、等が含まれる。ここで、本発明のポリエーテルは、末端のOH基が封止されている態様(例えばアルキルや糖類で化学修飾)をも包含する。ここで、当該ポリエーテルの役割(酸化カルシウム形成防止機能)は下記の通りと推定される。まず、リン酸カルシウムナノ粒子と結合したポリエーテルは、リン酸カルシウム粒子同士を弱く結び付けるよう機能すると考えられる{この結果、ナノ粒子(塊)同士の凝集が阻害されたスラリーを得ることができる}。このようにして得られたスラリーを加熱することにより、均質性が保たれたまま脱水が進行し、一次粒子が接近することで、融着が促進される。このような原理にて、通常の焼結温度よりも低い温度でリン酸カルシウムナノ粒子同士が接着するものと推察される。即ち、リン酸カルシウムが酸化カルシウムに変化する温度未満での加熱環境下では、リン酸カルシウム粒子同士は既に凝集しており緻密構造を採っているため、当該状況でもリン酸カルシウム粒子間での物質移動が起き難くなる(即ち、リン酸カルシウム→酸化カルシウムの分解が抑制)。この結果、酸化カルシウムが殆ど存在しないリン酸カルシウム均一相が形成されると考えられる。
リン酸カルシウムナノ粒子は、焼成でも未焼成のものでもよい。尚、リン酸カルシウムナノ粒子は、例えば、特許第5043436号の記載に従い製造し得る。ここで、リン酸カルシウムナノ粒子の平均粒子径としては、10nm〜500nmが好ましく、15nm〜450nmがさらに好ましく、20nm〜400nmが最も好ましい。また一次粒子からなる一次粒子群の粒子径の変動係数が、20%以下であることが好ましく、18%以下であることがさらに好ましく、15%以下であることが最も好ましい。尚、一次粒子の粒子径および変動係数は、動的光散乱法又は電子顕微鏡を用い、少なくとも100個以上の一次粒子について粒子径を測定して計算すればよい。ここで、「変動係数」は、標準偏差÷平均粒子径×100(%)で計算することができる粒子間の粒子径のバラツキを示す値である。
ポリエーテル変性のリン酸カルシウムナノ粒子は、例えば、ポリエーテル(例えばPEG)及びリン酸カルシウムナノ粒子(例えばハイドロキシアパタイトナノ粒子)を水中に添加し混合し、温度5〜100℃にて10〜120分撹拌し続けることにより製造し得る。
(膨潤度)
本発明に係る焼成リン酸カルシウム成形体の内、多孔質体を製造する際には、原料として熱分解性粒子を用いることが好適である。なお、熱分解性粒子とは、後述する焼成温度(例えば、200〜2000℃)において熱分解が生じる粒子を示す。ここで、本発明で使用される熱分解性粒子は、好適には親水性ポリマーである。ここで、親水性とは、一般的な意義での親水性であり、例えば、水に対する膨潤度が5〜10,000倍(質量比)であることを指す。尚、当該膨潤度の測定法は下記の通りである。膨潤前の熱分解性粒子の嵩密度を計測し、一方、熱分解性粒子に対して成形体作成時に加える条件にあわせ水に加え、生じた上澄みを除いた後、水を加える前後の質量変化をもって膨潤度とする。
好適な熱分解性粒子は、(メタ)アクリル酸ポリマー又はその誘導体である。ここで、誘導体には、(メタ)アクリル酸ポリマーのカルボキシル基を修飾したものや、(メタ)アクリル酸ポリマーの架橋体を挙げることができる。ここで、特に好適な熱分解性粒子は、アクリル酸ポリマーの糖類での架橋体である。
本発明に係る熱分解性粒子(乾燥状態)は、好適には、リン酸カルシウムナノ粒子の平均粒子径よりも大きい平均粒子径のものである。具体的には、本製造法で用いられる熱分解性粒子(乾燥状態)は、好適には平均粒子径が10〜1,000nmのもの、より好適には平均粒子径が50〜800nmのものである。尚、本明細書において「平均粒子径」とは、個々の粒子における径の、粒子群全体での平均を意図するが、ロッド状粒子やファイバー状粒子等のような、長径と短径とが異なる粒子の場合は、個々の粒子における長径の粒子群全体での平均をいう。
本発明に係る焼成リン酸カルシウム成型体の製造方法は、前記複合体(リン酸カルシウムナノ粒子とポリエーテルとの複合体)を少なくとも含有する組成物(多孔質体を製造する際には熱分解性粒子も含む)を加熱し、ポリエーテル(熱分解性粒子を含む場合には当該成分も)を熱分解しつつ、前記リン酸カルシウムナノ粒子同士を焼結させる工程を含む。この際、焼成温度は、200〜2000℃であることが好適であり、300〜1800℃であることがより好適である。更に、焼成時間は、0時間超24時間以下であることが好適であり、0時間超18時間以下であることがより好適である。また、多孔質体を製造する場合には、前記複合体と熱分解性粒子との合計:前記熱分解性粒子(質量比)は、10:1〜30:1であることが好適であり、15:1〜20:1であることがより好適である。
