Abstract
PURPOSE: A crude solution containing impurities is passed through a column filled with activated carbon and the adsorbate is eluted with an aqueous acetone of a specific or higher pH, then adsorbed on an ion-exchange resin and eluted, whereby the titled compound used as a clinical testing regent is purified with industrial advantage.
CONSTITUTION: Bread yeast is extracted with hot water and the copper salt of glutathione is separated from the extract. The supernatant, including nicotinic amide adenine dinucleotide (NAD) together with impurities, is brought into contact with activated carbon to adsorb NAD, which is eluted with an aqueous acetone kept 4 or higher in pH. The eluate is passed through a strong-acid type cation-exchange resin or weak-base type anion exchange resin so that NAD is adsorbed on it. The adsorbed NAD is eluted with an aqueous salt buffer to give the objective purified NAD.
