Claims (1)
- 【特許請求の範囲】 1.a)ホエーまたはその濃縮物とスルフィットイオンを生成する試薬と酸化 剤とを接触させてスルホン化酸塩を形成し、すなわち亜硫酸化及び酸化を行ない 、 b)ホエーまたはその濃縮物から亜硫酸化され、酸化されたホエータンパ ク質を酸性pHで析出し、 c)生成混合物から亜硫酸化され、かつ酸化された析出ホエータンパク質 を回収し、必要により後処理を行う、ホエーからタンパク質を分離するための方 法において、 工程a)で使用する酸化剤が実質的に触媒を有しない食品グレードの酸化化合 物であり、使用温度が25〜55℃の範囲内の温度であり、この範囲内において 酸化化合物が亜硫酸化されたホエータンパク質と直接反応することを特徴とする 、ホエーからタンパク質を分離するための方法。 2.工程a)で使用するホエー濃縮物はホエーと比較して4〜16倍、好まし くは約8倍の濃縮率を有し、この場合、そのホエータンパク質含有量は好ましく は約2〜7重量/容積%、より好ましくは4重量/容積%であることを特徴とす る、請求項1記載の方法。 3.工程a)で使用するスルフィットイオン生成試薬 がアルカリ金属スルフィットまたはアルカリ土類金属スルフィット、水素スルフ ィット、および/またはメタビスルフィット、好ましくはナトリウムスルフィッ ト、ナトリウム水素スルフィットおよび/またはナトリウムメタビスルフィット 、最も好ましくはナトリウム水素スルフィットであることを特徴とする、請求項 1または2記載の方法。 4.工程a)において、亜硫酸化混合物中のスルフィットの濃度が約0.02 〜0.20M、好ましくは0.05〜0.1Mの範囲内に入るよう、スルフィッ トイオン生成試薬の濃度となっていることを特徴とする、請求項1、2または3 記載の方法。 5.工程a)で使用する食品グレードの酸化化合物が食品グレードのペルオキ シド化合物および/またはハロゲン化物、好ましくはそれぞれ過酸化カルシウム CaO2および/または臭素酸カリウムKBrO3であることを特徴とする、上記 請求項のいずれかに記載の方法。 6.工程a)において、食品グレードの酸化化合物の含有量が約0.01・Oact 〜0.15・Oact重量/容積%(ここでOactは化合物中の活性酸素量を表 示する)であることを特徴とする、上記請求項のいずれかに記載の方法。 7.約5.0〜8.5のpHで工程a)を実行することを特徴とする、上記請 求項のいずれかに記載の方法。 8.約30〜50℃の温度で工程a)を実行することを特徴とする、上記請求 項のいずれかに記載の方法。 9. a1)ホエータンパク質を亜硫酸化するようにホエーまたはその濃縮物 をスルフィットイオン生成試薬に接触させ、 a2)亜硫酸化されたホエータンパク質を酸化するように工程a1)の亜 硫酸化されたホエータンパク質を食品グレードの酸化化合物に接触させるよう2 つのサブ工程で工程a)を実行することを特徴とする、上記請求項のいずれかに 記載の方法。 10.約6.0〜7.5のpH、好ましくは6.5〜7.0のpHでサブ工程 a1)を実行することを特徴とする、請求項9記載の方法。 11.サブ工程a1)の亜硫酸化を約10〜50分で実行することを特徴とす る、請求項9または10記載の方法。 12.約5.0〜7.0のpH、好ましくは5.5〜 6.5のpHでサブ工程a2)を実行することを特徴とする、請求項9〜11の いずれかに記載の方法。 13.サブ工程a2)の酸化を約15〜60分で実行することを特徴とする、 請求項9〜12のいずれかに記載の方法。 14.サブ工程a1)およびa2)において、ほぼ同じ温度、すなわち工程a) の温度を使用することを特徴とする、請求項9〜13のいずれかに記載の方法。 15.約2.5〜6.5のpH値、好ましくは3.0〜5.0のpH値を使用 することにより、亜硫酸化され酸化されたホエータンパク質を工程b)において 析出することを特徴とする、上記請求項のいずれかに記載の方法。 16.工程b)において、25〜55℃の温度、好ましくは30〜50℃の温 度で析出を実行することを特徴とする、請求項15記載の方法。 17.工程b)において、pH値を前記析出pH値にゆっくりと調節すること を特徴とする、請求項15または16記載の方法。 18.工程a)の亜硫酸化および酸化から工程b)の析出までの変更時のpH 値の調節を10〜40分以内に行うことを特徴とする、請求項15、16または 17記載の方法。 19.工程b)のpH調節の後に混合物の攪拌を実行し、この攪拌は好ましく は約10〜60分続けることを特徴とする、請求項15〜18のいずれかに記載 の方法。 20.少なくとも2つの容器内で交互に工程b)の析出を実行し、好ましくは 一方の反応器を空にし、再充填しながら、他方の反応器で亜硫酸化、酸化および 析出を実行するように前記少なくとも2つの反応器内で交互に工程a)の亜硫酸 化および酸化と工程b)の析出とを実行することを特徴とする、上記請求項のい ずれかに記載の方法。 21.工程b)において析出した、亜硫酸化され、酸化されたホエータンパク 質を工程c)において、好ましくはマイクロ濾過により濃縮し、洗浄することを 特徴とする、上記請求項のいずれかに記載の方法。 22.工程c)のマイクロ濾過の濾液のタンパク質を限外濾過により濃縮し、 洗浄し、噴霧乾燥により分離することを特徴とする、請求項21記載の方法。 23.工程c)において、遠心分離法または好ましくはバンド状またはドラム 状フィルターにおける濾過により析出濃縮物から水を除去し、この結果、タンパ ク質ペーストを得ることを特徴とする、請求項21または22記載の方法。 24.それぞれ好ましくはNaOHまたはNaOHとCa(OH)2とKOH との混合物である塩基または塩基混合物により、工程c)においてタンパク質ペ ーストのpHを6〜8の値に上げ、この場合、Ca(OH)2の量は少なくとも 元のCaの量と置換するような大きさであることが好ましいことを特徴とする、 請求項23記載の方法。 25.工程c)において、噴霧乾燥により析出濃縮物を乾燥し、好ましくは請 求項24と同じ塩基または塩基化合物により、乾燥前の析出濃縮物のpHが6〜 8の値に調節されていることが好ましいことを特徴とする、請求項21記載の方 法。
