また、核初期化プロトコルについても未だ改善の余地があり、種々の改良プロトコルが報告されている(非特許文献9−14)。
[1]T細胞抗原受容体遺伝子のα鎖領域がNKT細胞受容体特異的な態様で均一なVα-Jαに再構成されている体細胞に核初期化因子を接触させることにより得られるクローン化された細胞であって、T細胞抗原受容体遺伝子のα鎖領域がNKT細胞受容体特異的な態様で均一なVα-Jαに再構成されており、かつ分化多能性および自己複製能を有する細胞。
[2]体細胞がNKT細胞である、上記[1]記載の細胞。
[3]体細胞が線維芽細胞である、上記[1]記載の細胞。

これらの組み合わせの中で、得られるNKT-iPS細胞を治療用途に用いることを念頭においた場合、Oct3/4, Sox2及びKlf4の3因子の組み合わせが好ましい。一方、NKT-iPS細胞を治療用途に用いることを念頭に置かない場合(例えば、創薬スクリーニング等の研究ツールとして用いる場合など)は、Oct3/4, Klf4, Sox2及びc-Mycの4因子か、それにLin28またはNanogを加えた5因子が好ましい。特に好ましくは、本発明における核初期化因子は、Oct3/4, Klf4, Sox2及びc-Mycの4因子である。
PTDとしては、ショウジョウバエ由来のAntP、HIV由来のTAT、HSV由来のVP22等のタンパク質の細胞通過ドメインを用いたものが開発されている。核初期化因子のcDNAとPTD配列とを組み込んだ融合タンパク質発現ベクターを作製して組換え発現させ、融合タンパク質を回収して導入に用いる。導入は、タンパク質導入試薬を添加しない以外は上記と同様にして行うことができる。
マイクロインジェクションは、先端径1μm程度のガラス針にタンパク質溶液を入れ、細胞に穿刺導入する方法であり、確実に細胞内にタンパク質を導入することができる。
核初期化因子のcDNAは、宿主となるNKT細胞等の体細胞で機能し得るプロモーターを含む適当な発現ベクターに挿入される。発現ベクターとしては、例えば、レトロウイルス、レンチウイルス、アデノウイルス、アデノ随伴ウイルス、ヘルペスウイルスなどのウイルスベクター、動物細胞発現プラスミド(例、pA1-11,pXT1,pRc/CMV,pRc/RSV,pcDNAI/Neo)などが用いられ得る。
用いるベクターの種類は、得られるNKT-iPS細胞の用途に応じて適宜選択することができる。例えば、アデノウイルスベクター、プラスミドベクター、アデノ随伴ウイルスベクター、レトロウイルスベクター、レンチウイルスベクターなどが使用され得る。
核初期化因子である核酸を含む発現ベクターは、ベクターの種類に応じて、自体公知の手法により細胞に導入することができる。例えば、ウイルスベクターの場合、該核酸を含むプラスミドを適当なパッケージング細胞(例、Plat-E細胞)や相補細胞株(例、293細胞)に導入して、培養上清中に産生されるウイルスベクターを回収し、各ウイルスベクターに応じた適切な方法により、該ベクターを細胞に感染させる。一方、プラスミドベクターの場合には、リポフェクション法、リポソーム法、エレクトロポレーション法、リン酸カルシウム共沈殿法、DEAEデキストラン法、マイクロインジェクション法、遺伝子銃法などを用いて該ベクターを細胞に導入することができる。
後述する実施例に示すように、当該工程を含めることにより、より効率的にNKT-iPS細胞を樹立することが可能となる。
iPS細胞の樹立効率改善物質としては、例えば、ヒストンデアセチラーゼ(HDAC)阻害剤[例えば、バルプロ酸 (Nat. Biotechnol., 26(7): 795-797 (2008))、トリコスタチンA、酪酸ナトリウム、MC 1293、M344等の低分子阻害剤、HDACに対するsiRNAおよびshRNA(例、HDAC1 siRNA SmartpoolO (Millipore)、HuSH 29mer shRNA Constructs against HDAC1 (OriGene)等)等の核酸性発現阻害剤など]、G9aヒストンメチルトランスフェラーゼ阻害剤[例えば、BIX-01294 (Cell Stem Cell, 2: 525-528 (2008))等の低分子阻害剤、G9aに対するsiRNAおよびshRNA(例、G9a siRNA(human) (Santa Cruz Biotechnology)等)等の核酸性発現阻害剤など]等が挙げられるが、それらに限定されない。核酸性の発現阻害剤はsiRNAもしくはshRNAをコードするDNAを含む発現ベクターの形態であってもよい。
