Claims (1)
- 【特許請求の範囲】 1.以下の配列を含んで成るDNA構築体 5′-P-SP-(LP)n-PS-HP-3′ (式中、 Pはプロモーター配列、 SPは酵母アスパラギン酸プロテアーゼ3(YAP3)シグナルペプチドをコードす るDNA配列、 LPはリーダーペプチドをコードするDNA配列、 nは0又は1、 PSは酵母プロセシング部位を規定するペプチドをコードするDNA配列、そして HPは選択した宿主生物にとって異種であるポリペプチドをコードするDNA配列 である)。 2.前記プロモーター配列がサッカロマイセス・セレビジエのMFα1,TPI,A DH,BARIもしくはPGKプロモーター、又はシゾサッカロマイセス・ポンベのADHプ ロモーターから選ばれたものである、請求項1記載のDNA構築体。 3.前記YAP3シグナルペプチドが下記のDNA配列 又はYAP3シグナルペプチドに対して高度の相同性を有するペプチドをコードする その適当な改変体によりコードされる請求項1記載のDNA構築体。 4.nが1である請求項1記載のDNA構築体。 5.前記リーダーペプチドが酵母のMFα1リーダーペプチド又は合成リーダー ペプチドである、請求項5記載のDNA構築体。 6.PSがLys-Arg,Arg-Lys,Lys-Lys,Arg-Arg又はIle-Glu-Gly-Argをコード するDNA配列である、請求項1記載のDNA構築体。 7.異種ポリペプチドがアプロチニン、組織因子経路インヒビターもしくはそ の他のプロテアーゼインヒビター、インスリンもしくはインスリン前駆体、ヒト もしくはウシ成長ホルモン、インターロイキン、グルカゴン、グルカゴン様ペプ チド1、組織プラスミノゲンアクチベーター、トランスフォーミング成長因子α もしくはβ、血小板由来成長因子、酵素、又はその機能的類似体より成る群から 選ばれる、請求項1記載のDNA構築体。 8.転写終止配列を更に含んで成る、請求項1記載のDNA構築体。 9.前記転写終止配列がTPIターミネーターである、請求項8記載のDNA構築体 。 10.請求項1〜9のいづれか1項記載のDNA構築体を含んで成る組換発現ベク ター。 11.請求項10記載のベクターにより形質転換された細胞。 12.菌類細胞である、請求項11記載の細胞。 13.酵母細胞である、請求項12記載の細胞。 14.サッカロマイセス、シゾサッカロマイセス、クルイベロマイセス、ハンセ ヌラ 又はヤロイアの細胞である、請求項13記載の細胞。 15.サッカロマイセス・セレビジエ又はシゾサッカロマイセス・ポンベの細胞 である、請求項14記載の細胞。 16.異種ポリペプチドを製造する方法であって、異種ポリペプチドを発現する ことができ、且つ請求項1〜9のいづれか1項記載のDNA構築体により形質転換 された細胞を適当な培地の中で培養してこの異種ポリペプチドを発現及び分泌さ せ、次いでこの異種ポリペ プチドをこの培地から回収することを含んで成る方法。
