Abstract

PURPOSE: A bacterium belonging to Bacillus is cultured and the cell bodies are extracted to produce a heat-resistant enzyme that can keep higher than 80% activity of that of untreated enzyme even after the treatment in a buffef at about 50°C for about 15min, thue producing an inexpensive enzyme that acts on a coenzyme and kees high stability for a long period of time.
CONSTITUTION: A Bacillus bacterium such as Bacillus stearothermophilus is cultured, the culture mixture is centrifuged, the resultant cell bodies are suspended in a phosphoric acid buffer, the cells are crushed by means of a French press or the like and the crushed cell walls are removed to give a crude extract. The extract is purified to give the objective glucose-phosphate dehydrogenase. The resultant enzyme can keep about 80W100% activity of that of untreated one even after the treatment of heating in a phosphoric acid buffer solution at about 50°C for 15min. It is used as an enzyme with stably high activity for a long period of time for determining glucose, glucose-6-phosphate, hexonase or the like by using inexpensive nicotinamide adenin dinucleotide as a coenzyme in medical and food fields.