iPS細胞の樹立効率改善物質は、該物質の非存在下と比較してNKT細胞等の体細胞からのNKT-iPS細胞樹立効率が有意に改善される限り、核初期化因子と同時にNKT細胞等の体細胞に接触させてもよいし、また、どちらかを先に接触させてもよい。一実施態様において、例えば、核初期化因子がタンパク性因子をコードする核酸であり、iPS細胞の樹立効率改善物質が化学的阻害物質である場合には、前者は遺伝子導入処理からタンパク性因子を大量発現するまでに一定期間のラグがあるのに対し、後者は速やかに細胞に作用しうることから、遺伝子導入処理から一定期間細胞を培養した後に、iPS細胞の樹立効率改善物質を培地に添加することができる。別の実施態様において、例えば、核初期化因子とiPS細胞の樹立効率改善物質とがいずれもウイルスベクターやプラスミドベクターの形態で用いられる場合には、両者を同時に細胞に導入してもよい。
核初期化因子(及びiPS細胞の樹立効率改善物質)との接触に際し、例えば、カチオニックリポソームなど導入試薬を用いる場合には、導入効率の低下を防ぐため、無血清培地に交換しておくことが好ましい場合がある。核初期化因子(及びiPS細胞の樹立効率改善物質)を接触させた後、細胞を、例えばES細胞の培養に適した条件下で培養することができる。ヒト細胞の場合、通常の培地に分化抑制因子として塩基性線維芽細胞増殖因子(bFGF)を添加して培養を行うことが好ましい。一方、マウス細胞の場合には、bFGFの代わりにLeukemia Inhibitory Factor(LIF)を添加することが望ましい。また通常、細胞は、フィーダー細胞として、放射線や抗生物質で処理して細胞分裂を停止させたマウス胎仔由来の線維芽細胞(MEF)の共存下で培養される。MEFとしては、通常STO細胞等がよく使われるが、iPS細胞の誘導には、SNL細胞(McMahon, A. P. & Bradley, A. Cell 62, 1073-1085 (1990))等がよく使われている。
一方、α-GalCerの代わりにα-GalCerでパルスしたDCを用いる場合、活性化NKT細胞の作製において上記したような手法により調製されるα-GalCerでパルスしたDCを、上記水性液に約1.0×106-約1.0×107細胞/mLとなるように懸濁すればよい。
iPS-NKT細胞含有製剤とα-GalCer含有製剤とは、別個にもしくは用時混合して同時に投与することもできるし、iPS-NKT細胞含有製剤、次いでα-GalCer含有製剤の順で、経時的に投与することもできる。
T細胞抗原受容体α鎖(TCRα)領域が既にNKT細胞で用いられるTCR (NKT-TCR) に再構成しているC57BL/6マウスの脾臓細胞より、NKT細胞を調製した。NKT細胞はMHCクラスI様分子であるCD1dに提示される糖脂質抗原を認識することを特徴とするため、糖脂質抗原であるα-GalCerを挟んだ可溶化型CD1d-マウスIgG1組換え体 (BD Bioscience社製) をAPC標識した抗マウスIgG1抗体に反応性を有する細胞を、抗APC磁気ビーズを利用したMACS法(Miltenyi Biotech社製)を用いてポジティブ選択することにより、細胞を濃縮した。この操作により、α-GalCer loaded CD1d dimer陽性/TCRβ陽性の細胞として規定されるNKT細胞は90%以上の純度となった。濃縮したNKT細胞を、IL-2(10ng/ml)存在下、106個/mlの細胞密度で10%FCSを含むRPMI培地で24時間培養した後、Cell, 126: 663-676 (2006) に記載の方法に準じて、マウス由来の4因子(Oct3/4、Sox2、Klf4、c-Mycをコードする核酸)を含むレトロウイルス(106pfu/ml)に24時間感染させた。ウイルス感染から3-7日後に細胞を回収し、マウス胚性線維芽細胞(MEF)上への蒔き直しを行い、LIF存在下、ES細胞培養用培地で共培養した。出現したコロニーを形態的に判断し、ES様の形態をとるコロニーについてピックアップし、さらにLIF存在下MEF上で培養することにより、NKT細胞由来iPS細胞(NKT-iPS細胞)を4クローン樹立した(クローン名:2a, 5b, 5d, 5g)。
