Abstract
Description
下記実施例および比較例に示す重量平均分子量(カタログ値のものを除く)および動粘度について、各試料をそれぞれ以下のようにて測定することにより、予め確認したものを使用した。
蒸留水に溶解したポリ−γ−グルタミン酸ナトリウムを、下記条件でゲル透過クロマトグラフィーに供し、重量平均分子量を測定した。
カラム:Shodex Asahipak GF-710HQ(昭和電工株式会社製)
カラム温度:40℃
移動相:50mM塩化ナトリウム水溶液
流量:0.6mL/分
注入量:100μL
検出器:UV220nm
なお、標準物質としてプルランを使用した。
蒸留水に溶解したポリ−γ−グルタミン酸ナトリウム(濃度0.05%w/w)について、試料温度37℃にて粘度計ウベローデ(柴田科学株式会社製(型番)SU−00286)により動粘度(cSt)を測定した。当該動粘度については、得られた流出時間(秒)に対し、以下の式:
「動粘度(cSt)=粘度計定数×流出時間(秒)」
より算出した。
Donryuラット(雄、5〜8週齢)を、1群当たり4〜8体になるようにA群、B群、C群、D群およびE群の5群に分けた。各ラットについて、全身麻酔下にて腹部正中切開で開腹し、大腸の回盲部を腹腔外へ取り出し、その漿膜面を小出血班ができるまでガーゼによる擦過を行い、そしてドライヤーで温風を約10秒間当てた。次いで、大腸の回盲部直上の腹壁を約1cm×1cmの範囲に渡って切除し、ドライヤーで温風を約10秒間当てた。その後、約10分間室内気で乾燥させた。
1 鈍的に剥離可能な軽度の癒着が観察された
2 鋭的剥離が5割以下の割合で観察された
3 鋭的剥離が5割以上の割合で観察された
4 剥離時に盲腸の漿膜損傷が観察された
5 剥離時に盲腸の全層損傷が観察された
3つのポリスチレン(PSt)シャーレを用意し、各シャーレに80万の重量平均分子量および2.8cStの動粘度を有するポリ−γ−グルタミン酸ナトリウム粉末(実施例3)、150万〜250万(カタログ値)の重量平均分子量および5.5cStの動粘度を有するポリ−γ−グルタミン酸ナトリウム粉末(和光純薬工業株式会社製;実施例4)、20万〜50万(カタログ値)の重量平均分子量および1.1cStの動粘度を有するポリ−γ−グルタミン酸ナトリウム粉末(和光純薬工業株式会社製;比較例4)をそれぞれ20mgずつ塗布後、0.1mlの蒸留水を添加し、スパチュラでかき混ぜてゲルを作製した。
Claims (4)
- 60万から1300万の重量平均分子量を有するポリ−γ−グルタミン酸および/またはその塩を有効成分として含有する、癒着防止材。
- 前記ポリ−γ−グルタミン酸および/またはその塩を0.05質量%の濃度で蒸留水に溶解させた際の37℃での動粘度が2cStから15cStである、請求項1に記載の癒着防止材。
- 粉末またはゲルの形態に加工されている、請求項1または2に記載の癒着防止材。
- 術後の創部における臓器の癒着を防止するために用いられる、請求項1から3のいずれか一項に記載の癒着防止材。
