Description
Claims (1)
- 【特許請求の範囲】 1.下記式(I)で表される(S)−2−アリール−1,3−プロパンジオール モノカルバメート。 前記式(I)において、Rは (但し、Xは酸素原子又は硫黄原子であり、Yはハロゲン元素、三フッ化メチル 基又は炭素原子1〜3の低級アルキル基を示す。)である。 2.下記式(II)で表される(S)−アセトキシ−2−アリール−プロパノール カルバメート。 前記式(II)において、Rは (但し、Xは酸素原子又は硫黄原子であり、Yはハロゲン元素、三フッ化メチル 基又は炭素原子1〜3の低級アルキル基を示す。)である。 3.下記構造式(III)で表される(R)−3−アセトキシ−2−アリール−プ ロパノールを芳香族炭化水素とハロゲン化炭化水素の混合溶媒中でアミン系塩基 存在下、ホスゲンと反応させ、次いでアンモニアと反応させて短時間内に高い光 学純度が得られることを特徴とする下記式(II)で表される(S)−3−アセト キシ−2−アリールプロパノールカルバメートの製造方法。 (式(II)において、Rは (但し、Xは酸素原子又は硫黄原子であり、Yはハロゲン元素、三フッ化メチル 基又は炭素原子1〜3の低級アルキル基を示す。)である。) (式(III)において、Rは上記と同義である。) 4.前記混合溶媒はベンゼン、トルエン及びキシレンからなる群から選択される 芳香族炭化水素並びにクロロホルム、ジクロロメタン、1,2−ジクロロエタン 及びトリクロロエタンからなる群から選択されるハロゲン化炭化水素を含む請求 項3に記載の製造方法。 5.前記混合溶媒はトルエンとジクロロメタンとを含む請求項4に記載の製造方 法。 6.前記アミン系塩基はアンチピリン、ジイソプロピルエチルアミン、ジエチル アミン、トリエチルアミン及びピリジンからなる群から選択される請求項3に記 載の製造方法。 7.前記ホスゲンを反応させる時の温度が−30℃〜0℃であり、前記アンモニ アを反応させる時の温度が−70℃〜0℃である請求項3に記載の製造方法。 8.前記アンモニアは凝縮して液体状態とし、その使用量は10〜100当量過 剰とする請求項3に記載の製造方法。 9.下記式(II)で表される(S)−アセトキシ−2−アリール−プロパノール カルバメートを緩衝溶液に溶解させ、ここに激しく攪拌しながら加水分解酵素を 添加して、短時間内に高収率と高い光学純度を得る下記式(I)で表される(S )−2−アリール−1,3−プロパンジオールモノカルバメートの製造方法。 (式(I)において、Rは (但し、Xは酸素原子又は硫黄原子であり、Yはハロゲン元素、三フッ化メチル 基又は炭素原子1〜3の低級アルキル基を示す。)である。) (式と(II)において、Rは前記と同義である。) 10. 反応は0〜35℃の温度で、常圧下で行われる請求項9に記載の製造方法 。 11. 前記リン酸緩衝溶液は6〜9のpHと0.01〜0.1Mの濃度とを有する 請求項9に記載の製造方法。 12. 前記加水分解酵素は、豚の膵臓から抽出したリパーゼ、カンジダリパーゼ 、アスペルギルスリパーゼ、シュードモナスリパーゼ、及び豚の肝から抽出した エ ステラーゼからなる群から選択される請求項9に記載の製造方法。 13. 前記加水分解酵素は豚の肝から抽出したエステラーゼである請求項12に 記載の製造方法。