実施例1で樹立した4クローンのNKT-iPS細胞がNKT細胞由来であるか証明するために、NKT-TCRへの再構成が行なわれているか否かをgenomic PCRにより確認した。NKT-TCRを有する細胞では、TCRα領域の片方がVα14-Jα18に既に再構成していることから(図1)、下記に示すプライマーを用いて、各NKT-iPSクローンのゲノムを鋳型としてPCRを行なうことにより、再構成の有無を確認した。

[Vα14-Jα18に再構成していない(ワイルド)場合、プライマー1とプライマー2を用いたPCRにより、349bpのバンドが増幅される。一方、Vα14-Jα18に再構成している(NKT-TCR)場合、プライマー1とプライマー3を用いたPCRにより、317bpのバンドが増幅される。]
その結果、樹立された4クローンいずれもゲノムの片方がNKT-TCRに再構成していることが明らかとなり(図2)、樹立されたiPS細胞がNKT細胞由来であることが確認できた。
実施例2で確認されたNKT-iPS細胞がiPS様の細胞へリプログラミングされているか否かを確認するために、遺伝子発現プロファイリングを実施した。NKT-iPS細胞、NKT細胞の核移植胚由来のES細胞(NKT-ES細胞)、末梢のNKT細胞からそれぞれtotal RNAをフェノール-クロロホルム法により調製し、ES細胞において発現が知られている一連の遺伝子群の発現をone step RT-PCR法 (Invitrogen社製) により解析した。解析を行なった遺伝子と用いたプライマーの配列を下記に列記する。

その結果、解析した全ての遺伝子において、NKT-iPS細胞、NKT-ES細胞で発現が確認され、末梢のNKT細胞では発現が確認されなかった(図3)。この結果は、樹立したNKT-iPS細胞がiPS細胞様の機能を有している可能性を示唆している。
さらに、DNAマイクロアレイ (Affimetrix社製) により、NKT-iPS細胞、NKT-ES細胞、ES細胞、末梢のNKT細胞間での遺伝子発現プロファイルの相関を確認したところ、NKT-iPS細胞はNKT-ES細胞あるいはES細胞と非常に良く似た遺伝子発現パターンを有し、末梢のNKT細胞とは異なることが明らかとなった(図4)。
実施例2で樹立されたNKT-iPS細胞の形態を顕微鏡下で観察した。その結果、SSEA1やOct3/4の発現局在や細胞自身の形態がES細胞のそれらと酷似していることが明らかとなった(図5)。
実施例2で樹立されたC57BL/6マウス NKTクローン由来のNKT-iPS細胞と、Balb/c由来細胞から、アグリゲーション法によりキメラマウスを作製した。得られたキメラマウスの脾臓細胞について、細胞表面抗原の解析を行なった。C57BL/6由来の細胞とBalb/c由来の細胞とを、MHCクラスIで見分けることによりゲート(それぞれI-AbとI-Ad)し、NKT細胞の含有率を解析した。その結果、C57BL/6由来の細胞はほとんど全ての細胞が、α-GalCer loaded CD1d dimer陽性/TCRβ陽性のNKT細胞様の表面抗原を有しているのに比して、Balb/c由来の細胞は通常の頻度のT/NKT細胞の含有率であった(図6)。
