Claims (1)
- 【特許請求の範囲】 1. ガラニン作動性レセプター活性を有するポリペプチドをコードするヌク レオチド配列。 2. ヒトガラニン作動性レセプターをコードすることを特徴とする、請求の 範囲1に記載のヌクレオチド配列。 3. (a)配列番号1の配列もしくはその相補鎖の全部もしくは部分、 (b)配列(a)とハイブリダイズし、かつガラニン作動性レセプター 活性を有するポリペプチドをコードするいずれかの配列、ならびに (c)遺伝子コードの縮重の結果、配列(a)および(b)から取得さ れる配列: から選択されることを特徴とする、請求の範囲1もしくは2に記載のヌクレオチ ド配列。 4. 配列番号1のヌクレオチド配列もしくはその相補鎖の全部もしくは部分 を含むことを特徴とするヌクレオチド配列。 5. ゲノムDNA、cDNA、もしくはRNA配列、ハイブリッド配列、あ るいは合成もしくは半合成配列から選択されることを特徴とする、請求の範囲1 〜4に記載のヌクレオチド配列。 6. 前記ポリペプチドをコードする部分が、ある細胞宿主中でのその発現を 可能にさせるシグナルの制御下に置かれることを特徴とする、請求の範囲1〜5 の内の一つに記載の配列。 7. 既述の請求の範囲の内のいずれか一つに記載のヌクレオチド配列の発現 により生じるポリペプチド。 8. 配列番号1のペプチド配列もしくは後者の誘導体の全部もしくは部分を 含むポリペプチド。 9. 請求の範囲7および8のいずれかに記載のポリペプチドの産生を少なく とも部分的に阻害することが可能であるアンチセンス配列。 10. 請求の範囲3に記載の配列もしくはそれに対応するmRNAとハイブリ ダイズすることが可能なヌクレオチドプローブ。 11. ガラニン作動性レセプターの発現を検出するため;あるいは遺伝子異常 (スプライシングの過誤、多形性、および点突然変異など)を証明するため;あ るいはガラニン作動性レセプターに関連する神経学的、心臓血管的、内分泌学的 、もしくは精神医学的障害を同定するため;あるいは別法ではガラニン作動性ポ リペプチドをコードする相同核酸配列を証明および単離するための、請求の範囲 10に記載のプローブの使用。 12. ガラニン作動性レセプター活性を保持することを特徴とする、請求の範 囲7もしくは8に記載のポリペプチド。 13. 請求の範囲7、8、および12の内の一つに記載のポリペプチドをその 表面に発現することが可能な組みかえ細胞。 14. 真核生物および原核生物細胞から選択されることを特徴とする、請求の 範囲13に記載の方法。 15. 以下に示す段階、すなわち: − 一つの分子、もしくは様々な分子(適切な場合には、これらは未同定であ る)を含む混合物を、請求の範囲13に記載されその表面にガラニン作動性ポリ ペプチドを発現する組換え細胞に、前記ポリペプチドと前記分子との間の相互作 用を可能にさせる条件下(後者が前記ポリペプチドに対する親和性を有する予定 になっている場合に)で接触させる 段階、ならびに − 前記ポリペプチドに結合した分子を検出および/または単離する段階、 を実施することを特徴とする、ガラニン作動性レセプターのリガンドを証明およ び/または単離するための方法。 16. ガラニン作動性レセプターについてのアゴニストおよび/またはアンタ ゴニストの証明および/または単離のための、請求の範囲15に記載の方法。 17. 以下に示す段階、すなわち: − 一つの分子、もしくは様々な分子(適切な場合には、これらは未同定であ る)を含む混合物を、請求の範囲13に記載されその表面にガラニン作動性レセ プター活性を有するポリペプチドを発現する組換え細胞に、前記レセプターのリ ガンドの存在下、前記ポリペプチドとそのリガンドとの間の相互作用を可能にさ せる条件下で接触させる段階、および − 前記ポリペプチドに関するリガンドの活性の調節を行うことが可能な分子 を検出および/または単離する段階、 を実施することを特徴とする、ガラニン作動性レセプターについての調節因子を 証明および/または単離するための方法。 18. 請求の範囲15〜17の方法に従って取得され得る、請求の範囲7、8 、および12に特定されるポリペプチドについてのリガンドもしくは調節因子。 19. 請求の範囲12に記載のポリペプチドに作用する少なくとも一つの分子 を活性成分として含む医療用産物。 20. その分子が、請求の範囲15〜17の方法に従って同定および/または 単離されるリガンドもしくは調節因子であることを特徴とする、請求の範囲19 に記載の医療用産物。
