Claims (23)
- 1.ヒト又は動物患者のNMR画像形成における使用のための粒子であって、酸 化鉄のコア及び疎水性の尾部に結合された負電荷リン含有頭部をもつ少なくとも 1の両親媒性化合物の層及び1以上の界面活性剤を含んで成り、その両親媒性化 合物がミセル形態で存在し、そしてその層が、その両親媒性化合物がそのミセル 形態で存在することを生じさせる少なくとも1の界面活性剤を含んで成る3次元 シェルの形状をもつような、粒子。
- 2.その両親媒性化合物のリン含有頭部が酸化鉄のに向かって集中し、一方、そ の疎水性尾部がそれから突き出し、ウルチン様構造を形成するように、両親媒性 化合物の分子が、酸化鉄コアに関して有機的に構成されている、請求項1に記載 の粒子。
- 3.頭部が、両方の酸素原子がコアとの相互作用に利用されることができるホス ホリルの頭である、請求項1又は2に記載の粒子。
- 4.両親媒性化合物がリン酸のアルキル、アルケニル又はシクロアルキルのモノ ・エステル又はリン脂質である、請求項3に記載の粒子。
- 5.アルキル基又はアルケニル基が、少なくとも8炭素原子、好ましくは少なく とも10炭素原子、そしてより好ましくは少なくとも12炭素原子を含んで成る 、請求項3に記載の粒子。
- 6.ミセル形態のリン脂質が、置換又は部分的に置換されたポリアルコールのモ ノ−ホスフェート・エステルから成り、このポリアルコールの少なくとも1の他 のアルコール官能基が長鎖の飽和又は不飽和脂肪族脂肪酸によりエステル化され ているか、又は長鎖の飽和又は不飽和アルコールによりエーテル化されており、 そのリン酸の他の2の酸性官能基が遊離であるか又はアルカリ金属若しくはアル カリ土類金属により塩化されているかのいずれかで存在しているような、請求項 4に記載の粒子。
- 7.両親媒性化合物が、ジミリストイルホスファチド酸、ジバルミトイルホスフ ァチド酸、又はジステアロイルホスファチド酸から選ばれた脂肪酸グリセリドの モノホスフェートである、請求項1、2又は3に記載の粒子。
- 8.界面活性剤が両親媒性化合物によりからみ合わされた生理学的に許容される 非イオン界面活性剤である、請求項1、2又は3に記載の粒子。
- 9.非イオン界面活性剤が、ポリオキシエチレン及びポリオキシプロピレン・セ グメントをもつ少なくとも1のプロッタ−コポリマー又はポリエチレングリコー ルアルキルエーテルを含んで成る、請求項8に記載の粒子。
- 10.界面活性剤が、Pluronic(R)、Poloxamer(R)、P oloxamine(R)、Synperonic(R)又はBRIJ(R)で ある、請求項9に記載の粒子。
- 11.両親媒性化合物が、イオン性及び天然のリン脂質、リン酸のモノ・アルキ ル又はアルケニル・エステル及び/又はコレステロール、エルゴステロール、フ ィトステロール、シトステロール、ラノステロール、及びトコフェロールから選 ばれた2以上の化合物の混合物である、請求項1、2又は3に記載の粒子。
- 12.リン脂質対界面活性剤の重量比が1:10から10:1までにある、請求 項1、2又は3に記載の粒子。
- 13.外側の層内の両親媒性化合物及び界面活性剤とコアのものとの重量比が1 :1未満である、請求項1、2又は3に記載の粒子。
- 14.ヒト又は動物の体の器官のNMR画像形成のための、請求項1〜13のい ずれか1項に記載の酸化鉄粒子の使用。
- 15.生理学的に許容される液体担体中に分散される間に、NMR画像形成コン トラスト剤として有用な懸濁液を形成するであろう、請求項1〜13のいずれか 1項に記載の酸化鉄粒子を含んで成る粒状配合物。
- 16.ヒト又は動物の体のNMR画像形成におけるコントラスト剤として有用な 、生理学的に許容される液体担体中に懸濁された請求項1〜13のいずれか1項 に記載の酸化鉄粒子を含んで成る注射可能な水性懸濁液。
- 17.請求項1の記載の粒子の製造方法であって、以下の段階:a)選択し、そ して両親媒性化合物及び界面活性剤と共に酸化鉄粒子を水相中に懸濁させ、混合 物を形成し、及び、b)音波処理、マイクロ流動化又は加熱により上記混合物に エネルギーを与え、酸化鉄粒子の周りに3次元シェル様の層を作り出す、を特徴 とする方法。
- 18.音波処理、マイクロ流動化又は加熱の後に、混合物を、滅菌及び/又は凍 結乾燥する、請求項17に記載の方法。
- 19.エネルギー供与及び場合により音波処理、マイクロ流動化又は加熱を繰り 返した後に、酸化鉄と尚親媒性化合物との混合物に界面活性剤を添加する、請求 項17又は18に記載の方法。
- 20.界面活性剤が、ポリオキシエチレン及びポリオキシプロピレン・セグメン トをもつ少なくとも1のプロッタ−コポリマー又はポリエチレングリコールアル キルエーテルから成る生理学的に許容される非イオン界面活性剤である、請求項 17又は18に記載の方法。
- 21.両親媒性化合物が、界面活性剤によりミセル形態に変換されるリン脂質で ある、請求項17又は18に記載の方法。
- 22.ミセル形態のリン脂質が、置換又は部分的に置換されたポリアルコールの 有機モノ−ホスフェート・エステルから成り、このポリアルコールの少なくとも 1の他のアルコール官能基が長鎖の飽和又は不飽和脂肪族脂肪酸によりエステル 化されているか、又は長鎖の飽和又は不飽和アルコールによりエーテル化されて おり、そのリン酸の他の2の酸性官能基が遊離であるか又はアルカリ金属若しく はアルカリ土類金属により塩化されているかのいずれかで存在しているような、 請求項21に記載の方法。
- 23.リン脂質が、ジミリストイルホスファチド酸、ジパルミトイルホスファチ ド酸、又はジステアロイルホスファチド酸から選ばれた脂肪酸グリセリドのモノ ホスフェートである、請求項22に記載の方法。