ES細胞は、NotchリガンドであるDelta-like1(Dll-1)を強制発現したOP9ストローマ細胞(OP9/Dll-1)と、IL-7およびFlt3リガンド(FL)存在下で共培養することにより、CD4/CD8ダブルポジティブ(DP)のT細胞に分化誘導させることができる(Schmitt TM, de Pooter RF, Gronski MA, Cho SK, Ohashi PS, Zuniga-Pflucker JC. Induction of T cell development and establishment of T cell competence from embryonic stem cells differentiated in vitro. Nat Immunol. 2004 Apr;5(4):410-417.)。この知見に基づき、NKT-iPS細胞をOP9/Dll-1細胞とIL-7(1 ng/ml)、FL(5 ng/ml)存在下で20日間共培養したところ、CD4/CD8 DPのα-GalCer loaded CD1d dimer陽性/TCRβ陽性のNKT細胞様の細胞(NKT-iPS細胞由来DP-NKT細胞)が誘導されることが明らかとなった(図7)。さらに詳細に細胞表面抗原の発現解析を行なうと、DP-NKT細胞の表現型は、胸腺内のCD4/CD8 DP細胞のそれと酷似していることが明らかとなった(図8)。この結果は、NKT-iPS細胞がTCRα領域の遺伝子再構成の影響を受けて、NKT様の細胞に分化誘導されやすいことを示唆しているとともに、免疫細胞の遺伝子再構成という特徴を生かした新しいコンセプトを導入することで、目的の免疫細胞 (特にNKT細胞、T細胞、B細胞) を大量に調製する技術を確立できることを示している。
NKT-iPS細胞を培養14日目までOP9/Dll-1と共培養し、培養14日目から20日目までOP9と共培養することによってNKT細胞が誘導されるか否かを解析した。その結果、Lin陰性/CD44陽性/CD25陰性の所謂DN1しか出現していない分化段階早期においても、NKT細胞が出現しうること(図9)、分化誘導したNKT細胞の表面マーカーが末梢のNKT細胞に酷似していること(図9)を見出した。この結果は、NKT細胞が分化段階早期にNotchシグナルが欠損しても、成熟したNKT細胞に分化し得ることを示すものである。
実施例6で誘導が確認できたNKT-iPS細胞由来DP-NKT細胞を試験管内でさらに増やし成熟化させるために、種々のサイトカインの組み合わせでフィーダー細胞あり、なしの条件で5日間培養を行なった。その結果、OP9あるいはOP9/Dll-1との共培養で、IL-2/IL-15/FLあるいはIL-2/IL-7/IL-15/FLのサイトカインの組み合わせで培養することにより、さらに10倍以上に細胞を増やせることが明らかとなった(図10)。さらに、これら条件で培養した細胞はNK1.1の発現が誘導されており、より分化誘導が進んでいることが推測された(図11)。そこで、OP9あるいはOP9/Dll-1との共培養で、IL-2/IL-15/FLあるいはIL-2/IL-7/IL-15/FLのサイトカインの組み合わせで誘導して得られるNKT細胞について、骨髄細胞由来樹状細胞(GM-CSFで誘導)とα-GalCer存在下で共培養することにより、その増殖能およびサイトカイン産生能を確認した。その結果、OP9/Dll-1でさらに培養した場合に増殖能が確認されること(図12)、その中でもIL-2/IL-15/FLで培養した場合にTh1への偏りがあること(図12)が確認できた。
実施例8で誘導されたNKT-iPS細胞由来成熟NKT細胞(IL-7/FL存在下、OP9/Dll-1と20日間共培養した後、IL-2/IL-15/FL存在下、OP9/Dll-1とさらに5日間共培養したもの)が、生体内で機能的であるか否かを評価した。TAPノックアウトマウス由来脾細胞を10mg/mlの卵白アルブミン(OVA)と高張液下培養した後、アポトーシスを誘導し、アポトーシスした細胞2×107個を、2μgのα-GalCerとともに、NKT-iPS細胞由来成熟NKT細胞105個あるいは106個を1時間前に予め移入しておいたJα18ノックアウトマウス(NKT細胞欠損マウス)に移入した。7日後、該マウスより脾臓細胞を採取し、試験管内でOVAペプチド(257-264)により刺激し、細胞内染色によりIFN-γの産生を解析した。その結果、IFN-γ産生能を有するOVA抗原特異的CD8陽性T細胞が移入細胞数依存的に誘導できていることが確認され、NKT-iPS細胞由来成熟NKT細胞が生体内で機能し、強力なアジュバント効果を有することが明らかとなった(図13)。
