Abstract

PURPOSE: To obtain a novel and highly active glycerol dehydrogenase (GDH) by culturing a GDH-producing strain belonging to Bacillus genus in a medium containing glycerol, and extracting the GDH accumulated in the bacterial cells.
CONSTITUTION: G-1 Bacteria belonging to Bacillus genus are cultured in a medium containing 2% of glycerol to promote the DGH-producing activity and a Mn salt as an essential salt component, at 30°C for about 24hrs under agitation. After separation and grinding of the cultured bacterial cells, the objective enzyme is extracted, and purified by conventional process such as salting-out, etc. to obtain crude GDH, which can be purified by the adsorption and desorption with an ion exchange material. The purified GDH obtained by this process in high efficiency and useful for the clinical examination has the following characteristics: molecular weight, 85,000±5,000; optimum pH range for operation, 9.5W10; stable pH range, 5.0W9.0; optimum temperature for operation, about 45°C; stable temperature, ≤50°C; Km value for glycerol, 1.33×10-2M, Km value for NAD, 1.28×10-4M.
