Claims (1)
- 【特許請求の範囲】 1.突然変異体が、野生型SPE−A毒素に実質的に対応するタンパク質と比べ て、少なくとも1つのアミノ酸変化を有し、実質的に非致死性である、突然変異 体SPE−A毒素またはそのフラグメント。 2.突然変異体SPE−A毒素が、1〜6個のアミノ酸置換を含み、 置換されたアミノ酸の少なくとも1つが、N末端アルファらせん3、ド メインBベータ鎖1、ドメインBベータ鎖2、ドメインBベータ鎖3、ドメイン Aベータ鎖6、ドメインAベータ鎖8、ドメインAベータ鎖9、ドメインAベー タ鎖10に位置するか、またはシステインである、 請求項1に記載の突然変異体SPE−A毒素。 3.突然変異体SPE−A毒素が、1〜6個のアミノ酸置換を含み、 置換されたアミノ酸の少なくとも1つが、アスパラギン−20、リシン −157またはシステイン−98である、請求項1に記載の突然変異体SPE− A毒素。 4.少なくとも1つのアミノ酸置換が、アスパラギン酸、グルタミン酸、リシン またはアルギニンへのアスパラギン−20の置換、セリン、アラニン、グリシン 、またはトレオニンへのシステイン98の置換、あるいは、グルタミン酸または アスパラギン酸へのリシン−157の置換を含む、請求項3に記載の突然変異体 SPE−A毒素。 5.少なくとも1つのアミノ酸置換が、アスパラギン酸へのアスパラギン−20 、セリンへのシステイン98、グルタミン酸へのリシン−157の置換を含む、 請求項4に記載の突然変異体SPE−A毒素。 6.少なくとも1つのアミノ酸置換が、アスパラギン−20の置換を含む、請求 項3に記載の突然変異体SPE−A毒素。 7.置換が、アスパラギン酸へのアスパラギン−20である、請求項6に記載の 突然変異体SPE−A毒素。 8.システイン−98またはリシン−157の置換をさらに含む、請求項6に記 載の突然変異体SPE−A毒素。 9.置換が、セリンへのシステイン−98またはグルタミン酸へのリシン−15 7である、請求項8に記載の突然変異体SPE−A毒素。 10.突然変異体が、少なくとも1つの以下の特性を有する突然変異体SPE− A毒素であって、特性とは、突然変異体は、T細胞ための細胞分裂促進性の低下 を有すること、突然変異体は、内毒素ショックを実質的に高めないこと、突然変 異体は、致死性でないこと、または突然変異体は、非致死性であるが、野生型S PE−A毒素に匹敵する細胞分裂促進性を保持することである、請求項1に記載 の突然変異体SPE−A毒素。 11.野生型SPE−A毒素の少なくとも1つの生物活性に対して動物を防護す るワクチンであって、請求項1に記載の少なくとも1つの突然変異体SPE−A 毒素の有効量を含むワクチン。 12.生理学上許容可能な担体と混合された請求項1に記載の突然変異体SPE −A毒素を含む製剤組成物。 13.請求項1に記載の突然変異体SPE−A毒素をコードするDNA配列。 14.請求項13に記載のDNA配列を含む安定して形質転換された宿主細胞。 15.野生型SPE−A毒素の少なくとも1つの生物活性に対して動物を防護す る方法であって、請求項11に記載のワクチンを動物に投与することを含む方法 。 16.毒素ショックに伴う症状を軽減する方法であって、請求項11に記載のワ クチンを動物に投与することを含む方法。