実施例1−8で、純化したマウスNKT細胞集団からのNKT-iPS細胞の樹立と、該NKT-iPS細胞からのNKT細胞への分化誘導、該細胞がNKT細胞としての機能を有していることなどが明らかとなった。そこで、実施例1と同様の手順により、C57BL/6マウスの脾臓細胞より濃縮操作なしにNKT細胞を調製し、30-50%程度の純度のNKT細胞集団を得た。実施例1と同様に、IL-2(10ng/ml)存在下、106個/mlの細胞密度で24時間この細胞を培養した後、マウス由来の4因子(Oct3/4、Sox2、Klf4、c-Mycをコードする核酸)を含むレトロウイルス(106pfu/ml)に24時間感染させた。ウイルス感染から3-7日後に細胞を回収し、マウス胚性線維芽細胞(MEF)上への蒔き直しを行い、LIF存在下、ES細胞培養用培地で共培養した。出現したコロニーを形態的に判断し、ES様の形態をとるコロニーについてピックアップし、さらにLIF存在下MEF上で培養することにより、NKT-iPS細胞5クローンの樹立に成功した(クローン名:i12c, i12d, i13g, i13h, i13i)(図14)。
図15に示すように、細胞移入実験を行う目的でC57BL/6バックグラウンドのNKTクローンマウスを作製した。すなわち、C57BL/6マウスの脾臓由来のNKT細胞の核をBDF1マウスの脱核した卵母細胞に移入した。この細胞をBDF1×ICRのES細胞とフュージョンし、仮親の子宮に移入し、産仔であるキメラマウスを得た。得られたキメラマウス(雄)をC57BL/6マウス(雌)と交配することで得られた産仔をC57BL/6バックグラウンドのNKTクローンマウスとした。
この方法で樹立されたNKTクローンマウスは、元となる核が100%、C57BL/6マウスの脾臓由来のNKT細胞に由来するため、完全なC57BL/6であり、C57BL/6マウスとアロジェニックあるいはセミアロジェニックな反応を起こさない。C57BL/6バックグラウンドのNKTクローンマウスとC57BL/6マウスとのゲノム上の相違は、NKT細胞への分化に伴うT細胞レセプター領域の再構成による配列の相違のみと考えられる。遺伝子再構成を終えているC57BL/6バックグラウンドのNKTクローンマウスのT細胞レセプター領域の塩基配列を図16に示す。
従来の山中らにより確立されたマウスiPS細胞の樹立の方法は、胚性線維芽細胞(MEF)にOct3/4、Sox2、Klf4、c-Mycを導入することによるものである。そこで、作製例1において創生されたNKTクローンマウスのMEFから、iPS細胞の樹立を試み、NKT-iPS細胞7aおよび7gの樹立に成功した。樹立された7aおよび7gは、T細胞レセプター領域がNKT細胞のT細胞レセプターに遺伝子再構成を起こしていることが確認される(図17)とともに、ES細胞マーカーとなり得る遺伝子群の発現がES細胞と同等であることがRT-PCR法により確認された(図18)。また、DNAマイクロアレイによる網羅的な遺伝子発現解析とそのクラスター解析から、樹立された7a及び7gはES細胞に近く、元となるNKTクローンマウスのMEFとは遠いという結果を得た(図19)。さらに、その形態学的観察を行うと、7aおよび7gはES細胞のそれと非常に良く似ており、ES細胞マーカーであるSSEA1、Oct3/4、Nanogが発現していることも確認された(図20)。さらに、Oct3/4及びNanogの遺伝子発現調節領域のゲノムのメチル化傾向を解析したところ、7aおよび7gにおいてはES細胞と同等に非メチル化されており、十分にリプログラミングされていることが明らかとなった(図21)。また、7a及び7gからはキメラマウスも複数産まれ、C57BL/6マウスとの交配で得られた産仔は全て、NKT細胞のT細胞レセプターに遺伝子再構成を起こしていることから、子孫伝達も確認され、7aおよび7gが多能性を有することが示された(図22)。以上のことから、7aおよび7gはiPS細胞のクライテリアを満たしていると結論付けた。
