Abstract
PURPOSE: To separate L-isoleucine from an aqueous solution containing L-isoleucine and DL-α-aminobutyric acid, by passing the solution through a strongly acidic cation exchange resin, and treating the eluate first with an aqueous solution of dilute hydrochloric acid, and then with ammonia water.
CONSTITUTION: An aqueous solution containing L-isoleucine and DL-α-aminobutyric acid is passed through a column packed with a strongly acidic cation exchange resin (e.g. Diaion SK-1BS), to adsorb both amino acids to said resin. After washing off the non-adsorbed substances in the resin layer with deionized water, the DL-α- amino acid is eluted completely from the column by passing a 0.02W0.6N, pref. 0.05W0.07N dilute hydrochloric acid through the column. The hydrochloric acid in the resin is washed off with deionized water, and the remaining L-isoleucine is eluted separately from the DL-α-amino acid by passing dilute ammonia water through the column.
USE: An essential amino acid which plays important role in the nutrition.