焼成リン酸カルシウム成形体の内、ハイドロキシアパタイト成形体の製造プロセスを説明する。ハイドロキシアパタイト成形体の製造プロセスは、例えば、(1)原料として、ハイドロキシアパタイトナノ粒子とポリエーテルとの複合体を用い、300〜1400℃{ハイドロキシアパタイトの融点よりも低い温度}にて焼成する手法、(2)原料として、α−TCPナノ粒子とポリエーテルとの複合体を用い、300〜1800℃にて焼成して焼成体を得、その後、当該焼成体に水を加える手法、である。
本発明に係る焼成リン酸カルシウム成形体は、前記のように酸化カルシウムを実質的に含有しない。よって、焼成リン酸カルシウム成形体が水と接触した場合であっても、不均一相となることに起因した機械的強度の低下や酸化カルシウムが溶解することに起因したアルカリ発生の問題が無い。したがって、機械的強度が要求される分野やアルカリ発生が問題視される分野(例えば電気・電子材料分野では接合面の腐食の問題を招く)に有効である。具体的には、例えば、また、本発明に係る焼成リン酸カルシウム成形体の内、緻密体である態様は、センサ等の電極表面の保護皮膜、絶縁物、電池のセパレータ、イオン伝導体、ペプチドや生理活性物質の担体に使用し得る。他方、本発明に係る焼成リン酸カルシウム成形体の内、多孔質である態様は、前記の特性に加え、微細な孔(好適には、平均孔径=10〜100nm)と、大きな孔(好適には、平均孔径=50〜500μm)と、を好適には有している。即ち、比表面積が非常に大きい。更に、従来の焼成リン酸カルシウム成形体のような、ナノサイズを中心とした孔だけでなく、マイクロサイズの孔も有している。よって、以上の特性を考慮し、多孔質である態様は、例えばフィルタとして使用した場合、圧力損出の少なく、タンパク吸着能が高く、且つ、水不溶性のフィルタとなり得る。具体例としては、微生物やウイルス除去用フィルタや水浄化用フィルタを挙げることができる。また、電子や物質移動を検知するためのセンサに用いる隔膜としても有用である。更に、本発明に係る焼成リン酸カルシウム成形体がハイドロキシアパタイト成形体である場合、水に殆ど溶解しないため、例えば生体内に載置する医療器具等に適している。
連続オイル相としてドデカン〔CH3(CH2)10CH3〕、非イオン性界面活性剤として曇点31℃のペンタエチレングリコールドデシルエーテル〔CH3(CH2)10CH2O(CH2CH2O)4CH2CH2OH〕を用いた。室温において、上記非イオン性界面活性剤0.5gを含有している連続オイル相40mlを調製した。次に、上記で調製した連続オイル相に2.5mol/l水酸化カルシウム〔Ca(OH)2〕分散水溶液10mlを添加し、油中水滴型溶液(W/O溶液)を調製した。上記W/O溶液を撹拌しながら、そこに1.5mol/lリン酸二水素カリウム〔(KH2PO4)〕溶液を10ml添加した。そして、24時間、室温で撹拌しながら反応させた。次に、得られた反応物を遠心分離により分離洗浄することにより、ハイドロキシアパタイト(HAp)一次粒子群を取得した。当該ハイドロキシアパタイト(HAp)一次粒子群における一次粒子の粒子径は10nm〜100nm(平均粒子径23nm)であり、当該一次粒子群の粒子径の変動係数が11%以下であった。
窒素ガス雰囲気下、HApナノ粒子50gとポリエチレングリコール(PEG3400)50gとを脱イオン水500mlに添加して80℃にて約1時間保持し、HAp/PEG複合体を含有する分散液を得た。尚、熱重量測定を用いて、複合化前のHAp及びPEGと、HAp/PEG複合体と、を熱重量測定してそれぞれを比較した。ここで、図1は、複合化前のHApのチャートであり、図2は、複合化前のPEGのチャートであり、図3は、HAp/PEG複合体のチャートである。これらの図から分かるように、複合化前のHApやPEGと比較して、HAp/PEG複合体の発熱ピークが低温側にシフトした。このこと、即ち、HApとPEGが固有に示す特性とは異なる温度に発熱ピークが観察されたことから、複合化されたと理解される。
・製造例1(原料:HAp)