次に、MEFに由来する7aおよび7gから試験管内でNKT細胞を分化誘導することが可能であるか否かを検証した。7aおよび7gを、骨髄由来ストローマ細胞であるOP9にNotchリガンドであるDll-1を強制発現させた細胞株OP9/Dll-1と、図23に示すプロトコルで25日間共培養した。その結果、図24に示すように、α-GalCer/CD1d dimer陽性、TCRβ陽性のNKT細胞様の細胞(7a dif.および7g dif.)に分化誘導されることが明らかとなった。さらに、7a dif.および7g dif.は、その細胞表面マーカーの発現が、胸腺内に存在するCD4陽性CD8陽性の所謂DP細胞のそれと酷似していた。また、DNAマイクロアレイによる網羅的な遺伝子発現解析とそのクラスター解析から、7a dif.および7g dif.は分化誘導前の7aおよび7gとは遠く、末梢に存在する脾臓由来NKT細胞に近いという結果を得た(図25)。そこで、7a dif.および7g dif.が機能的にNKT細胞と同等であるか否かを評価するために、樹状細胞との共培養下、糖脂質リガンドであるα-GalCerで刺激し、増殖能およびサイトカイン産生能を調べた。その結果、7a dif.および7g dif.は、末梢のNKT細胞と同様に、顕著な増殖能を有し、大量のIFN-γやIL-4を産生する能力を有することが確認できた(図26)。
実施例12に記載のように7a dif.および7g dif.が末梢のNKT細胞と同等の機能を有することから、生体内において同様の効果があるか否かをNKT細胞欠損マウスを用いて解析した。NKT細胞欠損マウスに7a dif.および7g dif.を移入後、1週間および2週間における移入細胞の存在を確認すると、肝臓に、α-GalCer/CD1d dimer陽性、TCRβ陽性の7a dif.および7g dif.が存在することが明らかとなった(図27)。そこで図28に示すプロトコルに基づいて、抗原特異的CD8陽性T細胞の誘導とそれに伴う抗腫瘍効果を確認した。TAPノックアウトマウス由来脾細胞を10mg/mlの卵白アルブミン(OVA)と高張液下培養した後、アポトーシスを誘導し、アポトーシスした細胞2×107個を、2μgのα-GalCerとともに7a dif.および7g dif.を一週間前に移入したNKT細胞欠損マウスに移入した(TOG免疫)。7日後、該マウスより脾臓細胞を採取し、試験管内でOVAペプチド(257-264)により刺激し、細胞内染色により、IFN-γの産生を解析した。その結果、IFN-γ産生能を有するOVA抗原特異的CD8陽性T細胞が野生型マウスと同等に誘導できていることが確認され、7a dif.および7g dif.が生体内で機能し、強力なアジュバント効果を有することが明らかとなった(図29)。そこで、該マウスにC57BL/6 マウスの胸腺腫細胞系 EL4あるいはEG7(EL4 OVA- transfectant)を使用した悪性腫瘍拒絶モデルによる評価を行なった。その結果、OVA強制発現株であるEG7においては、TOG免疫を行なっていない野生型マウスでは悪性腫瘍の進行が見られるのに対して、TOG免疫を行なった7a dif.および7g dif.移入NKT細胞欠損マウスにおいては、TOG免疫を行なった野生型マウスと同様に進行が確認されなかった。EL4においては、TOG免疫を行なった野生型マウスとNKT細胞欠損マウスとで同様の進行が見られることから、移入した7a dif.および7g dif.による抗原特異的なアジュバント効果によってもたらされているものと結論付けた(図30)。
以上の結果は、如何なる細胞であれ、T細胞レセプターが再構成しているものであるならば、該T細胞レセプターに再構成している、機能的な免疫担当細胞に分化誘導させうることを示している。
実施例10に示すように野生型マウスの脾臓細胞由来のNKT細胞からのiPS細胞の樹立に成功しているが、より効率的なiPS細胞樹立方法を確立した。
すなわち、脾細胞よりMACSビーズを用いてCD1d拘束性のNKT細胞を濃縮後、FACSソーティングにより純度99.9%のNKT細胞を得る。該NKT細胞は、抗CD3抗体(10μg/ml)および抗CD28抗体(10μg/ml)による刺激下、IL-2(10ng/ml)及びIL-12(10ng/ml)を加えることにより、増殖サイクルに入れることができる。この条件下で、該NKT細胞をOct3/4、Sox2、Klf4、c-Mycをコードする核酸を含むレトロウイルスに感染させることで、実施例10よりも10倍以上効率的にiPS細胞を樹立することが可能であることが明らかとなり、NKT-iPSクローン14kの樹立に成功した(図31)。図32に示すように、14kはES細胞様の形態とES細胞マーカーの発現、さらにT細胞レセプター領域のNKT細胞T細胞レセプターの再構成が起こっている。また、実施例12に記載の方法により、NKT細胞様の細胞への分化がみられ、試験管内での糖脂質刺激によって増殖し、サイトカインを産生する機能的な細胞となった(図33)。
Claims (22)
- T細胞抗原受容体遺伝子のα鎖領域がNKT細胞受容体特異的な態様で均一なVα-Jαに再構成されている体細胞に核初期化因子を接触させることにより得られるクローン化された細胞であって、T細胞抗原受容体遺伝子のα鎖領域がNKT細胞受容体特異的な態様で均一なVα-Jαに再構成されており、かつ分化多能性および自己複製能を有する細胞。
- 体細胞がNKT細胞である、請求項1記載の細胞。
- 体細胞が線維芽細胞である、請求項1記載の細胞。
- 核初期化因子が、Oct3/4、Sox2、Klf4およびc-Mycまたはそれらをコードする核酸、あるいはOct3/4、Sox2およびKlf4またはそれらをコードする核酸である、請求項1〜3のいずれか1項に記載の細胞。
- 体細胞がヒト由来である、請求項1〜4のいずれか1項に記載の細胞。
- 請求項1〜5のいずれか1項に記載の細胞を生体外で分化させて得られる、単離されたNKT細胞。
- NK1.1ポジティブである、請求項6記載の細胞。
- α-ガラクトシルセラミドを提示した樹状細胞との接触によりTh1優位なサイトカイン産生が誘導される、請求項6記載の細胞。
- 請求項1〜5のいずれかに記載の細胞を、Notchリガンドを発現するストローマ細胞と共培養することを特徴とする、CD4/CD8ダブルポジティブNKT細胞の作製方法。
- 請求項9記載の方法により得られるCD4/CD8ダブルポジティブNKT細胞を、インターロイキン-2、インターロイキン-7、インターロイキン-15およびFlt3リガンドからなる群より選択される3以上のサイトカインの存在下、ストローマ細胞と共培養することを特徴とする、NKT細胞の増幅方法。
- サイトカインがインターロイキン-2、インターロイキン-15およびFlt3リガンドを含む、請求項10記載の方法。
- 請求項9記載の方法により得られるCD4/CD8ダブルポジティブNKT細胞を、Notchリガンドを発現しないストローマ細胞と共培養することを特徴とする、NK1.1ポジティブNKT細胞の作製方法。
- 培養がインターロイキン-15の存在下で行われることを特徴とする、請求項12記載の方法。
- 請求項9〜13のいずれか1項に記載の方法により得られるNKT細胞を、α-ガラクトシルセラミドを提示した樹状細胞と接触させることを特徴とする、活性化NKT細胞の作製方法。
- 請求項14記載の方法により得られる活性化NKT細胞を含有してなるNKT細胞療法剤。
- 請求項6〜8のいずれかに記載の細胞または請求項9〜14のいずれかに記載の方法により得られる細胞と、α-ガラクトシルセラミドとを組み合わせてなる、NKT細胞療法剤。
- T細胞抗原受容体遺伝子のα鎖領域がNKT細胞受容体特異的な態様で均一なVα-Jαに再構成されており、かつ分化多能性および自己複製能を有するクローン化された細胞の製造方法であって、T細胞抗原受容体遺伝子のα鎖領域がNKT細胞受容体特異的な態様で均一なVα-Jαに再構成されている体細胞に核初期化因子を接触させることを含む方法。
- 体細胞がNKT細胞である、請求項17記載の製造方法。
- 核初期化因子と接触されるNKT細胞が、IL-2およびIL-12の存在下で抗CD3抗体および抗CD28抗体によって刺激されたものである、請求項18記載の製造方法。
- 体細胞が線維芽細胞である、請求項17記載の細胞。