1gのアクリル酸ポリマー架橋体からなる粒子であるカーボポール{商品名:LubrizolAdvancedMaterials.Inc製)}を50mlの脱イオン水で溶解したものに、19gの前記分散液(HAp/PEG複合体を含有する分散液)を24℃にて12時間以上混合して撹拌した後、当該液を1200℃で4時間熱処理し、多孔質材料を得た。図4及び図5は、本多孔質材料のSEM写真である(倍率違い)。図4及び図5から、マイクロメートルオーダー及びナノメートルオーダーの孔が開いていることが確認できる。尚、マイクロメートルオーダー孔の平均孔径は280μmであり、ナノメートルオーダー孔の平均孔径は200nmであった。更に、気孔率は11%であった。また、図6は、焼成前後におけるX線回折チャートである(左が焼成後、右が焼成前)。当該図から、多少、β−TCP由来のピークが出ている(複数回実施した結果、β−TCPは最大でも20質量%)ものの、当該条件では大部分(80質量%以上)がハイドロキシアパタイトであることが確認できた。また、酸化カルシウムのピークは確認されなかった。
・製造例2(原料:HAp)
製造例1におけるカーボボールの使用量を0.1gに変更した以外は、製造例1と同様の手法にて多孔質材料を得た。図7は、本多孔質材料のSEM写真である。当該写真から、数百μmの孔が開いていることが分かる(また、図示しないが、ナノメートルオーダーの孔が開いていることが確認された)。気孔率は5%超であった。尚、当該多孔質材料におけるハイドロキシアパタイトの量は84質量%であり、β−TCPは16質量%であった。また、酸化カルシウムのピークは確認されなかった。
・製造例3(原料:HAp)
製造例1におけるカーボボールの使用量を2.5gに変更した以外は、製造例1と同様の手法にて多孔質材料を得た。図8は、本多孔質材料のSEM写真である。当該写真から、数百μmの孔が開いていることが分かる(また、図示しないが、ナノメートルオーダーの孔が開いていることが確認された)。気孔率は5%超であった。また、図示しないが、本多孔質材料においては、ハイドロキシアパタイト量を80質量%超含み、且つ、酸化カルシウムを包含していないことが確認された。
窒素ガス雰囲気下、α−TCP微粒子20gとポリエチレングリコール(PEG3400)20gとを脱イオン水200mlに添加して80℃にて約1時間保持し、α−TCP/PEG複合体を含有する分散液を得た。尚、製造例1での手法に準じ、α−TCP粒子とPEGとが化学結合していることが確認された。
1gのアクリル酸ポリマー架橋体からなる粒子であるカーボポール{商品名:LubrizolAdvancedMaterials.Inc製)}を50mlの脱イオン水で溶解したものに、19gの前記分散液(α−TCP/PEG複合体を含有する分散液)を24℃にて12時間以上混合して撹拌した後、当該液を1200℃で4時間熱処理し、目視において孔の形成が確認出来る状態の多孔質材料を得た(図示しないが、マイクロメートルオーダー及びナノメートルオーダーの孔が開いていることが確認された)。気孔率は5%超であった。
・製造例5
20gの前記分散液(HAp/PEG複合体を含有する分散液)を1200℃で4時間熱処理し、緻密体材料を得た。本緻密体材料の気孔率は、2%であった。また、図示しないが、本多孔質材料においては、ハイドロキシアパタイト量を80質量%超含み、且つ、酸化カルシウムを包含していないことが確認された。
Claims (7)
- リン酸カルシウム微粒子とポリエーテルとの複合体を少なくとも含む組成物を加熱し、前記リン酸カルシウム微粒子同士を焼結させる工程を含むことを特徴とする、焼成リン酸カルシウム成形体の製造方法。
- 前記リン酸カルシウム微粒子の平均粒子径が、10〜500nmである、請求項1記載の製造方法。
- 前記組成物が、熱分解性粒子を含む、請求項1又は2記載の製造方法。
- 前記熱分解性粒子が、親水性ポリマーである、請求項3記載の製造方法。
- 前記熱分解性粒子が、(メタ)アクリル酸ポリマー又はその誘導体である、請求項3又は4記載の製造方法。
- 前記焼成リン酸カルシウム成形体が、ハイドロキシアパタイトからなる、請求項1〜5のいずれか一項記載の製造方法。
- 前記焼成リン酸カルシウム成形体が、前記焼成リン酸カルシウム成形体の全質量を基準として、ハイドロキシアパタイトを80質量%以上含有する、請求項6記載の製造方法。
